Sex-Specific Adaptations in Placental Mitochondrial Biogenesis and Function in babies Born with Critical Congenital Heart Disease.

Congenital heart disease (CHD) is the leading cause of birth defects; most critical (c)CHD newborns will develop neurodevelopmental disorders. Abnormal placental development may be involved, as genetics explains only 30% of sporadic CHD cases. Growing evidence suggests that placental dysfunction may result in heart and brain defects, as it shares a common early development. We hypothesized that newborns with critical CHD exhibit sex-specific alterations in placental mitochondrial biogenesis and function which may affect the fetal growth trajectory. Term placenta and paired umbilical cord (UC)

Exploring lipid metabolism reprogramming in meningiomas through lipidomics of matched clinical specimens.

BACKGROUND: Meningiomas, among the most common primary intracranial tumors, present significant clinical challenges, particularly due to the propensity for recurrence in higher-grade variants and the paucity of effective non-surgical therapies.Lipid metabolism plays a critical role in tumor progression; however, the specific lipid dysregulation underlying meningioma biology remains incompletely understood. METHODS: In this study, meningioma tissues and patient-matched arachnoid membrane tissues were collected from 12 patients undergoing meningioma resection surgery. A comprehensive lipidomic

Rule-Based Ion Prediction with Orthogonal Constraints Reveals Bacterial Phospholipid Remodeling Signatures.

Background: Phospholipids are essential components of bacterial membranes and play central roles in membrane integrity and adaptation to antibiotic stress. However, confident annotation of phospholipid molecular species remains challenging due to the complexity of the lipidome and the limited structural constraints in conventional lipidomics workflows. Methods: Here, we present a bacterial phospholipidomic framework that integrates orthogonal structural evidence to achieve high-confidence and traceable annotation. Thin-layer chromatography (TLC) provides phospholipid headgroup assignment, gas

Targeting the ALCAT1 enzyme for the treatment of pressure-overload hypertrophy.

AIMS: Pressure overload-induced heart failure is a major cause of fatality in patients with heart diseases. At present, there exists no highly effective treatment for this incapacitating condition. Cardiolipin (CL) is a mitochondrial specific phospholipid that plays an essential role in cardiac health. Depletion of tetralinoleoyl CL (TLCL), the signature CL species, is implicated in human and animal models of heart failure. We investigated whether pathological CL remodelling by lysocardiolipin acyltransferase-1 (ALCAT1) promotes the progression of left ventricular (LV) hypertrophy induced by p

Time-resolved lipidomic atlas of the murine lung response to acute ionizing radiation.

The lung is among the most radiosensitive organs and a principal dose-limiting organ in thoracic radiotherapy, yet the acute metabolic consequences of irradiation at the lipidomic level remain poorly characterized. We constructed a time-resolved lipidomic atlas of the murine lung response to high-dose irradiation. Male C57BL/6 J mice received a single 10 Gy whole-body X-ray dose, and lungs were collected at 5 minutes, 1 hour, 4 hours, and 24 hours post-exposure (n = 5 per group) and analyzed by untargeted liquid chromatography-tandem mass spectrometry. Radiation triggered rapid and coordinated

Deep Venous Thrombosis Within Two Weeks of Initiating Oral Semaglutide (Rybelsus): A Case Report.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) are increasingly prescribed for type 2 diabetes mellitus and weight management, with growing off-label use in young adults. While GLP-1RAs are generally well tolerated, emerging evidence suggests potential associations with thromboembolic events. We report the case of a 16-year-old male with mild conventional risk factors for thrombosis [overweight body mass index (BMI) and prolonged sedentary behavior] who developed acute left lower extremity deep venous thrombosis (DVT) within two weeks of initiating oral semaglutide (Rybelsus, 7 mg daily)

Lipid metabolism in the aphid-Buchnera symbiosis: an underexplored dimension of insect physiology.

Lipids are essential biomolecules that function as energy reserves, signaling mediators, and structural components of cellular membranes, yet their roles in insect-microbe symbiosis remain poorly understood. In contrast to the well-established contributions of microbial symbionts to amino acid and vitamin metabolism, symbiotic lipid metabolism has received comparatively little attention. Here, we review current knowledge of lipid metabolism in insect symbiosis, focusing on the aphid-Buchnera association as a model of obligate mutualism. Physiological evidence indicates that aphids rely primari

Mitochondrial Dysfunction Predisposes Radioresistant Insect Cells to Ionizing Radiation by Impairing DNA Repair.

Radioresistant insect cell lines, such as Spodoptera frugiperda Sf9 cells, are well-established eukaryotic model systems for investigating cellular responses to ionizing radiation (IR). Mitochondria are central to these responses due to their roles in reactive oxygen species (ROS) production and in regulating apoptotic signaling pathways. In this study, we compared the radiation responses of parental Sf9 cells with those of a mitochondrial dysfunction variant, Sf9N, to elucidate the contribution of mitochondria to the intrinsic radioresistance of lepidopteran insect cells. Both cell types were

Membrane properties modulate methane oxidation by particulate methane monooxygenase.

The copper-dependent membrane monooxygenases particulate methane monooxygenase (pMMO) and ammonia monooxygenase (AMO) oxidize methane to methanol and ammonia to hydroxylamine, respectively. These enzymes, which are important targets for biotechnology, reside in intracytoplasmic membranes (ICMs) where they form densely packed hexagonal arrays. While cryoEM structures of pMMO and AMO in ICMs have revealed closely associated lipids, little is known about how specific lipids and membrane morphologies influence activity. Here we show through cryoelectron tomography (cryoET) that three species of me

Mitochondrial immunometabolism in sepsis: bridging immune cell dysfunction and organ failure.

