The effect of dutasteride on the efficacy of photoselective vaporization of the prostate: results of a randomized, placebo-controlled, double-blind study (DOP trial).

OBJECTIVES: To study the effect of Dutasteride on the efficacy of GreenLight photoselective vaporization of the prostate (PV). Some investigators have suggested that 5 alpha-reductase inhibitors may interfere with PV by reducing intraprostatic blood flow. Dutasteride offers the most complete blockade of the 5 alpha-reductase inhibitors, with minimal increase in side effects. METHODS: This is a prospective, placebo-controlled, randomized, double-blind study. A total of 59 patients were randomized to either dutasteride 0.5 mg or placebo for 3 months before and 12 months after PV. Surgical time,

Effect of progesterone on cervical shortening in women at risk for preterm birth: secondary analysis from a multinational, randomized, double-blind, placebo-controlled trial.

OBJECTIVES: To determine whether progesterone supplementation alters cervical shortening in women at increased risk for preterm birth. METHODS: We performed a planned secondary analysis from a large, multinational preterm birth prevention trial of daily intravaginal progesterone gel, 90 mg, compared with placebo in women with a history of spontaneous preterm birth or premature cervical shortening. Transvaginal cervical length measurements were obtained in all randomized patients at baseline (18 + 0 to 22 + 6 weeks' gestation) and at 28 weeks' gestation. For this secondary analysis, the differ

Treatment of psoriasis vulgaris with topical calcipotriol has no short-term effect on calcium or bone metabolism. A randomized, double-blind, placebo-controlled study.

The purpose of this double-blind, placebo-controlled study was to examine whether the vitamin D analogue calcipotriol, topically applied to psoriatic skin lesions, had any effect on calcium or bone metabolism. Thirty-four outpatients with psoriasis vulgaris were randomized to treatment with either calcipotriol ointment (50 micrograms/g) or vehicle ointment twice daily for 3 weeks. The patients were put on a calcium energy fixed diet and examined once weekly. The mean amount of calcipotriol ointment used was 40.3 g/week (range 8.2-95.4 g/week). The results of biochemical markers on calcium and

A randomised trial to evaluate the effect of infra-red stimulation of local trigger points, versus placebo, on the pain caused by cervical osteoarthrosis.

Claims have been made that local heat is particularly useful in treating osteoarthritis pain. This randomised trial evaluates the short term efficacy of local heat, on the pain caused by cervical osteoarthrosis. Two treatment groups were compared, one receiving mock TNS and the other heat from an infra-red gun (the IRS Medtec 100). Both the placebo and treatment groups received treatment on local tender trigger points. Each patient received four treatments. Pain was assessed before entry into the trial and after the completion of treatment; the parameters used for assessing pain were, analgesi

Multimodal examination of the acute effects of cannabis on subjective and physiological stress-related outcomes: a randomized placebo-controlled laboratory study.

People who use cannabis commonly report stress-relieving effects, yet no studies have simultaneously assessed the acute effects of cannabis on multiple stress systems under placebo-controlled conditions. We conducted a randomized, double-blind, placebo-controlled trial in which 120 people (56 men, 52 women, 12 non-binary/transgender) who regularly use cannabis were assigned to vape placebo, a moderate dose (20 mg Δ9-tetrahydrocannabinol [THC]), or a high dose (40 mg THC) of cannabis. Stress ratings and salivary cortisol samples were obtained before (T0), 5 minutes (T1), and 60 minutes (T2) aft

The effects of cannabidiol on sleep disturbances within a sample of high trait worriers: A double-blind, randomized placebo controlled trial.

Cannabidiol (CBD), a nonintoxicating molecule derived from the cannabis plant, is garnering attention as a novel sleep aid despite a dearth of empirical literature supporting its efficacy for sleep-related indications. The present study aimed to address this gap. Participants were 63 individuals (Mage = 29.27; SDage = 9.58) reporting elevated trait worry who were randomly assigned to take 300 mg CBD, 50 mg CBD, or placebo daily for 2 weeks. Results suggested 300 mg CBD decreased sleep disturbances and sleep quality significantly more than 50 mg CBD (B = -0.39, t = -2.59, p < .05, d = 0.08), bu

Effects of Omega-3 Fatty Acids on Tobacco Craving in Tobacco Users: A Single-blind, Randomized, Placebo-controlled Study.

