Gestational Aging on Fast Forward: The Epigenetic Link to Recurrent Pregnancy Loss.

PROBLEM: Recurrent pregnancy loss (RPL) is a distressing complication with poorly understood causes. Increasing evidence suggests that epige … a strong positive correlation between elevated EGA and RPL, and identified modules associated with critical pathways. CONCLUSION: Our findings suggest that premature epigenetic aging of the placenta and disrupted networks regulating apoptosis, DNA repair, oxidative stress, and immune responses may underlie RPL. These insights highlight the importance of epigeneti …

HypoxamiRs in pancreatic cancer: master regulators of the hypoxic tumor microenvironment.

Pancreatic cancer (PC) is characterized by aggressive progression, chemoresistance, and immune evasion, largely driven by its hypoxic tumor microenvironment (TME). The extensive desmoplastic reaction in PC, characterized by dense stromal fibrosis, exacerbates hypoxia by impairing blood flow, creating a hostile environment that limits therapeutic efficacy. Hypoxia-induced microRNAs (HypoxamiRs) have emerged as critical regulators of these processes, modulating cellular pathways that promote tumor survival and therapy resistance. HypoxamiRs exert their effects by interacting with hypoxia-inducib

The Role of Mitochondrial DNA in Modulating Chemoresistance in Esophageal Cancer: Mechanistic Insights and Therapeutic Potential.

Chemotherapy remains a cornerstone in the treatment of esophageal cancer (EC), yet chemoresistance remains a critical challenge, leading to poor outcomes and limited therapeutic success. Mitochondrial DNA (mtDNA) has emerged as a pivotal player in mediating these responses, influencing cellular metabolism, oxidative stress regulation, and apoptotic pathways. This review provides a comprehensive overview of the mechanisms by which mtDNA alterations, including mutations and copy number variations, drive chemoresistance in EC. Specific focus is given to the role of mtDNA in metabolic reprogrammin

Opportunities to encourage adoption of a biomarker-enabled care pathway for Alzheimer's in primary care.

UNLABELLED: Identification of early-stage Alzheimer's disease (AD) remains a challenge due to limited specialist availability, diagnostic access, disease awareness, and cultural factors. Blood-based biomarkers (BBBM) could play a critical role in the identification and referral of patients suspected of AD to specialty care. A multidisciplinary AD Biomarker Task Force was convened to evaluate current biomarker use cases, define an optimal biomarker-enabled AD diagnostic care pathway, and understand factors impacting adoption. The Task Force identified opportunities to support biomarker-enabled

GABAergic signaling by VIP interneurons gates running-dependent visual recovery in the adult brain.

Experience-dependent plasticity in the adult visual cortex is enhanced by locomotion, a process mediated by vasoactive intestinal peptide (VIP)-expressing interneurons. While VIP interneurons are known to signal through both Gamma-aminobutyric acid (GABA) and VIP peptide, the specific contributions of these pathways during different forms of plasticity remain unclear. Monocular deprivation (MD) in adult mice alters cortical responses, though more slowly and differently than during a critical period in early life. Here, we used two-photon calcium imaging in awake adult mice to dissect the roles

TLR2 and TLR4 bridge physiological and pathological inflammation in the reproductive system.

Toll-like receptors (TLRs), particularly TLR2 and TLR4, are critical components of the innate immune system that play significant roles in reproductive biology beyond their well-established functions in immune defense. These receptors recognize distinct pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs), triggering signaling cascades that impact diverse reproductive processes. TLR2-induced physiological inflammation, characterized by transient and precisely controlled inflammatory responses, is essential for facilitating sperm-triggered uterine clea

Developing zebrafish utilize taste-signaling pathways for oxygen chemoreception.

A fundamental requirement for all animals is to sense and respond to changes in environmental O2 availability. Low O2 (hypoxia) typically stimulates breathing, a universal and critical response termed the hypoxic ventilatory response (HVR). In this study, we test the hypothesis that taste-signaling pathways are used for O2 sensing and activation of the HVR. We show that Merkel-like cells (MLCs), which are part of the taste-bud complex, function as O2 chemoreceptor cells in larval zebrafish and that transduction of the O2 signal uses taste-signaling pathways. Specifically, MLCs responded to hyp

Pulmonary arterial hypertension.

Pulmonary arterial hypertension (PAH) is a rare and debilitating disease characterized by abnormal proliferation and contraction of pulmonary vascular smooth muscle cells. The resulting increase in pressure and pulmonary vascular resistance results in progressive right heart failure, low cardiac output, and ultimately death if left untreated. PAH is defined by a persistent elevation in pulmonary artery pressure with normal left-sided pressures, differentiating it from left-sided heart disease. Symptoms progress from shortness of breath and decreasing exercise tolerance to right heart failure,

Enhanced immunoreceptor tyrosine-based activation motif signaling is related to pathological bone resorption during critical illness.

Prolonged critically ill patients present with distinct alterations in calcium and bone metabolism. Circulating bone formation markers are reduced and bone resorption markers are substantially elevated, indicating an uncoupling between osteoclast and osteoblast activity, possibly resulting in pronounced bone loss, impaired traumatic or surgical fracture healing, and osteoporosis. In addition, we have previously shown that increased circulating osteoclast precursors in critically ill patients result in increased osteoclastogenesis in vitro, possibly through FcγRIII signaling. In the current stu

[Critically Ill prehospital patients : direct admission to intensive care unit ?].

