The Opioid System in Temporal Lobe Epilepsy: Functional Role and Therapeutic Potential.

Temporal lobe epilepsy is considered to be one of the most common and severe forms of focal epilepsies. Patients often develop cognitive def … , most probably through positive effects on mitochondrial function. Despite the supposed absence of endorphins in the hippocampus, exogenous activation of the mu opioid receptors (MOPr) induces pro-convulsant effects. Recently-expanded knowledge of the complex ways opioid receptors ligands elicit their effects (including biased agonism, mixed binding, and op …

Rostral ventrolateral medullary opioid receptor subtypes in the inhibitory effect of electroacupuncture on reflex autonomic response in cats.

Electroacupuncture (EA) is used in traditional Chinese medicine to treat arrhythmias, hypertension and myocardial ischemia. Our previous wor … id receptors in the rVLM. The endogenous neurotransmitters for mu- and delta-opioid receptors, beta-endorphins and enkephalins, in the rVLM, therefore appear to play a role in the EA-related modulation of cardiovascular reflex responses. Conversely, dynorphin is less likely to be involved in this response.

Intragonadal regulation of immune system functions.

Immune responses within the mammalian gonads, and in particular the testis, are deficient in spite of adequate lymphatic drainage and the presence of lymphocytes and MHC II+ macrophages. There is considerable evidence from in vivo and in vitro studies that this 'suppression' of the immune system may be due, at least in part, to localized inhibition or regulation of normal lymphocyte and/or macrophage functions within the gonads. In the testis, both steroidal and non-steroidal products of the Leydig cells, including androgens, endorphins, and inhibin-related proteins, have been implicated in me…

Periodic breathing triggered by hypoxia in normal awake adults. Modification by naloxone.

Breathing patterns in six normal awake subjects were monitored noninvasively during progressive hypoxia accomplished with the administration … administration of nitrogen. The irregularity in breathing appears to be mediated through release of endorphins, since the effect is blunted or eliminated by pretreatment or short-term treatment with naloxone.

Acupuncture for polycystic ovarian syndrome.

BACKGROUND: Polycystic ovarian syndrome (PCOS) is characterised by the clinical signs of oligo-amenorrhoea, infertility and hirsutism. Conventional treatment of PCOS includes a range of oral pharmacological agents, lifestyle changes and surgical modalities. Beta-endorphin is present in the follicular fluid of both normal and polycystic ovaries. It was demonstrated that the beta-endorphin levels in ovarian follicular fluid of otherwise healthy women who were undergoing ovulation were much higher than the levels measured in plasma. Given that acupuncture impacts on beta-endorphin production, whi

β-Edorphin predict pregnancy outcome of PCOS and DOR women after IVF-ET.

PURPOSE: The present study aims to explore whether β-EP in serum (sβ-EP) and follicular fluid (ffβ-EP) could predict the in vitro fertilization (IVF) outcomes of patients with polycystic ovary syndrome (PCOS) and diminished ovarian reserve (DOR). METHODS: 90 PCOS women, 50 DOR women, and 100 women with normal ovarian function (control group), who were all undergoing an IVF-embryo transfer trial, were included in the study. Biochemical characteristics, anti-Mullerian hormone (AMH), sβ-EP, ffβ-EP, embryo formation, and pregnancy indicators were assessed in all women. The correlations of AMH and

Autistic children display elevated urine levels of bovine casomorphin-7 immunoreactivity.

Elevated concentrations of circulating casomorphins (CM), the exogenous opioid peptides from milk casein, may contribute to the pathogenesis of autism in children. Because several mass spectrometry studies failed to detect casomorphins in autistic children, it was questioned whether these peptides can be detected in body fluids by mass spec. Here we demonstrated, using a novel high sensitivity ELISA method, that autistic children have significantly higher levels of urine CM-7 than control children. The severity of autistic symptoms correlated with concentrations of CM-7 in the urine. Because C

Recent developments in alcoholism:opioid peptides.

A large body of evidence indicates that the endogenous opioid system plays an important role in maintaining alcohol drinking behavior. Research is reviewed indicating that the reinforcing properties of alcohol which lead to continued and repeated bouts of drinking are due, in part, to alcohol-induced activation of the endogenous opioid system. Opioid receptor antagonists decrease alcohol craving, alcohol consumption, and loss of control over drinking. The potential of opioid receptor antagonists to improve treatment outcome in comprehensive relapse prevention programs is discussed.

Opioid peptidergic control of gonadotropin-releasing hormone pulse generator activity in the ovariectomized goat.

The role of endogenous opioid peptides in the control of gonadotropin-releasing hormone pulse generator activity was examined in long-term ovariectomized goats by recording the hypothalamic multiple-unit activity specifically associated with pulsatile luteinizing hormone secretion (MUA volleys). Administration of naloxone, an opioid receptor antagonist, at a rate of 1.0 mg/kg/h for 2 h resulted in shorter (p < 0.05) intervals between MUA volleys than during the control period (61.6 +/- 0.7 vs. 92.5 +/- 7.2 min) in animals treated with progesterone and estradiol that had reproduced a steroidal

(+)-cis-3-methylfentanyl and its analogs bind pseudoirreversibly to the mu opioid binding site: evidence for pseudoallosteric modulation.

