The retrovirus strain MC29 induces a variety of tumors in chickens, including myelocytomatosis and carcinomas of the kidney and liver. In addition, the virus can transform cultures of embryonic avian macrophages and fibroblasts. We have characterized the genome of MC29 virus and have identified nucleotide sequences that may encode the oncogenic potential ofthe virus. MC29 virus can replicate only with the assistance of a related helper virus. The defect in replication is apparently a consequence of a deletion in one or more viral genes: the haploid genome of the MC29 virus has a molecular weig

BACKGROUND: Recent advances have revealed oncogenic HER2 mutations as an alternative tumorigenic mechanism independent of HER2 overexpression. Certain HER2 mutations confer resistance to current HER2-targeted therapies.
OBJECTIVE: Based on our previous finding that neratinib promotes HER2 endocytic degradation, its inhibitory effects were examined on drug-resistant oncogenic HER2 mutations.
METHODS: The intracellular distribution of HER2 was observed by immunofluorescence. The expression levels of HER2, pAKT, and pMEK with or without neratinib treatment were investigated by Western blotting.

Colorectal cancer (CRC) remains a significant global health burden, with growing evidence highlighting microbial contributions to its pathogenesis. Certain genotoxigenic bacteria, such as Escherichia coli, Campylobacter jejuni, and Helicobacter pylori, produce virulence factors that induce DNA damage, genomic instability, and chronic inflammation-key features of carcinogenesis. At the same time, viruses such as JC polyomavirus (JCPyV), considered potentially oncogenic, and established oncogenic viruses like Epstein-Barr virus (EBV) and human papillomavirus (HPV) have been detected in colorecta

The cyclin D binding Myb-like Transcription Factor 1 (DMTF1) is a haploinsufficient tumor suppressor in various tumors. Alternative splicing generates a dominant negative, truncated version of full-length DMTF1α, named DMTF1β. DMTF1β has so far been described as an oncogene in breast cancer development. However, a clear understanding of how DMTF1β contributes to carcinogenesis remains unknown. Analyzing DMTF1β protein expression in breast cancer cell lines, as well as a highly metastatic prostate cancer cell line, confirmed a positive correlation between DMTF1β expression and tumorigenic poten

BACKGROUND: METTL3 is a pivotal N6-methyladenosine (m⁶A) regulator that is linked to a worse prognosis of colorectal cancer (CRC). However, METTL3's oncogenic molecular networks in CRC remain unclear. This study aims to establish a co-expression network of METTL3 and identify novel diagnostic and therapeutic targets for CRC.
METHODS: We integrated transcriptomic datasets with clinical cohorts and patient-derived organoids (PDOs) to evaluate the expression profiles of METTL3 and PRMT5. Mechanistic insights were gained through co-immunoprecipitation (Co-IP), m⁶A-methylated RNA immunoprecipitati

Long noncoding RNAs (lncRNAs), widely regarded as nonprotein-coding transcripts longer than 200 nucleotides in length, play active roles in tumorigenesis, including in colorectal cancer (CRC). The abnormally elevated lncRNA small nucleolar RNA host gene 9 (SNHG9) in CRC samples was observed in The Cancer Genome Atlas (TCGA) database. However, the biological role and potential mechanism of SNHG9 in CRC development remain elusive. Herein, 49 paired CRC tissues and matched adjacent normal tissues were obtained to examine SNHG9 levels. Biological functions related to cell proliferation, migration

DEAD-box (DDX) RNA helicases are essential regulators of RNA metabolism and gene expression. Among them, DDX10 remains poorly characterized despite growing evidence supporting its involvement in human diseases. This review provides a comprehensive analysis of DDX10, from its structural and functional features to its emerging roles in solid tumors and hematologic malignancies. We discuss how DDX10, through its conserved domains, contributes to pre-rRNA processing, ribosome biogenesis, and cell proliferation, and explore potential links between DDX10 and processes such as liquid-liquid phase sep

RATIONALE: Only subset of patients with Non-Small Cell Lung Cancer (NSCLC) benefit from immunotherapy and this is partly due to limited understanding of how oncogenic mutations shape the tumor microenvironment (TME).
OBJECTIVES: To define how EGFR and KRAS mutations influence the spatial organization and immune composition of the NSCLC TME.
METHODS: We conducted 38-marker high-plex immunofluorescence to 197 NSCLC tumors (>2 million cells) stratified by EGFR and KRAS genotypes. Quantitative phenotyping and spatial analyses, including cellular neighborhoods (CN), nearest neighbors (NNs = 5-20)

