Exploring propriospinal neuron-mediated neural circuit plasticity using recombinant viruses after spinal cord injury.

Propriospinal neurons (PSNs) play a crucial role in motor control and sensory processing and contribute to plastic reorganization of spinal circuits responsible for recovery from spinal cord injury (SCI). Due to their scattered distribution and various intersegmental projection patterns, it is challenging to dissect the function of PSNs within the neuronal network. New genetically encoded tools, particularly cell-type-specific transgene expression methods using recombinant viral vectors combined with other genetic, pharmacologic, and optogenetic approaches, have enormous potential for visualiz

Genome characterization of a novel fowl adenovirus serotype 8b isolate and construction of the reverse genetic system for rapid genome manipulation.

Inclusion body hepatitis (IBH) induced by fowl adenovirus serotype 8b (FAdV-8b) infection is an important avian infectious disease circulating around the globe, posing significant losses to the poultry industry. In this study, a FAdV-8b strain, CH/SDQD/2021, was isolated from IBH-affected chickens in Shandong province, China and the genetic properties of CH/SDQD/2021 were characterized. The full genome length of CH/SDQD/2021 is 44,000 bp, with a G+C content of 58 % and 32 open reading frames (ORF). Sequencing alignment and phylogenetic analysis indicated that the genome identity of CH/SDQD/202

Reductive modification of genetically encoded 3-nitrotyrosine sites in alpha synuclein expressed in E.coli.

Tyrosine nitration is a post-translational protein modification relevant to various pathophysiological processes. Chemical nitration procedures have been used to generate and study nitrated proteins, but these methods regularly lead to modifications at other amino acid residues. A novel strategy employs a genetic code modification that allows incorporation of 3-nitrotyrosine (3-NT) during ribosomal protein synthesis to generate a recombinant protein with defined 3-NT-sites, in the absence of other post-translational modifications. This approach was applied to study the generation and stability

Genomic epidemiology, evolution, and transmission dynamics of porcine sapelovirus associated with diarrheic piglets.

Porcine sapelovirus (PSV) is increasingly detected in swine enteric disease complexes, but its evolutionary dynamics and transmission patterns remain insufficiently characterized. In this study, we analyzed 327 fecal samples collected from diarrheic piglets between 2015 and 2022 and identified persistent PSV detection in northeastern China. Because PSV was detected in the context of mixed enteric viral infections in this dataset, our data do not establish PSV as an independent causative agent of diarrhea. Comparative whole-genome and evolutionary analyses of globally circulating PSV strains re

Construction of a versatile cloning vector for Agrobacterium tumefaciens-mediated transformation of Beauveria bassiana using benomyl for selection.

Beauveria bassiana is a filamentous fungus that has considerable potential to combat insect pests. Therefore, an easy to use genetic transformation tool helps to investigate which fungal genes play a role in virulence and host specificity. However, B. bassiana is often very resistant to antibiotics used as selective agents for genetic transformation. We developed a binary cloning vector, pMBE1, for Agrobacterium tumefaciens-mediated transformation of B. bassiana. Selection of fungal transformants is based on the benomyl resistance gene that was introduced in this vector, as B. bassiana is very

Recombination-center ablation of Igh and Igκ defines distinct developmental and immune phenotypes in mice.

Genetic disruption of immunoglobulin loci has long established the requirement for heavy- and light-chain rearrangement during B-cell development. Here, we generated Igh-KO, Igκ-KO, and Igh/Igκ double-KO mice by CRISPR/Cas9-mediated deletion of essential recombination-center elements and performed a coordinated side-by-side phenotypic analysis. Consistent with established heavy-chain-deficient models, Igh ablation caused a profound block in B-cell development beyond the pro-B stage, with marked loss of peripheral B cells and humoral effector populations. Consistent with classic Igκ-targeting s

Genetic Basis of Breast and Ovarian Cancer: Approaches and Lessons Learnt from Three Decades of Inherited Predisposition Testing.

