In a patient with primary hyperparathyroidism an attempt was made to ablate a middle mediastinal parathyroid gland by forceful staining with radiographic contrast material. The gland was stained on two separate occasions, two weeks apart. Both times the serum calcium level temporarily fell to the normal range but reverted to abnormal levels. The patient ultimately required surgery for correction of hypercalcemia. The mechanism of staining and possible reasons for failure as well as potential complications are discussed.

Known epidemiological and clinical features of the disease should arouse suspicion and prompt appropriate bacteriological studies. Thyroglobulin molecules from normal rats and rats treated with propylthiouracil (PTU) were studied in the electron microscope by the negative staining technique. Examination of 39 eyes with pterygium and 17 contralateral eyes revealed increased astigmatism with the rule (greater than 0.5 D in 46%, greater than or equal to 4 D in 13%) without associated impairment of vision. Feulgen-acriflavine-SO2 staining was performed on chicken erythrocytes and rat liver cells under different cytochemical conditions. The obtained results were compared to conventional Feulgen-pararosaniline-SO2 staining by means of scanning cytophotometry and microfluorometry. It was found that if acid ethanol rinsing was included in the Feulgen-acriflavine-SO2 procedure, both Feulgen procedures were fully comparable in specificity and quantitative aspects.

Methyl green-pyronin (MGP) was used in a one-step procedure to stain smears of urethral discharge from 169 men. Duplicate smears were stained by Gram's method and discharge was cultured for Neisseria gonorrhoeae. The organisms were isolated from 67 specimens and intracellular diplococci were seen in 74 smears after Gram staining and in 77 after staining by MGP. Furthermore, more extracellular and intracellular diplococci were seen in smears stained by MGP than by Gram's method and the proportion of polymorphonuclear leucocytes found to contain the organisms was greater after staining with MGP.

Non-specific esterase staining has been found useful in the detection of macrophages in prostatic fluid (PF). In the early stage of acute prostatitis most of the leucocytes in a smear of PF took up the stain, suggesting the dominant participation of macrophages in the inflammatory area in this stage of the disease. In chronic prostatitis, only a few leucocytes took up the stain. In female acute cystitis, none of the large numbers of leucocytes exhibited staining at any stage of the disease. In the course of studies on the histopathology and pathogenesis of trichomonal infections, specific antisera to Trichomonas vaginalis were produced. Using these antisera, conventional immunocytochemical techniques were modified and applied to a variety of cytologic specimens. With this methodology, trichomonal organisms were identified in specimens from usual and unexpected locations. These findings plus the potential applications of similar methodologies to other areas of cytopathology are reported.

Use of Shikata's stain was found to be a sensitive method for the demonstration of copper-protein complexes, but use of the rhodamine stain may be more reliable. Histochemical and chemical techniques have been used to identify, isolate and characterize elastin from certain bovine cartilages. The results strongly suggest that in addition to fibroblasts and smooth muscle cells, chondroblasts also synthesize elastin. Some of the possible functions of elastin in elastic cartilages are discussed and the possiblity of a new type of elastin perhaps unique to cartilage is suggested.

Histochemical studies were made of pigmented cutaneous lesions from three cases of lepromatous leprosy treated with B663 to determine the nature and histogenesis of the brown pigmentation which develops as a side effect of the drug. For application of the Bodian method to frozen sections, cut frozen peripheral nerve or muscle at 10 mum and mount. Fix for 4 days in 18 parts 80% ethanol, 1 part 10% formalin, and 1 part glacial acetic acid. Fix central nervous tissue in the same mixture prior to freezing and sectioning, and after mounting postfix for 4 days. Impregnate by the Bodian procedure. The results equal Bodian stains of paraffin sections. The technique is simple and reliable. The use of 10 mum frozen sections produces little artifact and allows alternate serial sections to be stained with other techniques.

Application of the Schiff's reagent on fresh and undecalcified cuticle of crustaceans such as Emerita asiatica and Ocypoda platytarsis showed positive results with their calcified layer which led to suggest that salts of calcium account for the staining. In vitro studies with carbonate and other salts furnished confirmatory evidence. It is suggested that in tests involving the Schiff's reagent proper removal of resident salts must be carried out.

The formation of sister chromatid exchanges has been postulated to depend upon the action of DNA repair enzymes. Our experiments with various human cell lines show that the yield of sister chromatid exchanges is within normal limits in both excision-repair-defective and post-replication-repair-defective cells from the autosomal recessive disease, xeroderma pigmentosum. These results indicate that hypotheses invoking known DNA repair processes to acconnt for the recombination of sister chromatids are inadequate and that the exact enzymatic processes are as yet unknown.
