Functional connectivity differences of the olfactory network in Parkinson's Disease, mild cognitive impairment and cognitively normal individuals: A resting-state fMRI study.

Olfactory dysfunction is an early sign of such neurodegenerative diseases as Parkinson's (PD) and Alzheimer's (AD), and is often present in Mild Cognitive Impairment (MCI), a precursor of AD. Understanding neuro-temporal relationships, i.e., functional connectivity, between olfactory eloquent structures in such disorders, could shed light on their basic pathophysiology. To this end, we employed region-based analyses using resting-state functional magnetic resonance imaging (rs-fMRI) obtained from cognitively normal (CN), MCI, and PD patients with cognitive impairment (PD-CogImp). Using machine

A voxel-based morphometry study of anosmic patients.

OBJECTIVE: The aim of our study was to compare volume change in grey matter (GM) and white matter (WM) in a group of subjects with anosmia and a healthy control group. We tried to find a regular pattern of atrophy within and between GM and WM and to determine whether any particular areas are more sensitive to olfactory injury. METHODS: There were 19 anosmic patients and 20 age- and sex-matched control subjects. We acquired MR images on a 3-T scanner and performed voxel-based morphometry using the VBM8 toolbox and SPM8 in a MATLAB® (MathWorks®, Natick, MA) environment. RESULTS: Patients with

The medial amygdala controls the coital access of female rats: a possible involvement of emotional responsiveness.

Partner preference and paced mating tests were accomplished in ovariectomized female rats following bilateral radiofrequency lesions in the medial amygdala. Open field behavior and passive avoidance learning were also examined to investigate the underlying behavioral mechanism. Partner preference was determined in a chamber located between castrated and sexually active males. Airborne olfactory cues were presented to the female through small holes on the partition. The lesion diminished preference for the odor of sexually active males over that of castrated males, even after injection with a h

Influence of the entorhinal cortex on accumbal and striatal dopaminergic responses in a latent inhibition paradigm.

The use of latent inhibition paradigms is one means of investigating the involvement of mesencephalic dopaminergic (DA) neurons in cognitive processes. We have shown recently that DA neurons reaching the core and the dorsomedial shell parts of the nucleus accumbens and the anterior part of the striatum are differentially involved in latent inhibition. In other respects, theoretical, behavioral and anatomo-functional data suggest that the entorhinal cortex (ENT) may control latent inhibition expression. In this study, using in vivo voltammetry in freely moving rats, we investigated the influenc

OCAM: A new member of the neural cell adhesion molecule family related to zone-to-zone projection of olfactory and vomeronasal axons.

Zone-to-zone projection of olfactory and vomeronasal sensory axons underlies the topographic and functional mapping of chemoreceptor expression zones of the sensory epithelia onto zonally arranged glomeruli in the main and accessory olfactory bulbs. Here we identified OCAM (R4B12 antigen), an axonal surface glycoprotein expressed by subsets of both olfactory and vomeronasal axons in a zone-specific manner. OCAM is a novel homophilic adhesion molecule belonging to the immunoglobulin superfamily with striking structural homology to neural cell adhesion molecule. In both the main and accessory ol

Interhemispheric approach for microsurgical removal of olfactory groove meningiomas.

Eighteen consecutive patients with olfactory groove meningiomas, with diameters ranging from 1.5 to 7 cm, underwent microsurgical tumour resection using a unilateral frontal interhemispheric approach. Unilateral frontal craniotomy, superior to the frontal sinus, exposing the superior sagittal sinus was performed. The ipsilateral frontal lobe was gently retracted laterally, and the tumour resected through the gap between the falx and the medial aspect of the frontal lobe, anteriorly to the genu of the corpus callosum. Gross total tumour resection was achieved in all the patients. There was no e

Direct evidence for the migration of LHRH neurons from the nasal region to the forebrain in the chick embryo: a carbocyanine dye analysis.