Reducing wait times for critical care is a major organizational challenge. Several studies suggest an association between delayed admission to the intensive care unit (ICU) and a poor prognosis in critically ill patients. The Swiss healthcare system, characterized by advanced prehospital medical care and specialized care pathways that are already operational, provides a favorable context for establishing direct or expedited pathways to ICUs starting from the prehospital care phase. However, this approach raises significant challenges related to over-triage, resource availability, and the safet

The role of nutrient stress in DNA damage.

Cells are constantly exposed to various stresses, including nutrient deprivation and genotoxic stress, which dynamically interact with cellular sensing pathways to influence metabolism, gene expression, and homeostasis. The integration of nutrient-sensing mechanisms and DNA damage response pathways is critical in cancer progression. While individual processes are well-characterized, their cross-regulatory mechanisms are just beginning to emerge. Deciphering the interplay between nutrient stress and DNA damage is crucial for elucidating the mechanisms underlying cellular responses to stress and

Micro- and Macrovascular Effects of Inflammation in Peripheral Artery Disease-Pathophysiology and Translational Therapeutic Approaches.

Inflammation has a critical role in the development and progression of atherosclerosis. On the molecular level, inflammatory pathways negatively impact endothelial barrier properties and thus, tissue homeostasis. Conformational changes and destruction of the glycocalyx further promote pro-inflammatory pathways also contributing to pro-coagulability and a prothrombotic state. In addition, changes in the extracellular matrix composition lead to (peri-)vascular remodelling and alterations of the vessel wall, e.g., aneurysm formation. Moreover, progressive fibrosis leads to reduced tissue perfusio

Peripheral T-Cell Lymphoma: Moving Toward Targeted Therapies.

Therapeutic advances for peripheral T-cell non-Hodgkin lymphoma (PTCL) have lagged behind their B-cell NHL counterparts in part because novel agents to treat PTCL have been developed empirically. The recent clinical success of brentuximab-vedotin suggests that novel therapies for PTCL can significantly improve outcomes when properly targeted. Aberrancies in T-cell receptor, Jak/STAT, and DNA methylation pathways play critical roles in T-NHL pathogenesis based on genomic studies and preclinical experimental validation. New strategies targeting these pathways in patients with PTCL are underway,

The Impact of Generative AI Dependence on University Students' Creativity: The Mediating Role of Critical Thinking and the Moderating Role of Thinking Orientation.

The rapid proliferation of generative artificial intelligence (AI) has raised concerns about its impact on creativity, yet mechanisms and boundary conditions remain underexplored. Using one survey and two experiments, we investigated how AI dependence affects university students' creativity. Study 1 (cross-sectional) found AI dependence was negatively related to creativity, fully mediated by critical thinking. Study 2 (between-subjects experiment) experimentally demonstrated that high AI dependence was associated with lower creativity than low dependence, with critical thinking again mediating

Immune shifts in inflammatory skin diseases.

Inflammatory skin diseases are driven by dominant immune programs, including T helper (Th)1, Th2, Th17, type I IFN, and neutrophilic pathways, which shape responsiveness to targeted immunotherapies. Molecular profiling has revealed that these diseases frequently harbor additional, coexisting immune programs and that dominance among pathways can shift over time, either spontaneously or in response to therapy. Such immune shifts, which limit therapeutic effectiveness, often occur in the absence of clinically evident morphological changes. In this review, we describe the molecular immune landscap

DNA Damage Repair: A Crucial Link to Neurological Disorders.

Environmental stressors and endogenous metabolites constantly threaten DNA integrity; therefore, terrestrial organisms have evolved sophisticated repair and maintenance pathways to eliminate DNA lesions and preserve genome stability. Defective or inefficient DNA repair mechanisms compromise genomic integrity and contribute to the onset and progression of various diseases. In recent years, numerous proteins and signaling pathways essential for DNA damage repair have been extensively characterized. Notably, many of these factors also play critical roles in brain development and are implicated in

Pathological Mechanisms Involved in HIV-Associated Lymphomagenesis: Novel Targeted Therapeutic Approaches.

The intricate interplay of direct and indirect mechanisms relating to immune dysfunction, chronic inflammation, and viral proteins represents a key factor of lymphomagenesis in HIV-infected patients. Indirect mechanisms based on cytokine dysregulation, HIV-induced immune dysfunction, and co-infections with oncogenic viruses induce chronic B-cell activation and generation of a prone environment for malignant transformation and tumor growth. Direct mechanisms arise from oncogenic influences of p17, Tat, and Nef HIV proteins, which generate genomic instability, alteration of cellular signaling, a

NAD in pathological cardiac remodeling: Metabolic regulation and beyond.

Nicotinamide adenine dinucleotide (NAD) coenzymes are carriers of high energy electrons in metabolism and also play critical roles in numerous signaling pathways. NAD metabolism is decreased in various cardiovascular diseases. Importantly, stimulation of NAD biosynthesis protects against heart disease under different pathological conditions. In this review, we describe pathways for both generation and catabolism of NAD coenzymes and the respective changes of these pathways in the heart under cardiac diseases, including pressure overload, myocardial infarction, cardiometabolic disease, cancer t

Transformation of macrophages into myofibroblasts in fibrosis-related diseases: emerging biological concepts and potential mechanism.

Macrophage-myofibroblast transformation (MMT) transforms macrophages into myofibroblasts in a specific inflammation or injury microenvironment. MMT is an essential biological process in fibrosis-related diseases involving the lung, heart, kidney, liver, skeletal muscle, and other organs and tissues. This process consists of interacting with various cells and molecules and activating different signal transduction pathways. This review deeply discussed the molecular mechanism of MMT, clarified crucial signal pathways, multiple cytokines, and growth factors, and formed a complex regulatory networ
