In this study scanning electron microscopy was used to examine the stereocilia and their cross-linkages in guinea pig cochleas 1, 2, 4 and 8 months after endolymphatic sac obliteration. Initial changes were restricted to the stereocilia of the outer hair cells and consisted of a disarrangement and bulging of the stereocilia with an interruption of their cross-linkage systems. Subsequently, the stereocilia became fused and atrophied. Cross-linkages of inner hair cells remained intact. We have studied the structure and morphogenesis of Dugbe (DUG) virus (Bunyaviridae, Nairovirus) in cultured porcine kidney (PS) cells and a tick cell line (Ra 243) using immunogold electron microscopy. The purpose of this study was to undertake a three-dimensional analysis of the vascular network of the lamina propria in the gingiva, alveolar mucosa, buccal mucosa, palate, and lingual mucosa of the dog. Using the corrosive resin casting technique, casts of the vascular network were prepared and examined by scanning electron microscopy. In the oral mucosa, larger arteries in the submucosa divide into smaller branches that enter the lamina propria. The distribution of acetylcholinesterase (AChE)-positive nerve fibers in the chicken pancreas was investigated with histochemical methods at the light and electron microscopic level. AChE-positive nerve bundles were found to run along the pancreaticoduodenal artery and their branches proceed into interlobular connective tissue, form a plexus around the interlobular secretory ducts and small arteries, and penetrate the exocrine parenchyma. G cells of the frog antral mucosa are described both in deplasticized semithin sections treated with antigastrin/CCK COOH terminal serum and in subsequent thin sections observed under the electron microscope. G cells are quite abundant in the antral mucosa, located between mucosecretory cells in the lateral aspects of glands. They bear irregular, occasionally round granules, with a 190-nm mean diameter.

We compared the ultrastructure of the intracytoplasmic inclusion substance present in Lafora bodies and the myocardium of patients with Lafora disease, with that in the hepatocytes and myocardium of patients with type IV glycogenosis. Fibril-like structures and associated electron-dense clumps (crumpled plates) are the main components of the deposits in these two diseases. Ultrastructurally, the inclusions in both diseases appeared quite similar. The use of SEM as an adjunct to TEM and electrophysiological examination of the cochlea is now well established. It has provided a relatively simple method of assessing the effect of noise, ototoxic drugs, electrical stimulation, etc. on the surface features of the organ of corti. In the present study we compared the accuracy of three different ultrastructural methods used to confirm clinically proved cases of mycosis fungoides (MF) from skin biopsy specimens. Method 1 is a qualitative method and confirmed the diagnosis of MF in only 44% of the patients with MF. Method 2 involved image analysis of lymphocytic nuclei and confirmed the diagnosis of MF in only 67% of the patients with MF. Digestion of the poly(ADP-ribose) with snake venom phosphodiesterase yielded phosphoribosyl-AMP and AMP in a ratio of 30 to 1, indicating the "chain length" of 30 if the poly(ADP-ribose) is a linear molecule as previously supposed. Gel filtration and gel electrophoresis, however, revealed a very heterogeneous size distribution, the average size being, much longer than (ADP-ribose)30. Neurosecretory PC12 cells were exposed in a variety of experimental conditions to nanomolar concentrations of alpha-latrotoxin purified from the venom of the black widow spider. A case of malignant carotid body chemodectoma with neighbouring lymph nodes metastases is reported. The case is noteworthy as it was accompanied by another jugulotympanic chemodectoma. There was also hereditary component as four of the patients forebears had been operated for cervical chemodectomas. An ultrastructural study showed the characteristic secretory granules. 32 other cases of tumors of carotid body and jugular glomus with metastases are reviewed.

A well-documented case of solitary extramedullary plasmacytoma located in the small intestine is presented. The patient underwent local resection of the tumor, and shows no signs of local recurrence or dissemination of the disease after a five-year follow-up. The pathologic nature of plasma cell tumors is discussed, and it is concluded that the demonstration of intracytoplasmatic immunoglobulin is essential for the exact classification of plasma cell neoplasms.

A method was developed for observing changes in the endothelial cells in rabbit ear veins in vivo by scanning electron microscopy. Injection of fatty acids into the ear vein caused damage to the endothelium. The first signs of damage seen were marked bulges in the nuclei and loss of the rhomboidal shape of the endothelial cells. More severe damage included loss of nuclei, leaving holes in the cytoplasm. Colposcopically directed punch biopsy specimens of the uterine cervix were taken from women in whom Papanicolaou smears indicated cytological abnormalities. Half of each specimen was processed for scanning electron microscopy and half for light microscopy. The surface morphology of normal cervical, metaplastic and frankly carcinomatous epithelia and epithelia of carcinoma in situ, as seen under the scanning electron microscope, was compared with the appearance revealed by Papanicolaou smears, and haematoxylin- and eosin-stained sections.

The influence of chenodesoxycholic acid on the gastro-intestinal tract of male Wistar rats was studied. Chenodesoxycholic acid was orally administered in a dosis of 20mg/kg body weight over a period of 5, 14, 20, 30 and 60 days. The electron microscope revealed only slight pathological changes compared to the control group of untreated animals. While stomach, duodenum and ascending colon were found to be non-specific, supranuclear fatty deposits were detected in the jejunum; in the terminal ileum and the jejunum swollen microvilli and a loosening of the terminal reticulum was observed.