BACKGROUND: Lung adenocarcinoma (LUAD) remains a leading cause of cancer-related mortality, underscoring the urgent need for novel prognostic biomarkers and therapeutic targets. Although PKMYT1, a serine/threonine protein kinase, is implicated in cell cycle regulation, its comprehensive role and clinical significance in LUAD remain poorly defined. This study aims to investigate the oncogenic function, prognostic value, and immunomodulatory role of PKMYT1 in LUAD.
METHODS: We performed integrated multi-omics analyses utilizing data from The Cancer Genome Atlas and Gene Expression Omnibus datab

Chaperone-mediated autophagy (CMA) is a critical regulator of cancer progression and antiviral responses, often modulated by post-translational modifications. SUMOylation, a reversible post-translational modification, is implicated in viral infection and tumorigenesis, yet its role in CMA regulation remains unclear. Here we demonstrate that oncogenic viruses hijack CMA by inducing nuclear localization and SUMOylation of lysosomal-associated membrane protein 2A (LAMP2A), a central lysosomal receptor. Specifically, tripartite motif containing 32 (TRIM32) mediates SUMO2/3 conjugation to LAMP2A at

C-terminal binding protein 1 (CTBP1) is an NAD(H)-dependent transcriptional corepressor that links cellular metabolic state to chromatin-based gene regulation. Since its initial identification, CTBP1 has been implicated in a broad range of developmental and pathological processes, including cancer. Increasing evidence suggests that CTBP1 integrates redox cues, epigenetic repression, and transcriptional networks controlling epithelial plasticity, survival, DNA damage responses, and therapy resistance across multiple malignancies. In this review, we critically examine the current literature on C

Biological heterogeneity among different breast cancer (BC) subtypes results in markedly varying clinical outcomes. Identification and analysis of key gene biomarkers that are differentially regulated during radiation therapy (RT) may pose multiple clinical challenges for BC treatment. The purpose of the study is to identify and analyze the expression of key gene biomarkers and their networks that are differentially regulated after hypofractionated RT. Patients with BC (cT0-T2, N0, M0) were treated with hypofractionated whole breast RT 25 Gy in five fractions, 4 to 8 weeks before breast conser

This study introduces a statistical mechanical framework for allosteric communication in proteins based on the spanning-tree ensemble of residue contact networks. By representingprotein backbones as weighted graphs, we identify each spanning tree as a topological microstate. The canonical partition function is evaluated analytically via the determinant of the reduced weighted Kirchhoff (Laplacian) matrix, allowing for the derivation of global thermodynamic functions (including Helmholtz free energy, internal energy, entropy, and heat capacity) without stochastic sampling. Allosteric channels b

Radiotherapy (RT) is a central treatment for prostate cancer (PCa), relying on the induction of DNA double-strand breaks (DSBs). Tumor ability to repair these breaks limits RT efficacy, making DSB repair inhibitors potential radiosensitizers. However, many of these inhibitors lack tumor specificity and harm normal cells. Therefore, tumor-specific radiosensitization strategies are critically needed for PCa. Approximately 50% of PCa cases harbor the TMPRSS2-ERG gene fusion, leading to overexpression of the ERG transcription factor (ERG+). In this study, we demonstrate that ERG+ tumors shift DSB

SIGNIFICANCE: Nicotine from tobacco products, secondhand smoke, and emerging delivery systems remains a major but underappreciated driver of gastric carcinogenesis (GC). Although reactive oxygen species (ROS) have long been implicated in tumor biology, current models incompletely explain how chronic nicotine selectively reprograms gastric epithelial signaling. This review advances the concept of redox rewiring, whereby nicotine establishes a persistent oxidative state that orchestrates multiple oncogenic programs via spatially compartmentalized NOX signaling.

Cancer arises from oncogenic clones, yet the dynamic mechanisms driving their stepwise evolution toward malignancy remain incompletely understood. Here, we establish the Atlas of Ras-driven Tumors in Drosophila (ART-D), a systematic, cross-species platform that dissects the molecular and phenotypic trajectories of tumorigenesis across ten genetically defined RasV12-driven models. By integrating longitudinal phenotypic profiling, we define three conserved stages of tumor development-initiation, promotion, and progression-distinguished by distinct shifts in tumor burden and tumor-induced cachexi
