Intranodal injection of PD1 antibody enhances memory T cell responses to improve the efficacy of neoantigen vaccines.

Cold tumors are resistant to neoantigen vaccines (NeoVac) combined with systemic anti-PD1 (aPD1) due to unalleviated T-cell exhaustion in tumor. Here, we found the exhaustion of T cells in tumor-draining lymph nodes (TdLNs) contributed greatly to the exhaustion of T cells in tumor, which could not be improved by systemic aPD1. Therefore, we developed a novel strategy for cold tumors: NeoVac combined with intranodal aPD1 injection (NeoVac + aPD1-i.n). This approach effectively impedes T cell exhaustion in TdLNs and promotes naive T-cell differentiation into central memory T cells (TCM), which f

T cell decision-making decodes the dynamic antigenic landscape.

The adaptive immune system continuously encounters antigens from a wide range of sources, including pathogens, self-tissues, tumors, and environmental agents. While extensive studies have characterized how lymphocytes respond to antigen binding, most experimental frameworks consider the antigenic environment as static. In reality, antigen levels can fluctuate dramatically across a wide range of temporal and spatial scales. In this review, we examine how the dynamics of antigen presentation, ranging from molecular binding events to organism-level exposure, affect T cell activation and fate. We

Mito-Specific "Trojan Horse" Nanotherapy: Synergistic Antitumor Immunotherapy via Dual Modulation Mitochondrial Metabolism and Glycolysis.

Metabolic reprogramming enables tumor cells to survive and proliferate in a nutrient-deficient environment. However, the immunosuppressive tumor microenvironment caused by metabolic reprogramming is often overlooked in current metabolism interventions. Herein, we developed a mito-specific "Trojan Horse" nanoplatform (2-pN@LNPs) coloaded with Niclosamide (Nic) and 2-deoxy-d-glucose (2-DG) to attack key metabolism pathways and synergistically ignite pyroptosis for restoring antitumor immunity. 2-pN@LNPs promoted proton influx across the inner mitochondrial membrane and caused oxidative phosphory

Immunophenotype associated with high sustained antibody titers against enzyme replacement therapy in infantile-onset Pompe disease.

INTRODUCTION: The efficacy of enzyme replacement therapy (ERT) with alglucosidase alfa for infantile-onset Pompe disease (IOPD) is limited in some patients due to the development of high and sustained antibody titers (HSAT; ≥12,800). METHODS: We carried out detailed immunophenotyping of IOPD patients (n=40), including analysis of circulating cell populations by flow cytometry and plasma cytokines by multiplex array, to determine whether patients with HSAT have unique immunological characteristics compared to those with low titers (LT; <12,800). RESULTS: Compared to patients with LT, patients

An electron microscopical and histochemical study on ferric nitrilotriacetate-induced "F1 amyloidosis".

Of a total of 80 offspring obtained by reciprocal crossing of ICR/JCL strain of mice of both sexes receiving 84-140 injections of Fe-NTA over 98-163 days before crossing, 52 (65%) showed severe generalized amyloidosis after 93-502 injections of Fe-NTA over 115-522 days, but not "hemochromatosis", which was a striking contrast to the findings of "hemochromatosis" without amyloidosis observed in the parent mice. Electron microscopic examinations revealed numerous bundles of non-branching, well-oriented amyloid fibrils radiated outward from the surface of cytoplasmic invaginations of the Kupffer

Atomistic TCR-pMHC interactions bias CD8 memory fate within a single antigen-specific repertoire.

Memory CD8 T cells provide durable protection against recurrent infection and cancer, but how individual TCR clonotypes contribute to distinct memory fates remains enigmatic. Here, we analyzed 242 murine CD8 TCRαβ clonotypes specific for an influenza NP366-374/H-2Db ligand (pMHC) by integrating single-cell transcriptomics and paired TCR sequencing with force-dependent biophysics, structural analysis, and in vivo retrogenic validation. Within this shared antigenic and inflammatory setting, clonotype identity was associated with central memory, effector memory, or bipolar transcriptional outcome

CD8+ T-cell differentiation and dysfunction inform treatment response in acute myeloid leukemia.

The interplay between T-cell states of differentiation, dysfunction, and treatment response in acute myeloid leukemia (AML) remains unclear. Here, we leveraged a multimodal approach encompassing high-dimensional flow cytometry and single-cell transcriptomics and found that early memory CD8+ T cells are associated with therapy response and exhibit a bifurcation into 2 distinct terminal end states. One state is enriched for markers of activation, whereas the other expresses natural killer (NK)-like and senescence markers. The skewed clonal differentiation trajectory toward CD8+ senescence was al

Boosting long-term immunity with bioengineered nanovaccines.

