Lithium carbonate was administered to six patients with severe Felty's syndrome, five of whom had problems with infection. Two patients had granulocyte increments that persisted after therapy was discontinued; in one of them problems with infections resolved. Tissue sections embedded in paraffin and fixed in formalin from 32 patients with Hodgkin's disease, representing the major histological subtypes, were studied using two granulocyte specific monoclonal antibodies (Leu-M1 and 3C4) and an HLA-D region specific monoclonal antibody (TAL-IB5). A 5-year-old child presented with pneumonia and agranulocytosis. A Wright-stained peripheral blood smear showed only cells which had the morphological appearance of lymphocytes, plus a few monocytes and eosinophils. During a three-year period, 26 neonates with bacterial infection and neutropenia were studied. In order to assess the marrow neutrophil reserves, bone marrow aspirates were obtained from each of these patients. Transient granulocytopenia develops during the first few minutes of hemodialysis, and is associated with a marked increase in granulocyte adherence as the cells marginate in the pulmonary circulation. The increased adherence is mediated by factors present in the plasma, thought to be complement components activated by the dialysis coil. In the present study, an assay of plasma for adherence-augmenting activity shows that it is maximal at 15 min after starting dialysis, and undetectable at 40 min. Medullary granulocyte progenitor (CFUc) cultures were grown in vitro from samples taken from 6 patients with toxic granulocytopenia caused by either thiamphenicol cephalothin or amidopyrine and who are now apparently cured. Rats made transiently neutropenic by intra-arterial administration of vinblastine (3 mg/kg) and infected with E. coli (6.02+/-0.45 X 10(8), per animal) have a mortality rate of 90% within 48 h post infection. Multiple transfusions of large numbers of granulocytes (harvested from Deca-Durabolin treated donor rats) protected the neutropenic animals from sepsis. Out of a group of 11 rats, 10 recovered completely after repeated granulocyte transfusions.

Granulopoiesis was studied in 20 cases of chronic granulocytopenia (adults and children; acquired and congenital).
