Short antisense-locked nucleic acids (all-LNAs) correct alternative splicing abnormalities in myotonic dystrophy.

Myotonic dystrophy type 1 (DM1) is an autosomal dominant multisystemic disorder caused by expansion of CTG triplet repeats in 3'-untranslated region of DMPK gene. The pathomechanism of DM1 is driven by accumulation of toxic transcripts containing expanded CUG repeats (CUG(exp)) in nuclear foci which sequester several factors regulating RNA metabolism, such as Muscleblind-like proteins (MBNLs). In this work, we utilized very short chemically modified antisense oligonucleotides composed exclusively of locked nucleic acids (all-LNAs) complementary to CUG repeats, as potential therapeutic agents a

Glycosphingolipid modification: structural diversity, functional and mechanistic integration of diabetes.

Glycosphingolipids (GSLs) are present in all mammalian cell plasma membranes and intracellular membrane structures. They are especially concentrated in plasma membrane lipid domains that are specialized for cell signaling. Plasma membranes have typical structures called rafts and caveola domain structures, with large amounts of sphingolipids, cholesterol, and sphingomyelin. GSLs are usually observed in many organs ubiquitously. However, GSLs, including over 400 derivatives, participate in diverse cellular functions. Several studies indicate that GSLs might have an effect on signal transduction

Reversibility of heterotopic proliferative glands in glandular stomach of Helicobacter pylori-infected Mongolian gerbils on eradication.

Helicobacter pylori (Hp) infection is an important factor in human gastric disorders. Mongolian gerbils can be easily infected with Hp and represent excellent experimental models to clarify the role of Hp in chronic active gastritis, peptic ulcers, intestinal metaplasia, and gastric carcinoma. We have proved the enhancing effects of Hp infection on all histological types of gastric cancers in Mongolian gerbils exposed to chemical carcinogens. Heterotopic proliferative glands (HPGs) also frequently develop with Hp infection in the glandular stomach of infected gerbils, with a slightly dysplasti

Sports-related and other soft-tissue injuries, tendinitis, bursitis, and occupation-related syndromes.

In this review, four areas are discussed: fluoroquinolone-induced tendinitis, volar flexor tenosynovitis (trigger finger), Achilles tendon lesions, and occupational medicine issues. The relationship of fluoroquinolone treatment to musculoskeletal lesions, especially Achilles tendinitis and tear, is most intriguing. The steady increase in reports of the association cannot be ignored. Although Achilles tendinitis and rupture have comprised the most frequently seen lesions, articles on additional sites of involvement, such as in lateral epicondylitis and De Quervain's tenosynovitis, are reviewed.

Current concepts of hyperuricemia and gout.

Recent studies have confirmed that gout is an inborn error of metabolism. It has now become evident that the hyperuricemia associated with gout might occur either due to overproduction of uric acid, underexcretion of uric acid or a combination of these processes. Furthermore, patients with excessive purine synthesis may have a specific enzyme defect resulting in altered feedback inhibition of purine synthesis. A neurological disease manifest by mental retardation, choreo-athetosis, aggressive behavior, lip-biting and self-mutilation and associated with decidedly increased purine biosynthesis s

Acteoside-containing caffeic acid is bioactive functional group of antifibrotic effect by suppressing inflammation via inhibiting AHR nuclear translocation in chronic kidney disease.

Chronic kidney disease (CKD) is a common disorder with increasing prevalence and morbidity worldwide. However, the present agents do not effectively inhibit pathological processes. Aryl hydrocarbon receptor (AHR) is a ligand-activated transcription factor that mediated various diseases. Acteoside (ATS) is a commercial ATP-competitive inhibitor of protein kinase C. Clinical study showed ATS mitigated Th22 cell disorder and proteinuria in patients with immunoglobulin A nephropathy. This study analyzed AHR and nuclear factor kappa B (NF-κB) p65 expression in CKD patients. We compared the effects

Design, synthesis, and pharmacological evaluation of novel PROTAC degraders targeting 11β-HSD1 for metabolic disease intervention.

Hydroxysteroid 11-beta dehydrogenase 1 (11β-HSD1) plays a critical role in metabolic homeostasis by catalyzing the intracellular conversion of cortisone to cortisol. Dysregulated 11β-HSD1 activity is closely associated with metabolic disorders such as type 2 diabetes mellitus, obesity, and glucocorticoid-related inflammation. While small-molecule inhibitors of 11β-HSD1 have shown promise, they primarily suppress enzymatic activity without modulating protein abundance. Here, we report the development of the 11β-HSD1-targeting PROTAC degraders. A series of bifunctional molecules were synthesized

MEK1/2 inhibitors suppress pathological α-synuclein and neurotoxicity in cell models and a humanized mouse model of Parkinson's disease.

The abnormal accumulation of misfolded proteins is a common hallmark of many neurodegenerative disorders. Among these proteins, α-synuclein (αsyn) is a well-characterized pathogenic protein in Parkinson's disease (PD) and other synucleinopathies. αsyn can be hyperphosphorylated and form pathological aggregates, leading to neurodegeneration. Thus, chemical modulators of pathological αsyn may suppress its downstream toxicity and provide entry points to therapeutic intervention. Here, we identified mitogen-activated protein kinase kinase 1/2 (MEK1/2) inhibitors as negative modulators of basal αsy

Cell-Based Small-Molecule Screening Identifying Proteostasis Regulators Enhancing Factor VIII Missense Mutant Secretion.

