Long noncoding RNA Malat1 is not essential for T cell development and response to LCMV infection.

Long noncoding RNAs (lncRNAs) are emerging as critical mediators of various biological processes in the immune system. The current data showed that the lncRNA Malat1 is highly expressed in T cell subsets, but the function of Malat1 in T cell remains unclear. In this study, we detected the T cell development and both CD8+ and CD4+ T cell response to LCMV infection using Malat1-/- mice model. To our surprise, there were no significant defects in thymocytes at different developmental stages and the peripheral T cell pool with ablation of Malat1. During LCMV infection, Malat1-/- mice exhibited nor

T-cell subsets defined by monoclonal antibodies in monoclonal gammopathy of undetermined significance (MGUS).

Peripheral T lymphocytes from 31 patients with monoclonal gammopathy of undetermined significance (MGUS), and from a group of controls of the same age range, were stained using monoclonal antibodies of the OKT series. The absolute number and the percentage of OKT3+ cells did not differ in patients compared with the controls. The percentage and absolute number of T-cell subsets with helper/inducer OKT4+ and suppressor/cytotoxic OKT8+ phenotype were not different from those of the controls, thus the OKT4/OKT8 ratio in the patients with MGUS was normal (1.60 versus 1.57 in normal controls). These

Distinctive localization of antigen-presenting cells in human lymph nodes.

Professional antigen-presenting cells (APCs) are sentinel cells of the immune system that present antigen to T lymphocytes and mediate an appropriate immune response. It is therefore surprising that knowledge of the professional APCs in human lymph nodes is limited. Using 3-color immunohistochemistry, we have identified APCs in human lymph nodes, excluding plasmacytoid APCs, that fall into 2 nonoverlapping classes: (1) CD209+ APCs, coexpressing combinations of CD206, CD14, and CD68, that occupied the medullary cords, lined the capsule and trabeculae and were also scattered throughout the diffu

Phenotypic characterization of the cells of the inflammatory response in ovine encephalitic listeriosis.

The brainstem (pons cerebri and medulla oblongata) of 22 sheep aged between 6 months and 3 years which had developed clinical signs of central nervous system dysfunction were examined. Histopathological changes characterized by microabscesses, focal gliosis and perivascular cuffing compatible with natural infection with Listeria monocytogenes were present. The brains were examined by lectin histochemistry and immunohistochemistry with markers for T lymphocytes (CD4+ and CD8+ subsets), B lymphocytes, mononuclear phagocytes (including macrophages, ramified microglia, activated microglia and amoe

Distinct landscapes of T-cell immunity and TCR repertoire between sepsis and pre-septic high-risk states.

Sepsis remains a leading cause of in-hospital mortality worldwide. In recognition of its substantial morbidity and mortality even with optimal treatment, the World Health Organization has declared sepsis a global health priority. The immunoregulatory mechanisms in sepsis are highly complex, and the immune status during the disease course is closely associated with both short- and long-term patient outcomes, making early recognition and intervention critical for survival. While single-cell RNA sequencing (scRNA-seq) has been widely applied to decipher innate immune responses in sepsis patients,

Reduced peripheral CD4+ memory T-cell counts mark a mortality-associated immunophenotype in anti-MDA5 dermatomyositis.

BACKGROUND: Anti-MDA5 dermatomyositis is often complicated by interstitial lung disease and early mortality. Peripheral lymphopenia has been linked to poor outcomes, but the routinely measured lymphocyte compartments underlying this signal remain incompletely defined. We used hierarchical flow-cytometric immunophenotyping to characterize the mortality-associated lymphopenic pattern. METHODS: In this exploratory retrospective cohort, we studied 64 adults with anti-MDA5 dermatomyositis who had baseline lymphocyte subset testing and 180-day follow-up. We compared major lymphocyte populations and

The predictive value of baseline symptom score and the peripheral CD4CD8 double-positive T cells in patients with AECOPD.

BACKGROUND: Accurate prediction of acute exacerbation helps select patients with chronic obstructive pulmonary disease (COPD) for individualized therapy. The potential of lymphocyte subsets to function as clinical predictive factors for acute exacerbations of chronic obstructive pulmonary disease (AECOPD) remains uncertain. METHODS: In this single-center prospective cohort study with a 2-year follow-up, 137 patients aged 51 to 79 with AECOPD were enrolled. We examined the prognostic indicators of AECOPD by analyzing lymphocyte subsets and baseline symptom score. Furthermore, a predictive mode

Immunological reconstitution after autologous stem cell transplantation in patients with refractory systemic autoimmune diseases.

OBJECTIVE: High-dose chemotherapy followed by autologous haematopoietic stem cell transplantation (AHSCT) can be a salvage therapy for patients with severe, refractory systemic autoimmune diseases. The function of the newly rebuilt immune system is important, but little is known about immune reconstitution after AHSCT in autoimmune disorders. Our aim was to investigate the repopulation of different lymphocyte subsets in patients with systemic autoimmune diseases after AHSCT. METHODS: Twelve patients with severe refractory, autoimmune diseases were enrolled in the study: four with rheumatoid a

[Comparison of immunological aspects in diffuse panbronchiolitis and cystic bronchiectasis].

