Role of Descending Serotonergic Fibers in the Development of Pathophysiology after Spinal Cord Injury (SCI): Contribution to Chronic Pain, Spasticity, and Autonomic Dysreflexia.

As the nervous system develops, nerve fibers from the brain form descending tracts that regulate the execution of motor behavior within the spinal cord, incoming sensory signals, and capacity to change (plasticity). How these fibers affect function depends upon the transmitter released, the receptor system engaged, and the pattern of neural innervation. The current review focuses upon the neurotransmitter serotonin (5-HT) and its capacity to dampen (inhibit) neural excitation. A brief review of key anatomical details, receptor types, and pharmacology is provided. The paper then considers how d

Intestinal lipid metabolism controls molecular and behavioral immunity through NHR-68 and gut-brain signaling in Caenorhabditis elegans.

Animals must allocate limited energetic resources across competing defense programs in response to infection. Here, we show that the conserved nuclear hormone receptor NHR-68 integrates fatty acid metabolism with the neural control of molecular and behavioral immunity in Caenorhabditis elegans. Acting in parallel with NHR-10, NHR-68 controls genes involved in polyunsaturated fatty acid (PUFA) metabolism. Loss of NHR-68 disrupts linoleic acid (LA) homeostasis, impairing pathogen avoidance behavior. Supplementation with LA restores avoidance, and fat-3 inhibition, which elevates LA, enhances pat

Crosstalk of NPY and TGFβ orchestrates the signaling to facilitate perineural invasion of oral squamous cell carcinoma.

BACKGROUND: Perineural invasion (PNI) frequently occurs in oral squamous cell carcinoma (OSCC) and predicts poor prognosis. Although PNI is increasingly recognised as a process driven by tumour-nerve crosstalk, the underlying molecular mechanisms remain unclear. We investigated the role of sympathetic nerve-derived neuropeptide Y (NPY) and its receptor NPY1R in OSCC PNI. METHODS: NPY/NPY1R expression was assessed in human OSCC tissues by immunostaining, qPCR, and TCGA data analysis. Functional studies using Cal27 and SCC9 cells included migration, invasion, and sphere assays. The causal role

Examination of associations between psychopathy and neural reinforcement sensitivity theory constructs.

We investigated psychopathy from the neurobiological perspective of reinforcement sensitivity theory (RST). In contrast to previous semantically derived self-report scales, we operationalised RST systems neurally with evoked electroencephalography (EEG). Participants were from a community sample weighted towards externalising psychopathology. We compared the Carver & White Behavioural Inhibition System (BIS)/Behavioural Approach System (BAS) scales with EEG responses associated with RST's systems of goal conflict (aka 'behavioural inhibition'), repulsion/outcome conflict (aka 'fight/flight/fre

Motor cortex dysfunction in problem gamblers.

Impairments in response inhibition have been implicated in gambling psychopathology. This behavioral impairment may suggest that the neural mechanisms involved in response inhibition, such as GABAA -mediated neurotransmission in the primary motor cortex (M1), are also impaired. The present study obtained paired-pulse transcranial magnetic stimulation markers of GABAA and glutamate receptor activity from the left M1 of three groups-problem gamblers (n = 17, 12 males), at-risk gamblers (n = 29, 19 males), and controls (n = 23, six males)-with each group matched for alcohol use, substance use, an

The cholinergic anti-inflammatory pathway revisited.

Inflammatory bowel disease negatively affects the quality of life of millions of patients around the world. Although the precise etiology of the disease remains elusive, aberrant immune system activation is an underlying cause. As such, therapies that selectively inhibit immune cell activation without broad immunosuppression are desired. Inhibition of immune cell activation preventing pro-inflammatory cytokine production through neural stimulation has emerged as one such treatment. These therapeutics are based on the discovery of the cholinergic anti-inflammatory pathway, a reflex arc that ind

ZIKV envelope protein is a strong blocker of early directional differentiation in the neural lineage.

