Standardized Pluchea Indica Leaf Extract Exhibited Antiproliferative Activity Against TGF-β-induced Prostate Stromal Cells (WPMY-1) Through G 1 Phase Cell Cycle Arrest via SMAD2/3 and ERK1/2 Signaling Pathways.

BACKGROUND/AIM: Benign prostatic hyperplasia (BPH) is characterized by the abnormal proliferation of prostate stromal cells, resulting in the enlargement of the prostate gland and the manifestation of troublesome symptoms, such as nocturia, urinary retention, and urinary incontinence. Dihydrotestosterone (DHT) and interleukin-17 (IL-17) are known to be key factors in promoting the overproduction of transforming growth factor-beta (TGF-β) in prostate stromal cells, contributing to their excessive proliferation, leading to BPH. MATERIALS AND METHODS: In this study, selected plant extracts tradi

Genetic variants of ADAM17 are implicated in the pathological process of Kawasaki disease and secondary coronary artery lesions via the TGF-β/SMAD3 signaling pathway.

UNLABELLED: Kawasaki disease (KD) is a systemic vasculitis childhood disease frequently complicating coronary artery lesions (CALs). Recently, the gene encoding a disintegrin and metalloprotease 17 (ADAM17) was found to modify vascular pathology in humans by differentially regulating the transforming growth factor-β (TGF-β) signaling pathway, which affects KD/CAL susceptibility. To explore the potential role of ADAM17 in KD occurrence and outcomes, we investigated the association of 28 single nucleotide polymorphisms (SNPs) in ADAM17 and three pathway genes of TGF-β signaling with KD phenotype

RhoA/Rho-kinase activation promotes lung fibrosis in an animal model of systemic sclerosis.

BACKGROUND: Systemic sclerosis (SSc) is a connective-tissue disease characterized by vascular injury, immune-system disorders, and excessive fibrosis of the skin and multiple internal organs. Recent reports found that RhoA/Rho-kinase (ROCK) pathway is implicated in various fibrogenic diseases. Intradermal injection of hypochlorous acid (HOCl)-generating solution induced inflammation, autoimmune activation, and fibrosis, mimicking the cutaneous diffuse form of SSc in humans. Our study aimed firstly to describe pulmonary inflammation and fibrosis induced by HOCl in mice, and secondly to determin

Is Growth Differentiation Factor 11 a Realistic Therapeutic for Aging-Dependent Muscle Defects?

This "Controversies in Cardiovascular Research" article evaluates the evidence for and against the hypothesis that the circulating blood level of growth differentiation factor 11 (GDF11) decreases in old age and that restoring normal GDF11 levels in old animals rejuvenates their skeletal muscle and reverses pathological cardiac hypertrophy and cardiac dysfunction. Studies supporting the original GDF11 hypothesis in skeletal and cardiac muscle have not been validated by several independent groups. These new studies have either found no effects of restoring normal GDF11 levels on cardiac structu

SMAD3 activation in pancreatic stellate cells correlates with PTGDR2 expression and favorable stromal features in pancreatic cancer.

BACKGROUND: Pancreatic cancer is characterized by a dense tumor microenvironment enriched with cancer-associated fibroblasts (CAFs), which display context-dependent tumor-promoting and tumor-restraining properties. However, the molecular basis of stromal heterogeneity remains incompletely understood. METHODS: We examined stromal responses of pancreatic stellate cells (PSCs), a major source of CAFs, using an orthotopic mouse co-implantation model, RNA sequencing, conditioned medium-based functional assays, and immunohistochemical analysis of resected pancreatic cancer specimens. RESULTS: Co-i

Ogg1 regulates TGF‑β-Smad3 signalling and fibrosis progression in chronic kidney disease.

