Stimulation of T-helper cell gamma interferon and immunoglobulin G responses specific for Babesia bovis rhoptry-associated protein 1 (RAP-1) or a RAP-1 protein lacking the carboxy-terminal repeat region is insufficient to provide protective immunity against virulent B. bovis challenge.

Rhoptry-associated protein 1 (RAP-1) is a targeted vaccine antigen for Babesia bovis and Babesia bigemina infections of cattle. The 60-kDa B. bovis RAP-1 is recognized by antibodies and T lymphocytes from cattle that recovered from infection and were immune to subsequent challenge. Immunization with native or recombinant protein was reported to reduce parasitemias in challenged animals. We recently reported that the NT domain of B. bovis RAP-1 contained immunodominant T-cell epitopes, whereas the repeat-rich CT domain was less immunostimulatory for T lymphocytes from cattle immune to B. bovis.

Identification of distinct gene and miRNA expression profiles in severe early-onset COPD providing novel insights in disease pathogenesis.

OBJECTIVE: Severe early-onset-chronic obstructive pulmonary disease (SEO-COPD) represents the most severe form of COPD that manifests at an earlier age than common COPD. We hypothesise that this is driven by a partially different disease pathogenesis and investigated whether SEO-COPD has a distinct gene and miRNA expression profile. METHODS: RNA and small RNA sequencing was performed on lung tissue from 18 SEO-COPD, 22 common COPD and 32 non-COPD control subjects. Differential gene expression analysis was performed to identify SEO-COPD-specific genes and miRNA signatures. This was followed by

Polygenic Risk Scores of Coronary Heart Disease in Chronic Kidney Disease Population: The CRIC Study.

BACKGROUND: Cardiovascular disease is a major cause of death in individuals with chronic kidney disease; thus, improving its prediction is critical for cardiovascular disease risk reduction. Studies have not assessed whether a polygenic risk score (PRS) of coronary heart disease (CHD) developed from genetic studies in the general population can improve cardiovascular disease risk prediction in individuals with chronic kidney disease, especially when PRS is added to the American Heart Association Predicting Risk of Cardiovascular Disease EVENTs (PREVENT) equations. METHODS: Focusing on a 10-ye

Genetic architecture and evolution of stripe rust resistance uncovered using diverse panels of wheat lines and North American Puccinia striiformis f. sp. tritici isolates.

Screening global wheat germplasm with a diverse collection of pathogen races expands the catalog of novel Yr loci and identifies new sources of broad-spectrum resistance against evolving Pst populations. Newly emerging highly virulent races of Puccinia striiformis f. sp. tritici (Pst) often defeat deployed resistance genes (Yr), highlighting the need for novel sources of durable resistance. A global diversity panel of 377 spring wheat (Triticum aestivum L.) lines was screened for all-stage resistance (ASR) against a panel of diverse 20 Pst isolates at the seedling stage and for adult-plant-sta

PDMD: A Comprehensive Repository of Plants Reported for Skeletal Muscle-related Ailments.

INTRODUCTION: Medicinal plants and phytocompounds targeting skeletal muscle wasting in humans are under-represented in the majority of databases reporting plant/herb-diseases association. However, a large body of literature exists wherein plant extracts or active pharmaceutical ingredients thereof demonstrate potential benefit in skeletal muscle wasting diseases across model organisms. Underscoring the relevance of a repertoire documenting such medicinal plants, we introduce PDMD (Plants Database for Muscle Wasting Diseases), a manually curated plants database reported for muscle wasting disea

Layered immune biomarker monitoring in IgA nephropathy: from mucosal IgA dysregulation to precision therapeutic response assessment.

IgA nephropathy (IgAN) is the most common primary glomerular disease worldwide and remains a major cause of chronic kidney disease and kidney failure. Conventional monitoring still depends largely on proteinuria, estimated glomerular filtration rate (eGFR), hematuria, blood pressure and biopsy-based pathological classification. These measures are clinically indispensable, but they mainly capture downstream kidney injury and may lag behind the mucosal and systemic immune events that drive active disease. In the targeted-therapy era, this limitation has become more consequential. Targeted-releas

Beyond silencing: integrative multi-omics and spatial profiling unravel the systems-level role of piRNAs in HBV-driven Hepatocarcinogenesis.

Hepatocellular carcinoma (HCC) driven by chronic hepatitis B virus (HBV) infection remains a global health challenge, with late diagnosis and limited therapeutic options underscoring the need for novel biomarkers and mechanistic insights. Once considered guardians of the germline, PIWI-interacting RNAs (piRNAs) are now emerging as key somatic regulators of chromatin, transcription, and signaling in cancer. This review considers HBV a plausible perturbant of the hepatic piRNA ecosystem, through viral integration, epigenetic remodeling, and altered intercellular communication. Unlike prior revie

Plant-derived nanocarriers & nanostructures for barrier-defined translation: a comprehensive review of mechanisms, targets, and translation.

Plant-derived nanocarriers (PDNs) constitute a heterogeneous family of bioinspired delivery platforms, including plant-derived extracellular vesicles, lipid-based nanovectors, and plant viral nanoparticles, that have attracted growing interest for applications in diseases constrained by biological barriers. A critical challenge in this field is distinguishing descriptive reports of barrier interaction from mechanistically and translationally meaningful evidence. This review provides a structured synthesis of plant-derived nanocarriers through a barrier-defined framework, rather than a platform

Identifying shared polygenic risk across cancers.

BACKGROUND: Shared genetic susceptibility across cancers has been reported but is generally modest at the genome-wide level. Whether such shared polygenic risk exhibits structured convergence at regional or functional levels remains unclear. We investigated shared genetic risk across cancers by integrating local genetic correlation analyses with cross-cancer polygenic risk score (PRS) associations. METHODS: We estimated pairwise local genetic correlations across 16 specific cancers and one pan-cancer phenotype using SUPERGNOVA. Genome regions harboring multiple cancers with mutually correlate

Modulating limbic circuits in temporal lobe epilepsy: impacts on seizures, memory, mood and sleep.

Temporal lobe epilepsy is a common neurological disease characterized by recurrent seizures that often originate within limbic networks involving amygdala and hippocampus. The limbic network is involved in crucial physiologic functions involving memory, emotion and sleep. Temporal lobe epilepsy is frequently drug-resistant, and people often experience comorbidities related to memory, mood and sleep. Deep brain stimulation targeting the anterior nucleus of the thalamus (ANT-DBS) is an established therapy for temporal lobe epilepsy. However, the optimal stimulation parameters and their impact on
