Interrelationships Among Psychological Conditions, Oral Health-related Quality of Life, and Clinical Characteristics in Chinese Patients with Minor Recurrent Aphthous Stomatitis.

AIMS: Recurrent aphthous stomatitis (RAS) is a common chronic inflammatory oral mucosal disorder with uncertain aetiology. This study aimed to investigate the psychological conditions (ie, depression, anxiety and stress) and oral health-related quality of life (OHRQoL) profiles in Chinese Minor RAS (MiRAS) patients. It also sought to explore the relationships between psychological status and OHRQoL and to identify the impact of clinical characteristics of MiRAS on them. METHODS: The descriptive cross-sectional study was conducted using a non-random consecutive sampling method. All recruited p…

Factors influencing mental health and oral health-related quality of life in population with recurrent aphthous stomatitis: a cross-sectional study.

BACKGROUND: Recurrent episodes-induced oral functional limitations exacerbate anxiety, depression, and oral health-related quality of life (OHRQoL) impairments in Recurrent aphthous stomatitis (RAS) patients. However, the specific factors driving these psychological and OHRQoL deteriorations remain incompletely understood. OBJECTIVES: We aimed to quantify psychological distress and OHRQoL impairments in RAS patients and to identify modifiable risk factors underlying these deteriorations to inform targeted interventions. METHODS: This cross-sectional study surveyed RAS patients via an online/…

The Single Amino Acid Change of R516K Enables Efficient Generation of Vesicular Stomatitis Virus-Based Crimean-Congo Hemorrhagic Fever Reporter Virus.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a medically important tick-borne virus, causing severe hemorrhagic diseases in humans. There are no approved vaccines and therapeutics for CCHFV infection. The study of CCHFV authentic virus requires biosafety level 3 facilities, hindering the research and development of antivirals. Here we report the generation of a recombinant vesicular stomatitis virus (VSV) bearing both CCHFV glycoprotein precursor (GPC) and EGFP reporter (rVSV-CCHFV-GFP). We also find that the acquisition of an unexpected single R516K mutation in the GPC protein enables the…

Vesicular stomatitis virus-based vaccine targeting plasmodium blood-stage antigens elicits immune response and protects against malaria with protein booster strategy.

Merozoite invasion of the erythrocytes in humans is a key step in the pathogenesis of malaria. The proteins involved in the merozoite invasion could be potential targets for the development of malaria vaccines. Novel viral-vector-based malaria vaccine regimens developed are currently under clinical trials. Vesicular stomatitis virus (VSV) is a single-stranded negative-strand RNA virus widely used as a vector for virus or cancer vaccines. Whether the VSV-based malarial vaccine is more effective than conventional vaccines based on proteins involved in parasitic invasion is still unclear. In this…

Mucosal Delivery of Recombinant Vesicular Stomatitis Virus Vectors Expressing Envelope Proteins of Respiratory Syncytial Virus Induces Protective Immunity in Cotton Rats.

Respiratory syncytial virus (RSV) is a major cause of lower respiratory tract (LRT) infections, with increased severity in high-risk human populations, such as infants, the immunocompromised, and the elderly. Although the virus was identified more than 60 years ago, there is still no licensed vaccine available. Over the years, several vaccine delivery strategies have been evaluated. In this study, we developed two recombinant vesicular stomatitis virus (rVSV) vector-based vaccine candidates expressing the RSV-G (attachment) protein (rVSV-G) or F (fusion) protein (rVSV-F). All vectors were eval…

SC75741 antagonizes vesicular stomatitis virus, duck Tembusu virus, and duck plague virus infection in duck cells through promoting innate immune responses.

Duck Tembusu virus (DTMUV) and duck plague virus (DPV) are typical DNA and RNA viruses of waterfowl, causing drastic economic losses to the duck farm industry in terms of high mortality and decreased egg production. These 2 viruses reappear from time to time because the available vaccines fail to provide complete immunity and no clinical antiviral drugs are available for them. In the present study, we evaluated the antiviral activity of SC75741 for DTMUV, DPV, and the model virus, vesicular stomatitis virus infection in duck cells. SC75741, a nuclear factor-kappa B (NF-κB)-specific inhibitor i…

Risk factors associated with clinical signs of vesicular stomatitis and seroconversion without clinical disease in Colorado horses during the 2014 outbreak.

