Influence of adenosine on cerebral blood flow during hypoxic hypoxia in the newborn piglet.

This study investigated the role of adenosine in the regulation of neonatal cerebral blood flow (CBF) during moderate (arterial PO2 = 47 +/- 9 Torr) and severe (arterial PO2 = 25 +/- 4 Torr) hypoxia. Twenty-eight anesthetized and ventilated newborn piglets were assigned to four groups: 8 were injected intravenously with the vehicle (controls, group 1); 13 received an intravenous injection of 8-phenyltheophylline (8-PT), a potent adenosine receptor blocker, either 4 mg/kg (group 2, n = 6, mean cerebrospinal fluid (CSF) levels less than 1 mg/l) or 8 mg/kg (group 3, n = 7, mean CSF levels less th

Postprandial neurohormonal control of gastric emptying.

The delay in gastric emptying after a meal is thought to result from nutrient distention-induced release of peptides and neurotransmitters in the gut. The potential contribution of the brain and central neuropeptides to the postprandial regulation of gastric emptying has not been studied adequately. The purpose of this study was twofold: to examine the relative importance and pure central stimulation (sham feeding) or pure peripheral stimulation (duodenal feeding) on gastric emptying, and to determine whether relevant neuropeptides act in the brain to slow gastric emptying of a meal. Six mongr

Effects of central administration of morphine on renal function in conscious rats.

Systemic administration of morphine in rats produces an anti-natriuretic effect that is at least partially dependent on renal nerves. The present studies were carried out in order to assess the renal response to central administration of morphine. Male Sprague-Dawley rats were surgically prepared with arterial, venous and bladder cannulas. In addition, a guide cannula was placed into the lateral ventrical and secured to the surface of the skull. Experiments were carried out at least 3 days after surgery. Renal clearance measurements were 30 min each. After a basal period, morphine sulfate (4 m

Site-dependent central effects of aldosterone in rats.

A relationship between the subcommissural organ (SCO) and the adrenal glands has long been suspected. This report provides further information about the effects of a continuous D-aldosterone infusion into the SCO area of conscious, adult male Sprague-Dawley rats. A 6-day aldosterone infusion (5 ng/h) increased urinary sodium excretion, decreased adrenal medullary cross-sectional area, elevated adrenal corticosterone content and terminal plasma epinephrine concentration. Mineralocorticoid infusions directly into a lateral cerebral ventricle did not affect these parameters but, unlike SCO area i

Effect of central naloxone on hormone and blood pressure responses to hemorrhage in conscious sheep.

The role of the brain opioid system in the control of hypothalamic-pituitary-adrenal activity was studied in 10 conscious sheep with an indwelling cannula in a cerebral lateral ventricle. On separate days, sheep received infusions of artificial CSF (control) and the opiate antagonist, naloxone (100 micrograms/hr) before and during acute moderate hemorrhage (15 ml/kg over 10 min). Infusion of naloxone before hemorrhage raised plasma ACTH and resulted in a significant increase in cortisol compared to the control infusion. In contrast, ACTH and cortisol responses to hemorrhage tended to be blunte

Effects of H2-receptor antagonists on prolactin secretion: specificity and mediation of the response.

The effects on prolactin secretion of histamine H2-receptor antagonists infused intracerebroventricularly were studied in urethane anaesthetized male rats. A dose of 1.6 mumol cimetidine stimulated basal prolactin secretion and did not affect the histamine-induced release, whereas 0.4 mumol cimetidine inhibited basal and histamine-stimulated prolactin secretion. 0.1 mumol cimetidine had no effect. The more potent H2-receptor antagonist ranitidine at doses of 0.1, 0.4, 1.6 mumol had no effect on basal prolactin secretion, whereas 0.4 and 1.6 mumol inhibited the histamine-stimulated secretion co

Mechanism of central hyperglycemic effect of cholinergic agonists in fasted rats.

The influence of cholinergic agonists on central nervous system (CNS) regulation of blood sugar homeostasis was studied in fasted rats. When carbachol, muscarine, bethanechol, methacholine, or neostigmine was injected into the third cerebral ventricle, it caused a dose-dependent increase in the hepatic venous plasma glucose concentration. However, in the case of 1,1-dimethylphenyl-4-piperazinium iodide (DMPP) or nicotine, the level of hepatic venous glucose did not differ from that of the saline-treated control rats. The increase in glucose level caused by neostigmine was dose-dependently supp

The effects of central injections of adenosine analogs on blood pressure and heart rate in the rat.

Rats were implanted with chronic indwelling cannulae into the lateral cerebral ventricle. After recovery from surgery, acute experiments on blood pressure were conducted under methoxyflurane/nitrous oxide anesthesia. Rats were injected intracerebroventricularly with two adenosine analogs, 5'-N-ethylcarboxaminidoadenosine (NECA) and (-)-N-(1-methyl-2-phenylethyl)adenosine(L-phenylisopropyladenosine) (L-PIA), and the effects on blood pressure and heart rate recorded. Both analogs produced dose-related reductions in blood pressure and heart rate with L-PIA producing a more potent depression of he

Alpha- and beta-adrenergic receptor blockers and the effects of noradrenaline on heat production and body temperature.

