UNLABELLED: In a world with an increase in human population, food consumption, and the generation of organic waste, insects are emerging as a promising tool to convert organic waste material into human food or animal feed. The insect microbiome is known to play a key role in the degradation of organic substrates, but little is known about the metabolic potential of the microbiome of industrially reared fly larvae. We investigated the microbial composition and metabolic potential of the house fly (Musca domestica) larva gut microbiome from larvae grown on three different waste and by-product-ba

UNLABELLED: The causative agent of dengue fever, dengue virus (DENV), is transmitted by mosquitoes, and as distribution of these insects has expanded, so has dengue-related disease. DENV is a member of the Flaviviridae family and has 4 distinct serotypes (DENV-1, -2, -3, and -4). No lasting cross protection is afforded to heterologous serotypes following infection by any one of the individual serotypes. The presence of nonneutralizing antibodies to one serotype can facilitate the occurrence of more-severe dengue hemorrhagic fever through immune enhancement upon infection with a second serotype

Anti-insect depressant toxins represent a subfamily of scorpion venom-derived β-toxins that are polypeptides composed of 61-65 amino acid residues stabilized by four disulfide bridges. These toxins affect the activation of voltage-sensitive sodium channels (NaScTx) and exhibit the preferential ability to induce flaccid paralysis in insect larvae. Here we demonstrate the recombinant expression of the novel cardiac inotropic peptide (Bj-IP) that was classified as an anti-insect depressant βNaScTx isolated from the venom of Hottentotta judaicus. By using "splicing by overlap extension" (SOE)-PCR,

BACKGROUND: Filarial nematodes, including Brugia malayi, the causative agent of lymphatic filariasis, undergo molting in both arthropod and mammalian hosts to complete their life cycles. An understanding of how these parasites cross developmental checkpoints may reveal potential targets for intervention. Pharmacological evidence suggests that ecdysteroids play a role in parasitic nematode molting and fertility although their specific function remains unknown. In insects, ecdysone triggers molting through the activation of the ecdysone receptor: a heterodimer of EcR (ecdysone receptor) and USP

Optimizing black soldier fly larvae (BSFL) production requires a better understanding of how diet composition shapes growth, nutrient allocation, and metabolic efficiency. This study investigated whether respiratory measurements could serve as predictive, non-invasive tools for BSFL performance on diets differing in protein-to-carbohydrate (P:C) ratio. Larvae were reared for seven days on four experimental diets, with growth, survival, gas exchange (O2 consumption, CO2 production, and respiratory exchange ratio (RER)), and body composition (crude protein and lipid contents) measured across sev

Vespa affinis (Asian wasp, Thai banded tiger wasp, or local name: Tor Hua Seua) causes the most frequent incidence of medically important Hymenoptera sting in South and Southeast Asia. However, data on the venom components attributable to the sting derived-clinical manifestations (local reactions, IgE mediated-anaphylaxis, or systemic envenomation) are lacking. This study provides the first set information on V. affinis venom proteome, allergenome, and IgE reactivity of individual venom components. From 2DE-gel based-proteomics, the venom revealed 93 protein spots, of which proteins in 51 spot

We report the characterization of a novel Drosophila circadian clock-regulated output gene, takeout (to). The to amino acid sequence shows similarity to two ligand binding proteins, including juvenile hormone binding protein. to mRNA is expressed in the head and the cardia, crop, and antennae-structures related to feeding. to expression is induced by starvation, which is blocked in all arrhythmic central clock mutants, suggesting a direct molecular link between the circadian clock and the feeding/starvation response. A to mutant has aberrant locomotor activity and dies rapidly in response to s

In vertebrates (deuterostomes), brain patterning depends on signals from adjacent tissues. For example, holoprosencephaly, the most common brain anomaly in humans, results from defects in signaling between the embryonic prechordal plate (consisting of the dorsal foregut endoderm and mesoderm) and the brain. I have examined whether a similar mechanism of brain development occurs in the protostome Drosophila, and find that the foregut and mesoderm act to pattern the fly embryonic brain. When the foregut and mesoderm of Drosophila are ablated, brain patterning is disrupted. The loss of Hedgehog e

Numerous plants, including cereals, contain seed proteins able to inhibit amylolytic enzymes. Some of these inhibitors, the CM-proteins (soluble in chloroform:methanol mixtures)-also referred to as cereal-type inhibitors (CTIs)-are the topic of this review. CM-proteins were first reported 75 years ago. They are small sulfur-rich proteins of the prolamine superfamily embracing bifunctional α-amylase/trypsin inhibitors (ATIs), α-amylase inhibitors (AIs), limit dextrinase inhibitors (LDIs), and serine protease inhibitors. Phylogenetically CM-proteins are predicted across poaceae genomes and many