Vesicular stomatitis (VS) is caused by a contagious rhabdovirus that affects horses, cattle, and swine. Clinical signs of vesicular stomatitis virus (VSV) infection in pigs and cattle are indistinguishable from foot-and-mouth disease (FMD), a foreign animal disease and reportable disease in the United States (Rodriguez et al., 2000). A VS epidemic occurred in the Rocky Mountain region in 2014-15. A study was conducted in Colorado to evaluate horse- and management-level factors associated with VS. For a horse to be considered a clinical VS horse, there were two requirements. First, clinical VS …

Postexposure Efficacy of Recombinant Vesicular Stomatitis Virus Vectors Against High and Low Doses of Marburg Virus Variant Angola in Nonhuman Primates.

A recombinant vesicular stomatitis virus (rVSV) expressing the Marburg virus (MARV) Musoke variant glycoprotein fully protects macaques against 2 MARV variants and Ravn virus as a preventive vaccine and MARV variant Musoke as a postexposure treatment. To evaluate postexposure efficacy against the most pathogenic MARV variant, Angola, we engineered rVSVs expressing homologous Angola glycoprotein. Macaques were challenged with high or low doses of variant Angola and treated 20-30 minutes after exposure. A total of 25% and 60%-75% of treated macaques survived the high-dose and low-dose challenges…

A recombinant vesicular stomatitis virus encoding CCR5-tropic HIV-1 receptors targets HIV-1-infected cells and controls HIV-1 infection.

Anti-retroviral therapy is useful to treat human immunodeficiency virus type 1 (HIV-1)-infected individuals, but has some major problems, such as the generation of multidrug-resistant viruses. To develop a novel supplemental or alternative therapeutic for CCR5-tropic (R5) HIV-1 infection, we generated a recombinant vesicular stomatitis virus (rVSV) in which the gene encoding its envelope glycoprotein (G) was replaced with the genes encoding R5 HIV-1 receptors (human CD4 and CCR5), designated VSVΔG-CC5. Our present data demonstrate that this rVSV specifically infects cells that are transiently …

Vesicular stomatitis virus-based vaccines expressing EV71 virus-like particles elicit strong immune responses and protect newborn mice from lethal challenges.

Enterovirus 71 (EV71) belonging to the Picornaviridae family is considered the most frequently detected causative agent in hand-foot-and-mouth disease (HFMD) and is a serious threat to public health in the Asia-Pacific region. There are currently no approved vaccines or effective drugs for EV71. In this study, using recombinant vesicular stomatitis virus (rVSV) expressing viral VP1 protein (mVP1) of EV71 as a control, we generated two types of rVSVs that can form EV71 virus-like particles (VLPs). First, we co-infected two rVSVs singly expressing P1 (mP1) and 3CD (m3CD) of EV71. Second, we inse…

Upon intranasal vesicular stomatitis virus infection, astrocytes in the olfactory bulb are important interferon Beta producers that protect from lethal encephalitis.

UNLABELLED: Previously we found that following intranasal (i.n.) infection with neurotropic vesicular stomatitis virus (VSV) type I interferon receptor (IFNAR) triggering of neuroectodermal cells was critically required to constrain intracerebral virus spread. To address whether locally active IFN-β was induced proximally, we studied spatiotemporal conditions of VSV-mediated IFN-β induction. To this end, we performed infection studies with IFN-β reporter mice. One day after intravenous (i.v.) VSV infection, luciferase induction was detected in lymph nodes. Upon i.n. infection, luciferase induc…

In vitro activities of new triazole antifungal agents, posaconazole and voriconazole, against oral Candida isolates from patients suffering from denture stomatitis.

Denture stomatitis is often treated with antifungal agents but recurrences or new episodes are common, and certain episodes can be resistant. New triazoles, such as posaconazole and voriconazole, may represent useful alternatives for management. In vitro activities of amphotericin B, nystatin, miconazole, fluconazole, itraconazole, posaconazole and voriconazole against 150 oral Candida (101 C. albicans, 18 C. tropicalis, 12 C. glabrata, 11 C. guilliermondii, 4 C. parapsilosis, 2 Saccharomyces cerevisiae, 1 C. dubliniensis and 1 C. krusei) from 100 denture wearers were tested by the CLSI M27-A3…

Association between recurrent aphthous stomatitis and inheritance of a single-nucleotide polymorphism of the NOS2 gene encoding inducible nitric oxide synthase.

