Effective perception of fear signals is crucial for human survival and the importance of the amygdala in this process is well documented. Animal, lesion and neuroimaging studies indicate that incoming sensory signals of fear travel from thalamus to amygdala via two neural pathways: a direct subcortical route and an indirect pathway via the sensory cortex. Other lines of research have demonstrated prefrontal modulation of the amygdala. However, no study to date has examined the prefrontal modulation of the thalamo-cortico-amygdala pathways in vivo. We used psychophysiological and physiophysiolo…

Dental phobic patients with reported traumatic experiences at onset of dental anxiety were compared to subjects with an indirect fear acquisition in their psychophysiological responses to video scenes showing dental procedures. It has been suggested that individuals with conditioned fear would respond with higher levels of physiological arousal in response to threat compared to subjects with indirect fear acquisition. Although the overall pattern indicates support for this hypothesis, subjects grouped on the basis of their reported fear-etiology did not statistically differ in recorded muscle …

Data from two large undergraduate samples were used to develop the phobic stimuli response scales (PSRS). Factor analyses of data from the first sample (N = 380) led to the creation of several scales, two of which (Social and Blood-Injection Fears) demonstrated good reliability and convergent validity. Analyses of data from a second sample (N = 330) were used to modify the three remaining scales (Animal, Bodily Harm, and Physical Confinement Fears), which also showed sound psychometric properties. As predicted, the PSRS were differentially correlated with neuroticism and extraversion, which ma…

In this study, brain-derived neurotrophic factor (BDNF) heterozygous knock-outs were tested on fear conditioning, and their wild-type littermates were used as controls. Results showed that BDNF(+/-) mice are impaired in contextual learning, whereas tone learning remains intact. Because BDNF is involved in synaptic transmission and contextual learning is hippocampal dependent, we hypothesized that this deficit is attributable to abnormal BDNF-modulated synaptic plasticity in the hippocampus. A "gain-of-function" experiment was performed next by infusing recombinant BDNF protein into the hippoca…

There is now considerable evidence that phobic responding is associated with a bias towards expecting aversive or traumatic outcomes following encounters with the phobic stimulus (e.g. Behavioural Brain Sci. 18 (1995) 289-325; Phobias: A Handbook of Theory, Research and Treatment. Wiley, Chichester, 1997). In terms of conditioning contingencies, this can be described as a bias towards expecting an aversive unconditioned stimulus (UCS) following a phobic conditioned stimulus (CS). The disease-avoidance model of animal fears (Anxiety Res. 4 (1992a) 314; Matchett and Davey, 1991) suggests that co…

Thirty participants who had been treated for aviophobia with virtual reality graded exposure therapy with physiological monitoring and visual feedback (VRGETpm), virtual reality graded exposure therapy with physiological monitoring only (VRGETno), or imaginal exposure therapy (visualization) with physiological monitoring only (IET) between January 1998 and January 1999 were contacted in January 2002 for a 3-year posttreatment follow-up assessment. Of the participants in the VRGETpm group who had flown successfully by the end of treatment, all had maintained their ability to fly at follow-up. O

Twenty-three drug-free patients with agoraphobia and panic disorder (DSM-III criteria) had, at rest, lower mean end-tidal PCO2 (32 v. 36 mmHg) and higher mean heart rate (92 v. 83 bpm) than did 18 controls. During 5 min of listening to fear talk, only eight (35%) patients and three (16%) controls panicked, but panic was associated with marked physiological changes in only two patients and one control. Patients said that breathlessness began slightly more often before than after panic. In 59% of patients the symptoms from voluntary hyperventilation (VHV) were very similar or identical to those …

Males with high negative attitudes toward male homosexuality are often referred to in research as homophobics, yet it is unknown whether high homonegative males actually exhibit physiological responses characteristic of phobics. In a series of studies, heart rate was monitored in males with high or low negative attitudes toward male homosexuality as they viewed slides of landscapes and slides depicting explicit sexual activity. If high homonegativity is equivalent to homophobia, high homonegative males should exhibit heart rate acceleration to slides of male-male sexual activity, but, like low

Standard extinction (SE) is the most widely implemented intervention for reducing conditioned and generalized fear, indexed by psychophysiological skin conductance responses, and subjective self-report responses such as expectancy and valence ratings; however, the possibility of fear return still remains. Hence, novel interventions may be used to augment the efficiency of SE. Imagery rescripting (ImRS), based on the premise of unconditioned stimulus (UCS) devaluation, encompasses the devaluation of the cognitive representation of the UCS in a positive direction. Imaginal extinction (IE), based…

Fear discrimination-the ability to distinguish threatening stimuli from non-threatening ones-is an essential adaptive behavior, yet the neural mechanisms underlying individual differences in discrimination precision remain unclear. Here, we show that parvalbumin-positive interneuron (PV-IN)-mediated feedforward inhibition to fear engram neurons (ENs) in the lateral amygdala is selectively enhanced following auditory fear conditioning, driven by increased postsynaptic expression of α1 subunit (Gabra1)-containing GABAA receptors. Disruption of PV-IN activity during fear retrieval-via optogenetic…

