Anorexigenic effects of central neuropeptide S involve the hypothalamus in chicks (Gallus gallus).

Neuropeptide S (NPS) affects appetite-related processes in mammals. However, its role in avian biology is unreported. We hypothesized that intracerebroventricular (ICV) NPS would cause anorexigenic effects in chicks (Gallus gallus). To evaluate this, Cobb-500 chicks were centrally injected with multiple doses (0, 0.313, 0.625 and 1.250 mug) of NPS. NPS-treated chicks responded with decreased feed and water intake. The effect on water intake was secondary to feed intake, because fasted NPS-treated chicks did not reduce water intake. ICV NPS injection also reduced plasma corticosterone concentra

Investigating the role of melanocortinergic, glutamatergic and neuropeptide Y systems on hypophagia caused by gastric inhibitory polypeptide (GIP) in broilers.

Appetite control and the metabolic rate of energy expenditure are under the influence of a broad suite of biochemical messengers, which include peptide-based signals, hormonal agents, and neurotransmitters. This research sought to examine the correlation between melanocortinergic, glutamatergic, and neuropeptide Y (NPY) systems with gastric inhibitory polypeptide (GIP) in the modulation of appetite and consummatory behavior. In experiment 1, broilers were injected with GIP at various doses (3, 6 and 12 nmol) in addition to saline. The experimental design for the second trial involved four dist

A systematic and meta-analytic review on the neural correlates of viewing high- and low-calorie foods among normal-weight adults.

In the context of current-day online shopping, people select foods based on pictures and using their visual systems. Although there are some reviews of previous neuroimaging studies on appetitive behaviors, the findings on neural activation in response to pictures of high- and low-calorie foods seem inconsistent. This study aims to systematically review, integrate, and meta-analyze neuroimaging evidence of viewing high- and low-calorie foods. There were 25 samples from 24 studies, totalizing 489 normal-weight participants (311 female, 160 male, and 18 of unknown sex). We conducted a systematic…

The VMAT-2 inhibitor tetrabenazine affects effort-related decision making in a progressive ratio/chow feeding choice task: reversal with antidepressant drugs.

Behavioral activation is a fundamental feature of motivation, and organisms frequently make effort-related decisions based upon evaluations of reinforcement value and response costs. Furthermore, people with major depression and other disorders often show anergia, psychomotor retardation, fatigue, and alterations in effort-related decision making. Tasks measuring effort-based decision making can be used as animal models of the motivational symptoms of depression, and the present studies characterized the effort-related effects of the vesicular monoamine transport (VMAT-2) inhibitor tetrabenazi

Combined stimulation of glucagon-like peptide-1 receptor and inhibition of cannabinoid CB1 receptor act synergistically to reduce food intake and body weight in the rat.

Pharmacological activation of the glucagon-like peptide-1 (GLP-1) receptor and inhibition of the cannabinoid CB1 receptor were found to reduce food intake and body weight in humans and animals. Since earlier studies revealed that endocannabinoids may interact with other neurotransmitters to affect feeding behavior, we have examined whether a stable GLP-1 agonist, exendin-4 and a CB1 receptor antagonist, AM 251, may reciprocally enhance their inhibitory effects on food consumption in the rat. Additionally, we have tested whether the blockade of the GLP-1 receptor by exendin (9-39) modifies AM 2

Maternal Microbiota Transfer Programs Offspring Eating Behavior.

Understanding the link between mother's obesity and regulation of the child's appetite is a prerequisite for the design of successful preventive strategies. Beyond the possible contributions of genetic heritage, family culture, and hormonal and metabolic environment during pregnancy, we investigate in the present paper the causal role of the transmission of the maternal microbiotas in obesity as microbiotas differ between lean and obese mothers, maternal microbiotas are the main determinants of a baby's gut colonization, and the intestinal microbiota resulting from the early colonization could

From hypercortisolism to remission: impact of Cushing's disease on eating patterns.

BACKGROUND: Cushing's disease (CD) is characterized by chronic hypercortisolism and is associated with persistent metabolic, psychological, and neurocognitive disturbances. While metabolic consequences are well described, the impact of disease activity and remission on eating behavior remains insufficiently explored. OBJECTIVE: This study aimed to compare multidimensional eating behavior patterns among patients with active Cushing's disease, patients in biochemical remission, and healthy controls, and to examine their associations with cortisol biomarkers. METHODS: In this cross-sectional st

Alteration, Reduction and Taste Loss: Main Causes and Potential Implications on Dietary Habits.

Our sense of taste arises from the sensory information generated after compounds in the oral cavity and oropharynx activate taste receptor cells situated on taste buds. This produces the perception of sweet, bitter, salty, sour, or umami stimuli, depending on the chemical nature of the tastant. Taste impairments (dysgeusia) are alterations of this normal gustatory functioning that may result in complete taste losses (ageusia), partial reductions (hypogeusia), or over-acuteness of the sense of taste (hypergeusia). Taste impairments are not life-threatening conditions, but they can cause suffici

Central administration of mesotocin inhibits feeding behavior in chicks.

Mesotocin (MT) is a neurohypophysis hormone in non-mammalian vertebrates including chickens, and homologous of oxytocin (OT) in mammals. Oxytocin (OT) is a well known reproductive hormone in mammals, but the physiological roles of MT in chickens have not been clarified well. OT is thought to regulate feeding behavior because central and peripheral injections of OT inhibit feeding behavior in mammals. In avian, on the other hand, the effect of MT on feeding regulation has not yet been clarified. Therefore, the present study was carried out to examine whether MT is related to the regulation of f

Interaction between behavioral and pharmacological treatment strategies to decrease cocaine choice in rhesus monkeys.

