DirectContacts2: a wiring diagram of human physical protein interactions.

Cellular function is driven by the activity of proteins in stable complexes. Protein complex assembly depends on the direct physical association of component proteins. Advances in macromolecular structure prediction with tools like AlphaFold and RoseTTAFold have greatly improved our ability to model these interactions in silico, but an all-by-all analysis of the human proteome's ~200 M possible pairs remains computationally intractable. A comprehensive cellular map of direct protein interactions will therefore be an invaluable resource to direct screening efforts. Here, we present DirectContac

From Jenner to genomics: the unfolding future of vaccinology.

The development of the smallpox vaccine two centuries ago laid the foundation for modern vaccinology. Smallpox, once a major killer in human history, was eradicated globally in 1980. This breakthrough encouraged the development of contemporary vaccine technologies, including inactivated, recombinant, viral vector, and mRNA platforms. Owing to developments in genetics, bioinformatics, and molecular biology, personalised vaccines have been made possible. Advances in the field of immunology facilitated an accelerated response to the COVID-19 pandemic. This perspective synthesises key developments

The contribution of private and public centres to innovation in fertility care: acknowledging the past and preserving the future.

The remarkable advances in the efficacy and safety of IVF treatment that have been achieved since the first IVF baby was born in 1978 stand comparison with any other branch of medicine. These advances have continued despite the withdrawal of public investment in clinical reproductive medicine and research, raising a number of questions. How is this global shift from public to largely private sector care provision affecting the quality and direction of research, innovation and clinical training? This Commentary reviews the implications, both positive and less so, for academic reproductive medic

Noninvasive Coronary Atherosclerotic Plaque Imaging.

Coronary artery disease is the leading cause of morbidity and mortality worldwide. Despite remarkable advances in the management of coronary artery disease, the prediction of adverse coronary events remains challenging. Over the preceding decades, considerable effort has been made to improve risk stratification using noninvasive imaging. Recently, these efforts have increasingly focused on the direct imaging of coronary atherosclerotic plaque. Modern imaging now allows imaging of coronary plaque burden, plaque type, atherosclerotic plaque activity, and plaque thrombosis, which have major poten

When form meets function: the cells and signals that shape the lymphatic vasculature during development.

The lymphatic vasculature is an integral component of the cardiovascular system. It is essential to maintain tissue fluid homeostasis, direct immune cell trafficking and absorb dietary lipids from the digestive tract. Major advances in our understanding of the genetic and cellular events important for constructing the lymphatic vasculature during development have recently been made. These include the identification of novel sources of lymphatic endothelial progenitor cells, the recognition of lymphatic endothelial cell specialisation and heterogeneity, and discovery of novel genes and signalli

Four-dimensional-flow Magnetic Resonance Imaging of the Aortic Valve and Thoracic Aorta.

Blood flow through the heart and great vessels is sensitive to time and multiple velocity directions. The assessment of its three-dimensional nature has been limited. Recent advances in magnetic resonance imaging (MRI) allow the comprehensive visualization and quantification of in vivo flow dynamics using four-dimensional (4D)-flow MRI. In addition, the technique provides the opportunity to obtain advanced hemodynamic measures. This article introduces 4D-flow MRI as it is currently used for blood flow visualization and quantification of cardiac hemodynamic parameters. It discusses its advantag

Prevention of chemotherapy-induced ovarian damage.

Recent advances in our understanding of the mechanisms underlying the impact of cytotoxic drugs on the ovary have opened up new directions for the protection of the ovary from chemotherapy-induced damage. These advances have spurred the investigation of pharmacological agents to prevent ovarian damage at the time of treatment. Prevention of ovarian damage and follicle loss would provide significant advantages over existing fertility preservation techniques. This manuscript reviews new methods for the prevention of chemotherapy-induced ovarian damage, including agents that act on the PI3K/PTEN/

Open reduction and internal fixation of proximal humerus fractures.

Open reduction of proximal humeral fractures has the advantage of providing direct control over each fracture fragment and permitting anatomic reduction and fixation with advanced devices. Modern fixed-angle locking plates designed specifically for proximal humerus fractures have allowed the expansion of surgical indications permitting surgeons to address more complicated fractures. Advanced preoperative imaging and fluoroscopy allow a better understanding of fracture patterns and permit the surgeon to use this knowledge intraoperatively. Research is required to further validate fracture class

CyberKnife robotic radiosurgery system for tumor treatment.

Defined by its high level of accuracy and rapid radiation dose fall-off, radiosurgery has emerged as an effective radiation technique over the past few decades. Although it was once limited to conditions of the brain, head and neck regions, technological advances in computing and imaging have allowed the application of radiosurgery to conditions throughout the entire body. Using advanced imaging and robotics, the CyberKnife (Accuray, Inc., Sunnyvale, CA, USA) is one of few systems capable of delivering radiosurgery with exquisite accuracy to tumors, cancers and other conditions throughout the

Advancing insights into methods for studying environment-health relationships: a multidisciplinary approach to understanding Legionnaires' disease.

