Two new species of Prosorhynchoides (Digenea: Bucephalidae) from Tylosurus crocodilus (Belonidae) from the great barrier reef and French Polynesia.

We surveyed 14 individuals of Tylosurus crocodilus Péron & Lesueur 1821 (Belonidae) collected from the waters around Lizard Island and Heron Island, Great Barrier Reef, Queensland, Australia, and the waters around Moorea, French Polynesia. We describe two new species of bucephaline trematodes from them, Prosorhynchoides galaktionovi n. sp. and P. kohnae n. sp. They are morphologically distinct from existing Prosorhynchoides spp., with molecular data from 28S and ITS-2 ribosomal DNA, as well as cox1 mitochondrial DNA, further supporting our morphological findings. Neither species has been obser

Assessment of the genetic relationship between Dictyocaulus species from Bos taurus and Cervus elaphus using complete mitochondrial genomic datasets.

BACKGROUND: Dictyocaulus species are strongylid nematodes of major veterinary significance in ruminants, such as cattle and cervids, and cause serious bronchitis or pneumonia (dictyocaulosis or "husk"). There has been ongoing controversy surrounding the validity of some Dictyocaulus species and their host specificity. Here, we sequenced and characterized the mitochondrial (mt) genomes of Dictyocaulus viviparus (from Bos taurus) with Dictyocaulus sp. cf. eckerti from red deer (Cervus elaphus), used mt datasets to assess the genetic relationship between these and related parasites, and predicted

Risk factors for human Anisakis infection and association between the geographic origins of Scomber japonicus and anisakid nematodes.

It has been reported that nearly all cases of anisakiasis in Japan are caused by Anisakis simplex sensu stricto. To elucidate this further, we investigated the presence of Anisakis type I larvae and Pseudoterranova decipiens in 218 Scomber japonicus fish collected from the seas of Japan. Anisakis type I larvae were detected in 74.3% (162/218) of the fish, and 99.8% of the Anisakis type I larvae comprised A. simplex sensu stricto and Anisakis pegreffii. Molecular identification techniques were used for 7.5% (360/4806) of the Anisakis type I larvae. The larvae found in the fish of the Pacific st

Identification of Trichinella isolates by polymerase chain reaction--restriction fragment length polymorphism of the mitochondrial cytochrome c-oxidase subunit I gene.

We developed a polymerase chain reaction based approach using restriction fragment length polymorphisms of the mitochondrial cytochrome c-oxidase subunit I to identify nine genotypes (Trichinella spiralis, Trichinella britovi-European strains, Trichinella britovi-Japanese strains, Trichinella nativa, Trichinella nelsoni, Trichinella T5, Trichinella T6, Trichinella T8 and Trichinella pseudospiralis) in the genus Trichinella. Partial mitochondrial cytochrome c-oxidase subunit I genes of nine genotypes were amplified by polymerase chain reaction, sequenced, and digested with three restriction end

Brugia malayi: recombinant antigens expressed by genomic DNA clones.

A Brugia malayi genomic DNA expression library was screened with rabbit antiserum generated against live infective larvae and 33 clones were identified. Five randomly selected clones were characterized in detail by Western blot, DNA and RNA analyses. The fusion proteins produced by each of these recombinant DNA clones are expressed by different genomic sequences. A profile of antigenic cross-reactivities of all 33 recombinant clones was compiled using a battery of antisera, including sera from humans infected with B. malayi. A high percentage of clones were cross-reactive with antisera against

Gastrointestinal parasite infections and self-medication in wild chimpanzees surviving in degraded forest fragments within an agricultural landscape mosaic in Uganda.

Monitoring health in wild great apes is integral to their conservation and is especially important where they share habitats with humans, given the potential for zoonotic pathogen exchange. We studied the intestinal parasites of wild chimpanzees (Pan troglodytes schweinfurthii) inhabiting degraded forest fragments amid farmland and villages in Bulindi, Uganda. We first identified protozoan and helminth parasites infecting this population. Sixteen taxa were demonstrated microscopically (9 protozoa, 5 nematodes, 1 cestode, and 1 trematode). DNA sequence analysis enabled more precise identificati

The complete mitochondrial genome sequence of the filarial nematode Wuchereria bancrofti from three geographic isolates provides evidence of complex demographic history.

