[Purinergic neuromodulation].

Adenosine and its nucleotides participate in the regulation of various functions in the nerve system and in some internal organs. These purines are released from a variety of nervous and non-nervous cellular sources. Adenosine receptors are situated extracellular; they mediate some effects of the adenosine and are coupled negatively (A1 adenosine receptors) and positively (A2 adenosine receptors) to adenylate cyclase. The physiological effects of adenosine are inhibitory; they are exerted synaptically and extrasynaptically. The main synaptic modulatory effect is the presynaptic inhibition of t

Basic mechanisms in vascular headache.

To better understand and treat painful conditions, one needs to identify the cause, discover the source, and develop knowledge of peripheral and central pain transmission; headaches are no exception. The development of appropriate animal models is important. Accordingly, we have reviewed the anatomy, neurochemistry, electrophysiology, and pharmacology of the trigeminovascular system in experimental animals and emphasized whenever possible the relevance of this final common pathway to migraine, cluster, and other headache syndromes in humans. For example, based on recent anatomic dissections, t

Neurons projecting rostrally in the hypogastric nerve of the cat.

Afferent, pre- and postganglionic nerve cell bodies projecting rostrally in the hypogastric nerve have been labelled retrogradely with horseradish peroxidase, in order to study number and location of these neurons. Labelled somata were found in the ganglia of the pelvic plexus, in sacral spinal ganglia and in the sacral spinal cord. They were located mainly ipsilateral with a maximum of afferent and preganglionic neurons at the segmental level S2. The numbers of labelled cells were small (less than 3.5% of all axons in the hypogastric nerve) and varied considerably between animals. Therefore t

Bell's palsy: factors affecting the prognosis in 200 patients with reference to hypertension and diabetes mellitus.

In an effort to refine and amplify the basis for clinical prognosis in Bell's palsy, 14 factors in a clinical study were analysed. From the results, it is still impossible to make a certain prognosis of recovery in the individual patient. However, it is obvious that, based on degree of paresis on assessment, and to a lesser degree the MAP value, at least a quantitative statement can be made.

Diencephalic seizures: responsiveness to bromocriptine and morphine.

Two patients with posttraumatic diencephalic seizures, characterized by autonomic dysfunction and extensor posturing, had partial responses to bromocriptine and complete responses to morphine. Probable synergism between the two agents was noted. These 2 cases suggest the potential effectiveness of this regimen for the treatment of diencephalic seizures, raise questions regarding the role of the dopaminergic and opioid systems in this disease entity, and support the hypothesis that diencephalic seizures represent a release phenomenon in the brain.

Calcium channels. Interactions with ethanol and other sedative-hypnotic drugs.

Biochemical and electrophysiological studies have shown that ethanol potently inhibits voltage-dependent calcium channels in presynaptic nerve terminals in brain. The ability of ethanol to inhibit calcium channels appears to vary from one brain region to another. Chronic ethanol administration results in adaptation of the calcium channels in certain brain regions such that they become tolerant to the inhibitory actions of ethanol. Other sedative-hypnotic drugs, such as barbiturates and benzodiazepines, also inhibit calcium channels but may differ from ethanol in their brain regional potency. T

Identification and purification of an irreversible presynaptic neurotoxin from the venom of the spider Hololena curta.

A search for potent toxins that inhibit neuronal calcium channels in Drosophila melanogaster has resulted in the identification of a presynaptic neurotoxin from the venom of the hunting spider Hololena curta. Using Drosophila neuromuscular junction as an assay, presynaptic inhibitory activity was purified using gel filtration and reverse-phase HPLC. Data from gel electrophoresis indicate that the toxin is composed of two different subunits of Mr 7000 and 9000. At nanomolar concentrations the toxin produced a complete and long-lasting inhibition of synaptic transmission at the Drosophila larval…

Serotoninergic reuptake mechanisms in the control of cataplexy.

