An ESIPT-based fluorescent probe for highly selective and ratiometric detection of mercury(II) in solution and in cells.

A novel ratiometric fluorescent Hg(2+) detecting system was rationally developed based on the typical excited state intramolecular proton transfer (ESIPT) characteristic of the latent fluorophore, 2-(1-(p-tolyl)-1H-phenanthro[9,10-d]imidazol-2-yl)phenol (Pol) and the Hg(2+)-mediated cleavage of the vinyl group. The probe responds selectively to Hg(2+) over various other metal ions with a larger bathochromic shift (∼100 nm). The sensing mechanism was investigated in detail by fluorescence spectroscopy, NMR spectra and mass spectrometry. Taking advantage of the enhancement effect of dichlorometh

Sensitive detection of polycyclic aromatic hydrocarbons using CdTe quantum dot-modified TiO₂ nanotube array through fluorescence resonance energy transfer.

CdTe quantum dots (QDs) are prepared on TiO(2) nanotubes (TiO(2) NTs), for the first time, with pulse electrodeposition. A novel single-drop optical sensor is prepared with the CdTe QDs-modified TiO(2) NTs, and applied for the detection of polycyclic aromatic hydrocarbons (PAHs) based on fluorescence resonance energy transfer (FRET). Excited at 270 nm, the sensor shows fluorescence emission at around 370 nm. As PAHs are with absorption/fluorescence emission at around 364/410 nm, FRET happens between the CdTe QDs and PAHs with the CdTe QDs as donors and PAHs as receptors. The sensitivity is dep

Single-Atom Switching for Constructing a Full-Spectrum Hypochlorite-Responsive Probe Library: Screening Candidates for Water Analysis.

Residual hypochlorite/hypochlorous acid (ClO-/HClO) is widely used in disinfection, yet simple assays integrating visual readout with quantitative fluorescence analysis remain limited. Herein, we report a single-atom-switching strategy for constructing a six-member, full-spectrum library of ClO--responsive probes. Aqueous screening identified Scou, Scou-Ph, and Scou-CN as selected candidates based on pronounced spectral responses and visual contrast. HRMS supports the ClO--triggered oxidative desulfurization of the thiocarbonyl moiety to the corresponding carbonyl fluorophore, while DFT calcul

Raspberry-shaped H-COF-based dual-mode fluorescent sensing probe for tetracycline antibiotics: standalone and electrospun mat-based point-of-care applications.

Two-dimensional fluorescent materials have emerged as promising candidates for applications in food safety, environmental monitoring, and disease diagnostics, particularly for point-of-care sensors. We report solvothermal development of an imine-linked 2D hydrogen-covalent hybridized organic framework (H-COF), DaTa-1, with novel raspberry-like morphology for the sensitive chemosensing of tetracyclines (TCs) in tap water and milk samples. The developed H-COF exhibited good thermal stability and strong fluorescence in aqueous suspension and in the solid state. DaTa-1 demonstrated excellent sensi

Bifunctional Bile Acid-Salicylaldehyde Based Fluorescent Probe for Excellent Detection of Hydrazine and pH, and Multiple Applications in Environmental Samples, Smartphone-Assisted System, Paper Strips, and Living Cells.

A new cholic acid-salicylaldehyde conjugate, probe CASA, was facilely constructed through one-step esterification. The probe sensitively and selectively detected both N2H4 and pH, with the salicylaldehyde group as the sensing site. Based on the combined AIE/ESIPT mechanism, the probe exhibited a yellow-green turn-on fluorescent response to N2H4 with a low detection limit of 26 nM and a rapid detection time of 10 min. Through the ICT process, the probe displayed a light blue turn-on fluorescence signal for neutral pH variations in a linear range of 5.0-7.8 with a pKa value of 6.26. The microstr

A ratiometric fluorescent probe based on peanut-shell-derived CDs and CuNCs for smartphone-assisted detection of methyl orange.

Methyl orange (MO), a refractory azo dye, can persist in aqueous environments, creating a need for rapid and reliable monitoring methods. In this work, naturally O/N-containing peanut-shell biomass was utilized as an agricultural solid-waste precursor to synthesize oxygen-containing carbon dots (CDs) via a hydrothermal method. The CDs were mixed with D-penicillamine (DPA)/bovine serum albumin (BSA)-stabilized copper nanoclusters (CuNCs) at a controlled volume ratio to form a dual-emission ratiometric fluorescent probe. TEM was used to characterize the nanoscale morphologies, while XPS confirme

Mitochondria-targeted near-infrared probe for Dual-Channel fluorescence imaging of H₂O₂ and viscosity in diabetic pathophysiology.

