A deep architecture based on attention mechanisms for effective end-to-end detection of early and mature malaria parasites in a realistic scenario.

BACKGROUND: Malaria is a critical and potentially fatal disease caused by the Plasmodium parasite and is responsible for more than 600,000 deaths globally. Early and accurate detection of malaria parasites is crucial for effective treatment, yet conventional microscopy faces limitations in variability and efficiency. METHODS: We propose a novel computer-aided detection framework based on deep learning and attention mechanisms, extending the YOLO-SPAM and YOLO-PAM models. Our approach facilitates the detection and classification of malaria parasites across all infection stages and supports mul

A Case of Lacunar Stroke in a Patient with Possible Normal Pressure Hydrocephalus (NPH).

BACKGROUND: Normal pressure hydrocephalus (NPH) is a condition seen in the elderly, characterized by gait disturbances, urinary incontinence, and cognitive impairment. However, sudden onset neurological deficits suggest a vascular event rather than NPH. METHODOLOGY: We report a case of a 65-year-old male who presented with sudden onset gait difficulty and speech impairment, initially suspected to be idiopathic NPH. The diagnostic process and management strategies are discussed. RESULTS: The patient's symptoms, including insidious gait difficulties, speech impairment, and memory loss, led to

Automated fibril structure calculations in Xplor-NIH.

Amyloid fibrils are protein assemblies that are pathologically linked to neurodegenerative diseases. Fibril structures can aid development of highly specific ligands for diagnostic imaging and therapeutics. Solid-state NMR (SSNMR) is a viable approach to solving fibril structures; however, most SSNMR protocols require manual analysis of extensive spectral data, presenting a major bottleneck to determining structures. Standard automation; routines fall short for symmetric multimeric assemblies like amyloids due to high cross peak degeneracy and the need to account for multiple protein subunits.

Computational Strategies for Assessing Adverse Outcome Pathways: Hepatic Steatosis as a Case Study.

The evolving landscape of chemical risk assessment is increasingly focused on developing tiered, mechanistically driven approaches that avoid the use of animal experiments. In this context, adverse outcome pathways have gained importance for evaluating various types of chemical-induced toxicity. Using hepatic steatosis as a case study, this review explores the use of diverse computational techniques, such as structure-activity relationship models, quantitative structure-activity relationship models, read-across methods, omics data analysis, and structure-based approaches to fill data gaps with

Neural Networks Based on Synthesized Training Data for the Automated Detection of Chemical Plumes in Passive Infrared Multispectral Images.

Automated detection of volatile organic compounds in the atmosphere can be achieved by applying pattern recognition analysis to passive infrared (IR) multispectral remote sensing data. However, obtaining analyte-active training data through field experiments is time-consuming and expensive. To address this issue, methodology has been developed for simulating radiance profiles acquired using a multispectral IR line-scanner mounted in a downward-looking position on a fixed-wing aircraft. The simulation strategy used Planck's radiation law and a radiometric model along with the laboratory spectru

Intensity-Borrowing Mechanisms Pertinent to Laser Cooling of Linear Polyatomic Molecules.

A study of the intensity-borrowing mechanisms important to optical cycling transitions in laser-coolable polyatomic molecules arising from non-adiabatic coupling, contributions beyond the Franck-Condon approximation, and Fermi resonances is reported. It has been shown to be necessary to include non-adiabatic coupling to obtain computational accuracy that is sufficient to be useful for laser cooling of molecules. The predicted vibronic branching ratios using perturbation theory based on the non-adiabatic mechanisms have been demonstrated to agree well with those obtained from variational discre

Revisiting mandibular symphyseal shape in juvenile early hominins and modern humans using a deformation-based approach.

The juvenile mandible is important in the investigation of ontogenetic and evolutionary changes among early hominins. We revisit the mandibular symphysis in juvenile specimens of Australopithecus africanus and Paranthropus robustus with two main contributions. First, we employ, for the first time, methods of computational anatomy to model complex symphyseal shape differences. Second, we present new fossil evidence from Kromdraai to improve our knowledge of symphyseal morphology. We describe differences between shapes by landmark-free diffeomorphism needed to align them. We assess which feature

Bus network decomposition for fault detection and isolation through power line communication.

This paper deals with fault detection and isolation (FDI) in complex embedded wired communication networks. The considered faults are soft faults which do not prevent the communication, but may evolve into hard faults, i.e. short or open circuit. A novel FDI method based on power line communication (PLC) transmission systems is proposed. In these PLC systems, the transmission coefficients between the source and each receiver are estimated for communication purposes using orthogonal frequency division multiplexing (OFDM). Health indicators and residuals are computed by comparing the online esti

Immunoinformatics Approaches in Designing Vaccines Against COVID-19.

Since the onset of the COVID-19 pandemic, a number of approaches have been adopted by the scientific communities for developing efficient vaccine candidate against SARS-CoV-2. Conventional approaches of developing a vaccine require a long time and a series of trials and errors which indeed limit the feasibility of such approaches for developing a dependable vaccine in an emergency situation like the COVID-19 pandemic. Hitherto, most of the available vaccines have been developed against a particular antigen of SARS-CoV, spike protein in most of the cases, and intriguingly, these vaccines are no

Oxidative Cross Dehydrogenative Coupling of N-Heterocycles with Aldehydes through C(sp3)-H Functionalization.