Missense mutations are the most prevalent alterations in genetic disorders such as hemophilia A (HA), which results from coagulation factor VIII (FVIII) deficiencies. These mutations disrupt protein biosynthesis, folding, secretion, and function. Current treatments for HA are extremely expensive and inconvenient for patients. Small molecule drugs offer a promising alternative or adjunctive strategy due to their lower cost and ease of administration, enhancing accessibility and patient compliance. By screening drug/chemical libraries with cells stably expressing FVIII-Gaussia luciferase fusion

Predictive Analysis and Activity Validation of Quality Markers in Erchen Pill.

The ErChen Pill (ECP), a classical traditional Chinese medicine (TCM), is clinically effective in treating dampness-phlegm-related disorders, including cough, nausea, vomiting, and excessive phlegm production. However, the complexity of the chemical composition of traditional Chinese medicine poses challenges to controlling the quality of ECP. This study aimed to predict and validate the quality markers (Q-markers) of ECP. First, we performed high-performance liquid chromatography (HPLC) to establish fingerprints of 26 batches of ECP and identify common peaks. Subsequently, quality-differentia

Gastrointestinal Bleeding in Hereditary Hemorrhagic Telangiectasia Accompanied with Very Severe Thrombocytopenia.

BACKGROUND Hereditary hemorrhagic telangiectasia (HHT) is a rare autosomal-dominant disorder characterized by recurrent epistaxis and gastrointestinal bleeding (GIB). To our knowledge, the management of GIB in HHT patients accompanied with very severe thrombocytopenia has not been previously reported. CASE REPORT A 66-year-old woman with HHT who experienced recurrent epistaxis for over 5 years was admitted to the hospital due to intermittent hematemesis and melena for 2 weeks. Following admission, her platelet count dropped significantly to 1×10⁹/L. Following supportive treatments, a gastrosco

Bisphenol A and Di-n-butyl phthalate disrupt bone homeostasis bidirectionally via CD36-mediated BMSCs autophagy inhibition and exosome-promoted osteoclastogenesis.

Bisphenol A (BPA) and di-n-butyl phthalate (DBP) are ubiquitous endocrine disruptors implicated in bone metabolism disorders, but their precise mechanisms remain unclear. Here, we demonstrated that BPA and DBP bidirectionally disrupt bone homeostasis by targeting CD36 in bone marrow-derived mesenchymal stem cells (BMSCs). Mechanistically, both chemicals upregulate CD36 expression, which sequesters ATG9a at the Golgi apparatus, inhibits autophagosome maturation, and thereby impairs osteogenic differentiation of BMSCs, as evidenced by reduced ALP and RUNX-2 levels. Exosomes from BPA-treated BMSC

An Overview of Dietary Flavonoids as a Nutraceutical Nanoformulation Approach to Life-threatening Diseases.

Obesity, heart disease, diabetes, and cancer are just a few chronic diseases for which proper nutrition has been demonstrated to be a crucial factor in prevention. Flavonoids, found in many fruits and vegetables, are a type of dietary polyphenol with potent antioxidant activity and anti-carcinogenic characteristics. Flavonoids and foods rich in flavonoids have been shown in recent years to have profound effects on cognitive function, aging, and neurodegenerative diseases like Alzheimer's. Foods high in flavonoids, such as green tea, chocolate, blueberries, and other fruits and vegetables, have

COVID-19 and severity of liver diseases: Possible crosstalk and clinical implications.

COVID-19-infected individuals and those who recovered from the infection have been demonstrated to have elevated liver enzymes or abnormal liver biochemistries, particularly with preexisting liver diseases, liver metabolic disorders, viral hepatitis, and other hepatic comorbidities. However, possible crosstalk and intricate interplay between COVID-19 and liver disease severity are still elusive, and the available data are murky and confined. Similarly, the syndemic of other blood-borne infectious diseases, chemical-induced liver injuries, and chronic hepatic diseases continued to take lives wh

Quantitative risk assessment of allergens leaching from menstrual hygiene products.

Allergic contact dermatitis (ACD) often associated with the topical use of perfumed products, remains one of the most common chronic skin disorders in Western countries. Since labelling of scented menstrual hygiene products (MHPs) is not mandatory, women might be unknowingly exposed to allergens. Given that vaginal mucosae lack the vital barrier function of the skin, skin allergens can easily penetrate and become systemically available and hence women may experience adverse effects in the anogenital region. The aim of this study was therefore to investigate whether women using scented MHPs are

A new series of small molecular weight compounds induce read through of all three types of nonsense mutations in the ATM gene.

Chemical-induced read through of premature stop codons might be exploited as a potential treatment strategy for genetic disorders caused by nonsense mutations. Despite the promise of this approach, only a few read-through compounds (RTCs) have been discovered to date. These include aminoglycosides (e.g., gentamicin and G418) and nonaminoglycosides (e.g., PTC124 and RTC13). The therapeutic benefits of these RTCs remain to be determined. In an effort to find new RTCs, we screened an additional ~36,000 small molecular weight compounds using a high-throughput screening (HTS) assay that we had prev