Kidney fibrosis is a hallmark of chronic kidney disease (CKD) and a major contributor to progression toward end-stage renal disease. Here, we identify 8-oxoguanine DNA glycosylase 1 (Ogg1), a DNA repair enzyme, as a regulator of kidney fibrosis. Ogg1 expression is upregulated in experimental fibrosis models in male mice, including unilateral ureteral obstruction (UUO) and aristolochic acid (AA) nephropathy, as well as in kidney tissues from CKD patients. Genetic deletion of Ogg1 reduces fibrosis, tubular atrophy and fibrotic marker expression, and improves renal function in AA nephropathy. Mec

Neutrophil extracellular traps in post-traumatic urethral microenvironment promote hypertrophic urethral stricture via the TLR9/NF-kB/Smad3/IL-8 axis.

INTRODUCTION: Post-traumatic urethral stricture, characterized by hypertrophic scar formation, poses significant therapeutic challenges. Infiltration of neutrophil extracellular traps (NETs) is a prominent feature of post-traumatic urethral microenvironment, but their roles remain unknown. OBJECTIVES: To investigate the mechanisms by which NETs mediate urethral fibrosis and explore potential therapeutic strategies targeting NETs. METHODS: Single-cell RNA sequencing was performed on urethral tissues from New Zealand rabbits with urethral injury to reveal the cell atlas of post-traumatic infla

BMSC-derived EVs alleviate doxorubicin-induced cardiotoxicity via miR-210-3p involving the ACVR1B-SMAD3-PGC-1α signaling axis.

BACKGROUND: Doxorubicin-induced cardiotoxicity (DIC) limits the clinical use of doxorubicin, yet no specific therapeutic interventions are available. Although the cardioprotective role of miR-210 and the therapeutic potential of stem cell-derived extracellular vesicles have been reported, the specific downstream effectors and pathways mediating the cardioprotective effects of BMSC-EVs against DIC remain poorly understood. METHODS: DIC models were established in HL-1s and C57BL/6 mice. High-throughput RNA sequencing combined with EVs uptake assay and miRNA FISH staining was performed to screen

Atopic asthma after rhinovirus-induced wheezing is associated with DNA methylation change in the SMAD3 gene promoter.

Children with rhinovirus-induced severe early wheezing have an increased risk of developing asthma later in life. The exact molecular mechanisms for this association are still mostly unknown. To identify potential changes in the transcriptional and epigenetic regulation in rhinovirus-associated atopic or nonatopic asthma, we analyzed a cohort of 5-year-old children (n = 45) according to the virus etiology of the first severe wheezing episode at the mean age of 13 months and to 5-year asthma outcome. The development of atopic asthma in children with early rhinovirus-induced wheezing was associa

SMAD3 rs17228212 gene polymorphism is associated with reduced risk to cerebrovascular accidents and subclinical atherosclerosis in anti-CCP negative Spanish rheumatoid arthritis patients.

UNLABELLED: Rheumatoid arthritis (RA) is a complex polygenic inflammatory disease associated with accelerated atherosclerosis and increased risk of cardiovascular (CV) disease. Previous genome-wide association studies have described SMAD3 rs17228212 polymorphism as an important signal associated with CV events. The aim of the present study was to evaluate for the first time the relationship between this gene polymorphism and the susceptibility to CV manifestations and its potential association with the presence of subclinical atherosclerosis assessed by the evaluation of carotid intima-media t

TGF-β-SMAD3 signaling mediates hepatic bile acid and phospholipid metabolism following lithocholic acid-induced liver injury.

Transforming growth factor-β (TGFβ) is activated as a result of liver injury, such as cholestasis. However, its influence on endogenous metabolism is not known. This study demonstrated that TGFβ regulates hepatic phospholipid and bile acid homeostasis through MAD homolog 3 (SMAD3) activation as revealed by lithocholic acid-induced experimental intrahepatic cholestasis. Lithocholic acid (LCA) induced expression of TGFB1 and the receptors TGFBR1 and TGFBR2 in the liver. In addition, immunohistochemistry revealed higher TGFβ expression around the portal vein after LCA exposure and diminished SMAD

The human homologue of Bub3 is required for kinetochore localization of Bub1 and a Mad3/Bub1-related protein kinase.

