Histone deacetylase 6 (HDAC6) is a pivotal epigenetic regulator and is involved in the tumorigenesis and progression. Our previous study revealed that HDAC6 is overactivated in hepatocellular carcinoma (HCC), and identified SelSA as a novel HDAC6 inhibitor (HDAC6i) for HCC therapy. In this work, we optimized this inhibitor and discovered 14a as a novel HDAC6i with better efficacy against HDAC6 and HepG2 cells than SelSA through structure-activity relationship (SAR) studies. The flow cytometry indicated that 14a markedly induced apoptosis and cell cycle arrest at the S phase in HepG2 cells. Mec

Glioblastoma (GBM) is a highly aggressive primary brain tumor characterized by profound molecular heterogeneity and poor clinical outcome. Although current prognostic stratification mainly relies on clinicopathological and molecular features, the contribution of histone modification-related programs to GBM progression and therapeutic vulnerability remains insufficiently defined. In this study, we systematically analyzed histone modification patterns in public GBM transcriptomic cohorts and constructed a histone modification-related prognostic signature (HMS). Based on curated histone modificat

Brain aging is a natural biological process characterized by gradual structural, molecular, and functional changes in the brain over time. Aging is the major risk factor for neurodegenerative diseases characterized by progressive neuronal loss, cognitive decline, and impaired motor function. In recent years, growing evidence has highlighted the importance of epigenetic regulation in the development and progression of these disorders. Among these epigenetic regulators, histone deacetylases (HDACs) have emerged as critical modulators of gene expression and neuronal function. Dysregulation of HDA

Terbuthylazine (TER), a triazine herbicide, can trigger excessive production of reactive oxygen species (ROS) in organisms, leading to liver damage in aquatic species. Under conditions of abnormally elevated ROS, increased glycolysis promotes the lactic acid accumulation, which in turn enhances histone lactylation and ultimately triggers pyroptosis. Meanwhile, N6-methyladenosine (m6A) methylation contributes to the pathogenesis of various diseases by regulating redox states. PURPOSE: H3 K27-altered diffuse midline glioma (DMG) is a molecularly defined, highly aggressive tumor entity since the 2016 revision of the World Health Organization (WHO) classification of tumors of the central nervous system (CNS). It has been most extensively characterized in children and adolescents and continues to pose significant challenges to treating physicians due to its growth along the midline structures of the brain and its ability to progress distantly leading to poor prognosis. Clinical and molecular determinants of distant progression and its impact on patient survival particu

BACKGROUND: Histone lactylation, a novel posttranslational modification driven by glycolytic lactate, has emerged as a key regulator of tumorigenesis. However, its role in osteosarcoma (OS) progression and the underlying metabolic‒epigenetic crosstalk remain poorly understood.
METHODS: Using immunohistochemistry, molecular biology, and functional assays in vitro and in vivo, we investigated lactylation levels, DHX9 regulation, Lactylation and glycolytic activity in osteosarcoma models. Techniques included CUT&Tag, LC-MS/MS, site-directed mutagenesis, and xenograft studies.
RESULTS: Here, we

Osteoporosis (OP) is a systemic metabolic bone disorder. The excessive activation of osteoclasts (OCs) leads to a decrease in bone mass and damage to the bone microstructure, which plays a crucial role in OP. β-Hydroxybutyrate (BHB), the main component of ketone bodies, not only serves as an ancillary fuel substituting for glucose but also induces anti-oxidative, anti-inflammatory, and cardioprotective features via binding to several target proteins, including histone β-hydroxybutyrylation (Kbhb). Recent research has found that BHB has a positive therapeutic effect on OP, but the underlying mo

The M1-to-M2 macrophage phenotypic switch is critical for resolving inflammation and restoring homeostasis in hypoxic lung injury, but the underlying molecular mechanisms are unclear. Here, we show that hypoxia-induced histone lactylation (Kla) upregulates FTO via HIF1a-P300 interaction; lactylation-driven FTO then promotes late-stage M2 polarization by stabilizing Arg1 mRNA through the m6A-YTHDF2 axis. Mechanistically, HIF1a/YY1 liquid-liquid phase separation (LLPS) regulates macrophage glycolysis and oxidative phosphorylation (OXPHOS) via the cGAS-STING pathway downstream of lactylation. Imp

UNLABELLED: Immune evasion driven by the tumor microenvironment is a major obstacle to effective immunotherapy in gastric cancer. To overcome this barrier, a detailed understanding of the mechanisms by which gastric cancer circumvents antitumor immunity is essential. In this study, we identified KDM5B as a pivotal epigenetic-metabolic orchestrator of immune evasion in gastric cancer. Clinically, KDM5B overexpression correlated with poor prognosis and diminished CD8+ T-cell infiltration. Mechanistically, KDM5B suppressed NLRP3-dependent pyroptosis via H3K4me3 demethylation and inhibited JAK1-ST

