PDE4 regulates tissue plasminogen activator expression of human brain microvascular endothelial cells.

INTRODUCTION: Factors regulating brain tissue plasminogen activator (tPA) are pertinent for stroke. Recent observations have suggested a rol … following inhibition of PDE4 and PDE4D, using HBEC monocultures and co-cultures with astrocytes and pericytes, and analyzed relevant signal transduction pathways. RESULTS: PDE4 inhibitor rolipram enhanced OGD effects on endothelial tPA release in endothelial monocultures and co-cultures with astrocytes; there was a 54±10% (p<0.001) reduction of tPA release …

Treatment of Recurrent Intracranial Hemangiopericytoma with SRC-Related Tyrosine Kinase Targeted Therapy: A Case Report.

Hemangiopericytoma (HPC) is a rare sarcomatous tumor arising from pericytes, a support cell found in blood vessels. These tumors can occur throughout the body, particularly in the lower extremities and retroperitoneum. In rare circumstances, HPCs can arise from the meninges. In these cases, they behave similar to meningiomas, in particular angiomatous meningiomas, but tend to be more aggressive and are likely to recur. Treatment usually focuses on surgical resection and radiotherapy with possible inclusion of chemotherapy for control of recurrent disease. We describe a case of recurrent right …

Hemangioblastoma of the corpus callosum: A case report and review of the literature on its origin.

A third case of corpus callosum hemangioblastoma (HB) is presented. With no preoperative embolization, surgery was uneventful and the postop … nd may originate from angiomesenchyma because of the expression of three cell types (stromal cells, pericytes, and endothelial cells) in vimentin. Their capacity for proliferation and differentiation in HB depends on the microenvironment.

Antiangiogenesis in haematological malignancies.

Angiogenesis, the growth of new capillary blood vessels, is a central regulator of cancer growth, and a validated target for cancer therapy. The antiangiogenic agents in clinical use target one or more cellular pathways involved in the cascade of vascular growth. In haematological malignancies, angiogenesis occurs within a bone marrow ecosystem comprised of closely apposed malignant cells, endothelial cells, pericytes, fibroblasts, endothelial progenitor cells, dendritic cells, and extracellular matrix. Inhibition of angiogenesis therefore blocks not only the delivery of oxygen and micronutrie…

Fine structural study of the posterior pituitary after destruction of the hypophysial stalk in the rat.

Electrolytic destruction of the pituitary stalk in rats markedly affected their water metabolism and induced ultrastructural changes in the … bodies. Phagocytic and excretory phenomena were observed in the cytoplasm of endothelial cells and pericytes. The nuclei of pituicytes became loosened, and 5 days following surgery the mitochondria showed swelling and ruptures. The Golgi apparatus and the RER were prominent at 14 days, while degenerative lipid droplets accumulated in the cytoplasm. The fin …

Striated muscle in four categories of leprosy. II. Fine structural changes.

Fine structural changes are described in 21 muscle biopsy specimens from patients with early and established tuberculoid or treated and untr … changes in the small intramuscular blood vessels with proliferation of their basement membrane and pericytes. The mean serum IgG level in lepromatous patients was significantly elevated. This, together with the vascular change, might perhaps be responsible for the degenerative change in the muscle fiber despite a paucity of M. leprae in the muscle.

Endothelial Cell TRIM47 Triggers Blood-Brain Barrier Disruption via CYLD Ubiquitination Degradation in an Intracerebral Hemorrhage Model.

Blood-brain barrier (BBB) disruption is a major driver of neurological deficits after intracerebral hemorrhage (ICH); however, its mechanisms remain unclear. Here, we found that TRIM47 was markedly upregulated after ICH and predominantly expressed in endothelial cells. Endothelial-specific Trim47 deletion normalized peri-hematoma vascular remodeling by reducing the abnormal vascular area and diameter and increasing pericyte coverage, thereby preserving BBB integrity, improving perfusion, reducing neuronal death, and enhancing neurological recovery. Mechanistically, TRIM47 binds to cylindromato

Identification of a pro-angiogenic functional role for FSP1-positive fibroblast subtype in wound healing.