Diabetes mellitus (DM) is a global health threat marked by metabolic dysfunction. Its pathogenesis involves redox imbalance, mitochondrial dysfunction, and overproduction of H₂O₂, with altered intracellular viscosity associated with disease progression. Thus, monitoring both H₂O₂ and viscosity is key to understanding DM progression. Herein, a near-infrared, mitochondria-targeted dual-responsive fluorescent probe, TPTY-Q-H₂O₂, was designed for concurrent detection of H₂O₂ and viscosity. The probe utilizes a pentafluorobenzenesulfonyl moiety for H₂O₂ recognition, enabling a turn-on fluorescence

A novel smartphone-assisted fluorescence probe based on benzothiazole-derived Schiff base enables accurate detection of Cu2+/Cu+ and H2S and its portable applications.

Copper ions and hydrogen sulfide pollution exert severe adverse effects on plants and other organisms. Consequently, the rapid, real-time, and accurate monitoring of Cu2+/Cu+ and H2S remains challenging. In this study, we designed and synthesized a novel benzothiazole-functionalized salicylaldehyde-based Schiff base (HDBS) for the reversible dual detection of Cu2+/Cu+ and H2S. Probe HDBS enables sequential, fluorescence- and colorimetric-based detection of Cu2+/Cu+ and H₂S in a MeOH-PBS solution system. The high selectivity and water solubility make it suitable for monitoring Cu2+ and H2S in r

A novel dual-lock-dual-key naphthalimide-based fluorescent probe for the detection of H2S and H2O2.

H2S and H2O2 participate in various biochemical reactions as active signaling molecules and maintain cellular homeostasis during oxidative stress. Current methods for detecting H2S and H2O2 mostly rely on their respective single-detection systems, while the research on dual-detection systems for both substances remains less explored. Herein, we rationally constructed a new naphthalimide-based fluorescence probe P1 for the simultaneous detection of H2S and H2O2 using the dual-lock strategy. P1 consists of naphthalimide fluorephore, the disulfide bond-containing ferrocene (first lock) and the bo

Balancing sensitivity and environmental safety in fluorescent probe design: improved ethanol-thermal carbon dots enable a leap in mercury(II) detection and zebrafish-assessed safety.

This study presents an improved ethanol-mediated solvothermal method for synthesizing malic acid/urea-derived carbon dots (MUCDs) for the highly selective and sensitive detection of mercury ions (Hg2+). The detection limit was enhanced by over an order of magnitude compared to previous methods employing similar precursors, achieving a limit of detection of 88.46 nmol L-1, which meets the European Union standard for mercury discharge in industrial wastewater (250 nmol L-1). Pyrophosphate (P2O74-) has been used as a highly effective masking agent for Fe3+, substantially boosting the probe's perf

Efficient design of benzothiazole-based fluorescent probes for benzodiazepines detection by precisely mimicking their binding interactions with the GABAA receptor.

To address the lack of visual detection methods for the screening of abused benzodiazepines (BZDs), this study proposes a biomimetic probe design strategy. Specifically, molecular docking simulations of the γ-aminobutyric acid type-A receptor and BZD complexes revealed multiple shared π … π interactions with the TYR-58, PHE-77, PHE-100, and TYR-160 residues, along with a hydrophobic interaction with the VAL-203. The emulation of these receptor-ligand π … π interactions guided the biomimetic design of the styrylbenzothiazole-based fluorescent probes featuring a large π-conjugation structure. Th

Dual-Component Ratiometric Fluorescent Platform Constructed by Gold Nanoclusters and Cerium-doped Terbium-based Metal-organic Framework for Microcystin-LR Detection.

Ratiometric fluorescence sensing is effective for environmental pollutant monitoring owing to self-calibration and anti-interference properties, but high-sensitivity and high-specificity probes for microcystin-LR (MC-LR) are still scarce. Herein, GP-AuNCs@Ce/Tb-BTC (Ce/Tb metal-organic framework based on benzene-1,3,5-tricarboxylic acid) was developed as a novel ratiometric fluorescent probe for aquatic MC-LR. Coordination interaction and suppressed non-radiative relaxation strengthen its fluorescence response, while dual-fluorescence ratiometric self-calibration optimizes detection stability.

