Scale-up for bulk production of vaccine against meningococcal disease.

At the Netherlands Vaccine Institute (NVI) a vaccine against Neisseria meningitidis serogroup B organisms based on different porA subtypes contained in outer membrane vesicles (OMVs) is in advanced stage of development and will be evaluated in clinical trial studies in the near future. In order to meet the expected demand for product, the current biopharmaceutical production process is being scaled-up. This study describes the scale-up approach for the upstream process and the resulting bioreactor design and operation strategy leading towards a feasible solution for bulk production of a vaccin

Biofouling control: the impact of biofilm dispersal and membrane flushing.

Pure culture studies have shown that biofilm dispersal can be triggered if the nutrient supply is discontinued by stopping the flow. Stimulating biofilm dispersal in this manner would provide a sustainable manner to control unwanted biofilm growth in industrial settings, for instance on synthetic membranes used to purify water. The response of multispecies biofilms to nutrient limitation has not been thoroughly studied. To assess biomass dispersal during nutrient limitation it is common practise to flush the biofilm after a stop-period. Hence, flow-stop-induced biomass removal could occur as a…

Specific growth rates and growth stoichiometries of Saccharomycotina yeasts on ethanol as sole carbon and energy substrate.

Emerging low-emission production technologies make ethanol an interesting substrate for yeast biotechnology, but information on growth rates and biomass yields of yeasts on ethanol is scarce. Strains of 52 Saccharomycotina yeasts were screened for growth on ethanol. The 21 fastest strains, among which representatives of the Phaffomycetales order were overrepresented, showed specific growth rates in ethanol-grown shake-flask cultures between 0.12 and 0.46 h-1. Seven strains were studied in aerobic, ethanol-limited chemostats (dilution rate 0.10 h-1). Saccharomyces cerevisiae and Kluyveromyces l…

Phosphorus additions imbalance terrestrial ecosystem C:N:P stoichiometry.

Carbon (C):nitrogen (N):phosphorus (P) stoichiometry in plants, soils, and microbial biomass influences productivity and nutrient cycling in terrestrial ecosystems. Anthropogenic inputs of P to ecosystems are increasing; however, our understanding of the impacts of P addition on terrestrial ecosystem C:N:P ratios remains elusive. By conducting a meta-analysis with 1413 paired observations from 121 publications, we showed that P addition significantly decreased plant, soil, and microbial biomass N:P and C:P ratios, but had negligible effects on C:N ratios. The reductions in N:P and C:P ratios b…

Phosphorus Competition in Bioinduced Vivianite Recovery from Wastewater.

Phosphorus undergoes a one-way flow from minerals to soil to water, which creates a phosphorus crisis as well as aquatic eutrophication. Dissimilatory metal reduction bacterial (DMRB)-induced vivianite recovery from wastewater is a promising route to solve these problems synthetically. In this study, phosphorus competition between biomass growth and bioinduced vivianite mineralization was investigated at the batch scale. Biomass growth leads to phosphorus utilization over vivianite mineralization. Geobacter was selected as the main functional microorganism and presented higher vivianite recove…

Could nitrogen compounds be indicators of tolerance to high doses of Cu and Fe in the cultivation of Leucaena leucocephala?

Nitrogen metabolism and the production of primary and secondary metabolites vary according to biotic and abiotic factors such as trace elements (TE) stress, and can, therefore, be considered biomarkers. The present study evaluated the effect of copper (Cu) and iron (Fe) TE, separately, on the metabolism of nitrogen compounds and biomass production, partitioned into shoot and roots of Leucaena leucocephala (Lam.) de Wit., and identified possible defense mechanisms linked to nitrogen metabolism. At 120 days of cultivation, the biomass production of L. leucocephala was higher when exposed to exce…

Effects of nitrogen deposition on soil microbial communities in temperate and subtropical forests in China.

Increasing nitrogen (N) deposition has aroused large concerns because of its potential negative effects on forest ecosystems. Although microorganisms play a vital role in ecosystem carbon (C) and nutrient cycling, the effect of N deposition on soil microbiota still remains unclear. In this study, we investigated the responses of microbial biomass C (MBC) and N (MBN) and microbial community composition to 4-5years of experimentally simulated N deposition in temperate needle-leaf forests and subtropical evergreen broadleaf forests in eastern China, using chloroform fumigation extraction and phos…

Acorus calamus recycled as an additional carbon source in a microbial fuel cell-constructed wetland for enhanced nitrogen removal.

