Effect of lactic acid bacteria on diarrheal diseases.

Microbial balance is an important factor in the maintenance of intestinal homeostasis, and yogurt or fermented milk supplementation has been proposed to control diarrheal diseases. A number of studies using animal models and clinical studies in humans have confirmed the beneficial effect of such fermented products in case of lactose intolerance, viral diarrhea or antibiotics-associated diarrhea. The mechanisms by which lactic acid bacteria exert their effects are multiple. Bacterial lactase improves the absorption of lactose, but fermented products slow down the intestinal transit facilitating

Fibre utilization by Kalahari dwelling subterranean Damara mole-rats (Cryptomys damarensis) when fed their natural diet of gemsbok cucumber tubers (Acanthosicyos naudinianus).

Kalahari dwelling Damara mole-rats (Cryptomys damarensis) naturally feed on a high fibre diet of underground gemsbok cucumber tubers, Acanthosicyos naudinianus. We investigated the degree of fibre utilization and fermentation on this diet by measuring caecal characteristics (namely temperature, pH and weight) and in vitro rates of gas and short chain fatty acid (SCFA) production in these underground dwelling hind-gut fermentors. Rectal temperatures (33.8 +/- 0.6 degrees C) were consistently higher than caecal temperatures (33.3 +/- 0.6 degrees C). Furthermore, a 0.8 degrees C gradient of tempe

Incidence and health risk assessment of hydrogen cyanide and multi-mycotoxins in Nigerian garri.

Garri is a granular, starchy food prepared by the fermentation of mashed cassava. Hydrogen cyanide (HCN) and mycotoxins are contaminants in certain foods at different points along the food value chain. The incidence and contamination levels of HCN and multi-mycotoxins in garri from five agroecological zones of Nigeria were determined using a spectrophotometric method and ultra-high-performance liquid chromatography-tandem mass spectroscopy (UHPLC-MS/MS), respectively. The health risk associated with the consumption of contaminated garri was assessed. The health risk assessment model was used t

Dose-dependent effects of a soluble dietary fibre (pectin) on food intake, adiposity, gut hypertrophy and gut satiety hormone secretion in rats.

Soluble fermentable dietary fibre elicits gut adaptations, increases satiety and potentially offers a natural sustainable means of body weight regulation. Here we aimed to quantify physiological responses to graded intakes of a specific dietary fibre (pectin) in an animal model. Four isocaloric semi-purified diets containing 0, 3.3%, 6.7% or 10% w/w apple pectin were offered ad libitum for 8 or 28 days to young adult male rats (n = 8/group). Measurements were made of voluntary food intake, body weight, initial and final body composition by magnetic resonance imaging, final gut regional weights

Fermentation-Induced Phenolic Remodeling of Aronia melanocarpa Alleviates Obesity-Induced Cognitive Dysfunction Associated With Coordinated Modulation of the AMPK/NF-κB/BDNF Axis and BBB Integrity.

Obesity-induced cognitive decline involves chronic inflammation and neuronal dysfunction. This study investigated the mechanistic associations of probiotic-fermented Aronia melanocarpa extract (LAB-A) on metabolic and cognitive dysfunction in high-fat diet (HFD)-fed mice. Fermentation doubled total polyphenol content and increased aglycones like quercetin and eriodictyol. LAB-A attenuated microglial neuroinflammation by inhibiting NF-κB translocation (94.4% reduction, p < 0.05) and protected neurons from apoptosis by reducing the BAX/BCL-2 ratio (89.3%, p < 0.05). In HFD-fed mice, LAB-A signif

Valorization of Ligusticum chuanxiong hydrosol via Eurotium cristatum biotransformation: mechanistic insights into flavor remodeling and functional enhancement.

Ligusticum chuanxiong hydrosol, a low-value distillation by-product, is usually discarded because of its offensive medicinal odor and pronounced bitterness. To overcome these sensory defects, this study employed Eurotium cristatum, a "golden flower" fungus renowned for flavor remodeling in traditional fermentation, to valorize this underexplored aqueous matrix. This represents the first systematic application of E. cristatum for hydrosol biotransformation. Under optimized conditions (39°C, pH 6.7, 7% inoculum), total phenolics reached 71.25 mg/L. Electronic nose and tongue analyses indicated c

Dietary Fiber from Baijiu Distillers' Grains Improves Glucose-Lipid Homeostasis via Gut-Liver Metabolic Remodeling.

Baijiu distillers' grains (BDG), a major fermented cereal by-product of baijiu production, represent an underutilized source of structurally modified dietary fiber with potential value for functional food development. Here, we found that BDG-derived dietary fiber (BDG-DF), mainly composed of mannose (34.83 ± 0.38%) and xylose (35.14 ± 0.25%), promoted short-chain fatty acid production during in vitro fermentation, and its fermentation supernatants reduced IL-1β and TNF-α levels and modestly decreased IL-6 production in a Caco-2/HepG2 co-culture model. In T2D mice, BDG-DF improved glucose toler

Engineering carboxyl methyltransferase for high-efficiency biosynthesis of methyl salicylate in Escherichia coli.

Methyl salicylate (MeSA) is a volatile methylated aromatic ester widely used in agriculture, food, and pharmaceuticals. Microbial biosynthesis offers a sustainable alternative to plant extraction and petrochemical synthesis; however, efficient MeSA production remains limited by the low catalytic efficiency of carboxyl methyltransferases, insufficient intracellular supply of S-adenosylmethionine (SAM), and severe product volatilization during fermentation. In this study, we systematically screened salicylate carboxymethyltransferase and identified CbSAMT as a relatively efficient catalyst for s

[Multidimensional metabolic engineering of Yarrowia lipolytica for β-carotene production].

β-carotene is a natural lipid-soluble carotenoid widely used in the food and pharmaceutical industries. Although metabolic engineering efforts for the production of β-carotene via microbial fermentation have achieved certain progress, there remains room for further improvement. Compared with Saccharomyces cerevisiae or Escherichia coli, Yarrowia lipolytica exhibits a pronounced lipid-flux orientation in its core metabolic logic, conferring a natural advantage for the production of lipid-soluble compounds. This study aims to engineer Y. lipolytica strains for efficient β-carotene production. Fi

Digestive Characteristics of Hericium erinaceus Polysaccharides and Their Positive Effects on Fecal Microbiota of Male and Female Volunteers During in vitro Fermentation.

Hericium erinaceus polysaccharides (HEPs) have attracted widespread attention in regulating gut microbiota (GM). To investigate digestibility and fermentation of HEPs and their effects on GM composition, three polysaccharide fractions, namely, HEP-30, HEP-50, and HEP-70, were fractionally precipitated with 30%, 50%, and 70% ethanol concentrations (v/v) from hot water-soluble extracts of Hericium erinaceus, respectively. Three kinds of prepared HEPs were structurally characterized and simulated gastrointestinal digestion, and their effects on human fecal microbiota fermentations of male and fem

Microbial dynamics of model Fabriano-like fermented sausages as affected by starter cultures, nitrates and nitrites.

The present study promotes the valorization of Fabriano-like fermented sausages, which are central-Italy salami with an origin that dates to the early 17th century, for the possible future selection of autochthonous starter cultures to be used with respect to local traditions. To the best of the authors' knowledge, this study represents the first attempt to define the microbial dynamics in Fabriano-like fermented sausage and the effect of nitrates/nitrites and starter cultures on its natural bacterial biota. Culture and RNA-based techniques (RT-PCR-DGGE and Illumina sequencing) were used to as
