The case for and concerns about continuous dopamine stimulation in Parkinson's disease.

Delivery of levodopa or other dopaminergic agents to the striatum is a critical determinant of the clinical response and may account for many of the fluctations in response ("on-off"). This should be manageable with innovative delivery systems for the drugs. However further studies are required to determine if continuous dopaminergic stimulation will be complicated by increased levels of drug metabolites or down regulation of dopaminergic neurotransmission.

Dopamine, acetylcholine, and glutamate interactions in aging. Behavioral and neurochemical correlates.

Aging, hypoxia, and thiamin deficiency diminish motor performance. Similar alterations of ACh, DA, and glutamate metabolism accompany hypoxia, thiamin deficiency, and aging. Both aging and hypoxia reduce ACh release and stimulate DA and glutamate release. Presynaptic enhancement of DA and glutamate release may be important in the production of cell damage that may contribute, in part, to age-related deficits in motor as well as cognitive function. The decline in ACh release may be important in the production of the cognitive deficits. An understanding of the interactions of neurotransmitters i

Narcotic antagonism of seizures induced by a dopamine-derived tetrahydroisoquinoline alkaloid.

This paper describes experiments designed to evaluate whether the narcotic antagonist naloxone significantly interferes with seizures induced by tetrahydroisoquinolines (TIQs). In these experiments we found that naloxone significantly reduced seizure scores induced by intra-cranially infusing mice with 50 micrograms of the dopamine-derived tetrahydroisoquinoline (TIQ) alkaloid, 6,7-dihydroxy TIQ. These findings support an opioid involvement in the actions of TIQs and may lead to further understanding of opioid-mediated novel excitatory receptors.

Experimental and clinical approaches to treatment of hypertension by dopamine receptor agonists.

The action of several dopamine agonists on blood pressure was investigated. In dogs, apomorphine induced a decrease in blood pressure and an increase in heart rate. These effects were suppressed by intravenous haloperidol or phentolamine. Intravenous but not intracisternal domperidone suppressed the hypotensive responses elicited by intravenous or intracisternal apomorphine. Furthermore, the hypotensive effect of bromocriptine or apomorphine was abolished in adrenal demedullated dogs. Apomorphine and bromocriptine decreased adrenal catecholamine levels. In men, the acute hypotensive property o…

Effect of dopamine agents on schedule- and deprivation-induced drinking in rats.

The dopamine agonist, apomorphine, or its antagonist, haloperidol, was administered to rats whose drinking was induced by fixed-interval schedules of pellet delivery or by water deprivation. The first study revealed that both drugs produced dose-dependent decreases in bar-pressing and schedule-induced polydipsia (SIP). At higher doses, haloperidol also depressed the rate of pellet delivery. The second study demonstrated that the suppression in SIP obtained in the first study was primarily due to the direct effect of the drugs and not to changes they produced on the underlying food reinforcemen…

Behavioural role of dopamine D1 receptors in the reserpine-treated mouse.

The effects of 2,3,4,5-tetrahydro-7,8-dihydroxy-1-phenyl-1H-3-benzazepine (SKF 38393) (D1 agonist) on the motor behaviour of mice rendered hypokinetic with reserpine, were studied in the absence and presence of additional treatment with N-n-propyl-N-phenylethyl-p(3-hydroxyphenyl)ethylamine hydrochloride (RU 24213), lisuride (D2 agonists) or apomorphine (mixed D1/D2 agonist). Three hours after reserpine (5 mg/kg) stimulating dopamine D2 receptors evoked slow, ponderous walking and head-down sniffing. SKF 38393 (1.5-15 mg/kg) had no direct effect of its own, but greatly amplified the D2 response…

Dopamine agonists in the treatment of hyperprolactinemia. Comparison between bromocriptine and lisuride.

27 patients (12 women and 15 men) with hyperprolactinemia were treated with the dopamine agonists lisuride (Dopergin) and bromocriptine on a cross-over basis for 3-6 months. By this treatment, plasma prolactin levels were reduced by 83 and 87%, and normalization was achieved in 13 patients in the lisuride group (average dosage 1 mg/d) and in 15 patients in the bromocriptine group (average dosage 10 mg/d). Side effects, consisting of orthostatic hypotension, vomiting and nausea, occurred in 11 patients treated with lisuride and 13 patients treated with bromocriptine, and 2 patients in the lisur…

Ventilatory effects of domperidone, a new dopamine antagonist, in anaesthetized rabbits.

The ventilatory effects of domperidone (DP), a dopamine antagonist that does not cross the blood-brain barrier, were studied on rabbits anaesthetized with pentobarbitone. DP induced hyperventilation (+20%), which was not dose-dependent, without any marked change in arterial blood gases. This effect was abolished by previous transection of both carotid sinus nerves. On an average, steady state hypoxia (PaO2 approximately 45 mmHg for 5 min) doubled ventilation before, and increased itself threefold after DP (0.10 mg kg-1 i.v.). Prazosin (PZ, 0.1 mg kg-1 i.v.), an alpha 1-adrenoceptor antagonist,…

EMD 23,448: effects of a putative dopamine autoreceptor agonist in chorea.

A novel dopamine agonist, which stimulates presynaptic dopamine receptors in the experimental animal, was administered acutely to four patients with chorea. Abnormal involuntary movements worsened, plasma prolactin levels fell, and plasma growth hormone concentrations increased in all patients tested. These effects resemble those associated with a postsynaptic rather than presynaptic dopamine agonist, and thus raise some question about the ability of conventional dopaminomimetic screens to predict clinical activity.