BACKGROUND: Recurrent aphthous stomatitis is a common ulcerative disease of the oral mucosa. Recurrent oral aphthous ulceration is also a feature of the more serious and systemic Behçet's disease. Nitric oxide is a free radical synthesized by one of a family of nitric oxide synthase (NOS) enzymes and is an important regulator of inflammation and immunity. Association of NOS3 gene polymorphisms encoding endothelial nitric oxide synthase has been reported in Behçet's disease but not recurrent aphthous stomatitis. The aim of this study was to investigate any association between NOS2 gene polymorp…

[Inhibition of 5-fluorouracil-cisplatin-induced stomatitis by oral cryotherapy: use of an ice-bar containing fibrinolysin and deoxyribonuclease combine (Elase)].

Stomatitis caused by a combined chemotherapy with 5-fluorouracil (5-FU) and cisplatin (CDDP) is a serious problem in the course of treatment for patients with oral carcinoma. In the present study, we proposed a form of cryotherapy using an ice-bar containing fibrinolysin and deoxyribonuclease (Elase) to inhibit the stomatitis. The therapeutic effect of the ice-bar cryotherapy was evaluated in 20 patients with oral squamous cell carcinoma who were undergoing 5-FU-CDDP chemotherapy. Nine of the 20 patients were given the ice-bar cryotherapy while the remaining patients were not. As a result, alt…

Mild alternate wetting and drying irrigation improves stomatal and mesophyll conductance of WDR under drought in association with anatomical and hormonal traits.

Alternate wetting and drying irrigation can improve the drought resistance of water-saving and drought-resistant rice (WDR). This response is associated with higher stomatal conductance (gs) and mesophyll conductance (gm), which corresponds to elevated net photosynthesis under drought stress. However, how anatomical and hormonal adjustments contribute to the responses of gs and gm under drought stress in WDR exposed to alternate wetting and drying irrigation remains largely unclear. A pot experiment was conducted under alternate wetting and drying irrigation using cultivars Hanyou73 (HY73, WDR

Linking drought strategy to climate-of-origin in a widespread C4 grass: hydraulic traits, xylem anatomy and stomatal behaviour in Themeda triandra.

Hydraulic, stomatal, and anatomical traits strongly influence plant performance under drought, but the extent to which these trait-types are coordinated and relate to climate-of-origin remains vastly underexplored in herbaceous species relative to woody plants. We quantified variation in these traits in eight populations of Themeda triandra selected from contrasting summer temperature and precipitation regimes across Australia, grown under common conditions and exposed to drought. Trait-climate-of-origin relationships were quantified using linear mixed models. Plants from drier climates exhibi

Using ΦPSII and leaf temperature as indicators of non-steady-state photosynthesis and stomatal conductance during stepwise changes in light intensity.

Quantifying the kinetics of net CO2 assimilation (A) and stomatal conductance (gs) under fluctuating light typically relies on gas exchange measurements, which are slow and thus unsuited for high-throughput phenotyping. As a result, faster, non-invasive phenotyping methods are needed to further evaluate these traits at a larger scale. However, first the relationship between non-steady-state parameters must be examined in greater detail. In this study, we aimed to determine whether variations in non-steady-state values of chlorophyll fluorescence and leaf temperature reflect differences in key

Role of stomatal and leaf anatomical features in defining plant performance under elevated carbon dioxide and ozone, in the changing climate scenario.

This research investigates the interactive effects of elevated ozone (eO3) and carbon dioxide (eCO2) on stomatal morphology and leaf anatomical characteristics in two wheat cultivars with varying O3 sensitivities. Elevated O3 increased stomatal density and conductance, causing oxidative stress and cellular damage, particularly in the O3-sensitive cultivar PBW-550 (PW), compared to HUW-55 (HW). Conversely, eCO2 reduced stomatal density and pore size, mitigating O3-induced damage by limiting O3 influx. Ultrastructural analysis showed that eO3 increased plastoglobule density and damaged chloropla

Alternative transcriptional initiation of OsβCA1 produces three distinct subcellular localization isoforms involved in stomatal response regulation and photosynthesis in rice.

Plants adjust the size of their stomatal openings to balance CO2 intake and water loss. Carbonic anhydrases (CAs) facilitate the conversion between CO2 and HCO3 -, and the OsβCA1 mutant in rice (Oryza sativa) shows similar traits in carbon fixation and stomatal response to CO2 as the dual βCA mutants in Arabidopsis thaliana. However, the exact role of OsβCA1 in these processes was unclear. We used gene editing, molecular biology, and plant physiology to study how OsβCA1 contributes to carbon fixation, stomatal opening, and CO2 responses. OsβCA1 produces three isoforms (OsβCA1A, OsβCA1B, and Os
