The effect of magnesium sulfate on fetal heart rate (FHR) variability and uterine activity is evaluated in 19 preeclamptic patients at term in active labor. Magnesium sulfate was given by intramuscular injection in nine patients and by intravenous infusion in 10 patients after an intravenous loading dose over a period of 15 minutes. No significant change was noted in FHR variability. The metabolism of sulfated glycosaminoglycans (GAG) in bone has been found to be altered in vitamin A deficiency. A neonatal rat calvarium model system was used to determine if these changes are related to catabolic defects. Two day old rat pup calvaria were cultured in media containing serum from A- or A+ rats and radiolabeled sulfate or glucosamine. Commercially available sodium heparinate has been sequentially treated with methanolic 0.06M hydrogen chloride and nitrous acid. ACTH stimulation tests were performed in 15 premature infants in a serial fashion at the ages of 5--10 days and 27--31 days. Six of the 15 had subsequent ACTH stimulation tests at 6--8 weeks and/or 12--13 weeks. We have studied the effects of magnesium sulfate (MgSO4) administration in chronically instrumented normotensive and hypertensive pregnant sheep. Animals of 90 to 144 days' gestation were divided in two groups; those with normotensive arterial pressure and those in which renovascular hypertension had been produced. In fibroblasts taken from patients witli Hurler's syndromne and grown in cultlure, dermatan sulfate constituted a larger percentage of the total sulfated glycosaminoglycans than it did in cultured fibroblasts from unaffected individuals. Moreover, the addition of ascorbic acid (vitamnin C) to the culture medium markedly increased the concentration of dermatan sulfate in the Hurler's fibroblasts but not in the normal fibroblasts. The biochemical phenotype of the Hurler's cells persisted during 28 weeks of serial culture.

Keratan sulfate (KS) is commonly elevated in urine samples from patients with mucopolysaccharidosis type IVA (MPS IVA) and is considered pathognomonic for the condition. Recently, a new method has been described by Martell et al. to detect and measure urinary KS utilizing LC-MS/MS.
