Anion exchange microcolumn chromatography is the ICSH (1978) recommended procedure for the quantitation of Hb A2 and Hb S. The use of isoele … evels were similar (r = +0.93, P less than 0.001). In addition, paired analyses of 30 specimens, 20 sickle cell trait and 10 with sickle cell trait combined with alpha thalassaemia, were quantitated for Hb S. The two groups were also well separated by both procedures and Hb S levels were similar (r = +0.94, P less than 0.001). In conclusion, IEF followed by laser d …

A range of estimates for sickle cell and beta thalassaemia have been derived for the different ethnic groups living in the U.K., reflecting uncertainty over the true population value in certain countries and the heterogeneity within and between countries of origin comprising the same ethnic group. These were validated against six community screening programmes, with the estimated range correctly predicting the number of affected births observed by the programmes. In England approximately 3000 affected babies (0.47%) carry sickle cell trait and 2800 (0.44%) carry beta thalassaemia trait annuall…

In a family of Low-Zaïre, a child with sickle cell beta+-thalassaemia disease, aged 7, presented 33.6% haemoglobin A. The clinical evolution was benign, except for the occurrence of an extensive bilateral ischemic necrosis of the ribs. This is the third case of sickle cell thalassaemia with the haemoglobin phenotype SFA reported in Zaïrians, whereas the SF phenotype has repeatedly been found: beta+-thalassaemia seems therefore much rarer in Zaïrians than betadegrees-thalassaemia. Besides the thalassaemia trait and the sickle cell trait, hereditary elliptocytosis was also segregating in this fa…

A 15 years old Muslim female presented with moderate anaemia, mild jaundice and hepatosplenomegaly with no history of blood transfusion in the past. Routine examination was suggestive of haemolytic jaundice. High pressure liquid chromatography (HPLC) electrophoresis of the patient's blood showed haemoglobin (Hb) SD disease. As it is a double heterozygous disease, family screening was done. It showed that the father was sickle cell trait. Mother was Hb D trait. Both the brothers were sickle cell trait and the only sister was normal. Hb SD disease is a very rare variety of haemoglobinopathy and …

In Colorado, newborn screening for hemoglobinopathies by cellulose acetate and citrate agar electrophoresis of dried capillary blood spots was established in 1979. We reviewed the results of screening 528,711 infants through 1988. Forty-seven infants with sickle cell diseases and 27 infants with other hemoglobin diseases were identified. The initial screening failed to detect sickle cell anemia in 4 infants, but the hemoglobinopathy in 3 of these infants was diagnosed correctly by routine retesting of those with suspected sickle cell trait. A total of 47 infants with sickle cell diseases were …

BACKGROUND: Infectious Pathogens (IP) remain a significant contributor to stillbirths and under-five deaths in resource-limited settings. Ch … valence among under-five deaths (51.1%, 95%CI:47.9-54.3). SCD was identified in 46 cases (3.5%) and sickle cell trait in 256 cases (19.2%), for a total of 302 cases (22.7%) with SCD/trait. IP prevalence was higher in SCD (60.9%, 95%CI: 46.3-73.9) than in sickle cell trait (25.8%, 95%CI: 20.6-31.6). Among SCD and sickle cell trait cases, the most frequently identifi …

Between 10 and 15 percent of blacks have sickle cell disease or sickle cell trait. Either is capable of a sickle cell crisis which could occur during a surgical procedure. Because of this, we as otolaryngologists should be aware of how to diagnose sickle cell disease and what specific measures should be followed in order to avoid a sickle cell crisis. This paper deals with specific diagnostic tests available, their interpretation, and our protocol for managment of these patients when they are in the hospital for a surgical procedure.

The γ(G)-158 (C→T) polymorphism plays important function in the disease severity of sickle cell anemia. The XmnI restriction site at -158 position of the γ(G)-gene is associated with increased expression of the γ(G)-globin gene and higher production of HbF. This study aims to determine the frequency of the different genotypes of the γ(G) Xmn I polymorphism in sickle cell anemia and sickle cell trait patients in Chhattisgarh and its association with high HbF level. The Xmn1 polymorphic site was determined by PCR-RFLP procedure. XmnI polymorphism were studied in 100 sickle cell patients (SS), 50…

A baby girl presented with symptomatic sickle cell disease exacerbated by mild hypoxemia, despite a newborn-screening diagnosis of sickle cell trait. DNA sequencing of the beta globin gene revealed that her maternal beta globin allele was normal. Her paternal allele had not only the expected sickle-trait mutation, betaGlu6Val, but also a second, charge-neutral mutation, betaLeu68Phe. Analysis of the patient's hemoglobin revealed that the double-mutant protein, which we called "hemoglobin Jamaica Plain," had severely reduced oxygen affinity. Structural modeling suggested destabilization of the …

BACKGROUND: Renal medullary carcinoma is a recently described, highly aggressive neoplasm that affects predominantly young African American males with a history of sickle cell trait. To the best of our knowledge, this is the first report of fine needle aspirate cytology (FNAC) findings of renal medullary carcinoma.
CASE: A 14-year-old, African American male with a history of sickle cell trait presented with the sudden onset of third cranial nerve palsy. Radiographic examination demonstrated possible tumor masses in the brain, thorax and left kidney. Ultrasound-guided fine needle aspiration wa…

OBJECTIVE: We review the cross-sectional imaging findings of six cases of pathologically proven renal medullary carcinoma in patients with sickle cell trait. MRI findings were available in three of the patients. To our knowledge, only one previous report has addressed MRI features of this rare disease.
CONCLUSION: In young patients with sickle cell trait, an infiltrative renal mass with associated retroperitoneal adenopathy and caliectasis are characteristic findings of renal medullary carcinoma on CT and MRI.

A cohort of 574 black Adults (mean age 47 years with a range of 28-73 years) with sickle cell trait (haemoglobin AS) which had been age and sex matched with 1148 controls (haemoglobin AA) was monitored for 7 years. We found no evidence of excess mortality or differential causes of mortality between cases and controls. There were no case deaths related to kidney disease, splenic infarcts, or pregnancy. Reports in the literature have been inconsistent with regard to the health effects of sickle cell trait. These findings suggest that discrimination against trait carriers in terms of life insuran…

We attempted prenatal diagnosis of hemoglobinopathies in 15 cases--11 for beta-thalassemia and four for sickle-cell disease. Fetoscopy was used in seven cases, and placental aspiration in eight. One premature labor, with fetal loss, followed placental aspiration. Globin synthesis was assessed by incubation of samples with 3H-leucine and chain separation on carboxymethylcellulose columns. Homozygous disease was predicted in two pregnancies, which were interrupted, and the diagnosis confirmed. In one case homozygosity was suspected. A repeat test was advised but not accepted. The fetus had thala