Previous studies demonstrated that preincubation of membranes from the brain of the rat with 1 microM (+)-cis-3-methylfentanyl produced a wash-resistant inhibition of mu receptor binding. The present study was designed to: (1) determine the mechanism by which (+)-cis-3-methylfentanyl produced a wash-resistant inhibition of mu receptor binding, and (2) to generate a structure-activity study, using wash-resistant inhibition as the end-point. Pretreatment of membranes with 500 nM (+)-cis-3-methylfentanyl increased the Kd of binding sites for [3H]ohmefentanyl, without altering the Bmax. The increa

Neuroendocrine profile in bulimia nervosa.

The neuroendocrinology of bulimia nervosa has only recently been investigated, with initial research suggesting some biological overlap with both anorexia nervosa (AN) and depression. Similarities among AN, depression, and bulimia include a nonsuppressed Dexamethasone Suppression Test and an abnormal growth hormone (GH) response to thyrotropin-releasing hormone (TRH). Bulimics and anorectics both tend to have a delayed thyrotropin (TSH) response to TRH and elevated basal GH levels. Bulimics, however, have a normal GH response to clonidine, a nonblunted TSH response to TRH, low basal prolactin

Role for opioid peptides in self-injurious behavior: dissociation from autonomic nervous system functioning.

The effects of acute, orally administered naltrexone (0.5, 1.0, 1.5 and 2.0 mg/kg), a potent opioid receptor antagonist, on self-injurious behavior (SIB), heart rate, and blood pressure in three males (one 10-year-old and two 17-year-olds) were investigated. Subjects were evaluated in a structured test session for SIB. The frequency of the most predominant type of SIB (head and face hitting) was significantly reduced by naltrexone (maximum was 71% at the 1.5 mg/kg dose), while self-biting was not significantly decreased at any dose. In contrast, naltrexone had no significant effect on heart ra

Opioid modulation of gonadotrophin release in Tourette's syndrome.

Currently the most prevailing hypothesis attempting to explain the pathophysiology of Tourette's syndrome (TS) suggests that the disease results from dopaminergic (DA) hyperactivity (Golden, 1986). Evidence for this hypothesis is indirect and includes the favorable response of these patients to haloperidol, exacerbation of symptoms with dopaminergic drugs (e.g., methylphenidate) and the findings of reduced DA metabolites in the CSF of some TS patients (Singer et al., 1982). We have recently suggested that deranged opioid functions may also be important in the pathophysiology of TS (Sandyk, 198

Long-lasting glucocorticoid suppression of opioid-induced antinociception.

The antinociceptive effect of morphine (5 mg/kg body weight i.p.) in rats subjected to various experimental manipulations of the pituitary-adrenocortical system was studied. The absence of adrenal steroids increased the sensitivity to morphine. The following findings suggest that glucocorticosteroids have a long-lasting influence on opioid-induced antinociception, even when the steroids have been removed by adrenalectomy. First, when rats were adrenalectomized in the morning under basal conditions of pituitary-adrenocortical activity (plasma corticosterone level less than 1 microgram %), the s

Endogenous opiate modulation of baroreflexes in normotensive and hypertensive rats.

It is evident that hypertension is associated with elevated endogenous opiates. This study was designed to examine the role of endogenous opiates in the development and/or maintenance of two-kidney renovascular hypertension and in baroreceptor reflex function in conscious hypertensive rats. Naloxone administration during the onset of hypertension significantly attenuated the rise in blood pressure. After one week, systolic blood pressure in naloxone-treated rats was 27 mmHg lower than in 0.9% NaCl-treated hypertensive rats. Acute naloxone infusions in chronic hypertensive animals also signific

Exercise and mental health.

This paper reviews the mood altering properties of exercise and its potential in the prevention and treatment of mental disorders. The role of the brain monoamines, opioid peptides, the sympathetic nervous system, and cognitive behavioural theory as mediating pathways for the psychological benefits of exercise is critically examined. Clinical trials on exercise are reviewed and suggestions are made for future research in this field.

Opiate antagonists do not alter neuronal responses to stimulation of opioid-containing pathways in rat hippocampus.

The opioid receptor antagonists, naloxone and beta-chlornaltrexamine, were used to determine whether activation of endogenous opioid peptide containing pathways produced pharmacologically reversible opioid actions. Extracellularly recorded responses of the hippocampal CA3 pyramidal cells were evoked by stimulation of the dynorphin-containing mossy fiber pathway. Neither naloxone nor beta-chlornaltrexamine pretreatment significantly changed the evoked response. However, both antagonists blocked the effect of applied dynorphin-A(1-17) on CA1 pyramidal cell evoked responses. Thus, our data demons

Thermoregulatory effects of (D-ala2)-methionine-enkephalinamide in the cat. Evidence for multiple naloxone-sensitive opioid receptors.

(D-ala2)-methionine-enkephalinamide (DAME) was injected into the third cerebral ventricle of unrestrained cats. At ambient temperature (Ta) = 22 degrees C, 3.1-50 micrograms caused dose-related hyperthermias. As dosage was increased, the hyperthermias diminished and in some cats hypothermia developed. Hyperthermia was not due to pyrogenic contamination or prostaglandin synthesis since it was not altered by pretreatment with a large IV dose of indomethacin. However, pretreatment with naloxone did cause a dose-related reduction in the hyperthermia. A low dose of DAME (12.5 micrograms) also cause

Enkephalins in large bowel malignancy and in acute appendicitis.

Leucine and methionine enkephalins were measured by biological assay in normal colon, adenocarcinoma of the colon, carcinoma of the caecum, and in normal and inflamed appendix. Methionine enkephalin levels in both the adenocarcinomas and inflamed appendices were significantly higher than in normal controls. No significant change occurred in leucine enkephalin levels. The caecal tumours were anaplastic and contained no demonstrable opioid activity.