Zimelidine, a selective inhibitor of serotonin (5-HT) reuptake in the CNS, was administered to narcoleptic patients. This medication has a potent anticataplectic action without improving daytime somnolence. These results suggest that 5-HT neuronal systems are involved in the physiopathology of cataplexy. Zimelidine, however, has no anticholinergic effect, so it is unlikely that cholinergic mechanisms thought to be important in animal cataplexy would play a major role in human cataplexy. In addition, zimelidine had no effect on nocturnal sleep patterns of these patients which is surprising cons

The mossy fiber projection of the nucleus reticularis tegmenti pontis to the flocculus and adjacent ventral paraflocculus in the cat.

The pontine projection of the flocculus and adjacent ventral paraflocculus was investigated with antegrade and retrograde axonal tracer techniques. Injections of horseradish peroxidase into the floccular complex revealed subsets of labeled neurons in the nucleus reticularis tegmenti pontis, the nucleus raphe pontis and the medial lemniscus. Following injections of tritiated leucine in these subsets, the topographical distribution of labeled mossy fibers in the floccular complex was studied. Cells clustered in the central part of the nucleus reticularis tegmenti pontis project to the rostral fl

Clinical electrophysiology of infantile botulism.

Infantile botulism is a recently recognized cause of acute hypotonic paresis and respiratory failure in young infants. Electrophysiological testing has proven useful in early diagnosis in suspected cases by demonstrating abnormal neuromuscular transmission as is known to occur in botulism. Twenty-five infants with bacteriologically proven botulism were studied by uniform methods in our laboratory and characteristic electrophysiological abnormalities were found. Repetitive stimulation at 20 and 50 Hz was the most specific single test; 23 patients (92%) showed incremental responses. Stimulation

Neuropharmacology of adrenergic neurons in teleost fish.

Although this brief review is based on relatively few types of experiments in few species of teleosts, it is possible to summarize some points of interest regarding the similarities and differences in the mechanisms of adrenergic neurotransmission in fish compared to the higher vertebrates. 1. There is a substantial mixing of cranial autonomic ("parasympathetic") and spinal autonomic ("sympathetic") pathways in the cranial nerves. This close relationship between the two systems and the differences in the nature of the neurons of cranial origin (cholinergic, and non-adrenergic, non-cholinergic)

Cholinergic motor control of sea urchin tube feet: evidence for chemical transmission without synapses.

Isolated tube feet of Strongylocentrotus franciscanus contract briefly when the outer epithelium is touched. Similar twitch-like contractions can be induced by electrical stimulation of the outer surface of the tube foot. These responses appear to be chemically mediated. The following evidence indicates that the transmitter substance may be acetylcholine (ACh): ACh causes muscle contraction. This effect and that of electrical stimuli is potentiated by anticholinesterase agents and is antagonized by cholinergic blocking agents. Anaesthesia with chloralhydrate or chloretone abolishes responsiven

"Prespondylosis" and some pain syndromes following denervation supersensitivity.

Pain is determined by the neurologic properties of receptor organs, neurons, and their interconnections. These may become supersensitive or hyperreactive following denervation (Cannon's Law). A common cause of denervation in the peripheral nervous system is neuropathy or radiculopathy as a sequel to spondylosis. Spondylosis in its early stage may be "asymptomatic" or painless and hency unsuspected, because small-diameter pain fibers may not initially be involved despite the attenuation of the other component fibers of the nerve. The term "prespondylosis" is introduced here to describe this pre

[Physiology of smell and taste].

The functional organization of olfaction and taste are briefly discussed in morphological, physiological, biochemical and behavioural terms. Olfaction in animals serves often for long range navigational purposes whereas taste acts as a close up "last moment food checking system". Special attention is given to the coding problems in both systems. In taste, the spatial coding mode is prevalent. In olfaction a very complex coding system exists, which used temporal as well as spatial means and in which a whole set of olfactory nerve fibers is activated during the transmission of any specific odor
