Phosphate metabolism and renal calcium stone disease.

The purpose of the present investigation was to study various aspects of phosphate metabolism in renal calcium stone patients with special reference to the renal handling of phosphate and its relationship to other renal tubular functions and calcium metabolism. Serum phosphate and the capacity for renal tubular reabsorption of phosphate were lower in stone patients than in controls and decreased with advancing age. Reduced tubular phosphate reabsorption was particularly evident in stone patients with other tubular dysfunctions. Absorptive hypercalciuria was common, but unrelated to the renal t…

Urate metabolism and renal calcium stone disease.

Both hyperuricaemia and hyperuricosuria have been considered to be common in renal calcium stone disease. Abnormalities in serum or urinary urate was not more common in 450 investigated patients than in 90 healthy controls, but when present it was often associated with a more complicated course of the disease, particularly in combination with renal acidification defects. Hyperuricosuria was not explained by a renal tubular dysfunction of urate transport. A decreased urate clearance in proximal renal acidification dysfunction was associated with a high tubular reabsorption of filtered urate. So…

Spinal calcium changes with 1alpha-hydroxyvitamin D3.

During treatment of renal osteodystrophy with 1alpha-hydroxyvitamin D3 in eleven patients, regional changes in the skeleton have been compared with long-term calcium balance as assessed by whole-body calcium. Radial bone changes did not correlated well with calcium balance, but spinal calcium changes were of a similar magnitude when changes were large. Bone alkaline phosphatase changes correlated well with changes in spinal calcium, but less well with changes in radial bone density.

Bovine factor v: a calcium-containing metalloprotein.

Although coagulation factor Xa requires Ca2+ for binding to phospholipid, factor V, the other protein component in the prothrombinase enzyme complex, binds tightly to phospholipid in the absence of Ca2+. To explore the possibility that calcium might be present in the fact V molecule, the effect of several chelators, including oxalate, citrate, pyrophosphate, and EDTA, on factor V activity has been studied. A time- and concentration-dependent inhibition of factor V which reflects the respective association constants for calcium of each chelator is observed. The inhibition can be prevented by th…

Calcium pyrophosphate dihydrate deposition disease without chondrocalcinosis.

The pseudogout syndrome is usually associated with radiographic evidence of articular cartilage calcification. Eight patients who had joints containing calcium pyrophosphate dihydrate crystals were studied. Extensive radiographic evaluation was obtained in seven patients and a limited evaluation in the other. None had evidence of chondrocalcinosis. Six had distinctive radiographic abnormalities of the wrists consisting of radiocarpal joint space narrowing and sclerosis, and subchondral cystic degeneration of the carpal bones. We conclude that calcium pyrophosphate dihydrate deposition disease …

Apparent digestibility and calcium and phosphorus in urine after feeding different combinations of calcium and phosphorus sources to adult dogs.

The present study aimed to investigate the effect of the combination of a water-soluble calcium (Ca) source (CaCl2) with a water-soluble phosphorus (P) source (NaH2PO4*2H2O, diet soluble, SOL) in comparison to a water-insoluble P source (CaHPO4*2H2O, diet insoluble, INS) on apparent digestibility and renal excretion of Ca and P in dogs. The Ca intake was 226 mg/kg bodyweight (bw), the Ca/P ratio 1.9/1 in SOL and 2.0/1 in INS. The percentage of Ca from CaCl2 was 60% in SOL and 33% in INS. Eight adult Foxhound-crossbred dogs FBI, (3-5 years, bw 24-32 kg) were available. Standard digestion trials…

ER calcium depletion as a key driver for impaired ER-to-mitochondria calcium transfer and mitochondrial dysfunction in Wolfram syndrome.

Wolfram syndrome is a rare genetic disease caused by mutations in the WFS1 or CISD2 gene. A primary defect in Wolfram syndrome involves poor ER Ca2+ handling, but how this disturbance leads to the disease is not known. The current study, performed in primary neurons, the most affected and disease-relevant cells, involving both Wolfram syndrome genes, explains how the disturbed ER Ca2+ handling compromises mitochondrial function and affects neuronal health. Loss of ER Ca2+ content and impaired ER-mitochondrial contact sites in the WFS1- or CISD2-deficient neurons is associated with lower IP3R-m

Effects of limestone particle size, phytate, calcium source, and phytase on standardized ileal calcium and phosphorus digestibility in broilers.