Fibrosis accompanying wound healing can drive the failure of many different organs. Activated fibroblasts are the principal determinants of post-injury pathological fibrosis along with physiological repair, making them a difficult therapeutic target. Although activated fibroblasts are phenotypically heterogeneous, they are not recognized as distinct functional entities. Using mice that express GFP under the FSP1 or αSMA promoter, we characterized two non-overlapping fibroblast subtypes from mouse hearts after myocardial infarction. Here, we report the identification of FSP1-GFP+ cells as a non

Paving the Way Toward Complex Blood-Brain Barrier Models Using Pluripotent Stem Cells.

A tissue with great need to be modeled in vitro is the blood-brain barrier (BBB). The BBB is a tight barrier that covers all blood vessels in the brain and separates the brain microenvironment from the blood system. It consists of three cell types [neurovascular unit (NVU)] that contribute to the unique tightness and selective permeability of the BBB and has been shown to be disrupted in many diseases and brain disorders, such as vascular dementia, stroke, multiple sclerosis, and Alzheimer's disease. Given the progress that pluripotent stem cells (PSCs) have made in the past two decades, it is

[Aortic calcification and calcium].

Vascular calcification occurred as the last step of arteriosclerosis makes a lot of disturbances on vascular function and should influence on the worsening of the vascular diseases. Calcium is the main component of the vascular calcification like bone, and one of causes of vascular calcification should be the hypoparathyroidism due to the lowering of serum calcium and the following calcium paradox seen in osteoporosis. Bone calcium must shift to the arterial wall from the bone. Medial calcification could be formed under the molecular regulatory control like in bone by differentiated osteoblast

Diabetes and retinal vascular disorders: role of the renin-angiotensin system.

Angiotensin II, the effector peptide of the renin-angiotensin system (RAS), has potent growth factor properties in a variety of organs. In the retina, a complete RAS exists, with components residing in the vasculature, neurons and glia. There is increasing interest in a pathogenetic role for angiotensin II in ischaemic retinopathies such as diabetic retinopathy and retinopathy of prematurity. In these situations, the retinal RAS becomes activated and stimulates growth factors such as vascular endothelial growth factor, which contribute to vascular leakage, pericyte migration, angiogenesis and

A simple method for the in vitro culture of human retinal capillary endothelial cells.

A simple method for the culture of human retinal capillary endothelial cells in vitro is described in this report. Through a process of gentle mechanical disruption and sieving, a sufficient yield of retinal microvessels was obtained from one or two human eyes to allow the culture of endothelial cells in abundance. The cells were factor VIII-positive and demonstrated typical vascular endothelial morphology. Pericyte contamination was prevented by using human platelet-poor serum in primary culture and passaging cells with a low concentration of trypsin. Proliferation assays revealed that the ce

Diabeteslike preproliferative retinal changes in galactose-fed dogs.

Retinal vessel changes were experimentally investigated by a combination of color fundus photography, fluorescein angiography, and histologic studies in beagles that were fed a 30% galactose diet for up to 66 months. Previously, we have described the appearance of pericyte ghosts, microaneurysms, acellular capillaries, and intraretinal hemorrhages in dogs fed a galactose diet for up to 36 months. These disorders were similar to those observed in humans with background diabetic retinopathy. We report herein that dogs fed galactose for 48 to 60 months experience retinal changes associated with t

Dual faces of angiogenesis: Mechanisms and therapeutic applications.

Angiogenesis is a highly coordinated process essential for development, tissue homeostasis, and repair. In physiologic settings, endothelial cells remain quiescent until transient hypoxia- or injury-induced surges of VEGF (vascular endothelial growth factor), FGF (fibroblast growth factor), and other pro-angiogenic cues activate receptor tyrosine kinases, triggering MAPK (mitogen-activated protein kinase) and PI3K-AKT (phosphoinositide 3-kinase-AKT) cascades that drive sprouting, proliferation, migration, and vessel stabilization via pericyte recruitment and balanced Ang (angiopoietin)-Tie2 (t