Acorus calamus was recycled as an additional carbon source in microbial fuel cell-constructed wetlands (MFC-CWs), for efficient nitrogen removal of low carbon wastewater. The pretreatment methods, adding positions, and nitrogen transformations were investigated. Results indicated that alkali-pretreatment cleaved the benzene rings in dominant released organics, producing chemical oxygen demand of 164.5 mg from per gram of A. calamus. Pretreated biomass addition in the anode of MFC-CW attained the maximum total nitrogen removal of 97.6% and power generation of 12.5 mW/m2, which were higher than …

Application and reactivation of magnetic nanoparticles in Microcystis aeruginosa harvesting.

This study developed a magnetic nanoparticles (MNPs) harvesting and reactivation technique for rapid cyanobacteria Microcystis aeruginosa separation. The harvesting of raw MNPs achieved high efficiency of 99.6% with the MNPs dosage of 0.58g MNPs/g dry-biomass, but gradually decreased to 59.1% when directly reused 5 times. With extra ultrasonic chloroform:methanol solvent treatment, the MNPs can be effectively reactivated for M. aeruginosa harvesting with 60% efficiency after 5 times reactivation and the separation efficiency kept above 93% with 0.20g MNPs/g dry-biomass dosage. The cyanobacteri…

Waste yeast of the genus Saccharomyces from the beer, wine, and sugarcane ethanol industries: valorization pathways and perspectives for the production of food ingredients.

Yeast plays a central role in several industrial fermentation processes, including large-scale beer, wine, and ethanol production. Although widely recognized for its fermentative efficiency and robustness, its residual biomass-often discarded or underutilized-represents a rich source of valuable compounds. These include cell wall components such as β-glucans and mannans, which exhibit significant nutritional, functional, and technological properties. The expansion of these fermentation processes has resulted in an abundance of excreted yeast biomass, much of which is used in animal feed or dis…

Projecting Impacts of Marine Heatwaves and Their Effects on Long-Term Changes in Marine Ecosystems Over the 21st Century.

Marine heatwaves (MHWs) are increasing in frequency, duration and intensity under climate change. Although current MHWs trigger broad ecological impacts, such as coral bleaching, mass mortality of seagrass meadows, and declines in important fish stocks, the cascading effects of MHWs on ecosystem structure and functions in the 21st century remain unclear. In this study, we utilise a global biomass flow modelling framework to analyse these impacts. Leveraging climate projections from two Earth system models (ESMs), we project marine food webs dynamics at a spatial resolution of 1° latitude and l…

Application of the brown macroalga Saccharina latissima (Laminariales, Phaeophyceae) as a feed ingredient for livestock: A review.

In recent years, marine macroalgae have been recognized as potential alternative and sustainable feeding resources for livestock. Differences in nutritional values and biomass yield across macroalgal species are critical factors while aiming to utilize them as animal feed components. A brown macroalga, Saccharina latissima, also known as sugar kelp, has a promising biomass yield and high nutritional and bioactive compounds that can benefit both ruminant and monogastric animals. For example, the dietary inclusion of S. latissima in dairy and beef cattle can enhance milk yield, meat quality, and…

The prospect of bamboo and non-fodder rice husk for sustainable bioethanol production.

Due to the rapid industrialization globally, there is a constant increase in demand for energy, which drives up fuel prices and contributes to the depletion of fossil fuels. The rise in the use of fossil fuels results in increasing greenhouse gas emissions contributing to biodiversity loss. Thus, the development of alternative green biofuel (e.g., bioethanol) from renewable and surplus biomass resources has taken center stage as our attention is drawn to environmental concerns and energy security. However, till now, the studies pertaining to the process optimization and techno-economic analysi…

Solubility of toluene, benzene and TCE in high-microbial concentration systems.

We report measurements of solubility limits for benzene, toluene, and TCE in systems that contain varying levels of biomass up to 0.13 g mL(-1) for TCE and 0.25 g mL(-1) for benzene and toluene. The solubility limit increased from 21 to 48 mM when biomass (in the form of yeast) was added to aqueous batch systems containing benzene. The toluene solubility limit increased from 4.9 to greater than 20mM. For TCE, the solubility increased from 8mM to more than 1000 mM. Solubility for TCE (trichloroethylene) was most heavily impacted by biomass levels, changing by two orders of magnitude as the micr…