Zika virus (ZIKV) infection is an international concern of public health emergency and has been associated with severe neurodevelopmental abnormalities. ZIKV-encoded envelope protein was virulent to neural cells, but its role in the neurodifferentiation process is not well known. Thus, we used mouse embryonic stem cells (mESCs) to mimic early neural differentiation in vitro and comprehensively studied the role of ZIKV envelope protein in the early directional differentiation of the neural lineage. We found that the ZIKV envelope protein does not affect the pluripotency of mESCs, but is a stron

Pineal progenitors originate from a non-neural territory limited by FGF signalling.

The embryonic development of the pineal organ, a neuroendocrine gland on top of the diencephalon, remains enigmatic. Classic fate-mapping studies suggested that pineal progenitors originate from the lateral border of the anterior neural plate. We show here, using gene expression and fate mapping/lineage tracing in zebrafish, that pineal progenitors originate, at least in part, from the non-neural ectoderm. Gene expression in chick indicates that this non-neural origin of pineal progenitors is conserved in amniotes. Genetic repression of placodal, but not neural crest, cell fate results in pine

[Effect of beta-adrenergic blocker propranolol during reflex renal degeneration induced by sciatic nerve transection].

We present the data on functional status of the mineralocorticoid receptor system in rat kidney in the course of renal reflex degeneration induced by sciatic nerve transection at the background of both renal denervation and injections of beta blocker propranolol. According to the status of renal mineralocorticoid receptors, simultaneous blocking of both neural and humoral pathological stimuli incoming to the kidney after the nerve injury to a higher extent inhibited cytochemical changes in the organ as compared to blocking the neural transmission alone. We propose that both the central neural

The data suggest that failure of neural inhibition induced by FG-7142 engages NMDA receptor processes to produce lasting potentiation of transmission in neural circuits that mediate defensive response with behavioral consequences. Since FG-7142 interferes with GABA mediated neural inhibition and is …

Changes in Aperiodic (1/f Slope) Activity During a Picture-Word Interference Task: Effects of Congruency and Sequence Manipulations.

Aperiodic neural activity (1/f EEG) has been proposed to reflect the balance between excitatory and inhibitory (E/I) inputs, with steeper spectral slopes reflecting increased inhibition and flatter slopes indicating excitation. This activity is also thought to reflect the temporal coordination of neural firing, offering insights into fundamental brain dynamics. Recent studies have shown that the 1/f slope is sensitive to stimulus onset, characterized by initial inhibitory shifts followed by excitatory rebounds, which may reflect cognitive control mechanisms involved in suppressing distractions

Cognitive Inflexibility Shares Across Schizophrenia and Obsessive-Compulsive Disorder: A Task-Based Functional MRI Study.

BACKGROUND AND HYPOTHESIS: Schizo-obsessive comorbidity (SOC), defined as obsessive-compulsive symptoms (OCS) in schizophrenia (SCZ), is linked to severe psychopathology and poor prognosis. Schizophrenia and obsessive-compulsive disorder (OCD) share cognitive impairments, particularly in inhibition and cognitive flexibility, which may underlie SOC. However, little is known regarding the underlying neural mechanisms of SOC. We aimed to directly compare the inhibition- and cognitive flexibility-related neural activations between patients with SOC, SCZ, OCD, and healthy controls (HCs). STUDY DES

Hyperthermia and Serotonin: The Quest for a "Better Cyproheptadine".

Fine temperature control is essential in homeothermic animals. Both hyper- and hypothermia can have deleterious effects. Multiple, efficient and partly redundant mechanisms of adjusting the body temperature to the value set by the internal thermostat exist. The neural circuitry of temperature control and the neurotransmitters involved are reviewed. The GABAergic inhibitory output from the brain thermostat in the preoptic area POA to subaltern neural circuitry of temperature control (Nucleus Raphe Dorsalis and Nucleus Raphe Pallidus) is a function of the balance between the (opposite) effects m

Shared Response Inhibition Deficits but Distinct Error Processing Capacities Between Schizophrenia and Obsessive-Compulsive Disorder Patients Revealed by Event-Related Potentials and Oscillations During a Stop Signal Task.