To examine the possible migration of luteinizing hormone-releasing hormone (LHRH) neurons from the olfactory placode to the brain, the epithelial cells of the olfactory placode of the chick embryos were labeled with a fluorescent carbocyanine dye DiI at embryonic days 3.5-4.0. The embryos treated with DiI were incubated in ovo for 1 to 7 days. The DiI-labeled cells were first detected in the olfactory epithelium and olfactory nerve one day after the application of DiI. Two to four days after the application of the dye, the labeled cells sequentially appeared in the rostral and medial forebrain

Acetylcholinesterase and luteinizing hormone-releasing hormone distinguish separate populations of terminal nerve neurons.

The terminal nerve is composed of a morphologically heterogeneous population of unipolar, bipolar and multipolar neurons located in the nasal and intracranial cavities of vertebrates. The question has arisen as to whether these neurons are neurochemically heterogeneous and therefore possibly functionally different as well. Among the substances localized in the terminal nerve are acetylcholinesterase and luteinizing hormone-releasing hormone-like immunoreactive material. We have developed a double-label procedure, combining immunocytochemistry and enzyme histochemistry to determine whether thes

Prenatal and Early Postnatal Odorant Exposure Heightens Odor-Evoked Mitral Cell Responses in the Mouse Olfactory Bulb.

Early sensory experience shapes the anatomy and function of sensory circuits. In the mouse olfactory bulb (OB), prenatal and early postnatal odorant exposure through odorized food (food/odorant pairing) not only increases the volume of activated glomeruli but also increases the number of mitral and tufted cells (M/TCs) connected to activated glomeruli. Given the importance of M/TCs in OB output and in mediating lateral inhibitory networks, increasing the number of M/TCs connected to a single glomerulus may significantly change odorant representation by increasing the total output of that glome

The Diabetic Nose: A Narrative Review of Rhinologic Involvement in Diabetes (1973-2025).

Background: Although diabetes mellitus is traditionally viewed as a systemic metabolic disorder, growing evidence indicates that it also affects the upper airways through vascular, inflammatory, and neuro-sensory mechanisms. The sinonasal mucosa, highly vascularized and immunologically active, may represent an early target of diabetic microangiopathy and immune-metabolic imbalance. Objectives: Our objectives are to synthesize current evidence on the rhinologic manifestations of DM, with a focus on chronic rhinosinusitis, olfactory dysfunction, and other nasal disorders, and to identify the mai

Chemosensory tobacco product toxicology part 1: sensory mechanisms.

Chemosensory systems detect and discriminate a wide variety of molecules to monitor internal and external chemical environments. They initiate olfactory, gustatory, and chemesthetic sensations; influence human brain cognition and emotion; and guide a wide variety of behaviors essential for survival, including protective reactions, such as avoidance of contaminated foods and potential toxicants. Electronic nicotine delivery systems (ENDS) aerosolize e-liquids for inhaled consumption that typically contain flavorants, propylene glycol, vegetable glycerin, and nicotine. E-liquid aerosols also con

Aberrant DNA methylation profile of hepatitis B virus infection.

We aimed to study the aberrant DNA methylation profile associated with hepatitis B virus (HBV) infection and, to identify key genes and pathways associated with the HBV infection stage. A total of 54 antiviral treatment-naïve HBV-infected patients and six healthy controls were included. Genome-wide methylated DNA immunoprecipitation analysis was performed, as previously described, after which the chip data were preprocessed. Subsequently, Cytoscape software was used for the construction of a protein-protein interaction network, and a database for annotation, visualization, and integrated disco

Relationship of migrating luteinizing hormone-releasing hormone neurons to unique olfactory system glycoconjugates in embryonic rats.

