Association between ambient temperature and thrombosis-related signaling lipids: Implications for early cardiovascular risk.

BACKGROUND: Nonoptimal temperatures significantly contribute to cardiovascular mortality, a burden expected to increase with climate change. Sensitive biomarkers for early cardiovascular risks remain scarce. OBJECTIVE: We examined how thrombosis-related signaling lipids respond to ambient temperature, analyzing dose-response and lag patterns over a 30-day period, and assessed whether lipid changes mediate the association between temperature and thrombosis biomarkers using mediation analyses. METHODS: We conducted a prospective panel study of 112 participants (aged 42-80) in Beijing (2016-201…

Adaptive Plasma-Membrane Nanodomains in Plants: Integrating Lipids, Receptors, Mechanics and Trafficking During Stress.

The plant plasma membrane is a chemically asymmetric, mechanically coupled and actively renewed interface in which lipids, receptors, ion channels, transporters and scaffolds form transient nanoscale assemblies. Direct evidence from a limited number of plant systems shows that membrane organisation can alter protein mobility, trafficking and physiological output, including remorin-dependent PIN2 hyperclustering, phosphatidylserine-dependent ROP6 nano-organisation and stimulus-dependent receptor or channel redistribution. These findings do not establish a universal molecular network or a direct…

An experimental approach to investigate extracellular vesicle-mediated transfer of lipids between fungal cells.

Extracellular vesicles (EVs) are key mediators of communication in fungal populations, but the mechanisms underlying lipid exchange through … ical consequences of this process remain underexplored. Here, we provide experimental evidence that lipids can be exchanged between fungal cells via EVs, visualized using the lipophilic dye FM 1-43. This approach allows tracking of lipid trafficking between cells of the same or different species, even in the absence of direct contact. Understanding EV-me …

ABO Blood Groups, Lipids, and Coronary CT Imaging in A Japanese Single-Center Cohort.

Background and Objectives: Non-O ABO blood groups have been linked to higher coronary risk, plausibly via hemostatic and lipid pathways. However, evidence in Japanese populations and imaging-defined disease is limited. We examined whether ABO status relates to serum lipids and coronary CT imaging findings in Japanese adults. Materials and Methods: We reviewed adults who underwent coronary CT angiography (CCTA) at our institution. After prespecified exclusions, 865 patients comprised the imaging cohort. For lipid analyses, we excluded patients receiving lipid-lowering therapy at the time of blo…

Retinoic-Acid-Derived Ionizable Lipids Enable Spleen-Selective Antigen Expression for mRNA Cancer Vaccination.

Effective mRNA cancer vaccination requires productive antigen expression in lymphoid antigen-presenting cells rather than high total antigen output. Here, we report a retinoic-acid-inspired ionizable lipid platform for spleen-selective functional mRNA expression. Systematic variation of the ionizable scaffold, linker architecture, and retinoid-derived hydrophobic domain generated a modular 40-member library of retinoic-acid-derived ionizable lipids (RAILs). Paired in vitro and in vivo screening identified AT4-13CRA LNPs as the lead formulation, with the highest spleen-to-liver reporter-express…

Sex-specific APOE4-dependent innate immunity regulates meningeal lymphatics, brain lipids, neuroinflammation, and cognition.

Sex and apolipoprotein E ε4 (APOE4) interact to alter the risk for Alzheimer's disease and other neurodegenerative disorders. Herein, we show sex-specific differences in immune activation and lymphatic function in the meningeal dura of humanized female and male mice expressing two alleles of APOE4 (E4/E4), when compared with their respective sex-matched E3/E3 controls. We also describe distinct effects of APOE4 on brain lipid composition and inflammation in females and males that were partially reverted upon colony-stimulating factor 1 receptor (CSF1R) inhibition. Suppressing innate immunity r

An adenylyl cyclase switch reroutes carbon from growth to virulence lipids in Mycobacterium tuberculosis.

Anti-TB drugs act non-uniformly on Mycobacterium tuberculosis (Mtb) because of its distinct physiological states across diverse infection niches shaped by host-derived nutrients and stresses. Agonists of the adenylyl cyclase Rv1625c are a novel drug class that selectively inhibits Mtb growth in macrophages and cholesterol-rich conditions by an unknown mechanism. Combining condition-resolved transcriptomics, genome-scale metabolic modeling, and genetic perturbation, we show these agonists cause a blockade in the electron transport chain, which likely activates Rv1625c. The elevated cAMP in turn

Circulating lipids uncover early membrane disruption as a primary event preceding Alzheimer's disease onset.

