Systematic druggable genome-wide Mendelian randomization identifies therapeutic targets for hyperemesis gravidarum.

BACKGROUND: Hyperemesis gravidarum (HG), excessive vomiting in pregnancy, occurs in 0.3-10.8% of pregnancies and is associated with maternal and fetal morbidity. Despite the existence of several off-label treatment options that have shown clinical effectiveness in managing HG symptoms, the variability in treatment response highlights the need for more effective therapies. Our study aims to identify novel therapeutic targets that could lead to the development of additional, more effective treatment options. METHODS: A two-sample Mendelian randomization (MR) analysis was performed to estimate t

Mendelian Randomization Analysis of Psoriasis and Multiparameter Renal Function Trajectories.

INTRODUCTION: While observational studies suggest that psoriasis may increase the risk of renal impairment, evidence supporting a causal relationship remains inconclusive. This Mendelian randomization (MR) study systematically investigates the genetic causality between psoriasis and multidimensional renal function trajectories, encompassing both static measures and dynamic progression patterns. METHODS: We utilized genetic variants associated with psoriasis identified in a Finnish population- based genome-wide study of 373,338 participants. Using a two-sample MR framework, we examined potenti

Are neurodegenerative diseases associated with an increased risk of inflammatory bowel disease? A two-sample Mendelian randomization study.

BACKGROUND: Several studies have shown that neurodegenerative diseases (e.g., Parkinson's disease [PD] and Alzheimer's disease [AD]) are associated with inflammatory bowel disease (IBD), but the causality and direction of their associations remain unclear. Mendelian randomization (MR) studies have explored the causal effects of IBD on PD and AD. However, only a few studies examined this reverse association. Thus, this study aimed to explore whether there are causal associations of genetically predicted PD and AD with IBD, using a two-sample MR study. METHODS: Summary statistics for IBD, ulcer

Aerobic Fitness and Health-Related Phenotypes: A Two-Stage Phenome-Wide Mendelian Randomization Study.

PURPOSE: We investigated potentially causal associations between genetically predicted aerobic fitness and multiple health phenotypes using a two-stage phenome-wide Mendelian randomization (MR) study. METHODS: Genetically determined aerobic fitness, as operationalized by Cai et al., served as the exposure instrument. We screened 712 health-related phenotypes as outcomes using publicly available European-ancestry genome-wide association studies (GWAS) summary statistics from OpenGWAS (Discovery GWAS n > 5000), prioritizing non-UK Biobank/non-FinnGen datasets for Discovery when available and se

Associations between reproductive milestones and Alzheimer's disease risk: a Mendelian randomization study.

BACKGROUND: Alzheimer's disease (AD) is a prevalent neurodegenerative disorder. Previous studies have suggested associations between female reproductive history and AD risk, but the causal nature of these relationships remains unclear. This study aimed to investigate the causal relationships between key female reproductive milestones (age at menarche, age at first birth, and menopause) and AD risk using Mendelian randomization (MR) and multivariate MR analysis. METHODS: Two-sample MR and multivariate MR analyses were conducted using pooled data from large-scale genome-wide association studies

Aortic Stenosis and Renal Function: A Bidirectional Mendelian Randomization Analysis.

BACKGROUND: Large observational studies have demonstrated a clear inverse association between renal function and risk of aortic stenosis (AS). Whether this represents a causal, reverse causal or correlative relationship remains unclear. We investigated this using a bidirectional 2-sample Mendelian randomization approach. METHODS AND RESULTS: We collected summary statistics for the primary analysis of chronic kidney disease (CKD) and AS from genome-wide association study meta-analyses including 480 698 and 653 867 participants, respectively. We collected further genome-wide association study s

Pleiotropic loci for cannabis use disorder severity in multi-ancestry high-risk populations.

Cannabis use disorder (CUD) is common and has in part a genetic basis. The risk factors underlying its development likely involve multiple genes that are polygenetic and interact with each other and the environment to ultimately lead to the disorder. Co-morbidity and genetic correlations have been identified between CUD and other disorders and traits in select populations primarily of European descent. If two or more traits, such as CUD and another disorder, are affected by the same genetic locus, they are said to be pleiotropic. The present study aimed to identify specific pleiotropic loci fo

Vitamin D Levels and Risk of Juvenile Idiopathic Arthritis: A Mendelian Randomization Study.