Behavioral and pharmacotherapeutic approaches constitute two prominent strategies for treating cocaine dependence. This study investigated interactions between behavioral and pharmacological strategies in a preclinical model of cocaine vs food choice. Six rhesus monkeys, implanted with a chronic indwelling double-lumen venous catheter, initially responded under a concurrent schedule of food delivery (1-g pellets, fixed-ratio (FR) 100 schedule) and cocaine injections (0-0.1 mg/kg/injection, FR 10 schedule) during continuous 7-day treatment periods with saline or the agonist medication phenmetra

Weight loss in obese African American and Caucasian adolescents: secondary analysis of a randomized clinical trial of behavioral therapy plus sibutramine.

BACKGROUND: Obesity, a major risk factor for cardiovascular disease, is more prevalent in African American adolescents than in their Caucasian counterparts. Effective weight loss treatment options are required to aid in the reduction of this health disparity. The purpose of this study, a secondary analysis of data collected as part of a double-blind, randomized clinical trial of behavioral and pharmacologic weight loss, was to examine and compare changes in weight loss and cardiometabolic risk factors in African American and Caucasian adolescents. METHODS: Analysis included 34 African America

Orexigenic/anorexigenic signals in bulimia nervosa.

Bulimia nervosa (BN) and Anorexia Nervosa (AN) are currently classified as eating disorders (ED). Both disorders are the product of complex interaction between physiological and psychological and social processes; they are characterized by abnormal eating behavior. However, patients with BN differ from AN in their nutritional state and response of treatment with serotonin-selective reuptake inhibitor (SSRI) as well as frequency of comorbidity of mood and anxiety disorders. Although biological mechanisms of both BN and AN are largely unknown, excess of both feeding-stimulatory and feeding inhib

Combining behavioral and pharmacological treatments for obesity.

Weight-loss medications are currently recommended for use only as an adjunct to diet, exercise, and behavior modification. Little, however, is known about the benefits of combining behavioral and pharmacological therapies or about the mechanisms that would make these combined approaches more effective than either used alone. This article reviews the effects of adding pharmacotherapy (i.e., principally sibutramine and orlistat) to a modest program of lifestyle modification. Studies revealed that the addition of medication typically improved short- and long-term weight loss compared with lifesty

Add-on combination and maintenance treatment: case series of five obese patients with different eating behavior.

Obesity is a general medical condition associated with an increase in morbidity and mortality. Although it would be desirable to use efficacious prevention programs, the success rates reported to date have been rather disappointing. In this observational study, a new drug treatment regimen was evaluated in five obese patients with a mean age of 39.6 +/- 4.2 years and an initial body mass index between 34.5 and 38.3 kg/m for a period of 96 weeks. The patients showed restrained and unrestrained eating patterns according to a German version of the Three-Factor Eating Questionnaire and were treate

Lessons from obesity management programmes: greater initial weight loss improves long-term maintenance.

It is a common belief that weight loss achieved at a slow rate is better preserved than if the weight is lost more rapidly. However, the literature shows that initial weight loss is positively, not negatively, related to long-term weight maintenance. There is evidence from randomised intervention trials to support that a greater initial weight loss induced without changes in lifestyle (e.g. liquid formula diets or anorectic drugs) improves long-term weight maintenance, providing it is followed by a 1-2 years integrated weight maintenance programme consisting of lifestyle interventions involvin

[Behavioral changes induced by repeated administration of mazindol, an anorexiant, in rats].

Behavioral changes in locomotor activity and stereotyped behaviors induced by daily administration (for 7 days) of mazindol (5 and 10 mg/kg, po) were compared with those induced by methamphetamine (10 mg/kg) in rats. On day 1, mazindol increased locomotor activity, which was enhanced by daily administrations. Stereotyped behaviors were also induced by mazindol, which became more marked following daily administrations. Methamphetamine markedly increased stereotyped behavior rather than locomotor activity on day 1, and the effects were not enhanced by daily administrations. The increased locomot

Effect of the 5-HT2 antagonist ketanserin on salt appetite in the rat.

The 5-HT2 antagonist ketanserin inhibited salt appetite induced by subchronic deoxycorticosterone acetate (DOCA) treatment, as well as salt appetite induced by sodium depletion (which is governed by the synergy of aldosterone and angiotensin in the brain). The effect of ketanserin was more evident following intraperitoneal than intracerebroventricular injection. On the other hand, ketanserin did not inhibit either salt intake induced by intracranial renin injection, or the need-free salt intake of the multidepleted female rat, which is not dependent on the renin-angiotensin-aldosterone system.

Metabolic mechanism of the anorectic and leptogenic effects of the serotonin agonist fenfluramine.

The purpose of these experiments was to investigate the action of a 5HT-agonist, D-fenfluramine (D-FF) upon energy expenditure in addition to its known anorectic action. Experiment 1 showed that body weight (BW) loss was more than predicted by the anorectic action of D-FF. In pair pattern feeding, D-FF induced a similar BW loss in treated and untreated partners despite the sedation of the former and agitation of the latter. Metabolic measurements (oxygen, carbon dioxide, respiratory quotient and locomotor activity (LA] revealed that D-FF enhances mobilization and intense utilization of endogen

1-Deoxy-glucosamine initiates, then effectively suppresses feeding in the rat.

The deoxy analogues of D-glucose, 1-deoxy-D-glucosamine and D-glucosamine, are biochemically and structurally similar to 1-deoxy-D-glucose, so their direct effects on food intake were studied. Both 12 and 24 mumol 1-deoxy-glucosamine potently decreased feeding and body weight after an initial transient elicitation of food intake. The suppression included decreased meal size and prolonged postprandial intermeal interval which persisted for at least 3 days after injection. Ambulatory activity was unaffected. The initial elicitation of feeding was not accompanied by drinking episodes, and subsequ