BACKGROUND: Circulating tumor DNA (ctDNA) is a novel prognostic biomarker indicating minimal residual disease and high recurrence risk. In three randomised DYNAMIC trials, ctDNA was used to guide adjuvant chemotherapy (ACT) for early-stage colorectal cancer. While clinical applications of ctDNA are being established, the psychosocial impact, particularly on fear of cancer recurrence (FCR), is less understood.
OBJECTIVE: To explore the psychosocial impact, especially FCR, of ctDNA-informed ACT decisions among a subset of DYNAMIC trial participants.
METHODS: A qualitative study using semi-stru…

BACKGROUND AND PURPOSE: TRPM3 is best known for its role as a peripheral sensor of pain and heat. Recent studies reporting gain-of-function … sion in excitatory neurons of the lateral nucleus of the amygdala (LA), a brain region critical for fear learning also implicated in neuropsychiatric and epileptic disorders. We demonstrate that TRPM3 is functionally active in LA neurons and regulates their intrinsic electrophysiological properties. Consistent with this role, genetic ablation of TRPM3 …

The COVID-19 pandemic severely affected university students' mental health. The objective of this study was to evaluate the level of fear associated with COVID-19 in university students from the Coast, Sierra, and the Jungle regions of Peru between April and May 2020. The sample consisted of 915 university students from the three regions of Peru. The Fear of COVID-19 Scale was used to assess fear and its dimensions (psychological and somatic symptoms). Data were analyzed using analysis of variance (ANOVA) and Tukey's post hoc test to compare group means. Additionally, multiple linear regressio…

Trait mindfulness refers to an individual’s inherent capacity to maintain sustained awareness and focus on present experiences. This characteristic is naturally present in the population, even without intentional mindfulness training, and is significantly negatively correlated with negative emotions, anxiety, and depression. Fear is an adaptive response to threat, but excessive generalization of fear is a key underlying factor in anxiety disorders. Fear generalization occurs when an individual, having learned to fear a specific threat, extends this fear response to stimuli that share similarit…

Prediction errors (PE) are thought to drive associative learning. While neural signals consistent with PE encoding have been identified, the expression of PE in psychophysiological indices remains debated. Here, we sought to fill this gap by investigating responses to unconditioned stimulus (US) occurrence and probability in skin conductance responses (SCR), pupil size responses (PSR), heart period responses (HPR), and respiration amplitude responses (RAR). Data set 1 consisted of eight published studies (N1 = 264) using differential fear conditioning with partial reinforcement (50%), and nove…

Associating events separated in time depends on the CA1, subiculum (SUB), and retrosplenial cortex (RSP). The degree to which their connectivity and underlying circuit mechanisms contribute to the association of such temporally discontiguous events is not known. Here we showed, using trace fear conditioning (TFC), wherein mice learn to associate tone and shock separated by a temporal trace, that molecularly distinct excitatory VGluT1+ and VGluT2+ SUB→RSP projections subserve the associative and temporal components of TFC. During trace memory formation, VGluT2+ SUB→RSP projections showed increa…

Passive risks are risks that arise or are amplified by inaction, such as failing to undergo a medical screening test. Because their consequences are often delayed and diffuse, such risks may receive limited attention. The present research examined whether directing attention to passive risks is associated with differences in evoked fear, perceived severity, and prioritization for mitigation. In Experiment 1, participants were shown images of various passive risks, with attention consistently directed to one of them. In Experiments 2 and 3, verbal labels of the risks were used instead of images…

OBJECTIVE: To characterize the prevalence of fear of cancer recurrence (FCR) and examine demographic, clinical, and psychosocial factors associated with high FCR among low-income, racially and ethnically diverse prostate cancer survivors treated with definitive therapy for localized disease.
METHODS: We conducted a retrospective, cross-sectional analysis of 150 participants enrolled in the Men's Health Study (2001-2007) who completed the FCR Survey at baseline. High FCR was defined as a score ≤14 (lower tertile). Demographic, clinical, and patient-reported outcomes were assessed, including he…

Fear memory generalization is a fundamental hallmark of post-traumatic stress disorder (PTSD) that enables animals to use past experience to adapt to changing conditions. The infralimbic cortex (IL) is implicated in suppressing generalized fear, but the underlying molecular mechanisms remain unknown. Here, we demonstrate that S-nitrosylation of Dexras1 (SNO-Dexras1) in the IL drives fear generalization. Dexras1 is activated by nitric oxide (NO) donors as well as by N-methyl-D-aspartic acid (NMDA) receptor-stimulated NO synthesis in cortical neurons. It is found that the level of SNO-Dexras1 is…

INTRODUCTION: There is substantial burden associated with mild traumatic brain injury (mTBI), particularly for people who experience persistent postconcussion symptoms (PPCS). Research has explored the role in PPCS of fear avoidance as a psychogenic contributing factor; however, understanding remains limited. The present study employed experimental methods to investigate the strength and direction of the relationship between fear avoidance and PPCS.
METHOD: Participants (N = 143) were randomly assigned to a fear avoidance (n = 74) or control group (n = 69). They viewed a mTBI vignette with or…