Existing methodologies for metal-catalyzed cross-couplings typically rely on preinstallation of reactive functional groups on both reaction partners. In contrast, C-H functionalization approaches offer promise in simplification of the requisite substrates; however, challenges from low reactivity and similar reactivity of various C-H bonds introduce considerable complexity. Herein, the oxidative cross dehydrogenative coupling of α-amino C(sp3)-H bonds and aldehydes to produce ketone derivatives is described using an unusual reaction medium that incorporates the simultaneous use of di-tert-butyl

A Comparison of Vessel Patch Materials in Tetralogy of Fallot Patients Using Virtual Surgery Techniques.

Tetralogy of Fallot (ToF) is characterized by stenosis causing partial obstruction of the right ventricular outflow tract, typically alleviated through the surgical application of a vessel patch made from a biocompatible material. In this study, we use computational simulations to compare the mechanical performance of four patch materials for various stenosis locations. Nine idealized pre-operative ToF geometries were created by imposing symmetrical stenoses on each of three anatomical sub-regions of the pulmonary arteries of three patients with previously repaired ToF. A virtual surgery metho

[Calcific tendinitis].

CLINICAL ISSUE: Calcific tendinitis (TC) is a common-usually self-limiting-musculoskeletal disease, histopathologically characterized by both deposition and subsequent inflammatory breakdown of calcium crystals in tendons. The disease can cause acute, sometimes excruciating pain and restricted movement in the shoulder joint. Furthermore, 10-30% of patients have a complicated course of the disease. STANDARD RADIOLOGICAL METHODS: Imaging-based assessment by X‑ray and ultrasound is required to establish the initial diagnosis and differential diagnosis as well as for follow-up. METHODOLOGICAL IN

The Use of Data Collection and Big Data Analysis in Neurodegenerative Disease Prevention.

Data collection and analysis is becoming more and more important in our everyday lives. It is widely believed that with the power of data collection, almost every problem can be given a possible solution. Machine learning and artificial intelligence algorithms along with the use of the Alzheimer's Disease Neuroimaging Initiative (ADNI) database that collects data from subjects suffering from Alzheimer's disease (AD) can be utilised for recognising biomarkers' patterns that suggest degenerative behavior. Thus, allowing for early prevention. Computers are able to distinguish patterns over millio

Consciousness Detection in a Complete Locked-in Syndrome Patient through Multiscale Approach Analysis.

Completely locked-in state (CLIS) patients are unable to speak and have lost all muscle movement. From the external view, the internal brain activity of such patients cannot be easily perceived, but CLIS patients are considered to still be conscious and cognitively active. Detecting the current state of consciousness of CLIS patients is non-trivial, and it is difficult to ascertain whether CLIS patients are conscious or not. Thus, it is important to find alternative ways to re-establish communication with these patients during periods of awareness, and one such alternative is through a brain-c

Computational Tool to Study Perturbations in Muscle Regulation and Its Application to Heart Disease.

Striated muscle contraction occurs when myosin thick filaments bind to thin filaments in the sarcomere and generate pulling forces. This process is regulated by calcium, and it can be perturbed by pathological conditions (e.g., myopathies), physiological adaptations (e.g., β-adrenergic stimulation), and pharmacological interventions. Therefore, it is important to have a methodology to robustly determine the impact of these perturbations and statistically evaluate their effects. Here, we present an approach to measure the equilibrium constants that govern muscle activation, estimate uncertainty

Accidental awareness while under general anaesthesia.

Accidental awareness during general anaesthesia may cause many intraoperative discomforts and bring further moderate to severe long-term symptoms including flashbacks, nightmares, hyperarousal or post-traumatic stress disorder. The incidence of awareness varied from 0.017% to 4% among studies. The relatively reliable incidence of intraoperative awareness with postoperative recall is 0.02%. The reason causing awareness was unclear. Insufficient anaesthetic dosing was thought as the principal cause. Even awareness was not comprehensively understood, some endeavors have been raised to prevent or

Cardiovascular models for personalised medicine: Where now and where next?

The aim of this position paper is to provide a brief overview of the current status of cardiovascular modelling and of the processes required and some of the challenges to be addressed to see wider exploitation in both personal health management and clinical practice. In most branches of engineering the concept of the digital twin, informed by extensive and continuous monitoring and coupled with robust data assimilation and simulation techniques, is gaining traction: the Gartner Group listed it as one of the top ten digital trends in 2018. The cardiovascular modelling community is starting to

Cyclic Peptide Design Guided by Residual Dipolar Couplings, J-Couplings, and Intramolecular Hydrogen Bond Analysis.

Cyclic peptides have long tantalized drug designers with their potential ability to combine the best attributes of antibodies and small molecules. An ideal cyclic peptide drug candidate would be able to recognize a protein surface like an antibody while achieving the oral bioavailability of a small molecule. It has been hypothesized that such cyclic peptides balance permeability and solubility using their solvent-dependent conformational flexibility. Herein we report a conformational deconvolution NMR methodology that combines residual dipolar couplings, J-couplings, and intramolecular hydroge