Green solvent boosts fluorescence of copper nanoclusters: Achieving ultra-fast fabrication of a universal sensing platform for H2O2-related biomarkers detection.

BACKGROUND: Copper nanoclusters (Cu NCs) exhibit considerable potential for biosensing applications. However, their practical application is hindered by low fluorescence emission intensity and inadequate stability. Conventional aqueous-based synthesis methods have shown limitations in addressing these issues effectively. Consequently, there is an urgent need to develop efficient strategies that can enhance the fluorescence performance of Cu NCs while maintaining the simplicity of preparation. This study proposes the use of polyethylene glycol 200 (PEG 200) as a green solvent for fabricating Cu

Instantaneous Detection of Trichlorinated Carbon via Photo-Induced Electron Transfer toward Chemosensor for Toxic Organochlorides.

Despite the usefulness of organochlorides as raw materials for organic synthesis, they cause several issues in the human body, such as hepatic dysfunction, tumor, and heavy damage to the central nervous system. Especially when organochlorides contain three or more chlorinated carbons, they tend to be more toxic to the human body possibly owing to relatively high reactivity. Several electron donors (TPCAs) are designed to devise a novel detection system for toxic organochlorides containing trichlorinated carbons, and the detection mechanism of the devised sensor system is systematically identif

Tuning of resonance energy transfer from 4',6-diamidino-2-phenylindole to an ultrasmall silver nanocluster across the lipid bilayer.

Herein we have developed a simple liposome-based donor-acceptor system across the lipid bilayer using 4',6-diamidino-2-phenylindole (DAPI) as the donor and an ultrasmall ligand-capped silver nanocluster (Ag NC) as the acceptor. The process of Förster resonance energy transfer (FRET) between DAPI and the Ag NC across the liposome bilayer has been demonstrated using steady-state and time-resolved fluorescence spectroscopy. The synthesized Ag NCs with a majority of Ag4 and Ag5 cores have been characterized using FTIR, mass spectrometry, HRTEM, UV-Vis and PL spectroscopy. Scanning electron microsc

Time-Resolved Visualisation of Nearly-Native Influenza A Virus Progeny Ribonucleoproteins and Their Individual Components in Live Infected Cells.

Influenza viruses are a global health concern because of the permanent threat of novel emerging strains potentially capable of causing pandemics. Viral ribonucleoproteins (vRNPs) containing genomic RNA segments, nucleoprotein oligomers, and the viral polymerase, play a central role in the viral replication cycle. Our knowledge about critical events such as vRNP assembly and interactions with other viral and cellular proteins is poor and could be substantially improved by time lapse imaging of the infected cells. However, such studies are limited by the difficulty to achieve live-cell compatibl

A Blue-Light-Emitting BODIPY Probe for Lipid Membranes.

Here we describe a new BODIPY-based membrane probe (1) that provides an alternative to dialkylcarbocyanine dyes, such as DiI-C18, that can be excited in the blue spectral region. Compound 1 has unbranched octadecyl chains at the 3,5-positions and a meso-amino function. In organic solvents, the absorption and emission maxima of 1 are determined mainly by solvent acidity and dipolarity. The fluorescence quantum yield is high and reaches 0.93 in 2-propanol. The fluorescence decays are well fitted with a single-exponential in pure solvents and in small and giant unilamellar vesicles (GUV) with a l

Facile ratiometric fluorapatite nanoprobes for rapid and sensitive bacterial spore biomarker detection.

Since bacterial spores, such as Bacillus anthracis spores, are extremely hazardous to human beings and animals, efforts have focused on the development of bacterial spore detector with rapid response and high selectivity and sensitivity. Therefore, we reported a facile one-step chelating-reagent-assisted hydrothermal synthesis of lanthanide-doped fluorapatite (FA) nanoprobes for detecting the biomarker of bacterial spores. In FA synthesis, ethylenediaminetetraacetic acid (EDTA) can serve not only as a shape controller and a stabilizer but also as a chelating reagent for lanthanide ions. After
