Any benefits of probiotics in allergic disorders?

Development of the child's immune system tends to be directed toward a T-helper 2 (Th2) phenotype in infants. To prevent development of childhood allergic/atopic diseases, immature Th2-dominant neonatal responses must undergo environment-driven maturation via microbial contact in the early postnatal period. Lactic acid bacteria and bifidobacteria are found more commonly in the composition of the intestinal flora of nonallergic children. Epidemiological data also showed that atopic children have a different intestinal flora from healthy children. Probiotics are ingested live health-promoting mi

Tick-borne encephalitis virus and the immune response of the mammalian host.

Tick-borne encephalitis (TBE) is caused by Tick-borne encephalitis virus (TBEV), one of the most prevalent arboviruses in Europe and in many parts of Asia. Transmission of TBEV to humans usually occurs by bite of an infected tick or rarely by ingestion of unpasteurized milk products of infected livestock. TBEV infection induces an innate and adaptive immune response, nevertheless it is able to replicate in several cell types of the immune system at the same time which probably contributes to the spread of the virus in the human host. Furthermore, TBEV can enter the central nervous system (CNS)

Cellular and molecular biology of Neisseria meningitidis colonization and invasive disease.

The human species is the only natural host of Neisseria meningitidis, an important cause of bacterial meningitis globally, and, despite its association with devastating diseases, N. meningitidis is a commensal organism found frequently in the respiratory tract of healthy individuals. To date, antibiotic resistance is relatively uncommon in N. meningitidis isolates but, due to the rapid onset of disease in susceptible hosts, the mortality rate remains approx. 10%. Additionally, patients who survive meningococcal disease often endure numerous debilitating sequelae. N. meningitidis strains are cl

A new paradigm of bacteria-gut interplay brought through the study of Shigella.

Bacteria-gut epithelial interplay and the mucosal immune response are the most critical issues in determining the fate of bacterial infection and the severity of diseases. Shigella species (abbreviated here as Shigella), the causative agent of bacillary dysentery (shigellosis), are highly adapted human pathogens that are capable of invading and colonizing the intestinal epithelium, which results in severe inflammatory colitis. Shigella secrete a large and diverse number (more then 50) of effectors via the type III secretion system (TTSS) during infection, some of which are delivered into the s

Differential immunoproteomics enables identification of Metarhizium anisopliae proteins related to Rhipicephalus microplus infection.

Differential immunoproteomics was applied to identify proteins secreted by Metarhizium anisopliae induced by the Rhipicephalus microplus cuticle. In addition, IgG anti-spore surface proteins were used for searching for proteins possibly involved in early stages of fungus versus tick infection. LC-MS/MS of differentially secreted proteins led to the identification of proteases (carboxypeptidase and Pr1A), chitinase, carboxylic acid transport and proline-rich protein. Differential immunoproteomics strategy facilitated the detection and the identification of new proteins related to M. anisopliae

Potential of Lecanicillium spp. for management of insects, nematodes and plant diseases.

Fungi in the genus Lecanicillium (formerly classified as the single species Verticillium lecanii) are important pathogens of insects and some have been developed as commercial biopesticides. Some isolates are also active against phytoparasitic nematodes or fungi. Lecanicillium spp. use both mechanical forces and hydrolytic enzymes to directly penetrate the insect integument and the cell wall of the fungal plant pathogen. In addition to mycoparasitism of the plant pathogen, the mode of action is linked to colonization of host plant tissues, triggering an induced systemic resistance. Recently it

Chlamydia trachomatis Inc Ct226 is vital for FLI1 and LRRF1 recruitment to the chlamydial inclusion.

The obligate intracellular pathogen, Chlamydia trachomatis, establishes an intracellular niche within a host membrane-derived vacuole called the chlamydial inclusion. From within this inclusion, C. trachomatis orchestrates numerous host-pathogen interactions, in part, by utilizing a family of type III secreted effectors, termed inclusion membrane proteins (Incs). Incs are embedded within the inclusion membrane, and some function to recruit host proteins to the inclusion. Two such recruited host proteins are leucine rich repeat Flightless-1 interacting protein 1 (LRRF1/LRRFIP1) and its binding …

Mapping the drivers of within-host pathogen evolution using massive data sets.

Differences among hosts, resulting from genetic variation in the immune system or heterogeneity in drug treatment, can impact within-host pathogen evolution. Genetic association studies can potentially identify such interactions. However, extensive and correlated genetic population structure in hosts and pathogens presents a substantial risk of confounding analyses. Moreover, the multiple testing burden of interaction scanning can potentially limit power. We present a Bayesian approach for detecting host influences on pathogen evolution that exploits vast existing data sets of pathogen diversi

Structure-Based Design and Synthesis of Covalent Inhibitors for Deubiquitinase and Acetyltransferase ChlaDUB1 of Chlamydia trachomatis.

Upon infection by an intracellular pathogen, host cells activate apoptotic pathways to limit pathogen replication. Consequently, efficient proliferation of the obligate intracellular pathogen Chlamydia trachomatis, a major cause of trachoma and sexually transmitted diseases, depends on the suppression of host cell apoptosis. C. trachomatis secretes deubiquitinase ChlaDUB1 into the host cell, leading among other interactions to the stabilization of antiapoptotic proteins and, thus, suppression of host cell apoptosis. Targeting the bacterial effector protein may, therefore, lead to new therapeut

Differential gene-expression and host-response profiles against avian influenza virus within the chicken lung due to anatomy and airflow.