Sepsis, defined as life-threatening organ dysfunction caused by a dysregulated host response to infection, remains a leading cause of mortality in critical care, and sepsis-associated multiple organ failure continues to defy effective therapy. Increasing evidence positions mitochondria at the interface of cellular bioenergetics and innate immune signaling, making mitochondrial immunometabolism a compelling framework for understanding sepsis pathophysiology. In this mini-review, we synthesize how mitochondrial bioenergetic dysfunction shapes immune cell function across the dynamic course of sep

SLC25A51 and mitochondrial NAD⁺ transport in acute myeloid leukemia: mechanisms, therapeutic potential, and translational perspectives.

Acute myeloid leukemia (AML) remains a highly lethal hematologic malignancy characterized by metabolic reprogramming, therapeutic resistance, and poor survival, particularly in older patients. Nicotinamide adenine dinucleotide (NAD⁺) metabolism has emerged as a central driver of AML progression, and recent studies have identified solute carrier family 25 member 51 (SLC25A51) as the primary mitochondrial NAD⁺ transporter in mammalian cells. SLC25A51 regulates mitochondrial redox balance, oxidative phosphorylation, and tricarboxylic acid (TCA) cycle activity, thereby sustaining leukemic prolifer

Diaphragmatic Mitochondrial Myopathy in a Patient-Derived Mouse Model of Barth Syndrome.

Barth syndrome (BTHS) is a rare, X-linked genetic disorder caused by mutations in the enzyme TAFAZZIN (TAZ), resulting in insufficient cardiolipin (CL) remodeling and mitochondrial dysfunction. While BTHS respiratory distress and breathing difficulties are commonly reported, the precise role of intrinsic respiratory tissue vulnerabilities has only recently begun to be appreciated. Historically, BTHS respiratory distress is frequently attributed to secondary consequences like cardiomyopathy or generalized skeletal myopathy, leaving the intrinsic vulnerability of vital respiratory muscles poorly

Mitochondria-targeted NIR-II organic probes for imaging-guided photodynamic therapy.

Fluorescence imaging-guided photodynamic therapy (PDT) is a promising modality for precision cancer treatment, yet most organic photosensitizers emitting in the visible or first near-infrared (NIR-I) region suffer from limited penetration, strong photon scattering, and high tissue autofluorescence. Organic fluorophores with second near-infrared emission (NIR-II, >1000 nm) mitigate these constraints, enabling deeper imaging, improved spatial resolution, and higher signal-to-background ratios for accurate tumor delineation, real-time therapeutic guidance and longitudinal monitoring of PDT respon

NheABC is a pH-dependent cytotoxin that targets mitochondria and contributes to the virulence of Bacillus cereus in wound infections.

Members of the Bacillus cereus group are Gram-positive, spore-forming, facultative anaerobic bacteria widely distributed in the environment. Pathogenic B. cereus group strains have been linked to a range of illnesses and infections, including severe non-gastrointestinal infections, such as wound infections. In the present study, we show that, among the pore-forming toxin (PFT)-producing bacterial strains tested, B. cereus group strain 75/95 induced pronounced membrane permeabilization in epithelial cells, prompting us to investigate the contribution of its tripartite NheABC toxin to host cell

Lupus Anticoagulant-hypoprothrombinemia Syndrome: A Review Enriched by a New Particular Patient.

Lupus anticoagulant-hypoprothrombinemia syndrome (LAHS) is a rare hemorrhagic disorder that should be differentiated from classical antiphospholipid syndrome. Literature review shows that LAHS may affect people at any age but approximately 40% are children younger than 10 years. Autoimmune and infectious diseases are the most frequent triggering causes, and the laboratory profile is characterized by a prolongation of prothrombin time (PT) and activated partial thromboplastin time (aPTT) with a mild to severe reduction in factor II levels. In more than half the patients, the other coagulation f

Antiphospholipid antibodies in patients with COVID-19: a systematic review and meta-analysis.

COVID-19 patients are at an increased risk of developing thrombotic events, with venous thromboembolism (VTE) occurring in 16% and arterial thrombosis in 11.1% of cases. This systematic review and meta-analysis aimed to explore the prevalence of anti-phospholipid antibodies (aPLs) in COVID-19 patients and their potential role in thrombotic complications. A comprehensive literature search across PubMed, Scopus, and Web of Science identified 48 studies from an initial pool of 1,819 articles, which met the inclusion criteria and were rigorously evaluated for methodological quality. The analysis o

Novel MAFG-METTL14-SCD1 axis regulates lipid metabolism mediating choroidal melanoma distant metastasis.

BACKGROUND: Tumor invasion and metastasis are strongly influenced by cell membrane fluidity, regulated by lipid metabolism. In choroidal melanoma (CM), a highly metastatic cancer, the relationship between lipid metabolism, membrane fluidity, and metastatic mechanisms remains unclear. METHODS: We examined m6A methylation in CM patient samples. Lipidomic profiling was performed in control, METTL14-silenced, or SCD1-silenced CM cells. Transcriptomics were analyzed after METTL14 manipulation. Transmission electron microscopy assessed ultrastructural changes, while multiplex immunohistochemistry v

Glycerophospholipids: Roles in Cell Trafficking and Associated Inborn Errors.

Glycerophospholipids (GPLs) are the main lipid components of cellular membranes. They are implicated in membrane structure, vesicle trafficking, neurotransmission, and cell signalling. GPL molecules are amphiphilic, organized around the three carbons of glycerol. Positions sn-1 and sn-2 are each esterified to a fatty acid (FA). At position sn-3, a phosphate group is linked, which in turn can bind a polar head group, the most prevalent classes being phosphatidic acid (PA, phosphate alone as head group), phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylserine (PS), phosphatid