Alzheimer's disease (AD) is not a single entity but a biologically heterogeneous disorder, with substantial inter-individual variability in clinical presentation, molecular drivers, and disease trajectories. This heterogeneity limits current diagnostic and therapeutic strategies, which largely rely on late-stage protein biomarkers that, while sensitive to prodromal impairment, remain insufficient for primary prevention and patient stratification. Lipids are fundamental to brain structure and function, yet their role in the earliest stages of AD remains poorly defined. Here, we identify a robus…

Spatial Tail Design in Ionizable Lipids Enhances the Safety and Efficacy of mRNA Delivery.

Messenger RNA (mRNA) delivery critically depends on the molecular architecture of ionizable lipids within lipid nanoparticles (LNPs). We report a modular platform of sterol-containing ionizable lipids designed to probe how sterol motifs and branched dialkyl tails govern in vivo performance. Each lipid incorporates a sterol-derived (e.g., cholesterol) tail and a dialkyl tail with tunable branching and spatial arrangement, enabling systematic control over packing behavior, protonation characteristics, and nanoparticle assembly. The resulting LNPs exhibited favorable apparent pKa values (6.2-6.8)…

Sn-2 palmitate-enriched structured lipids attenuate early aging phenotypes in senescence-accelerated mice.

Aging is characterized by progressive cognitive decline and metabolic dysregulation, contributing to increased vulnerability to age-related disorders. β-position palmitic acid-containing structured lipids (sn-2 palmitate), naturally enriched in human milk fat, have been reported to modulate inflammation and lipid metabolism. However, whether sn-2 palmitate-rich lipids exert beneficial effects beyond early development, particularly in the context of aging-related decline, remains unclear; further, their effects in aged populations or senescence models remain largely unknown. Therefore, we inves…

Mendelian Randomization and Transcriptomics Analysis Reveal Potential Relationships Between Lipids, Immune Cells and PCOS.

BACKGROUND: It has been noted that plasma lipid levels are closely associated with polycystic ovary syndrome (PCOS). Immune cells have also been recognized as critical indicators for evaluating the impact of lipids on disease progression. However, there is still a lack of conclusive causal evidence as to whether immune cells mediate the relationship between lipids and PCOS. METHODS: This study used a two-step two-sample Mendelian randomisation (TSMR) analysis to explore the causal relationship between plasma lipid groups and PCOS, and the mediating role of immune cells in this relationship. T…

Resolving the mRNA Encapsulation-Release Trade-off via Compensatory Forces in Engineered Ionizable Lipids.

A critical challenge in lipid nanoparticle (LNP) delivery of messenger RNA (mRNA) is the inherent trade-off between stable encapsulation and efficient intracellular release. Here, this challenge is addressed through rationally engineering compensatory forces between mRNA and LNP, leveraging short-range intermolecular interactions (van der Waals, hydrogen bonding) to dynamically balance long-range Coulombic binding. Guided by a computational-experimental framework, a "contact number" metric is developed to decipher mRNA and LNP binding hierarchies, enabling the strategic incorporation of short-

Mendelian randomization, lipids, and coronary artery disease: trade-offs between study designs and assumptions.

Mendelian randomization (MR) studies have been described as naturally occurring randomized trials. However, MR studies often deviate from appropriate trial design principles, and many use two-sample designs with strong assumptions not required in randomized trials. Using data from the Million Veteran Program, we empirically evaluated the impact of study design choices and two-sample approaches in an MR study of lipids and coronary artery disease. We designed an MR study of participants of European descent with no history of coronary artery disease and no contraindications to low-density lipopr…

Engineered Hybrid Nanovesicles Combining Macrophage Membranes and Artificial Lipids for Abdominal Aortic Aneurysm Therapy.

BACKGROUND AND AIMS: Abdominal aortic aneurysm (AAA) is a vascular condition with high mortality for which no pharmacological treatments have been approved. Targeting endothelial dysfunction as a primary disease initiator, the vascular endothelial cell (VEC)- protective compound Senkyunolide I (SEI) demonstrates therapeutic promise through robust antiapoptotic activity. Nevertheless, SEI's clinical translation faces limitations due to systemic toxicity, necessitating development of safer therapeutic alternatives. RESULTS: This study presents an engineered biomimetic nanoplatform (Lipo-MM nano