Noradrenaline (NA) administered systemically or into the lateral cerebral ventricle (ICV) in appropriate doses increased heat production and colonic temperature in 1-12 day-old guinea pigs. The effect of systemically applied NA could be blocked by systemically applied beta-adrenergic receptor blockers, while beta-blockers administered centrally or alpha-adrenergic blockers injected systemically or centrally had no effect on the action of systemically applied NA. Accordingly, the effect of systemically applied NA was mediated by peripheral beta-adrenergic receptors. The effect of centrally appl

The effect of 6-hydroxydopamine infused into the third cerebral ventricle on the plasma cortisol concentration in sheep subjected to repeated and prolonged stress stimuli.

It has previously been observed that sheep subjected to repeated and prolonged stress stimuli showed biphasic cortisol responses. On the first and second day of stimulation an elevation was observed, while on the subsequent days and on the day after the stimulation a marked suppression of plasma cortisol level and a disappearance of its circadian rhythm was noted. It was hypothesized that these changes in the secretion of the hormone were caused by the alteration of catecholaminergic systems in the CNS. To verify this suggestion chemical lesions of the catecholergic systems of the diencephalon

Motor hypoactivity induced by neurotensin and related peptides in mice.

The tridecapeptide neurotensin (NT) induces a variety of behavioral changes in animals. The present study characterizes the behavioral hypoactivity observed after intracerebroventricular (ICV) injection in mice. At doses higher than 25 ng, NT induced a reduction of general motor activity and increases in immobility which lasted for about one hour. The NT-related amphibian skin peptide xenopsin was about 70-fold more potent than NT itself. After repeated NT-injections, tolerance developed within 2-4 days and disappeared within 2-4 days after cessation of the treatment. The motor hypoactivity in

Central gastric antisecretory action of prostaglandin E2: the involvement of the pituitary gland.

The mechanism of the gastric antisecretory action of intracerebroventricularly administered prostaglandin (PG)E2 was investigated in conscious, pylorus-ligated rats. It was found that intracerebroventricular administration of 3 micrograms of PGE2 inhibited carbachol-stimulated gastric secretion (acid, pepsin and volume) in vagotomized rats and spontaneous secretion in rats with intact vagi to the same degree. Hypophysectomy prevented the antisecretory effect of PGE2, whereas neither adrenalectomy nor gastric sympathectomy (extirpation of the ganglion coeliacum and mesentericus superius) did mo

Calcitonin gene-related peptide acts within the central nervous system to inhibit gastric acid secretion.

The central nervous system effect of calcitonin gene-related peptide (CGRP) on gastric acid secretion was studied in conscious freely moving rats. CGRP (220 fmol to 2.2 nmol) injected into the lateral cerebral ventricle or intravenously inhibited gastric acid secretion. Intravenous passive immunization with CGRP antiserum prevented the inhibitory effect of CGRP following intravenous but not intracerebroventricular administration. Adrenalectomy and noradrenergic blockade with bretylium tosylate did not significantly alter the inhibitory action of CGRP given intracerebroventricularly on gastric

[The effects of the intracerebroventricular administration of 5-hydroxytryptamine on hemodynamics in conscious rats. Participation of the serotonin receptor and the sympathetic nervous system].

We have previously reported that the intracerebroventricular (i.c.v.) administration of 5-hydroxytryptamine (5-HT) in conscious rats elicited significant pressor response, which was accompanied with an increase in plasma norepinephrine and the pressor response, was abolished by peripheral phenoxybenzamine pretreatment. For further investigation of the relationship between the serotonergic nervous system and the sympathetic nervous system, the effect of either i.c.v. or intravenous (i.v.) serotonin receptor antagonist, methysergide pretreatment on hemodynamic response to centrally administrated

An inhibitory role for morphine on the release of dopamine into hypophysial portal blood and on the synthesis of dopamine in tuberoinfundibular neurons.

The intracerebroventricular administration of morphine to ovariectomized rats resulted in a marked decrease in the concentration of dopamine in plasma of hypophysial portal blood. A 90% reduction in the rate of release of hypothalamic dopamine into hypophysial portal blood occurred during the 60 min following the intraventricular administration of 60 ng of morphine sulfate. A dose-related decrease in the rate of release of dopamine into the portal vasculature was observed between 7.5 ng and 60 ng of morphine sulfate. Regardless of the quantity of morphine sulfate (1-500 ng) given to the animal

Behavioral and electrocortical effects of beta-endorphin after intracerebral infusion in rats.

In rats with cannulae chronically implanted into specific areas of the brain, the effects of beta-endorphin were studied on behavior and electrocortical activity. It has been shown that analgesia and catatonia were evoked after infusing beta-endorphin into the III cerebral ventricle or into the hypothalamus. On the contrary, beta-endorphin microinjected into the caudate nucleus or into the substantia nigra produced an intense pattern of stereotyped movements, occasional contralateral circling, contralateral myoclonic jerks and asymmetric posture. Stereotyped gnawing, grooming and "wet-dog" syn

[Infusion test in the normal pressure hydrocephalus (author's transl)].

Continuous monitoring of the intracranial pressure and ventricular infusion test were carried out in 13 cases in which normal pressure hydrocephalus (NPH) was suspected. The infusion test was performed by intraventricular bolus injection of 8 ml of saline. The result was collated with the effect of shunting operation, which was judged by means of the improvement in the clinical symptoms and the amplitude of contingent negative variation. The excellent effect of shunting operation was obtained in the patients who showed the T of more than 200 sec, the compliance of less than 1.0 ml/mmHg, the co