Effects of dopamine and serotonin on eructation rate and ruminal motility in sheep.

The relationships between forestomach motility and eructation rate were studied during dopamine infusion (2 sheep) and serotonin infusion (2 sheep). The sheep were chronically fitted with strain gauges on the reticulorumen and with a cannula in the dorsal sac of the rumen. A tracheotomy was performed to intercept the eructated gases and to permit measurement of their volume. To maintain a regular rate of eructation during the control periods, experiments were performed with a moderately increased intraruminal pressure obtained by continuous ruminal insufflation of nitrogen. Dopamine and seroto…

The dopamine agonist bromocriptine induces hypotension by venous and arteriolar dilation.

Acute bromocriptine administration reduced sitting and standing blood pressure and produced severe orthostatic hypotension in 12 normal subjects. Concomitantly, there was an increase in venous distensibility and basal blood flow, and a decrease in peripheral vascular resistance, as determined by forearm plethysmography. After administration of bromocriptine, plasma norepinephrine concentration decreased. Bromocriptine lowers blood pressure by dilating arterioles and veins, at least in part by means of dopaminergic inhibition of sympathetic nervous system activity.

The long-acting dopamine agonist pergolide mesylate for treatment of hyperprolactinaemia.

Seven hyperprolactinaemic women were treated with the new, long-acting dopamine agonist pergolide mesylate. The treatment resulted in normalization of the prolactin secretion in four of the seven women and six of them experienced regular uterine bleedings. Four of the patients had previously discontinued bromocriptine because of adverse effects but had no problems to tolerate pergolide. One bromocriptine-resistant woman was unresponsive also to pergolide therapy. Reported side-effects in the seven women were mild and transient. Pergolide mesylate may be a valuable alternative to bromocriptine …

Resistance to barbiturate is changed by developmental alteration of dopamine receptor sensitivity.

Adult rats whose dopamine receptor number was reduced via prenatal haloperidol exposure were less resistant than controls to pentobarbital narcosis. This altered resistance was functional (CNS) and not metabolic, as shown by pentobarbital brain levels. These results are consistent with our previous experiments, indicating that changes in the number of dopamine receptors may underlie relative susceptibility to barbiturates.

[Sulpiride (stereoisomers, racemic) and dopamine: actions and interactions on renal circulation].

Interaction of sulpiride - both 1- and d- isomers as well as racemic- - with Dopamine (DA, subpressor dosage 0.1 microgram X kg -1 X min -1), on the renal hemodynamic, was studied in DOCA-pretreated men during hypotonic polyuria. P.A.H. and creatinine clearance and renal vascular resistances were determined. In the presence of d-Sulpiride, DA - induced renal vasodilation is carried out gradually and finally reaches similar levels as in the absence of d-Sulpiride. However no glomerular filtration rate increase is produced by DA. In the presence of 1-Sulpiride, DA vasodilating effect is suppress…

[Sulpiride (stereoisomers, racemic) and dopamine: actions and interactions on tubular reabsorption].

In DOCA-pretreated men during hypotonic polyuria Dopamine (DA) infusion in a subpressor dosage (0.1 microgram X kg -1 X min -1) produces renal hyperemia, inhibition of isosmotic sodium reabsorption, increase in sodium distal load (s. d.l.) and finally inhibition of anisosmotic sodium reabsorption as a percentage of s.d.l. Such DA effects are variously modified by sulpiride isomers. Sodium distal load increase is still apparent in the presence of racemic, but not in the presence of either d- or l-Sulpiride isomers. However DA inhibition of anisosmotic sodium reabsorption % s.d.l. is unaffected …

Behavioural correlates to the dopamine D-1 and D-2 antagonists.

The acute dopamine (DA) receptor blockade of neuroleptics can be demonstrated in mice by antagonism of stereotypies induced by the DA-agonist methylphenidate and in rats by antagonism of stereotypies induced by the DA-agonists amphetamine or apomorphine. Neuroleptics such as the thioxanthene, cis(Z)-flupentixol, the phenothiazine, fluphenazine, the butyrophenone, haloperidol and the benzamide clebopride are equipotent behaviourally as well as clinically. Also the D-1 receptor-antagonist SCH 23390 has the same pharmacological effects. In a series of experiments where the methylphenidate-induced…

Actions of GABAergic agents on dopamine metabolism in the nigrostriatal pathway of the rat.

The gamma-aminobutyric acid (GABA) transaminase inhibitor aminoxyacetic acid, diazepam and the GABA agonists muscimol, 4,5,6,7-tetrahydroisoxazolo-[5,4-C]-pyridin-3-ol, kojic amine and SL-76002 all depressed nigrostriatal dopamine release as measured by decreased 3-methoxytyramine after parenteral administration. Experiments with intrastriatal and intranigral injections as well as parenteral injections in acutely hemisected and kainate lesioned animals demonstrated that parenteral muscimol inhibits the nigrostriatal pathway at the level of the substantia nigra. In contrast, parenteral 4,5,6,7-…

Effects of CDP-choline on striatal dopamine level and behavior in rats.

To further assess the effects of CDP (cytidine diphosphate)-choline on Parkinsonian symptoms, striatal dopamine (DA) was measured fluorometrically in rats after injection of CDP-choline. CDP-choline (300 mg/kg, i.p.) increased the DA content in the striatum (p less than 0.05) one hour after injection. The behavioral effect of CDP-choline was then tested in rats in which the unilateral nigro-striatal DA neurons had degenerated following an intranigral injection of 6-hydroxydopamine (6-OHDA). CDP-choline alone did not produce behavioral changes in these rats. However, pretreatment with a single …
