Amyloid beta (Aβ) oligomers are thought to play an important role during development and progression of Alzheimer's disease (AD). Previously, we determined the Aβ oligomer concentrations in various AD mouse models and in human brain tissues of former AD patients. Here, we investigate which proteins are part of these Aβ oligomers, apart from Aβ itself. Because several oligomer-associated proteins have been implicated in mechanisms leading to AD pathology, identification of the Aβ oligomer proteome may provide insights into the formation of Aβ oligomers in vivo and may reveal novel targets for d

Sperm selection by density-gradient centrifugation (DGC) is widely used prior to bovine in vitro fertilization (IVF) but may affect sperm DNA integrity. Microfluidic sperm selection (MFSS) is a less invasive alternative; however, whether its effects depend on the initial level of sperm DNA fragmentation remains unclear. This study examined the effects of MFSS on sperm quality, fertilization status, first cleavage pattern, blastocyst development, ploidy, and bioenergetics using cryopreserved semen from Simmental bulls with low or high DNA fragmentation index (%DFI). Semen from 11 bulls was clas

In vitro embryo production (IVEP) has become an essential tool for livestock breeding, genetic dissemination, and research. Despite its widespread use, overall efficiency remains limited, as only approximately 25% of fertilized oocytes develop into transferable embryos, and pregnancy outcomes are consistently lower than with in vivo-derived embryos. While oocyte quality is a major determinant, fertilization success and early embryonic development are also strongly influenced by sperm functional competence, which is only partially captured by conventional selection methods based primarily on mo

Extracellular vesicles (EVs) have dominated research on intercellular communication for decades. However, the discovery of exomeres in 2018 … and structural features of NVEPs. It then compares the respective limitations of differential ultracentrifugation, density gradient centrifugation, size-exclusion chromatography, and flow field-flow fractionation for NVEP isolation, together with strategies for their combined use. Furthermore, the functional mechanisms that distinguish NVEPs from vesicular EVs are discussed at t …

BACKGROUND: Chronic hepatitis B virus (HBV) infection is characterized by selective impairment of virus-specific immune responses. However, progressive liver disease, particularly cirrhosis, is associated with broader immune remodeling that may influence B cell phenotype and function. CD72 is a negative regulator of B cell receptor (BCR) signaling but its role in CD5+B cell regulation under these conditions remains unclear.
METHODS: Peripheral Blood Mononuclear cells (PBMCs) were isolated from 51 HBV-infected patients at differing disease stages and 24 healthy controls by density gradient cen

We have established and optimized a robust protocol to isolate, culture, and study primary human brain myeloid cells (BrMCs) from rapid-autopsy postmortem brain tissues obtained from people with or without HIV-1 (HIV), in which >97.3% of BrMCs are brain microglia (MG). Therefore, we refer to these isolated BrMCs as MG or BrMCs/MG to better represent the cell populations. This method enables comprehensive ex vivo investigation of microglial biology, including their roles in maintaining HIV reservoirs in the brain, mediating neuroinflammation, and contributing to neuronal injury, as well as the

The current clinical application of the boron drug boronophenylalanine (BPA) in BNCT faces various issues owing to its low boron loading (approximately 5%), which limits its therapeutic efficacy. Therefore, the development of boron drugs with higher boron contents is essential. Enhancing the boron content in boron drug materials is the focus of this study. Two-dimensional (2D) boron nitride-doped nano graphene (BNNG) with a high boron content of 24.97% ± 1.14% w/w was synthesized via the chemical vapor deposition (CVD) method. The size of BNNG was controlled through gradient density centrifuga

Considering the challenges surrounding causative factors in male infertility, rather than relying on standard semen analysis, the assessment of sperm subpopulations and functional characteristics essential for fertilization is paramount. Furthermore, the diagnostic value of sperm interactions with biological components in the female reproductive tract may improve our understanding of subfertility and provide applications in assisted reproductive techniques. We investigated the response of two sperm motility subpopulations (mimicking the functionality of potentially fertile and sub-fertile seme

Isolation and analysis of circulating rare cells is a promising approach for early detection of cancer and other diseases and for prenatal diagnosis. Isolation of rare cells is usually difficult due to their heterogeneity as well as their low abundance in peripheral blood. We previously reported a two-stage ensemble-decision aliquot ranking platform (S-eDAR) for isolating circulating tumor cells from whole blood with high throughput, high recovery rate (>90%), and good purity (>70%), allowing detection of low surface antigen-expressing cancer cells linked to metastasis. However, due to the sca

BACKGROUND AND AIMS: The Hepatitis E virus hijacks the endosomal system for its release. These structures are highly dependent on cholesterol. Hence, this study investigates the impact of HEV on cholesterol-metabolism, the effect of intracellular cholesterol content on HEV-release and the potential of cholesterol-modulators to serve as antivirals.
METHODS: Intracellular cholesterol-content of cells was modulated and impacts on HEV were monitored using qPCR, Western blot, microscopy, virus-titration and density-gradient centrifugation. Blood-lipids and HEV-RNA were routinely quantified in chro

BACKGROUND: Small cell lung cancer (SCLC) is highly aggressive and is associated with a dismal prognosis. However, there are no clinically recognized biomarkers for early diagnosis. In this study, we used quantitative proteomics to build differential mitochondrial protein profiles that may be used for early diagnosis and investigated the pathogenesis of lung cancer.
METHODS: We cultured SCLC cells (NCI-H446) and normal human bronchial epithelial cells (16-HBE); mitochondria were extracted and purified using differential and Percoll density gradient centrifugation. Subsequently, we used Wester

BACKGROUND/AIMS: Tonsillectomy may be an important method to achieve a long-term remission of IgAN, but patients' physical status may limit their access to this surgery. We proposed an encouraging solution through inhibiting GADD34 expression in order to promote tonsillar mononuclear cells (TMCs) apoptosis and reduce nephropathic IgA secretion.
METHODS: A total of 12 IgAN and 9 non-IgAN patients were involved from March 2015 to May 2016. After TMCs were extracted by density gradient centrifugation and stimulated by inactivated hemolytic streptococcus, the mRNA and protein expression of GADD34

BACKGROUND: Blood-brain barrier impairment is a major indicator of endothelial dysfunction in diabetes. Studies showed that endothelial progenitor cell (EPC) transplantation promoted angiogenesis and improved function recovery after hind limb ischemia in diabetic mice. The effect of EPC transplantation on blood-brain barrier integrity after cerebral ischemia in diabetic animals is unknown. The aim of this study is to explore the effect of EPC transplantation on the integrity of the blood-brain barrier after cerebral ischemia in diabetic mice.
METHODS: EPCs were isolated by density gradient ce
