Genomic Analysis of Trichotillomania.

Trichotillomania (TTM) is a psychiatric condition in which people feel an overwhelming urge to pull out their hair, resulting in noticeable hair loss and significant distress. Twin, family and candidate gene studies suggest that TTM is at least partly genetic, but no genome-wide analyses have been completed. To fill the gap in this field, we have conducted a case-control study of genotype array data from 101 European ancestry TTM cases and 488 ancestry-matched unaffected controls. TTM cases were ascertained in the USA through web-based recruitment, patient support groups, and conferences organ

Genome Editing of Rat.

Many genetically engineered rat strains have been produced by the development of genome editing technology, although it used to be technical difficulty and low production efficiency. Knockout and knock-in strains can be simple and quick produced using zinc finger nuclease (ZFN), transcription activator-like effector nuclease (TALEN), or clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9. Presently, genome edited strains have been produced by microinjection and a new electroporation method named technique for animal knockout system by electroporation (TAKE). This chapter pr

Plague in the genomic area.

With plague being not only a subject of interest for historians, but still a disease of public health concern in several countries, mainly in Africa, there were hopes that analyses of the Yersinia pestis genomes would put an end to this deadly epidemic pathogen. Genomics revealed that Y. pestis isolates evolved from Yersinia pseudotuberculosis in Central Asia some millennia ago, after the acquisition of two Y. pestis-specific plasmids balanced genomic reduction parallel with the expansion of insertion sequences, illustrating the modern concept that, except for the acquisition of plasmid-borne …

Vaccinology in the genome era.

Vaccination has played a significant role in controlling and eliminating life-threatening infectious diseases throughout the world, and yet currently licensed vaccines represent only the tip of the iceberg in terms of controlling human pathogens. However, as we discuss in this Review, the arrival of the genome era has revolutionized vaccine development and catalyzed a shift from conventional culture-based approaches to genome-based vaccinology. The availability of complete bacterial genomes has led to the development and application of high-throughput analyses that enable rapid targeted identi

Meningococcal genome dynamics.

Neisseria meningitidis (the meningococcus) is an important commensal, pathogen and model organism that faces up to the environment in its exclusive human host with a small but hyperdynamic genome. Compared with Escherichia coli, several DNA-repair genes are absent in N. meningitidis, whereas the gene products of others interact differently. Instead of responding to external stimuli, the meningococcus spontaneously produces a plethora of genetic variants. The frequent genomic alterations and polymorphisms have profound consequences for the interaction of this microorganism with its host, impact

Genome of bacteriophage P1.

P1 is a bacteriophage of Escherichia coli and other enteric bacteria. It lysogenizes its hosts as a circular, low-copy-number plasmid. We have determined the complete nucleotide sequences of two strains of a P1 thermoinducible mutant, P1 c1-100. The P1 genome (93,601 bp) contains at least 117 genes, of which almost two-thirds had not been sequenced previously and 49 have no homologs in other organisms. Protein-coding genes occupy 92% of the genome and are organized in 45 operons, of which four are decisive for the choice between lysis and lysogeny. Four others ensure plasmid maintenance. The m

Genome-based vaccines.

Vaccination is an effective possibility to prevent many bacterial or viral infections, but for several important pathogens still no vaccines are available. The sequences of complete genomes are now decoded for an increasing number of bacterial pathogens and offer the possibility for comprehensive screenings to identify targets for vaccine development. In this article current genomic approaches to identify antigenic proteins of Neisseria meningitidis, Streptococcus pneumoniae, Staphylococcus aureus, and Chlamydia pneumoniae are summarized.

The genome of Brucella melitensis.

The genome of Brucella melitensis strain 16M was sequenced and contained 3,294,931 bp distributed over two circular chromosomes. Chromosome I was composed of 2,117,144 bp and chromosome II has 1,177,787 bp. A total of 3,198 ORFs were predicted. The origins of replication of the chromosomes are similar to each other and to those of other alpha-proteobacteria. Housekeeping genes such as those that encode for DNA replication, protein synthesis, core metabolism, and cell-wall biosynthesis were found on both chromosomes. Genes encoding adhesins, invasins, and hemolysins were also identified.

The genome of echovirus 11.

Echoviruses are the largest enterovirus subgroup consisting of 32 serotypes. They are common human pathogens causing, for example, meningitis, encephalitis and exanthema, but in spite of their clinical importance, relatively little is known about their biology. To illuminate the molecular characteristics of echoviruses, we have completed the genomic sequence of serotype 11. The RNA genome is 7438 nucleotides in length and it codes for a 2195 amino acid long polyprotein. When compared to other sequenced enteroviruses, echovirus 11 (EV11) shows remarkable similarity with coxsackie B viruses (CBV

[The mitochondrial genome in medicine].