Work was carried out to determine the effects of limestone (LM) geometric mean diameter (GMD), phytate, Ca source, and phytase on standardized ileal digestibility (SID) of Ca and P in broilers. Twelve treatments (TRT) were tested. One basal corn and corn germ-based diet was prepared without adding inorganic Ca or P (TRT1, 0.02% Ca). Limestone from the same source (800 or 151 μm GMD) and bone (151 μm GMD) were added to TRT 2-4, (0.7% Ca). TRT5 was a corn/soybean meal (SBM) diet with 800 μm GMD LM (0.77% Ca). Buttiauxella sp. phytase was added (1,000 U/kg) to TRT 1-5 to prepare TRT 6-10, respect

Comparative study of calcium and calcium-related enzymes with differentiation markers in different ages and muscle types in mdx mice.

Sarcolemma instability and increased calcium influx in muscle fibers are characteristics of the Duchenne muscular dystrophy. Excessive calcium activates calcium-dependent enzymes, such as calpains (CAPN) and matrix metalloproteases (MMP). Here, we analyzed calcium deposits, the activity of CAPN and MMP and the expression of Myh, SERCA and myogenic regulatory factors in different skeletal muscles during myonecrosis (4-weeks) and regeneration (12-weeks) phases of the mdx muscular pathology. Alizarin red staining was used to assess calcium deposits, casein and gelatin zymography were performed to…

Role of Extracellular Calcium and Calcium Sensitization in the Anti-Contractile Effect of Perivascular Adipose Tissue in Pregnant Rat Aorta.

Previous studies have shown that the anti-contractile effect of the perivascular adipose tissue (PVAT) is attenuated in pregnancy. In the present investigation, we have examined the possibility that this loss of anti-contractile effect could be due to changes in calcium mobilization. PVAT exerted anti-contractile effect against 5-hydroxytryptamine (5-HT)-induced contractions of aorta segments from pregnant and non-pregnant rats and this anti-contractile effect was attenuated in segments from pregnant rats. Nifedipine (10-6 mol/L), an inhibitor of L-type dihydropyridine calcium channels, signif…

Efficacy of calcium excretion and calcium/creatinine clearance ratio in the differential diagnosis of familial hypocalciuric hypercalcemia and primary hyperparathyroidism.

BACKGROUND: Twenty-four-hour renal calcium-excretion (CE) and calcium/creatinine-clearance-ratio (CCCR), respectively, are widely used to rule out familial hypocalciuric hypercalcemia (FHH) in patients with suspected primary hyperparathyroidism before surgery. The aim was to evaluate the practicability of CE compared to CCCR. PATIENTS AND METHODS: We analyzed biochemical parameters, surgical treatment, gene mutation results, and long-term follow-up data of 198 patients (including 14 patients with FHH) and the discriminative power of CE and CCCR. RESULTS: Twenty four patients (12.1%) had a lo…

Endurance swimming stimulates transepithelial calcium transport and alters the expression of genes related to calcium absorption in the intestine of rats.

Endurance impact exercise, e.g., running, is known to enhance the intestinal calcium absorption. However, nonimpact exercise, e.g., swimming, is more appropriate for osteoporotic patients with cardiovascular diseases or disorders of bone and joint, but the effect of swimming on the intestinal calcium transport was unknown. This study, therefore, aimed to investigate the transepithelial calcium transport and the expression of related genes in the intestine of rats trained to swim nonstop 1 h/day, 5 days/wk for 2 wk. We found that endurance swimming stimulated calcium transport in the duodenum, …

Reevaluation of the normal epidermal calcium gradient, and analysis of calcium levels and ATP receptors in Hailey-Hailey and Darier epidermis.

Electron probe microanalysis was used to analyze elemental content of human epidermis. The results revealed that the calcium content of the basal keratinocyte layer was higher than that of the lowest spinous cell layer in normal epidermis. This was surprising, as it is generally accepted that the calcium level increases with cellular differentiation from the proliferative basal layer to the stratum corneum. Hailey-Hailey disease (HHD) and Darier disease (DD) are caused by mutations in Ca(2+)-ATPases with the end result of desmosomal disruption and suprabasal acantholysis. The results demonstra…