BACKGROUND: Tobacco use and its smoke produces oxidative stress in the body, which eventually triggers cell damage by lipid peroxidation. Smokers report lower levels of omega-3 fatty acids (FAs) in their serum as compared to nonsmokers. Omega-3 deficiency impairs neurotransmission, resulting in hypofunctioning of the mesocortical system, which is a reward and dependency system that can raise tobacco cravings, disrupting tobacco quitting efforts. Omega-3 polyunsaturated fatty acid (PUFA) regulates stress, anxiety, and negative emotions that are associated with tobacco urges. Limited research ha

Effects of Pep19 on adipose tissue distribution and metabolic parameters in adults with obesity: A randomized, double-blind, placebo-controlled trial.

Whether modulation of adipose tissue distribution alone can improve insulin sensitivity independently of weight loss remains unclear. In this randomized, double-blind, placebo-controlled 90-day trial, we investigated whether adults with obesity could achieve metabolic improvement through modulation of adipose tissue distribution independent of weight loss. Fifty-six participants received oral placebo or Pep19 (5 or 10 mg daily). Regional fat distribution and metabolic parameters were assessed longitudinally. Pep19 at 10 mg reduced central adiposity, as demonstrated by a reduced android-to-gyno

Effects of noradrenergic blockade on emotional working memory and its neural underpinnings: A randomized, double-blind, placebo-controlled trial.

The neurotransmitter noradrenaline (NA) has been previously associated with brain-wide regulatory functions for various cognitive as well as affective processes. Clonidine, an adrenoceptor agonist, predominantly stimulates α2-receptors, reducing NA release. However, the specific effects of clonidine, as well as NA's general impact on neural mechanisms underlying cognitive and affective processing remain incompletely understood. 74 healthy men were included in a double-blind, randomized, parallel-group functional magnetic resonance imaging (fMRI) study with two treatment conditions: single-dose

A randomised, double-blind, placebo-controlled trial examining the dose-dependent effects of acute alcohol intoxication on neurocognitive performance.

RATIONALE: Acute alcohol intoxication impairs cognitive and motor control, elevating risk in safety-critical contexts. While blood alcohol concentration (BAC) quantifies exposure, it does not reflect real-time functional impairment. Mobile, performance-based assessments may offer scalable detection tools, but their sensitivity under controlled alcohol challenge requires further validation. OBJECTIVES: To evaluate the sensitivity of a mobile cognitive assessment (DRUID®) to alcohol-induced impairment across low and moderate intoxication levels and examined objective cognitive performance and s

Effects of Protocatechuic Acid on Endothelial Function and HDL Functionality in Hypercholesterolemic Subjects: A Randomized Double-Blind Placebo-Controlled Trial.

Background: High-density lipoprotein (HDL) exerts atheroprotective properties, partially by maintaining endothelial function. Protocatechuic acid (PCA) has been shown to improve endothelial function in mice. However, whether PCA improves endothelial function in humans is currently unknown. Our studies aimed to evaluate the effects of PCA on endothelial function and HDL concentration, subclass distribution, and functionality in humans. Methods: A double-blind, randomized, placebo-controlled trial was conducted in 150 hypercholesterolemic participants (aged 35-65 y) daily receiving 20 mg PCA or

Placebo-Referenced Class-Level Treatment Effects on Chronic Kidney Disease Progression in Patients With Diabetes: A Network Meta-Analysis.

BACKGROUND AND AIM: Multiple pharmacologic therapies reduce chronic kidney disease (CKD) progression in patients with diabetes; however, the absence of head-to-head trials and heterogeneity across studies limits the interpretation of indirect comparisons. We evaluated class-level treatment effects within a predominantly placebo-centred evidence network. METHODS: We conducted a systematic review and class-level frequentist random-effects network meta-analysis of randomised controlled trials evaluating major pharmacologic therapy classes for CKD progression in patients with diabetes. Effects of Shuhe granules on chronic pain in patients with rheumatoid arthritis and fibromyalgia: a randomized, double-blind, placebo-controlled trial.

BACKGROUND: Patients with rheumatoid arthritis (RA) and concomitant fibromyalgia (FM) frequently experience severe, refractory pain, elevated disease activity scores, and substantial psychological distress. Conventional treatments frequently fail to adequately address this complex pain syndrome, and evidence-based traditional Chinese medicine (TCM) interventions for this specific comorbidity remain limited. This pilot study aimed to assess the feasibility of a definitive trial and preliminarily evaluate the efficacy and safety of Shuhe granules (SHG), a Chinese herbal formulation, for alleviat