Recent years have witnessed important advances in the analysis of spatially referenced health data. This paper uses GIS and point pattern modelling to address previously unanswered questions regarding the spatial epidemiology of Legionnaires' disease. We demonstrate a distance effect for proximity of residence to cooling towers; mixed support for a directional effect; and some evidence relating to multiple sources. In uncovering complex conceptual and technical problems in the spatial modelling of infection risk we also extended the limits of existing point pattern techniques. We advocate furt

Three-dimensional hindfoot motion in adolescents with surgically treated unilateral clubfoot.

Advances in imaging and computerized analyses have enabled three-dimensional bone motion in the treated clubfoot to be measured precisely. Three-dimensional translations and rotations of the talus, calcaneus, navicular, and cuboid of surgically treated clubfeet were less in magnitude and sometimes different in direction (or without motion in a specific plane) compared with the contralateral normal feet. Surgical techniques used for clubfoot treatment do not restore normal hindfoot bone motion when examined with high-resolution magnetic resonance imaging, computer reconstruction, and image anal

Far-advanced otosclerosis. Cochlear implantation vs stapedectomy.

In 1981, our group, as part of a Food and Drug Administration investigator team directed by William House, performed Illinois' first cochlear implantation. We have since performed cochlear implantations in ten other persons. Since May 1980, approximately 35 severely to profoundly deaf persons per year have been referred to our clinic for evaluation. A small number of these patients had far-advanced otosclerosis. In these cases, exploratory tympanotomy and stapedectomy followed by rehabilitation with a hearing aid was a more appropriate management than cochlear implantation. Case histories and

Retrograde endoscopic balloon dilation of chemotherapy- and radiation-induced esophageal stenosis under direct visualization.

INTRODUCTION: Esophageal stricture is a common complication following combined chemotherapy and radiation for advanced oropharyngeal cancer and severely compromises patients' quality of life. The severity of the stenosis after concomitant therapy, combined with the proximal location of these strictures, renders standard bougienage techniques difficult, and the risk of perforation significant. Retrograde endoscopic dilation has recently been described as a safe alternative to rigid endoscopic dilation or unguided bougienage. However, the near complete, or complete, stenosis seen in some of thes

Histone methylation machinery in gliomas: From enzymatic mechanisms to inhibitor development.

Glioma is the most common primary malignant tumor of the central nervous system (CNS). Advances in molecular pathology have established a robust causal link between histone modifications and glioma genesis. In addition to prior findings, recent empirical evidence has unequivocally substantiated the intricate and close relationship between histone modifications and temozolomide (TMZ) resistance, thereby advancing our understanding in this field. This article systematically reviews the roles of histone methylation modifications in glioma pathogenesis and their implications for the mechanisms of

What's New in Imaging for Multiple Myeloma.

Multiple myeloma is a haematological malignancy of plasma cells, typically characterised by a monoclonal immunoglobulin protein detectable in the blood. Myeloma requires treatment when it affects specific end-organs, including lytic bone disease. The accurate detection of bone disease is therefore crucial in the diagnosis and management of myeloma, and advanced imaging with whole-body magnetic resonance imaging or positron emission tomography is now recommended as the standard of care. For patients with non-secretory myeloma and those with extramedullary disease, imaging is the only way to mon

[Immunotherapy: progress and challenges of a revolutionary treatment for gastric cancer].

Gastric cancer is a common malignant tumor, ranking fifth in incidence and mortality among all malignancies globally. The lack of early symptoms or the non-specific nature of symptoms means that most patients are diagnosed at an advanced stage. The unique high heterogeneity of gastric cancer largely limits the effectiveness of traditional therapies, resulting in poor prognosis for patients. In recent years, immunotherapy has emerged as a new treatment option for advanced gastric cancer. The immunotherapy era has driven gastric cancer treatment towards more personalized and precise directions.

New tool in the toolbox: patient selection for zolbetuximab in advanced treatment-naïve gastric/gastroesophageal junction adenocarcinoma.

The treatment landscape for advanced gastroesophageal junction (GEJ) and gastric adenocarcinoma has rapidly evolved over the last decade. The introduction of human epidermal growth factor receptor-2 (HER2)-targeted therapies and immune checkpoint inhibitors in combination with chemotherapy has led to significant improvements in overall survival in biomarker-selected patient populations. The recent Food and Drug Administration approval of zolbetuximab for patients with HER2-negative and Claudin-18 isoform 2 (CLDN18.2)-positive advanced or inoperable GEJ/gastric adenocarcinoma has introduced fur