Germline variants occurring in BRCA1 and BRCA2 give rise to hereditary breast and ovarian cancer (HBOC) syndrome, predisposing to breast, ovarian, fallopian tube, and peritoneal cancers marked by elevated incidences of genomic aberrations that correspond to poor prognoses. These genes are in fact involved in genetic integrity, particularly in the process of homologous recombination (HR) DNA repair, a high-fidelity repair system for mending DNA double-strand breaks. In addition to its implication in HBOC pathogenesis, the impairment of HR has become a prime target for therapeutic intervention u

A genome-wide collection of barcoded single-gene deletion mutants in Salmonella enterica serovar Typhimurium.

Genetic screening of pools of mutants can reveal genetic determinants involved in complex biological interactions, processes, and systems. We previously constructed two single-gene deletion resources for Salmonella enterica serovar Typhimurium 14028s in which kanamycin (KanR) and chloramphenicol (CamR) cassettes were used to replace non-essential genes. We have now used lambda-red recombination to convert the antibiotic cassettes in these resources into a tetracycline-resistant (TetR) version where each mutant contains a different 21-base barcode flanked by Illumina Read1 and Read2 primer sequ

Revertant mosaicism in a human skin fragility disorder results from slipped mispairing and mitotic recombination.

Spontaneous gene repair, also called revertant mosaicism, has been documented in several genetic disorders involving organs that undergo self-regeneration, including the skin. Genetic reversion may occur through different mechanisms, and in a single individual, the mutation can be repaired in various ways. Here we describe a disseminated pattern of revertant mosaicism observed in 6 patients with Kindler syndrome (KS), a genodermatosis caused by loss of kindlin-1 (encoded by FERMT1) and clinically characterized by patchy skin pigmentation and atrophy. All patients presented duplication mutation

Bone marrow stem cells and liver regeneration.

Development of new approaches to treat patients with hepatic diseases that can eliminate the need for liver transplantation is imperative. Use of cell therapy as a means of repopulating the liver has several advantages over whole-organ transplantation because it would be less invasive, less immunogenic, and would allow the use, in some instances, of autologous-derived cells. Stem/progenitor cells that would be ideal for liver repopulation would need to have characteristics such as availability and ease of isolation, the ability to be expanded in vitro, ensuring adequate numbers of cells, susce

Genetic heterogeneity of Russian, Estonian and Finnish field rabies viruses.

Thirty-five field rabies virus strains were collected in recent years in different regions of the Russian Federation in order to characterize their genetic heterogeneity and to study their molecular epidemiology. In addition to the Russian viruses, seven archive samples from Estonia and Finland and two Russian vaccine strains were also included in the study. The viruses collected were subjected to two different reverse transcription-polymerase chain reaction tests, the amplicons were sequenced and the sequences were analysed phylogenetically. Among the field viruses studied, two main phylogene

Chromosome breakage syndromes and cancer.

There exist numerous genetic disorders, marked by chromosome instability, that are strikingly associated with various cancers. Both the chromosomal instabilities and neoplastic outcomes are related to abnormalities of DNA metabolism, DNA repair, cell-cycle governance, or control of apoptosis. Among these diseases are ataxia telangectasia and Nijmegen breakage syndrome, with increased incidences of lymphomas. Bloom syndrome, Werner syndrome, and Rothmund-Thompson syndrome, each characterized by a DNA helicase defect, are associated with early incidences of different cancers. Other diseases comb

hKCNN3 which maps to chromosome 1q21 is not the causative gene in periodic catatonia, a familial subtype of schizophrenia.

The human calcium-activated potassium channel gene (hKCNN3, hSKCa3) contains two tandemly arranged, multiallelic CAG repeats located in exon 1 which result in short to moderate polyglutamine stretches of unknown functional significance. Case-control and family-based association studies suggested an association of hKCNN3 repeats with susceptibility for schizophrenia. Twelve multiplex pedigrees with periodic catatonia, a schizophrenia subtype with major gene effect and patterns of anticipation, were genotyped using the multiallelic hKCNN3 repeat polymorphism. Using a dominant model of inheritanc

Refined localization of the Batten disease gene (CLN3) by haplotype and linkage disequilibrium mapping to D16S288-D16S383 and exclusion from this region of a variant form of Batten disease with granular osmiophilic deposits.