A feedback control mechanism, or cell cycle checkpoint, delays the onset of anaphase until all the chromosomes are correctly aligned on the mitotic spindle. Previously, we showed that the murine homologue of Bub1 is not only required for checkpoint response to spindle damage, but also restrains progression through a normal mitosis (Taylor, S.S., and F. McKeon. 1997. Cell. 89:727-735). Here, we describe the identification of a human homologue of Bub3, a 37-kD protein with four WD repeats. Like Bub1, Bub3 localizes to kinetochores before chromosome alignment. In addition, Bub3 and Bub1 interact

Prophylactic systemic low-dose pirfenidone attenuates intrauterine adhesion by inhibiting the TGF-β1/Smad3 pathway.

BACKGROUND: Intrauterine adhesion (IUA) is a severe fibrotic disorder lacking effective non-surgical treatments. This study explored pirfenidone (PFD), an antifibrotic drug approved for treating lung fibrosis, as a potential therapy for IUA. METHODS: Human endometrial stromal cells (HESCs) were treated with 10 ng/mL transforming growth factor-beta 1 (TGF-β1) ± PFD (0.2 or 0.5 mg/mL) for 48 h to assess fibrosis, proliferation, and migration. In vivo, IUA was induced in BALB/c mice via endometrial scraping and lipopolysaccharide (LPS) exposure, followed by oral PFD administration (150 or 300 mg

Decidualization-empowered ECM hydrogel integrating sustained Tβ4 release drives endometrial regeneration in intrauterine adhesions.

Intrauterine adhesions (IUA), a leading cause of female infertility, result from a pathological switch in the uterine injury response from regeneration to fibrotic scarring. Current treatments are often inadequate as they fail to address this fundamental shift. Here, we report a "decidualization-empowered" hydrogel that reverses this pathology by synergistically combining a bioactive extracellular matrix from decidualized endometrium (DEndo-UdECM) with the sustained release of the anti-fibrotic peptide Thymosin β4 (Tβ4). In a murine IUA model, a single administration of the hydrogel restores e

Herbacetin as a novel therapeutic agent for pulmonary and renal fibrosis by targeting TGFBR2 for degradation.

Rhodiola rosea L. is a traditional Chinese medicine used clinically to treat pulmonary fibrosis. Due to the presence of multicomponents in Rhodiola rosea L., the mechanism of its anti-fibrotic effect remains unclear. Here, we identified that herbacetin from Rhodiola rosea L. is an active molecule with anti-fibrotic effect in two mouse models of bleomycin-induced pulmonary fibrosis and unilateral ureteral obstruction-induced renal fibrosis. We also uncovered the molecular mechanism through which herbacetin exerted its anti-fibrotic effect by inhibiting transforming growth factor beta (TGF-β)/Sm

Activation of SIRT1 by Resveratrol Alleviates Pressure Overload-Induced Cardiac Hypertrophy via Suppression of TGF-β1 Signaling.

INTRODUCTION: Silent information regulator 1 (SIRT1) has been extensively investigated in the cardiovascular system and has been shown to play a pivotal role in mediating cell death/survival, energy production, and oxidative stress. However, the functional role of SIRT1 in pressure overload-induced cardiac hypertrophy and dysfunction remains unclear. Resveratrol (Rsv), a widely used activator of SIRT1, has been reported to protect against cardiovascular disease. We here examine whether activation of SIRT1 by Rsv attenuate pressure overload-induced cardiac hypertrophy and to identify the underl

An unanticipated copy number variant of chromosome 15 disrupting SMAD3 reveals a three-generation family at serious risk for aortic dissection.

Several genes involved in the familial appearance of thoracic aortic aneurysms and dissections (FTAAD) have been characterized recently, one of which is SMAD3. Mutations of SMAD3 cause a new syndromic form of aortic aneurysms and dissections associated with skeletal abnormalities. We discovered a small interstitial deletion of chromosome 15, leading to disruption of SMAD3, in a boy with mild mental retardation, behavioral problems and revealed features of the aneurysms-osteoarthritis syndrome (AOS). Several family members carried the same deletion and showed features including aortic aneurysms