Analysis of the mitochondrial genome contributed towards the elucidation of a number of deviations-deletions or point mutations in mtDNA which are considered the cause of some pathological conditions, usually neuro- and myopathies. The distribution of defective mtDNA in the affected organism is uneven not only at the tissue or organ level but there are also marked differences between individual cells. Maternal heredity assumed for the mitochondrial genome is apparent only in some families, frequently also autosomal dominant heredity is found.

Comparative Genomics and Functional Characterization Reveal FaERF4 as a Key Regulator of Proanthocyanidin Biosynthesis in Strawberry.

Strawberry (Fragaria × ananassa) is an economically important berry rich in bioactive proanthocyanidins (PAs) with healthy benefits. The AP2/ERF gene family, one of the largest transcription factors (TFs) in plants, regulates diverse biological processes. However, its role in PA biosynthesis in strawberry remains unclear. In this study, comparative genomics of 39 species revealed lineage-specific expansion of AP2/ERF, primarily driven by whole-genome/segmental duplication. Integrated transcriptomic and metabolomic analyses of a high-PA natural mutant identified FaERF4 as a key positive regulat…

Longitudinal Clinical Foundation Models Augmented with Genomics for Early Detection and Risk Stratification of Inherited Cardiomyopathy.

Hypertrophic and dilated cardiomyopathy (HCM and DCM) carry substantial morbidity and mortality, yet diagnosis may be delayed, particularly when presentation is nonspecific. Existing machine-learning approaches to cardiomyopathy phenotyping, genotype prediction, and risk stratification commonly rely on disease-specific, hand-engineered features drawn from echocardiography, cardiac MRI, ECG, or curated clinical variables. We evaluated whether a general-purpose clinical foundation model, CLMBR-T-base, pre-trained via next-clinical-event prediction with no cardiomyopathy-specific supervision, cou

Dynamic Fusion of Genomics and Functional Network Connectivity in UK Biobank Reveals Schizophrenia-Related SNP Manifolds.

Many mental disorders show strong genetic influence. In parallel, dynamic functional network connectivity (dFNC) has shown high sensitivity to brain changes related to mental disorders. However, previous studies linking dFNC to genetics largely follow a paradigm to identify associations between one set of genetic factors and multiple sets of connectivity features from different dFNC states, ignoring the potential variability in genetic correlates across states. We propose a novel joint ICA (jICA)-based "dynamic fusion" framework to identify dynamically tuned genetic manifolds. A sliding window

Combining Genomics With Lipid and Inflammatory Biomarkers to Predict Coronary Artery Disease Risk: UK Biobank Study.

BACKGROUND: Coronary artery disease (CAD) polygenic risk score (PRS), low-density-lipoprotein cholesterol (LDL-C), lipoprotein(a) (Lp(a)), and high-sensitivity C-reactive protein (hsCRP) are biomarkers that predict CAD. It is unclear whether integrating genomics with lipid and inflammatory biomarkers could complement traditional risk scores in identifying people at risk of CAD. OBJECTIVES: This study assesses the predictive value of CAD PRS, LDL-C, Lp(a), and hsCRP for incident CAD across different age and sex groups. METHODS: Participants (n = 215,695) from the UK Biobank aged 40 to 69 year…

Toward precision hernia surgery: integrating biomarkers, genomics, quantitative imaging, and artificial intelligence in abdominal wall repair.

PURPOSE: Despite technical advances, the expansion of minimally invasive approaches and the development of novel biomaterials, recurrence and complications following abdominal wall hernia (AWH) repair remain frequent. This variability likely reflects anatomical and technical factors and host biological heterogeneity. Precision herniology is emerging as a translational paradigm integrating biomarkers, genetics, quantitative imaging, and artificial intelligence to refine risk stratification and therapeutic planning. METHODS: We conducted a critical narrative review with translational scope. Pub

Newborn Screening: Equity for Aboriginal and Torres Strait Islander Families in the Context of Emerging Genomics.

Australia's newborn bloodspot screening (NBS) program is offered to every newborn. It screens for 34 rare conditions with the potential for hundreds more to be added using genomics. Despite NBS being available in Australia since the 1960s, there is a lack of evidence regarding the participation and experiences of Aboriginal and Torres Strait Islander peoples in NBS. As Australia considers a future where genomics might be used in NBS, there is a critical window of opportunity to understand and prioritise the perspectives, hopes and fears of Aboriginal and Torres Strait Islander peoples regardin…