OBJECTIVES: Observational studies report mixed findings regarding the association between vitamin D and juvenile idiopathic arthritis (JIA) incidence or activity; however, such studies are susceptible to considerable bias. Because low vitamin D levels are common within the general population and easily corrected, there is potential public health benefit in identifying a causal association between vitamin D insufficiency and JIA incidence. To limit bias due to confounding and reverse causation, we examined the causal effect of the major circulating form of vitamin D, 25-hydroxy vitamin D (25-[O

The causal effect of matrix metalloproteinase-3 on ankylosing spondylitis: Evidence from Mendelian randomization.

Previous investigations through observation have found that matrix metalloproteinase-3 (MMP-3) has benefits for ankylosing spondylitis (AS) but it is uncertain whether there is a true positive causal connection. Our goal was to demonstrate the relationship between AS and MMP-3. We executed Mendelian randomization (MR) research utilizing genome-wide association studies genetic data (n = 21,758) for MMP-3 publicly available from IEU Open and genome-wide association studies data for AS (n = 297,932) from FinnGen Biobank. The specific MR protocols were weighted median, weighted mode, MR-Egger, and

DNA Methylation at cg18095732 Modulates ZDHHC20 Expression and Decreases Acne Vulgaris Risk.

BACKGROUND: Acne vulgaris is a common chronic inflammatory skin disorder involving lipid metabolism and immune dysregulation. Protein S-palmitoylation regulates lipid homeostasis, while DNA methylation has emerged as a potential contributor to acne pathogenesis. Yet, how DNA methylation and palmitoylation intersect in acne remains unclear. METHODS: The objective of this study was to investigate whether palmitoylation-related genes are causally linked to acne by integrating large-scale genetic and epigenetic datasets through Mendelian randomization and complementary analyses. RESULTS: Our MR

Association between brain resting-state functional activities and migraine: a bidirectional mendelian randomization study.

Researchers have conducted extensive research on the correlation between brain resting-state functional activities (RSFA) and migraine. However, we still do not fully understand the exact nature of the causal relationship between these RSFA and migraine. We conducted a bidirectional two-sample Mendelian randomization (MR) study to investigate the causal association between migraine and RSFA. We gathered summary statistics from genome-wide association studies for 191 resting-state functional magnetic resonance imaging phenotypes. We employed various analytical methods for bidirectional two-samp

Causal association between mTOR-dependent circulating protein levels and central precocious puberty: a Mendelian randomization study.

BACKGROUND: The mechanistic target of rapamycin (mTOR) signaling pathway has a significant effect on central precocious puberty (CPP). However, the causality between mTOR-dependent circulating protein levels and CPP is still unclear. Our aim is to assess the effects of seven mTOR-dependent circulating protein levels on CPP using Mendelian randomization (MR). METHODS: Instrumental variables (IVs) for mTOR-dependent circulating protein levels were retrieved from the proteomics-GWAS INTERVAL study and eQTLGen. The summary-level genetic datasets for CPP outcome were obtained from the FinnGen Cons

Identifying the causal relationship between immune factors and osteonecrosis: a two-sample Mendelian randomization study.

A wealth of evidence intimates a profound connection between the immune system and osteonecrosis, albeit the specific immune factors underlying this connection remain largely veiled. A bidirectional Mendelian randomization (MR) study was conducted based on genome-wide association study summary data to identify causal links between 731 immune factors and osteonecrosis including drug-induced osteonecrosis. Preliminary MR analysis was accomplished utilizing the inverse-variance weighted method under a multiplicative random effects model, and heterogeneity and potential horizontal pleiotropy were

Causal relationship between obesity and iron deficiency anemia: a two-sample Mendelian randomization study.

BACKGROUND: Observational studies have suggested an association between obesity and iron deficiency anemia, but such studies are susceptible to reverse causation and residual confounding. Here we used Mendelian randomization to assess whether the association might be causal. METHODS: Data on single-nucleotide polymorphisms that might be associated with various anthropometric indicators of obesity were extracted as instrumental variables from genome-wide association studies in the UK Biobank. Data on genetic variants in iron deficiency anemia were extracted from a genome-wide association study

An integrated genome and phenome-wide association study approach to understanding Alzheimer's disease predisposition.

Genome-wide association studies (GWAS) have identified common single nucleotide polymorphisms (SNPs) that increase late-onset Alzheimer's disease (LOAD) risk. To identify additional LOAD-associated variants and provide insight into underlying disease biology, we performed a phenome-wide association study on 23 known LOAD-associated SNPs and 4:1 matched control SNPs using UK Biobank data. LOAD-associated SNPs were significantly enriched for associations with 8/778 queried traits, including 3 platelet traits. The strongest enrichment was for platelet distribution width (PDW) (p = 1.2 × 10-5), bu