Gametocyte-specific factor 1 (Gtsf1) plays a pivotal role in the gonadal development by transposon silencing via the piRNA pathway in Drosophila melanogaster and Bombyx mori, and is implicated in the female sex determination in B. mori. However, its function remains elusive in other insect species, including Plutella xylostella, a highly destructive pest in cruciferous crops. In this study, we characterized PxGtsf1, encoding a 27.84 kDa protein containing two conserved U11-48K-like CHHC zinc-finger domains. PxGtsf1 was predominantly expressed in male pupae, males and testes. Using CRISPR/Cas9

In Southeast Asia, Conopomorpha cramerella (Snellen) which is commonly known as the cocoa pod borer (CPB) moth has been identified as the most detrimental pest of Theobroma cacao L. Apart from the various side effects on human health and non-target organisms, heavily relying on synthetic pyrethroid insecticides to control CPB infestations also increases the environmental contamination risks. Thus, developing biorational insecticides that minimally affect the non-target organism and environment by targeting the insect growth regulation process is needed to manage the pest population. In insects

BACKGROUND: Edible insects are a novel source of animal protein. Moreover, edible insects contain iron concentrations similar to meat, potentially making them a valuable iron source for human consumers. Yet, it is unknown to what extent iron from insects is absorbed in humans.
OBJECTIVES: In this exploratory study, we assessed fractional iron absorption from house crickets (Acheta domesticus) consumed with refined (low-phytate, noninhibiting) or nonrefined (high-phytate, inhibiting) meals.
METHODS: Intrinsically [57Fe]-labeled and control crickets were reared. Six iron-balanced experimental

Reptalus artemisiae (Becker, 1865) is a vector of economically important plant pathogens. To date, no complete mitochondrial genome has been available for Pentastirini. We present the first complete mitogenome of R. artemisiae, sequenced using ONT MinION long reads. The circular genome is 15,476 bp long, comprises 13 protein-coding, two rRNA, 22 tRNA genes, and an AT-rich putative noncoding control region. Gene content and organization are consistent with the ancestral insect mitochondrial genome. This dataset provides a crucial reference for phylogenetic studies for both Reptalus and Pentasti

The growing demand for sustainable food systems has accelerated research into alternative proteins derived from plant sources, microorganisms, insects, and cultured animal cells. These protein systems offer promising solutions to environmental, ethical, and nutritional challenges associated with conventional livestock production. However, designing and optimizing alternative proteins requires deep understanding of molecular structures, biochemical properties, and production processes. Computational biology has emerged as a powerful approach to address these challenges by enabling predictive mo

Polycomb group (PcG) proteins repress homeotic genes in cells where these genes must remain inactive during development. This repression requires cis-acting silencers, also called PcG response elements. Currently, these silencers are ill-defined sequences and it is not known how PcG proteins associate with DNA. Here, we show that the Drosophila PcG protein Pleiohomeotic binds to specific sites in a silencer of the homeotic gene Ultrabithorax. In an Ultrabithorax reporter gene, point mutations in these Pleiohomeotic binding sites abolish PcG repression in vivo. Hence, DNA-bound Pleiohomeotic pr

Global temperature increases require the reevaluation of pesticide efficacy under thermal stress conditions; however, most studies have focused on acute high-temperature exposure, neglecting the effects of natural diurnal temperature fluctuations and cumulative acclimation. This study investigated the effect of thermal acclimation during developmental on the sensitivity of Mythimna separata (Walker) (Lepidoptera: Noctuidae) to emamectin benzoate (EB). Larvae were reared under cyclic thermal regimes (25→34/38→25 °C), experiencing daily high-temperature exposures of 2, 4, or 6 hours from the egg

Nitric oxide (NO), a small yet highly reactive gaseous molecule, has emerged as a crucial signaling component in plant biology, mainly in mediating its defense responses against biotic stress. Apart from mammalian role of NO, it also actively involved in the broad physiological, biochemical, and immunological processes in plants. NO is synthesized through a set of multiple enzymatic and non-enzymatic pathways, whereupon it rapidly accumulates upon pathogen recognition. NO is implicated in plant immunity by working in synergy with reactive oxygen species (ROS) in triggering the hypersensitive r

Cetacean morbillivirus (CeMV) is an enveloped, non-segmented, negative-stranded RNA virus that infects marine mammals, spreading across species and causing lethal disease outbreaks worldwide. Among the eight proteins encoded by the CeMV genome, the haemagglutinin (H) glycoprotein is responsible for the virus attachment to host cell receptors. CeMV H represents an attractive target for antiviral and diagnostic research, yet the elucidation of the molecular mechanisms underlying its role in infection and inter-species transmission was hampered thus far due to the unavailability of recombinant ve

Plasmodium ookinetes use an invasive apparatus to invade mosquito midguts, and tubulins are the major structural proteins of this apical complex. We examined the role of tubulins in malaria transmission to mosquitoes. Our results demonstrate that the rabbit polyclonal antibodies (pAb) against human α-tubulin significantly reduced the number of P. falciparum oocysts in Anopheles gambiae midguts, while rabbit pAb against human β-tubulin did not. Further studies showed that pAb, specifically against P. falciparum α-tubulin-1, also significantly limited P. falciparum transmission to mosquitoes. We