Mitochondrial (mt) genome sequences have enabled comparison of population genetics and evolution for numerous free-living and parasitic nematodes. Here we define the complete mt genome of Wuchereria bancrofti through analysis of isolates from Papua New Guinea, India and West Africa. Sequences were assembled for each isolate and annotated with reference to the mt genome sequence for Brugia malayi. The length of the W. bancrofti mt genome is approximately 13,637 nucleotides, contains 2 ribosomal RNAs (rrns), 22 transfer RNAs (trns), 12 protein-coding genes, and is characterized by a 74.6% AT con

Real-time PCR as a surveillance tool for the detection of Trichinella infection in muscle samples from wildlife.

Trichinella nematodes are the causative agent of trichinellosis, a meat-borne zoonosis acquired by consuming undercooked, infected meat. Although most human infections are sourced from the domestic environment, the majority of Trichinella parasites circulate in the natural environment in carnivorous and scavenging wildlife. Surveillance using reliable and accurate diagnostic tools to detect Trichinella parasites in wildlife hosts is necessary to evaluate the prevalence and risk of transmission from wildlife to humans. Real-time PCR assays have previously been developed for the detection of Eur

Purification and characterization of a 7-kDa protein from Clonorchis sinensis adult worms.

A 7-kDa protein was purified from extracts of adult Clonorchis sinensis by a combination of ammonium sulfate precipitation, anion exchange chromatography, cation exchange chromatography, gel-filtration chromatography, and reversed-phase FPLC. The 7-kDa protein exists in the excretory-secretory products of adult C. sinensis, but not in extracts of adult Paragonimus westermani. Also, the 7-kDa protein reacted with the sera of patients with clonorchiasis but not with paragonimiasis or normal human sera. To observe the localization of the 7-kDa protein in the tissue of adult C. sinensis, an immuno

Molecular cloning, expression and enzymatic activity of a thioredoxin peroxidase from Dirofilaria immitis.

A Dirofilaria immitis cDNA clone encoding a nucleic acid homolog of thioredoxin peroxidase (nDiTPx) was isolated from a fourth-stage larval cDNA library, using serum from dogs vaccinated by chemotherapeutically-abbreviated D. immitis larval infections. The protein encoded by nDiTPx had a predicted molecular mass of 22.1 kDa and the deduced amino acid sequence was homologous to thioredoxin peroxidase-like sequences described in other filarial nematodes, yeast, bacteria and mammals. As is true for other members of this peroxiredoxin family, the nDiTPx-encoded protein had the conserved cysteine n

Genetic diversity, population structure, and benzimidazole resistance profiling of Ascaris lumbricoides in northwestern Thailand.

Ascaris lumbricoides remains one of the most prevalent soil-transmitted helminths worldwide, with ongoing transmission in endemic regions despite long-standing control programs. This study investigated the genetic diversity, population structure, and benzimidazole resistance of A. lumbricoides across three endemic regions of Thailand using mitochondrial (cox1 and nad1) and nuclear (β-tubulin) markers. Haplotype network analysis identified 13 haplotypes within the Thailand dataset, including three newly identified haplotypes, indicating measurable genetic diversity at the local scale. When anal

REDESCRIPTION OF PARABENEDENIELLA POSTEROCOLPA (HARGIS, 1955) BRULE AND BULLARD N. GEN. ET N. COMB. (MONOGENOIDEA: CAPSALIDAE: ENTOBDELLINAE) FROM INFECTIONS ON THREE BATOID HOSTS (RHINOPTERIDAE AND MOBULIDAE) WITH PHYLOGENETIC ANALYSIS AND COMMENTS ABOUT CAPSALID HOST SPECIFICITY.

We herein propose Parabenedeniella Brule and Bullard n. gen. to accommodate Parabenedeniella posterocolpa n. comb., formerly of Benedeniella Johnston, 1929 (both Capsalidae: Entobdellinae). Parabenedeniella differs from all other entobdelline genera by the combination of having a U-shaped (not looped) proximal portion of the cirrus sac, a vagina that is directed posterolaterad, a tube-like proximal portion of the vagina that is approximately one-third of the total length of the vagina, a thick-walled and muscular distal portion of the vagina that is approximately two-thirds of the total length

Morphology and molecular characterization of a new species of Tereancistrum parasite from the gills of Brycon nattereri.

A new species of Tereancistrum Kritsky, Thatcher & Kayton, 1980, collected from the gills of Brycon nattereri (Bryconidae) in the Cerrado, in the state of Goiás, Brazil, is described based on morphological and molecular data, based on the mitochondrial cytochrome c oxidase subunit 1 (COI) gene region. Tereancistrum campanum n. sp. is distinguished from all its congeners by the unique 'bell-shaped' accessory piece in the male copulatory complex. Molecular analysis revealed that the specimens analyzed formed a distinct group from other Tereancistrum sequences and that the sequences of Tereancist

Multi-marker analysis of Fasciola gigantica from cattle and buffalo across Pakistan reveals high levels of genetic diversity and novel haplotypes.