Background: Schizophrenia (SCH) patients are at high risk for obsessive-compulsive syndrome, which can lead to difficulty in differential diagnosis between SCH and obsessive-compulsive disorder (OCD). It would be of great clinical value to identify objective markers for these diseases based on behavioral or neurological manifestations. Deficient response inhibition has been reported in both SCH and OCD; however, it is unclear if common or distinct neural abnormalities underlie this impairment. Methods: To address this question, we compared Stop signal task performance and associated event-rela

Regulation of cell surface protease matriptase by HAI2 is essential for placental development, neural tube closure and embryonic survival in mice.

Hypomorphic mutations in the human SPINT2 gene cause a broad spectrum of abnormalities in organogenesis, including organ and digit duplications, atresia, fistulas, hypertelorism, cleft palate and hamartoma. SPINT2 encodes the transmembrane serine protease inhibitor HAI2 (placental bikunin), and the severe developmental effects of decreased HAI2 activity can be hypothesized to be a consequence of excess pericellular proteolytic activity. Indeed, we show here that HAI2 is a potent regulator of protease-guided cellular responses, including motogenic activity and transepithelial resistance of epit

Mammalian D-Cysteine: A new addition to the growing family of biologically relevant D-amino acids.

Mammalian D-Cysteine is racemized from L-cysteine by serine racemase, a pyridoxal phosphate (PLP)-dependent enzyme. Endogenous D-Cysteine plays a role in neural development by inhibiting proliferation of neural progenitor cells (NPCs) via protein kinase B (AKT) signaling mediated by the FoxO family of transcription factors. D-Cysteine binds to Myristoylated Alanine Rich C Kinase Substrate (MARCKS) and alters phosphorylation at Ser 159/163 and its translocation from the membrane. By racemizing serine and cysteine, mammalian serine racemase may play important roles in neural development highligh

Enhanced response inhibition and reduced midfrontal theta activity in experienced Vipassana meditators.

Response inhibition - the ability to suppress inappropriate thoughts and actions - is a fundamental aspect of cognitive control. Recent research suggests that mental training by meditation may improve cognitive control. Yet, it is still unclear if and how, at the neural level, long-term meditation practice may affect (emotional) response inhibition. The present study aimed to address this outstanding question, and used an emotional Go/Nogo task and electroencephalography (EEG) to examine possible differences in behavioral and electrophysiological indices of response inhibition between Vipassan

Post-inhibitory rebound firing promotes hypothalamic activation during male mating in mice.

During male mating, activation of the medial preoptic area (mPOA) and inhibitory input from the bed nucleus of the stria terminalis (BNST) are both required for mount initiation. How behaviorally relevant neural activation emerges in this limbic-hypothalamic circuit dominated by inhibition remains unclear. Here, using in vivo recordings, we show that mount-active mPOA neurons are suppressed before behavioral onset and that this pre-behavioral inhibition depends on BNST activity. Moreover, the magnitude of this inhibition predicts subsequent activation, consistent with post-inhibitory rebound f

Competing mechanisms of plasticity impair compensatory responses to repetitive apnoea.

KEY POINTS: Intermittent reductions in respiratory neural activity, a characteristic of many ventilatory disorders, leads to inadequate ventilation and arterial hypoxia. Both intermittent reductions in respiratory neural activity and intermittent hypoxia trigger compensatory enhancements in inspiratory output when experienced separately, forms of plasticity called inactivity-induced inspiratory motor facilitation (iMF) and long-term facilitation (LTF), respectively. Reductions in respiratory neural activity that lead to moderate, but not mild, arterial hypoxia occludes plasticity expression, i

Effects of Short-Term Synaptic Plasticity in Feedforward Inhibitory Circuits on Cerebellar Responses to Repetitive Sensory Input.

Short-term plasticity occurs at synapses throughout the cerebellum, raising the question of how such plasticity affects cerebellar processing in vivo. To address this issue, we recorded responses of molecular layer interneurons (MLIs), Purkinje cells, neurons of the cerebellar nuclei (CbN), and mossy fibers in the CbN, in awake head-fixed female mice. During short trains of air puffs applied to the whisker pad with intervals from 25 to 200 ms, the first puff generated brief suppressions of spike probability in Purkinje cells and brief elevations in all other cell types, resulting in coincident