The efficacy of the host antiviral response against Influenza A virus (IAV), a leading cause of global pandemics, hinges upon the rapid recognition of the pathogen and the prompt activation of immune mechanisms. Nevertheless, the epigenetic landscape that orchestrates this antiviral response remains largely elusive. Here, we identify histone demethylase JMJD2D as a critical regulator in defense against IAV infection. A significant upregulation of JMJD2D expression was observed clinically in response to IAV infection, indicating that JMJD2D may play a role in regulating IAV infection. Indeed, J

Aspirin, as the most widely used drug in the world, has been reported to exert antiplatelet, anti-inflammatory, and anti-tumor effects. However, apart from the hydrolyzed acetyl group acetylating proteins, and salicylic acid functioning as a classic COX enzyme inhibitor, it is unknown whether the salicylic group of aspirin could participate in the salicylation of histone lysine. Herein, we discover and characterize lysine salicylation (Ksa) in histone and 39 Ksa sites of histone were mapped in 3 cell lines. Ksa could be stimulated by aspirin via direct action and conversion to salicyl-CoA, and

Microglia, the brain's resident immune cells, rely on histone modifications and chromatin remodeling to sculpt their identity across the trajectory from development through aging. Recent studies have identified enhancer rewiring and metabolic-epigenetic coupling-exemplified by histone lactylation-as central drivers of microglial plasticity. Disruption of this regulatory balance drives the transition from homeostatic microglia toward disease-associated microglia (DAM) in Alzheimer's and Parkinson's diseases. This Review examines emerging concepts such as trained innate immunity, state-specific

Malaria is a life-threatening infectious disease caused by Plasmodium parasites, and the emergence of widespread resistance to existing therapies underscores the urgent need for novel antimalarial agents. Quisinostat was identified as a Plasmodium falciparum histone deacetylase 1 (PfHDAC1) inhibitor against drug-resistant malaria parasites but suffered from intolerable toxicity. To achieve a potent and safe antimalarial agent, we hypothesized a scaffold hopping and linker optimization strategy, in which a total of 33 new structural γ-tetrahydrocarboline derivatives were synthesized and subject

Oncogenic viruses can induce epigenetic changes in host cells, which may contribute to cell transformation and tumorigenesis by altering gene expression patterns. Human adenoviruses, while primarily known for their lytic infections, have also been implicated in oncogenic transformation in experimental settings, but the mechanisms underlying adenovirus oncoprotein-induced epigenetic dysregulation remain poorly defined. Building on our recent findings that the adenoviral oncoprotein E1B-55K coordinates cellular transformation through interactions with DNA-bound host transcription factors, we nex

Naegleria fowleri is a free-living amoeba that causes a rare but almost always fatal brain infection known as primary amoebic meningoencephalitis (PAM). Although amphotericin B (AmB) is considered the primary treatment for PAM, it is often recommended in combination with several other drugs due to side effects associated with high concentrations. In this study, we investigated whether histone deacetylase inhibitors (HDACis) enhance the amoebicidal activity of AmB against N. fowleri. Anti-proliferative and cytotoxic activities were assessed using the cell counting kit and lactate dehydrogenase

The review covers research on dual-target antitumor agents over the past five years. One of the targets is histone deacetylases (HDACs), while the second potential target is a protein group located both on the membrane surface (phosphatidylinositol 3-kinase (PI3K), anaplastic lymphoma kinase (ALK), receptor tyrosine kinase (AXL), tyrosine protein kinase (HER2), FMS-like tyrosine kinase (FLT3), and vascular endothelial growth factor receptor (VEGFR2)) and in the nucleus (serine/threonine protein kinase Wee1, DNA methyltransferase (DNMT), dual-specificity phosphatase (CDC25A), an enzyme from the

Epigenetic dysregulation is a defining feature of solid tumors. Among epigenetic regulators, the nuclear receptor-binding SET domain (NSD) family of histone methyltransferases, comprising NSD1, NSD2, and NSD3, have emerged as critical mediators of oncogenic chromatin remodeling and transcriptional regulation. These enzymes primarily catalyze histone H3 lysine 36 (H3K36) methylation, thereby regulating chromatin accessibility, transcriptional programs, DNA damage repair, and genome stability. Aberrant expression, mutation, amplification, and chromosomal rearrangement of NSD family members have

Histone deacetylases (HDACs) are crucial enzymes involved in the regulation of gene expression through chromatin remodeling, impacting numerous cellular processes, including cell proliferation, differentiation, and survival. In recent years, HDACs have emerged as therapeutic targets for neurodegenerative diseases (NDDs), such as Alzheimer's disease, Parkinson's disease, and Huntington's disease, given their role in modulating neuronal plasticity, neuroinflammation, and neuronal survival. HDAC inhibitors (HDACi) are small molecules that prevent the deacetylation of histones, thereby promoting a…