Following their birth in olfactory placode, luteinizing hormone-releasing hormone (LHRH)-containing neurons migrate across the developing cribriform plate and form a dispersed population in the mammalian basal forebrain. The present study reveals the colocalization of unique glycoconjugate antigens (detected with monoclonal antibody CC2) on a subset of LHRH-immunoreactive (LHRHir) cell bodies and growth cones in the rostral forebrain during embryonic development in rats. In addition, LHRHir neurons were found along CC2-immunoreactive (CC2ir) fibers in the nasal cavity, across the cribriform pl

Digitized canine olfaction and multimodal biosensing for breath-based multi-cancer detection: a hypothesis-driven perspective.

Multi-cancer early detection (MCED) is critical for reducing cancer mortality, however current screening technologies have limitations in accessibility, cost, and early stage sensitivity. Breath-based detection using volatile organic compounds (VOCs) offers a non-invasive and scalable alternative, with trained detection dogs demonstrating exceptional olfactory sensitivity in clinical studies. However, the widespread deployment of canine scent detection has been hindered by subjectivity and lack of standardization. This perspective article proposes a novel canine olfaction based MCED platform t

Characterization of carboxypeptidase A6, an extracellular matrix peptidase.

Carboxypeptidase A6 (CPA6) is a member of the M14 metallocarboxypeptidase family that is highly expressed in the adult mouse olfactory bulb and broadly expressed in embryonic brain and other tissues. A disruption in the human CPA6 gene is linked to Duane syndrome, a defect in the abducens nerve/lateral rectus muscle connection. In this study the cellular distribution, processing, and substrate specificity of human CPA6 were investigated. The 50-kDa pro-CPA6 is routed through the constitutive secretory pathway, processed by furin or a furin-like enzyme into the 37-kDa active form, and secreted

CRSwNP in the biologics era: evaluating patient-reported quality of life outcomes.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is a prevalent type 2 inflammatory disorder that substantially impairs patient quality of life (QoL) through cardinal symptoms of nasal obstruction, olfactory dysfunction, and sleep disturbance, further compounded by frequent comorbidities including asthma and non-steroidal anti-inflammatory drug-exacerbated respiratory disease (NERD). While conventional therapies with intranasal corticosteroids, systemic corticosteroids, and endoscopic sinus surgery remain foundational, their limitations in achieving durable disease control have prompted the e

The Body, the Brain, the Environment, and Parkinson's Disease.

The brain- and body-first models of Lewy body disorders predict that aggregated alpha-synuclein pathology usually begins in either the olfactory system or the enteric nervous system. In both scenarios the pathology seems to arise in structures that are closely connected to the outside world. Environmental toxicants, including certain pesticides, industrial chemicals, and air pollution are therefore plausible trigger mechanisms for Parkinson's disease and dementia with Lewy bodies. Here, we propose that toxicants inhaled through the nose can lead to pathological changes in alpha-synuclein in th

TRPA1 channel in the airway underlies protection against airborne threats by modulating respiration and behaviour.

Transient receptor potential ankyrin 1 (TRPA1), a member of the TRP superfamily of cation channels, is broadly expressed in sensory neural pathways, including the trigeminal neurons innervating the nasal cavity and vagal neurons innervating the trachea and the lung. TRPA1 acts as a detector of various irritant chemicals as well as hypoxia and hyperoxia. For the past 15 years, we have characterised its role in respiratory and behavioural modulation in vivo using Trpa1 knockout (KO) mice and wild-type (WT) littermates. Trpa1 KO mice failed to detect, wake up from sleeping, and escape from formal

Environmental CO2 inhibits Caenorhabditis elegans egg-laying by modulating olfactory neurons and evokes widespread changes in neural activity.

Carbon dioxide (CO2) gradients are ubiquitous and provide animals with information about their environment, such as the potential presence of prey or predators. The nematode Caenorhabditis elegans avoids elevated CO2, and previous work identified three neuron pairs called "BAG," "AFD," and "ASE" that respond to CO2 stimuli. Using in vivo Ca(2+) imaging and behavioral analysis, we show that C. elegans can detect CO2 independently of these sensory pathways. Many of the C. elegans sensory neurons we examined, including the AWC olfactory neurons, the ASJ and ASK gustatory neurons, and the ASH and
