Antibiotic-Resistant Salmonella in Animal Products Jeopardize Human Health.

Despite the significance of antibiotics in treating bacterial infections, antibiotic resistance is continuously increasing, thus posing a significant threat. In addition to strains resistant to individual drugs, multidrug-resistant (MDR) and pandrug-resistant strains, are emerging. Salmonella, a primary cause of global foodborne illness, is often transmitted through animal products. Antibiotic treatment is crucial for immunocompromised individuals, such as older adults and patients with weakened immune systems, due to their increased susceptibility to severe effects. MDR Salmonella, which can

Mechanism of eravacycline resistance in Acinetobacter baumannii mediated by a deletion mutation in the sensor kinase adeS, leading to elevated expression of the efflux pump AdeABC.

Acinetobacter baumannii is an important pathogen and presents a major burden in healthcare as strains frequently cause hospital associated opportunistic infections with high mortality rates. Due to increasing numbers of drug resistant A. baumannii strains, newly developed antibiotics are being used to treat infections caused by such strains. One novel synthetic antibiotic of the tetracycline class with activity against A. baumannii is eravacycline. To investigate possible mechanisms of eravacycline resistance, we performed an in vitro evolution experiment to select for an eravacycline resistan

Characterization of the Genetic Environment of the blaVEB-4 Gene, Associated with a Transposable Region in a Proteus mirabilis Clinical Isolate.

Proteus mirabilis is the second most common cause of urinary tract infections and is also an important cause of nosocomial infections. TEM-type and CTX-M-type extended-spectrum β-lactamases (ESBLs) are the most widely distributed in this bacterial species, but minor ESBLs such as the VEB-type have also been identified. The aim of this study was to analyze the genetic environment of the blaVEB-4 gene found in a P. mirabilis clinical isolate recovered in Spain. P. mirabilis N2231 showed resistance to penicillins, cephalosporins, and aminoglycosides, remaining susceptible to imipenem, cefoxitin,

Multilocus interactions at maternal tumor necrosis factor-alpha, tumor necrosis factor receptors, interleukin-6 and interleukin-6 receptor genes predict spontaneous preterm labor in European-American women.

OBJECTIVE: We hypothesize that genetic variations (single nucleotide polymorphisms-SNPs) in the tumor necrosis factor-alpha (TNF-alpha), TNF receptors (TNFRI and TNFRII), interleukin-6 (IL-6) and IL-6 receptor (IL-6R) genes predict high-risk status for spontaneous preterm birth (sPTB) in European-American women. In this study we examine the allelic and genotypic variations and the gene-gene interactions in the TNF-alpha, TNFRs, IL-6, and IL-6R genes in maternal DNA samples by using a case-control model. STUDY DESIGN: Maternal DNA from cases of sPTB after preterm labor (n = 101) and controls (

Bactericidal activity of gallic acid against multi-drug resistance Escherichia coli.

The continuous emergence of multidrug-resistant (MDR) bacteria has posed an increasingly serious public health threat which urges people to develop some alternatives. Gallic acid (GA) is a natural ingredient in many traditional Chinese medicines, which has many biological activities, such as antibacterial, and antiseptic. Here, clinical isolates of MDR Escherichia coli (E. coli) were used to evaluate the antibacterial effect of GA and the underlying mechanism. The results revealed that GA exerted bactericidal activity and inhibited the formation of bacterial biofilm. GA enhanced the activities

Management of Multidrug-Resistant Infections in ICUs: A Narrative Review of Pharmacological and Non-pharmacological Interventions.

Multidrug-resistant (MDR) infections in ICUs are a growing global health challenge associated with increased morbidity, mortality, prolonged hospitalization, and rising healthcare costs. Critically ill patients are particularly vulnerable because of immune dysfunction, invasive procedures, prolonged ICU stays, and frequent exposure to broad-spectrum antibiotics, all of which increase susceptibility to MDR infections and antimicrobial selection pressure. Common MDR pathogens in ICU settings include Acinetobacter baumannii, Klebsiella pneumoniae, Pseudomonas aeruginosa, methicillin-resistant Sta

Genomic global epidemiology and evolution of Escherichia coli ST131 clade A.

During the 2010s, Escherichia coli ST131 clade A tested sensitive to most antibiotics. In the 2020s, Europe experienced outbreaks of fluoroquinolone-resistant ST131-A that contained CTX-M β-lactamases and OXA-48-like carbapenemases. We investigated the acquisition of different antimicrobial resistance determinants in the emergence of multidrug-resistant (MDR) ST131-A. Phylogenetic analysis and Bayesian inference were utilized to define subclades and clusters (within subclades) and investigate the evolutionary history of ST131-A, using sequenced genomes and publicly available genomic data (n=2,

A novel chimeric antimicrobial peptide B5PL dual-targets bacterial cell membrane and neutrophils to combat multidrug-resistant bacterial infections.

Methicillin-resistant Staphylococcus aureus (MRSA) and multidrug-resistant Klebsiella pneumoniae (MDR-KP) pose severe threats to global public health due to lack of effective therapeutic strategies. Conventional antibacterial agents exhibit limited potency for modulating host immune defense responses. This study designed a novel chimeric antimicrobial peptide B5PL by fusing a truncated bovine β-defensin 5 (B5) domain with a PL-5 derivative peptide (PL). Results showed that B5 exhibited weak direct bactericidal activity but enhanced the phagocytic and bactericidal functions of macrophages in a

Character Virulence Association Factors and Gene Mutation Mediating Multidrug Resistance Phenotypes in Candidozyma duobushaemulonii.

Candidiasis, traditionally dominated by Candida albicans infection, now faces a paradigm shift with the emergence of non-C. albicans infections, such as the multidrug-resistant (MDR) Candidozyma (formerly Candida) species. Critically, the emergence of multidrug resistance (MDR) Candidozyma duobushaemulonii (also known as Candida duobushaemulonii) poses critical challenges to antifungal therapy. Here, in this comparative study, we analyzed four clinical C. duobushaemulonii isolates (cd, cd1, cd2, and cd3) to establish strain-specific virulence and antifungal resistance profiles. Dose-and regimen-dependent therapeutic efficacy of the lytic Klebsiella phage TRK61 in a Galleria mellonella infection model.

Multidrug-resistant (MDR) Klebsiella infections pose a significant challenge and necessitate alternative antimicrobial approaches. Bacteriophage therapy represents a promising option; however, therapeutic outcomes depend on phage dose, dosing strategy, and host-pathogen interactions. In this study, we isolated and characterized a lytic Klebsiella phage, vB_KpnS_TRK61 (TRK61), from municipal wastewater. Electron microscopy revealed that TRK61 has an icosahedral head and a long, flexible tail, consistent with members of the family Demerecviridae. Host range analysis demonstrated lytic activity a