We examined immunological parameters in 35 patients with diffuse panbronchiolitis (DPB) and 20 patients with cystic bronchiectasis (CBE) to analyze the relation between local immune responses and subsequent clinical courses. Patients with DPB showed more impaired cell-mediated immunity than those with CBE. On analysis of peripheral blood lymphocytes from DPB and CBE, the significant increase in CD4 positive T-cells and Fc alpha R positive T-cells and decrease in Leu-11 positive cells were recognized. The observation of humoral immunity revealed a tendency for local immunity to be accelerated s

Distribution of naive and memory T-cells in photoprovoked and spontaneous skin lesions of discoid lupus erythematosus and polymorphous light eruption.

Immune response to ultraviolet radiation-modified skin antigens has been suggested as a pathomechanism of skin lesions in discoid lupus erythematosus and polymorphous light eruption. In order to elucidate the role of T-lymphocyte subsets in this response, we studied the distribution of CD45RO+, CD45RA+ and CD31+ cells and the endothelial expression of adhesion molecules E-selectin/P-selectin, intercellular adhesion molecule-1 and CD31 antigen in photoprovoked and spontaneous skin lesions. Typically, CD45RA+ cells were the prevailing inflammatory cell population of discoid lupus erythematosus, …

Loss of chromosome Y in regulatory T cells.

BACKGROUND: Mosaic loss of chromosome Y (LOY) in leukocytes is the most prevalent somatic aneuploidy in aging humans. Men with LOY have increased risks of all-cause mortality and the major causes of death, including many forms of cancer. It has been suggested that the association between LOY and disease risk depends on what type of leukocyte is affected with Y loss, with prostate cancer patients showing higher levels of LOY in CD4 + T lymphocytes. In previous studies, Y loss has however been observed at relatively low levels in this cell type. This motivated us to investigate whether specific

Immunohistochemical characterization of hepatic lymphocytes in acute hepatitis A, B, and C.

The distinctive histologic findings in acute hepatitis A, B, and C suggest that different immunological mechanisms are involved in the pathogenesis of these diseases. This study was undertaken to define the immune response in each type of acute hepatitis by identification of the intrahepatic lymphocyte subsets. Thirty paraffin-embedded liver biopsy specimens from 10 patients with acute hepatitis A, 10 patients with acute hepatitis B, and 10 patients with acute hepatitis C were evaluated. Immunohistochemical staining was performed by the indirect immunoperoxidase technique using the following m

Lymphocyte Perturbations in Malawian Children with Severe and Uncomplicated Malaria.

Lymphocytes are implicated in immunity and pathogenesis of severe malaria. Since lymphocyte subsets vary with age, assessment of their contribution to different etiologies can be difficult. We immunophenotyped peripheral blood from Malawian children presenting with cerebral malaria, severe malarial anemia, and uncomplicated malaria (n = 113) and healthy aparasitemic children (n = 42) in Blantyre, Malawi, and investigated lymphocyte subset counts, activation, and memory status. Children with cerebral malaria were older than those with severe malarial anemia. We found panlymphopenia in children

Leucocytes in human milk and lymphocyte subsets in cow's milk-allergic infants.

The breast-fed infant ingests an average of 108 leucocytes per day, with breast-feeding often continuing for several months. The precise role of human milk leucocytes is still unresolved. Breast-feeding has been recommended for infants at high risk of allergy to prevent or delay the development of food allergies and atopic eczema. However, studies dealing with distinct immunologic factors in the mother's milk, and their effect on health status or development of allergies in the infant, are scarce. We evaluated the relationship between the cellular composition of human milk and the development

Expression of perforin and cytolytic potential of human peripheral blood lymphocyte subpopulations.

To verify the physiological role of the pore-forming protein perforin in vivo, its expression in subpopulations of human peripheral blood lymphocytes was examined by immunocytochemical staining and their cytolytic potentials compared. In addition to NK cells and gamma delta T cells, which uniformly expressed abundant perforin in their cytoplasmic granules, only a small subpopulation of CD8+ alpha beta T cells contained perforin, namely the CD11b+ subset. However, in vitro activation with an anti-CD3 antibody and IL-2 induced perforin expression in approximately 50% of the CD8+CD11b- T cells an

Effects of a Low Dose of T-2 Toxin on the Percentage of T and B Lymphocytes and Cytokine Secretion in the Porcine Ileal Wall.

Plant materials used in the production of pig feed are frequently contaminated with mycotoxins. T-2 toxin is a secondary metabolite of selected Fusarium species, and it can exert a harmful influence on living organisms. Most mycotoxins enter the body via the gastrointestinal tract, and they can modulate the gut-associated lymphoid tissue (GALT) function. However, little is known about the influence of low T-2 toxin doses on GALT. Therefore, the aim of this study was to evaluate the effect of T-2 toxin administered at 50% of the lowest-observed-adverse-effect level (LOAEL) on the percentage of