Haplotype analysis in a collaborative collection of 143 families with juvenile-onset neuronal ceroid lipofuscinosis (JNCL) or Batten (Spielmeyer-Vogt-Sjögren) disease has permitted refined localization of the disease gene, CLN3, which was assigned to chromosome 16 in 1989. Recombination events in four maternal meioses delimit new flanking genetic markers for CLN3 which localize the gene to the chromosome interval 16p12.1-11.2 between microsatellite markers D16S288 and D16S383. This narrows the position of CLN3 to a region of 2.1 cM, a significant reduction from the previous best interval. Usin

Reverse Genetics System for Rift Valley Fever Virus.

Rift Valley fever virus (RVFV) is an important mosquito-borne virus that can cause severe disease manifestations in humans including ocular damage, vision loss, late-onset encephalitis, and hemorrhagic fever. In ruminants, RVFV can cause high mortality rates in young animals and high rates of abortion in pregnant animals resulting in an enormous negative impact on the economy of affected regions. To date, no licensed vaccines in humans or anti-RVFV therapeutics for animal or human use are available. The development of reverse genetics has facilitated the generation of recombinant infectious vi

Reverse Genetics Systems for Emerging and Re-Emerging Swine Coronaviruses and Applications.

Emerging and re-emerging swine coronaviruses (CoVs), including porcine epidemic diarrhea virus (PEDV), porcine deltacoronavirus (PDCoV), and swine acute diarrhea syndrome-CoV (SADS-CoV), cause severe diarrhea in neonatal piglets, and CoV infection is associated with significant economic losses for the swine industry worldwide. Reverse genetics systems realize the manipulation of RNA virus genome and facilitate the development of new vaccines. Thus far, five reverse genetics approaches have been successfully applied to engineer the swine CoV genome: targeted RNA recombination, in vitro ligation

Targeting polysialic acid-abundant cancers using oncolytic adenoviruses with fibers fused to active bacteriophage borne endosialidase.

Genetic replacement of adenoviral fiber knobs by ligands that enable tumor specific targeting of oncolytic adenoviruses is challenging because the fiber knob contributes to virus assembly. Here, we present a novel concept by describing stable recombinant adenoviruses with tumor specific infection mode. The fiber knob was replaced by endosialidaseNF (endoNF), the tailspike protein of bacteriophage K1F. EndoNF recognizes polysialic acid, an oncofetal antigen characteristic for high malignant tumors of neuroendocrine origin. An intramolecular chaperone contained in endoNF warrants folding and com

Recent advances in the development of adenovirus- and poxvirus-vectored tuberculosis vaccines.

Tuberculosis vaccine research began with the search for a vaccine that might be better than, and thus could replace, the current Bacillus Calmette Guérin (BCG) vaccine. Over the last fifteen years or so, intense research effort has led to the identification of a number of novel tuberculosis (TB) vaccines which can be divided into 4 categories: genetically modified mycobacteria, protein, plasmid DNA and viral. However, it is increasingly believed that the current BCG vaccine will continue to be used as a childhood vaccine and that more effort should be directed to developing appropriate boostin

A Dual-Plasmid CRISPR/Cas9-Based Genome Editing System for Efficient Gene Knockout and Large DNA Integration in Cronobacter malonaticus.

Species of Cronobacter are emerging foodborne pathogens that pose a significant threat to neonates. Functional genomic studies in Cronobacter have been hindered by the lack of efficient genetic manipulation tools. Here, we established a CRISPR/Cas9-based genome editing platform for Cronobacter. We developed a dual-plasmid platform, pAmpCRISPR/pCasCm, by integrating CRISPR/Cas9 with the λ-Red recombination machinery. This platform enables scarless genome editing in Cronobacter malonaticus and proved effective for gene knockout and large-fragment integration under the tested conditions. Furtherm

Can a Y Chromosome Degenerate in an Evolutionary Instant? A Commentary on Fong et al. 2023.

It is well known that the Y chromosomes of Drosophila and mammals and the W chromosomes of birds carry only small fractions of the genes carried by the homologous X or Z chromosomes, and this "genetic degeneration" is associated with loss of recombination between the sex chromosome pair. However, it is still not known how much evolutionary time is needed to reach such nearly complete degeneration. The XY pair of species in a group of closely related poecilid fish is homologous but has been found to have either nondegenerated or completely degenerated Y chromosomes. We evaluate evidence describ
