Patho-physiologic findings after chronic tryptophan deficiency in rats: a model for delayed growth and aging.

Long-Evans female rats three weeks, three months and 13-14 months of age were placed on tryptophan-deficient diets for periods ranging from a few months to nearly two years. Growth was interupted during the period of tryptophan-deficiency, but when the animals were returned to a complete diet, they gained weight and grew to normal size. Ability to reproduce, as indicated by litter production, was present at 17-28 months of age in rats which had been deprived of tryptophan, whereas no controls over 17 months of age produced any offspring. Other signs of delayed aging in the experimental group i…

A thin layer chromatographic method for the separation and identification of the tryptophan metabolites present in whole human urine.

This paper reports a new thin layer chromatographic system which is able to separate a total of thirtyeight (38) tryptophan metabolites from a synthetic mixture in a period of three hours. The system consists of thin layers of silica gel GF254 which are eluted in two dimensions, employing in the first dimension a mixture of propanone, 2-propanol, water, ammonia (0.88) (50 : 40 : 7 : 3 V/V) and in the second dimension a mixture of chloroform, glacial acetic acid, methanol and water (65 : 20 : 10 : 5 V/V). A method whereby the separated tryptophan metabolites may be unambiguously identified in s…

The microbiota-tryptophan-brain axis in neurodegenerative diseases: pathogenic mechanisms, disease-specific roles, and translational therapeutics.

The pathogenesis of neurodegenerative diseases (NDDs) such as Alzheimer's disease (AD) and Parkinson's disease (PD) is very complex. Recent studies have shown that gut microbiota and their metabolites play a key role in the progression of these diseases. Tryptophan (Trp) is an essential amino acid, which mainly produces a variety of biologically active compounds in the intestine through the metabolism of indole pathway, Kynurenine pathway (KP) and serotonin pathway, including indole derivatives, Kynurenine (KYN) and serotonin (5-HT). These metabolites affect the central nervous system (CNS) th…

Theabrownin from Fu Brick Tea Ameliorates Obesity via Modulating Tryptophan Metabolism and Downregulating Intestinal Lipid Transporters.

This study aims to explore the underlying mechanism by which theabrownin extracted from Fu brick tea (FBTB) alleviates obesity induced by a high-fat diet (HFD) in mice. Results showed that FBTB intervention ameliorated metabolic disorders, suppressed jejunal lipid accumulation, and enhanced fecal lipid excretion. 16S rRNA gene sequencing demonstrated that FBTB reversed gut microbiota dysbiosis and increased the abundance of potentially beneficial genera such as Dubosiella and Lactobacillus. Metabolomics revealed that FBTB altered tryptophan metabolism and significantly increased cecal levels o…

Exploring the molecular interaction between tryptophan and β-cyclodextrin modified ZnS quantum dots using spectroscopic analysis.

The luminescent properties of water-soluble β-cyclodextrin surface-modified capped ZnS QDs (β-CD/ZnS QDs) were investigated for host-guest interactions. The optical properties of the β-CD/ZnS QDs complex revealed selectivity for L-tryptophan (L-Trp) through aggregation-induced emission. Fluorescence titration and Dynamic Light Scattering analysis revealed the fluorescence enhancement effect and aggregation state of β-CD/ZnS QDs in the presence of L-Trp, respectively. The apparent binding constant was determined 1.75x10-8M-1 determine, along an LOD of 420 nM for L-Trp. UV-Visible, Raman, Fourie…

Proline/serine-rich coiled-coil 1 alleviates atherosclerosis via remodeling tryptophan metabolism mediated by Akkermansia muciniphila.

Genome-wide association studies have implicated proline/serine-rich coiled-coil 1 (PSRC1) in coronary artery disease (CAD) pathogenesis. Our previous studies demonstrated that Psrc1 deficiency accelerates atherosclerosis via gut microbial dysbiosis, characterized by a substantial depletion of Akkermansia muciniphila. Recent studies implicate microbiome-dependent tryptophan metabolism as a novel checkpoint in atherosclerosis, with specific microbial taxa regulating metabolite-driven immune responses. The mechanism by which Psrc1 modulates atherosclerosis through A. muciniphila and its regulatio…

Microbiome-Derived Tryptophan Metabolites Regulate AhR Signaling to Restore Epithelial Barrier Integrity in Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD) is characterized by chronic intestinal inflammation and compromised epithelial barrier integrity. Emerging evidence demonstrates that gut microbiota-derived tryptophan metabolites serve as endogenous ligands for the aryl hydrocarbon receptor (AhR), initiating protective signaling cascades that restore mucosal homeostasis. This review synthesizes current mechanistic insights into how microbial tryptophan catabolites including indole-3-aldehyde, indole-3-propionic acid, indole-3-lactic acid, and indole-3-acetic acid activate AhR to enhance epithelial barrier func…

Burn-Induced Gut Microbiota Dysbiosis Aggravates Skeletal Muscle Atrophy by Tryptophan-Kynurenine Mediated AHR Pathway Activation.

The hypermetabolic response associated with burns is characterized by skeletal muscle atrophy and an increased incidence of disability and death. Significant remodeling of the gut microbiota occurs after severe burn trauma. However, the specific mechanisms by which gut microbiota contribute to burn-induced muscle atrophy remain unexplored. The results showed that the disruption of the gut microbiota exacerbated skeletal muscle atrophy. Fecal metabolite analysis revealed perturbations, primarily within the tryptophan (Trp) metabolic pathway. Animal models further demonstrated that gut microbiot…