The development of vaccine platforms capable of inducing durable immunity remains a crucial frontier. While conventional vaccines offer proven efficacy, their often limited durability poses challenges for rapidly evolving pathogens and cancer immunotherapy. Recent advances in biomaterials and nanotechnology have enabled the rational design of bioengineered nanovaccines. These systems leverage engineered polymeric, lipid-based, and hybrid materials to achieve controlled antigen release, targeted lymph node delivery, and enhanced cross-presentation. Their "bioengineered" functionality allows the

Differential Immune Cell Distribution Between Arterial and Venous Blood in Healthy Rats.

We previously identified differentially expressed miRNAs between arterial and venous blood in rats; however, whether immune cell composition and transcriptional states differ along the arterial-venous axis remains unclear. Here, single-cell RNA sequencing of 119 481 PBMCs, complemented by flow cytometry and protein-level validation, was performed to characterize immune heterogeneity between arterial and venous blood. Cell composition analysis showed enrichment of T cells in arterial blood, whereas B cells, NK cells, and monocytes were more abundant in venous blood. At the subset level, naïve T

Induction of therapeutic immunity and cancer eradication through biofunctionalized liposome-like nanovesicles derived from irradiated-cancer cells.

Immunotherapy has revolutionized the treatment of cancer. However, its efficacy remains to be optimized. There are at least two major challenges in effectively eradicating cancer cells by immunotherapy. Firstly, cancer cells evade immune cell killing by down-regulating cell surface immune sensors. Secondly, immune cell dysfunction impairs their ability to execute anti-cancer functions. Radiotherapy, one of the cornerstones of cancer treatment, has the potential to enhance the immunogenicity of cancer cells and trigger an anti-tumor immune response. Inspired by this, we fabricate biofunctionali

Novel pathogenic variants in CTLA4 and LRBA immune dysregulation: Reduced CTLA-4 expression with normal expression of co-stimulatory surface molecules.

BACKGROUND: Genetic variants in Cytotoxic T-Lymphocyte-associated protein 4 (CTLA4) and LPS responsive beige-like anchor protein (LRBA), involved in the same biological pathways, are implicated as monogenic causes of Common Variable Immunodeficiency Disorder (CVID). The pitfall in the recognition of CVID possibly related to CTLA-4 haploinsufficiency or LRBA deficiency is a clinical picture that is very heterogeneous. In the present study, we illustrate this challenge by means of clinical and immunological analysis of five patients with novel genetic variants in CTLA4 and LRBA. METHODS: Whole-

Immune senescence: relative contributions of age and cytomegalovirus infection.

Immune senescence, defined as the age-associated dysregulation and dysfunction of the immune system, is characterised by impaired protective immunity and decreased efficacy of vaccines. Recent clinical, epidemiological and immunological studies suggest that Cytomegalovirus (CMV) infection may be associated with accelerated immune senescence, possibly by restricting the naïve T cell repertoire. However, direct evidence whether and how CMV-infection is implicated in immune senescence is still lacking. In this study, we have investigated whether latent mouse CMV (MCMV) infection with or without t

Dual TCR-expressing T lymphocytes in health and disease.

The authors briefly review recent experimental advances in elucidating the role of dual T cell receptor (TCR)-expressing lymphocytes in the development of diseases with special emphasis on autoimmunity. Moreover, they summarize present knowledge about these cells concerning their proportion among peripheral blood mononuclear cells, their functionality, and their impact on allorecognition and memory both in humans and in mice. Finally, they describe disease-associated clonal expansions of dual TCR-expressing cells in humans, most of which have been observed in peripheral T cell malignancies. Ot

Dendritic cell redundancy enables priming of anti-tumor CD4+ T cells in pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) is resistant to current immunotherapies and lacks effective anti-tumor CD8+ T cells, which is potentially due to insufficient cross-presentation by cDC1s. Here, we combine a STING agonist with anti-CTLA-4 and anti-PD-1 to achieve durable remissions and immunologic memory in multiple mouse models of poorly immunogenic PDAC. We find that tumor control does not depend on CD8+ T cells or tumor cell MHC expression but instead requires IFNγ-producing CD4+ T cells (Th1s) that are primed by dendritic cells in lymph nodes. The triple combination immunotherapy ind