Sampling the complete organ instead of defined parts might affect analysis at both the cellular and transcriptional levels. We defined host responses to H9N2 avian influenza virus (AIV) in trachea and different parts of the lung. Chickens were spray-inoculated with either saline or H9N2 AIV. Trachea and lung were sampled at 1 and 3 days post-inoculation (p.i.) for immunocytochemistry, real-time quantitative RT-PCR and gene-expression profiling. The trachea was divided into upper and lower parts and the lung into four segments, according to anatomy and airflow. Two segments contained the primar

Toxoplasma gondii secretory proteins and their role in invasion and pathogenesis.

T. gondii is a major opportunistic pathogen chronically infecting nearly one third of the world's population. Due to the high infection and mortality rates in immunocompromised patients and newborns, the extent or magnitude of T. gondii pathogenesis is determined mainly by host-pathogen interactions. T. gondii utilizes specialized secretory proteins to modify host cellular factors and facilitate invasion and replication. This review provides update on the recent progress in this field of research with particular emphasis on the T. gondii secretory proteins and their role in invasion and pathog…

Inside Enemy Lines: Adhesion, Invasion, and Intracellular Persistence of Acinetobacter baumannii in the Respiratory Epithelium.

Acinetobacter baumannii is a critical pathogen and a leading cause of hospital-acquired pneumonia, especially in immunocompromised patients. Although most research has focused on antimicrobial resistance, growing evidence shows that A. baumannii can efficiently adhere to, invade, and persist within human airway epithelial cells. Thus, the aim of this review is to summarize current knowledge on the mechanisms used by A. baumannii to establish infection, highlighting the bacterial traits responsible for attachment to airway epithelia, entry into host cells, manipulation of intracellular traffick

Host factors in parainfluenza virus replication: from entry to innate immunity evasion.

Human parainfluenza viruses (HPIVs)are significant pathogens responsible for acute respiratory infections in infants, the elderly, and immunocompromised individuals. Among children under five, HPIVs are the second most common viral cause of acute lower respiratory tract infections (ALRIs), surpassed only by respiratory syncytial virus (RSV). Despite their significant health impact, there are currently no licensed vaccines or specific antiviral treatments for HPIV infection. Recent advances in functional genomics, including CRISPR-Cas9 screening, cDNA library screening, and multi-omics approach

Mechanical forces in bacterial pathogenesis: Sensing, tropism, and intervention.

Bacterial pathogenesis unfolds within mechanically complex host niches, yet physical forces are frequently overlooked as active drivers of virulence. Here, we argue that mechanical cues-shear, confinement, and matrix rheology-serve as essential regulatory inputs that dynamically reprogram bacterial physiology. We propose a multi-scale framework connecting force-dependent receptor kinetics and envelope-stress signaling to community-level biofilm viscoelasticity. This synthesis elucidates how bacteria interpret the specific force landscapes of human tissues, providing a mechanistic rationale for

When the host is not healthy: Rethinking tuberculosis drug targets in comorbid conditions.

Tuberculosis (TB) frequently coexists with chronic conditions such as diabetes, HIV, malnutrition, sarcoidosis, and lung cancer. These comorbidities profoundly reshape the host immune, metabolic, and tissue environments, disrupting pathways that govern Mycobacterium tuberculosis (Mtb) containment and altering responses to both antimicrobial and host-directed therapies. Yet, most TB drug discovery pipelines continue to rely on "healthy-host" experimental models, overlooking the biological heterogeneity of the populations most affected by the disease. In this review, we argue that TB should be r

Autophagy in tuberculosis: advances, translational gaps, and the promise of host-directed therapies.

Tuberculosis (TB) is still a major global health threat, worsened by the advancement of multidrug-resistant and extensively drug-resistant strains of Mycobacterium tuberculosis (Mtb). Autophagy is a natural degradation and immune process that plays a critical role in the defence against intracellular pathogens, including Mtb. However, Mtb has developed complex mechanisms to evade autophagy by modulating phagosome-lysosome fusion, inhibiting xenophagy, and modifying host signaling pathways, including mTORC1 and AMPK, thereby suppressing epigenetically autophagy-related genes. These immune evasi

CsrA Enhances Cyclic-di-GMP Biosynthesis and Yersinia pestis Biofilm Blockage of the Flea Foregut by Alleviating Hfq-Dependent Repression of the hmsT mRNA.

Plague-causing Yersinia pestis is transmitted through regurgitation when it forms a biofilm-mediated blockage in the foregut of its flea vector. This biofilm is composed of an extracellular polysaccharide substance (EPS) produced when cyclic-di-GMP (c-di-GMP) levels are elevated. The Y. pestis diguanylate cyclase enzymes HmsD and HmsT synthesize c-di-GMP. HmsD is required for biofilm blockage formation but contributes minimally to in vitro biofilms. HmsT, however, is necessary for in vitro biofilms and contributes to intermediate rates of biofilm blockage. C-di-GMP synthesis is regulated at th
