Myocardial injury without heart failure following envenomation by the scorpion Leiurus quinquestriatus in children.

The enzymatic activity of creatine kinase-MB isoenzyme (CK-MB), a sensitive and specific marker of myocardial damage, was measured in 32 children following scorpion envenomation. CK-MB activity, total creatine phosphokinase (CPK) and serum glutamine oxalacetic transaminase (SGOT) levels were examined for relationship with electrocardiographic (ECG) results and the clinical state of the children. Twenty-seven out of the 32 children had signs of systemic intoxication ("symptomatic" cases), while the other five children had only local signs ("asymptomatic" cases). Thirteen out of the 27 symptomat

Activation of the ryanodine receptor Ca2+ release channel of sarcoplasmic reticulum by a novel scorpion venom.

We identified a peptide fraction from the venom of the scorpion Buthotus hottentota that stimulated binding of [3H]ryanodine to ryanodine receptors of skeletal and cardiac sarcoplasmic reticulum and brain microsomes in a highly specific manner. Activity was concentrated in a peptide fraction of Mr 5,000-8,000. Assuming a single active peptide in this fraction, we estimated a dissociation constant of 20-30 nM for the interaction of the peptide with the ryanodine receptor. The whole venom and the purified fraction activated skeletal ryanodine receptor Ca2+ release channels incorporated into plan

Voltage-sensitive Na+ channels in mammalian peripheral nerves detected using scorpion toxins.

The localization of voltage-sensitive sodium channels was investigated in mouse, rat and rabbit sciatic nerves using iodinated alpha- and beta-Scorpion toxins (ScTx) as specific probes. Saturable specific binding for a beta-ScTx was detected in mouse sciatic nerve homogenates (Kd = 90 pM, binding site capacity = 90 fmol mg-1 protein). LM autoradiographic studies demonstrated that the two types of ScTx stained the Ranvier nodes of the myelinated fibres, and also showed a clear but weaker labelling of the unmyelinated Remak bundles. In the sciatic nerve, which is widely considered as a model 'my

Structure/activity relationships of scorpion alpha-toxins. Multiple residues contribute to the interaction with receptors.

Chemical modifications of tyrosine and tryptophan residues of scorpion alpha-neurotoxins II and III from Androctonus australis Hector were performed as well as modification of the two arginines and the alpha-amino group of toxin I. The pharmacological potencies of each derivative were assessed in vivo by LD50 measurement and in vitro by competition experiments with 125I-toxin for synaptosomal receptors. Arginine residues in positions 2 and 60 and the alpha-amino group of Androctonus toxin I were derivatized by p-hydroxyphenylglyoxal; the corresponding modified toxins exhibit low pharmacologica

Isolation and characterization of two toxins from the Mexican scorpion Centruroides limpidus limpidus Karsch.

1. Several toxic polypeptides were found in the venom of the scorpion Centruroides limpidus limpidus. Comparative studies of the potency of the venom in different strains of mice were conducted. 2. A new type of toxin (component II.9), specific for crustaceans (crayfish and isopods), was isolated from this scorpion and was shown to have the following N-terminal amino acid sequence: Lys-Lys-Asp-Gly-Tyr-Leu-Val-Asn-Lys-Tyr-Thr-Gly-Cys-Lys-Val-Asn-Cys- Tyr-Lys-Leu-Gly-Glu-Asn-Lys-Phe-Cys-Asn-Arg-Glu-. 3. A polypeptide toxic to mice (component II.6) from this venom was shown to have the following

Scorpion venom inhibits carbamylcholine-induced 86rubidium efflux from rat parotid acini.

The muscarinic agonist carbamylcholine stimulated 5-fold 86Rb efflux from preloaded rat parotid acini. Apamin was without effect on this carbamylcholine-induced 86Rb efflux. By contrast, the venom from Leiurus quinquestriatus (a scorpion from Israel) inhibited non-competitively this efflux while being without effect on the carbamylcholine-stimulated 45Ca efflux and amylase release. This heat-resistant inhibitory effect of the venom was destroyed by boiling in the presence of dithiothreitol. These results suggest that the venom from L. quinquestriatus contains a toxin capable to block apamin-in

Pandinus imperator scorpion venom blocks voltage-gated potassium channels in nerve fibers.

We have examined the effects of venom from the scorpion Pandinus imperator on the membrane currents of voltage-clamped frog myelinated nerve fibers using the Vaseline-gap method. Crude venom, applied externally in concentrations from 50 to 500 micrograms/ml, selectively blocked the voltage-gated potassium currents without affecting nodal sodium currents or resting conductances. Block of potassium channels by Pandinus venom was highly dependent on the membrane voltage, being greater at negative potentials than at positive potentials. The blocking effects of Pandinus venom were irreversible on t

Impact of scorpion Heterometrus fulvipes venom on cholinesterase rhythmicity in the tropical mouse Mus booduga.