Molecular analyses of geographically dispersed Fasciola spp. isolates based on ribosomal, mitochondrial and nuclear molecular markers have revealed high levels of genetic diversity within liver fluke populations. To investigate the Fasciola population substructure across Pakistan 4 molecular markers were compared (fatty acid binding protein, fabp; phosphoenolpyruvate carboxykinase, pepck; random amplified polymorphic DNA, RAPD; mitochondrial NADH dehydrogenase, mt-nd1). Adult parasites (n = 595) were collected from buffalo and cattle across 4 provinces in Pakistan (Baluchistan, Gilgit Baltista

PHYLOGENY AND SYSTEMATICS OF CYATHOCOTYLID DIGENEANS (DIGENEA: DIPLOSTOMOIDEA) PARASITIZING SNAKES WITH DESCRIPTION OF THREE NEW SPECIES OF GOGATEA FROM AUSTRALIA AND VIETNAM.

The Cyathocotylidae Mühling, 1896 is a small but broadly distributed family of digeneans parasitic in a wide range of vertebrate definitive hosts, from fish to mammals. Despite existing taxonomic questions, only a few studies have generated DNA sequence data from cyathocotylids, and only a single publication included sequence data from a cyathocotylid parasitic in snakes. Four genera are known to infect snakes: GogateaLutz, 1935, Szidatia Dubois, 1938, MesostephanoidesDubois, 1951, and SerpentostephanusSudarikov, 1961. Members of these genera were known from only Asia and Africa. In the presen

Crossroad between the sylvatic and domestic cycles of Mesocestoides litteratus: mesocestodiasis in dogs adopted from shelters in Türkiye.

This study aimed to report the presence of Mesocestoides litteratus in dogs adopted from shelters in Türkiye. Gravid segments were examined microscopically in the faeces of dogs from different shelters located in Ankara and Kirikkale provinces in the central region of Türkiye. Then, genomic DNA obtained from these segments, a 446-bp fragment of the mitochondrial cytochrome C oxidase subunit 1 gene, and a 350-bp fragment of mitochondrial 12S rRNA were amplified and sequenced. BLASTn search was performed. During light microscopic examination, an egg-filled paruterine organ was observed in the mi

Hiding in plain sight: discovery and phylogeography of a cryptic species of Trichinella (Nematoda: Trichinellidae) in wolverine (Gulo gulo).

Understanding parasite diversity and distribution is essential in managing the potential impact of parasitic diseases in animals and people. Imperfect diagnostic methods, however, may conceal cryptic species. Here, we report the discovery and phylogeography of a previously unrecognized species of Trichinella in wolverine (Gulo gulo) from northwestern Canada that was indistinguishable from T. nativa using the standard multiplex PCR assay based on the expansion segment 5 (ESV) of ribosomal DNA. The novel genotype, designated as T13, was discovered when sequencing the mitochondrial genome. Phylog

Rodents of Senegal and their role as intermediate hosts of Hydatigera spp. (Cestoda: Taeniidae).

Hydatigera (Cestoda: Taeniidae) is a recently resurrected genus including species seldom investigated in sub-Saharan Africa. We surveyed wild small mammal populations in the areas of Richard Toll and Lake Guiers, Senegal, with the objective to evaluate their potential role as intermediate hosts of larval taeniid stages (i.e. metacestodes). Based on genetic sequences of a segment of the mitochondrial DNA gene cytochrome c oxidase subunit 1 (COI), we identified Hydatigera parva metacestodes in 19 out of 172 (11.0%) Hubert's multimammate mice (Mastomys huberti) and one out of six (16.7%) gerbils

Species delimitation based on mtDNA genes suggests the occurrence of new species of Mesocestoides in the Mediterranean region.

BACKGROUND: This study is the first contribution to the molecular taxonomy of Mesocestoides spp. from domestic and wild carnivores in the Mediterranean area. A total of 13 adult worms and 13 larval stages of Mesocestoides spp. were collected from domestic and wild carnivore hosts in Italy and Tunisia. Samples collected in the Slovak Republic were used as comparative samples from outside the Mediterranean. The genes cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogenase subunit 1 (nad1) of the mitochondrial genome were used as molecular markers to investigate the presence of cryptic Mesoc