Rhythmicities of acetylcholine (ACh) and acetylcholinesterase (AChE) were studied in the mouse, Mus booduga (Gray), following intramuscular injection of scorpion, Heterometrus fulvipes (C. Koch) venom. Envenomation inhibited the activity levels of ACh and AChE in all the tissues selected for experimentation. Control animals exhibited diel rhythmicity in ACh and AChE while envenomated animals showed fluctuation in the Phase (delta phi), amplitude (A), Acrophase (phi) and the extreme activity hours (X and X1).

Voltage clamp analysis of sodium channels in normal and scorpion toxin-resistant neuroblastoma cells.

Sodium currents mediated by voltage-sensitive sodium channels in normal and scorpion toxin-resistant neuroblastoma cells were measured using a giga-ohm seal recording method in the whole cell patch configuration. The voltage and time dependence of sodium currents were similar in normal and mutant cell lines. Half-maximal activation occurred for test depolarizations in the range of -7 to -11 mV. Half-maximal inactivation occurred for pre-pulses in the range of -62 to -69 mV. Scorpion toxin from Leiurus quinquestriatus (100 to 200 nM) increased the time constant for sodium channel inactivation 6

Alpha-scorpion neurotoxin derivatives suitable as potential markers of sodium channels. Preparation and characterization.

Modified scorpion neurotoxins, i.e. mono-biotinylated and mono-azido derivatives either on lysine 58 or lysine 60 have been characterized both at the structural level (sequence and circular dichroism) and by their pharmacological activity (toxicity to mice and ability to displace 125I-Androctonus australis Hector toxin II from its receptor sites. The results allowed us to pinpoint a region of the molecule including lysine residues 58 and 60 that is important for neurotoxin receptor interaction. Furthermore, as these derivatives retain, after 125I labeling, high binding capacities to synaptosom

Effects of tityustoxin (TsTX) from scorpion venom on the release and synthesis of acetylcholine in brain slices.

Tityustoxin (TsTX) increased the release and synthesis of acetylcholine (ACh) in slices of rat brain hippocampus, hypothalamus, thalamus, cortex and striatum. The effect was highest in slices of hippocampus and cortex and smallest in hypothalamus. These effects of TsTX were dependent on the presence of Na+ and Ca2+ in the incubation medium. EGTA, 0.1 mM, blocked the effect of TsTX in all areas, except for the hippocampus and frontal striatum, where a concentration of 10 mM was required. Tetrodotoxin blocked the increase in the release and synthesis of ACh induced by tityustoxin.

Abnormal cardiovascular and electrocardiographic profiles and cardiac glycogen content in rabbits injected with scorpion venom.

Cardiovascular and ECG abnormalities were studied following injection of venom of Buthus tamulus, a common scorpion found in South India. Venom was administered in doses of 2 and 4 mg/kg body weight. Subsequent estimation of glycogen content of different chambers of the heart, showed a significant reduction in tissue glycogen levels in both atria and ventricles in animals treated with a venom dose of 2 mg/kg. However, a significant reduction occurred only in atria with a higher venom dose of 4 mg/kg.

Potential-dependent effects of sea anemone toxins and scorpion venom on crayfish giant axon.

Effects of two kinds of sea anemone toxin (Parasicyonis actinostoloides and Anemonia sulcata) and scorpion venom (Leiurus quinquestriatus) on crayfish giant axons were examined electrophysiologically. All toxins acted on the axon in a similar manner to prolong the falling phase of the action potential. In all cases the development of toxicity was reduced when the nerve membrane was depolarized by a current injection. However, the ranges of membrane potential where the significant reduction in toxicity took place were different for each toxin. The action of Parasicyonis toxin was also suppresse

Three-dimensional structure of a protein from scorpion venom: a new structural class of neurotoxins.

The three-dimensional crystal structure of variant-3 toxin from the scorpion Centruroides sculpturatus Ewing has been determined at 3 A resolution. Phases were obtained by use of K2PtCl4 and K2IrCl6 derivatives. The most prominent secondary structural features are two and a half turns of alpha-helix and a three-strand stretch of antiparallel beta-sheet, which runs parallel to the alpha-helix. The helix is connected to the middle strand of the beta-sheet by two disulfide bridges; a third disulfide bridge is located nearby. Several loops extend out of this dense core of secondary structure. Studies in energetics and population dynamics of the water scorpion, Nepa rubra L. (Insecta: Hemiptera).

There is little information on the energy turnover of carnivorous water insects. This study concerns the energy flow through an imaginal population of the water scorpion, Nepra rubra, a water bug common in the lake "Neusiedler See" in eastern Austria.Population dynamics, production and respiration were determined from 1972 to 1975 under conditions as natural as possible. The cumulative sum of production energy is small, comprising 179.3 J for ♂♂ and 223.8 J for ♀♀, with an additional 241.6 J for reproduction. Food reserves, particularly fat accumulated in the fat body during prediapause, are u
