Light-dependent alteration of serotonin-1A receptor binding in cortical and subcortical limbic regions in the human brain.

OBJECTIVE: Climate, in particular sunshine, influences mood and energy levels, creating a positive upswing of mood on bright, sunny days and negative downswing in cold, dark winter seasons. Higher serotonin transporter availability in healthy human subjects in times of lesser light exposure and lower serotonin levels have been shown in winter. METHODS: We examined the light-dependent variations in serotonin-1A receptor binding in limbic regions in 36 drug-naive healthy human subjects. Receptor binding was quantified using positron emission tomography and the radioligand [carbonyl-¹¹C]WAY-1006

Increased involvement of the parahippocampal gyri in a sad mood predicts future depressive symptoms.

Behavioral studies suggest a relationship between autobiographical memory, rumination and depression. The objective of this study was to determine whether remitted depressed patients show alterations in connectivity of the posterior cingulate cortex (PCC, a node in the default mode network) with the parahippocampal gyri (PHG, a region associated with autobiographical memory) while intensively recalling negative memories and whether this is related to daily life symptoms and to the further course of depression. Sad mood was induced with keywords of personal negative life events in participants

Alterations in patients with major depressive disorder before and after electroconvulsive therapy measured by fractional amplitude of low-frequency fluctuations (fALFF).

BACKGROUND: Electroconvulsive therapy (ECT) is an important treatment option for patients with major depressive disorder (MDD). However, the mechanisms of ECT in MDD are still unclear. METHODS: Twenty-four patients with severe MDD and 14 healthy controls were enrolled in this study. Eight ECT sessions were conducted for MDD patients using brief-pulse square-wave signal at bitemporal locations. To investigate the regional cerebral blood flow in MDD patients before and after ECT treatments by resting-state functional magnetic resonance imaging (rs-fMRI), the patients were scanned twice (before

Resting-state functional connectivity of the nucleus accumbens in auditory and visual hallucinations in schizophrenia.

Both auditory hallucinations (AH) and visual hallucinations may occur in schizophrenia. One of the main hypotheses underlying their occurrence involves the increased activity of the mesolimbic pathway, which links the ventral tegmental area (VTA) and the nucleus accumbens (NAcc). However, the precise contribution of the mesolimbic pathway in hallucinations across various sensory modalities has not yet been explored. We compared the resting-state functional connectivity (rs-FC) of the NAcc among 16 schizophrenia patients with pure AH, 15 with both visuoauditory hallucinations (VAH), and 14 with

High-intensity transcranial alternating current stimulation enhances functional connectivity of subcortical regions in major depressive disorder.

BACKGROUND: Major depressive disorder (MDD) is associated with dysregulation of large-scale brain networks, particularly those supporting emotional regulation and cognitive control. Transcranial alternating current stimulation (tACS) has shown antidepressant potential, but its neurophysiological effects remain incompletely understood. This study investigates the effects of 77.5 Hz high-intensity tACS on functional connectivity (FC) within subcortical-limbic circuits in MDD. METHODS: In a double-blind, randomized, sham-controlled study, 66 MDD patients were randomized to receive either active

DNMT3a-mediated GluN2B desilencing in the anterior cingulate cortex underlies chemotherapy-induced neuropathic pain.

Chemotherapy-induced neuropathic pain (CINP) involves chemotherapy toxicity to cortical tissues. DNA methylation, a key epigenetic mechanism for gene regulation, has been implicated in neuropathic pain pathogenesis. The anterior cingulate cortex (ACC) is critical for pain processing, yet its susceptibility to DNA methylation-mediated epigenetic regulation remains unexplored. In a mouse model of paclitaxel (PTX)-induced CINP, we found that PTX downregulated DNA methyltransferase 3a (DNMT3a) in the ACC and induced mechanical and thermal pain hypersensitivity. Overexpression of DNMT3a specificall

Diazepam modulates anterior cingulate glutamate levels in people at clinical high-risk for psychosis.

OBJECTIVE: Preclinical evidence suggests that modulating neural excitation through administration of diazepam, a positive allosteric modulator of GABAA receptors, can prevent the emergence of behavioral and neurobiological alterations relevant to psychosis in adulthood. DESIGN AND PARTICIPANTS: Here, we examine this neurochemical mechanism in individuals at clinical high risk for psychosis in a randomized, double-blind, placebo-controlled crossover study. Twenty-four individuals (15 female and 9 male) aged 18-35 were scanned twice using proton magnetic resonance spectroscopy to measure anteri

Glutamate- and GABA-targeted drugs for Cc-occurring bipolar and alcohol use disorders: a randomized, double-blind, placebo-controlled, crossover study of N-acetylcysteine, gabapentin, and placebo.

Forty percent of people with bipolar disorder (BD) develop alcohol use disorder (AUD), with co-occurring AUD substantially worsening the course of BD. Though efficacious treatments remain elusive, frontal glutamate and GABA dysregulation represent promising targets for intervention. This study evaluated the ability of two adjunctive medications, N-acetylcysteine (NAC) and gabapentin, to modulate glutamate and GABA (primary) and to reduce heavy drinking and depressive symptoms (exploratory) in people with BD + AUD. Fifty-four individuals meeting DSM-5 criteria for BD + AUD were enrolled in a ra

Motor circuits and beyond: Functional connectivity related to psychomotor syndromes in depression.

BACKGROUND: Psychomotor disturbance (PmD) is prevalent in major depressive disorder (MDD), with neural substrates implicated in disrupted motor circuits and the interaction to non-motor cortex. Our objective is to explore the functional connectivity pattern underlying PmD using functional magnetic resonance imaging (fMRI). METHODS: A total of 150 patients with MDD and 91 healthy controls (HCs) were included in this study. The patients were categorized into psychomotor (pMDD, n = 107) and non-psychomotor (npMDD, n = 43) groups based on the Hamilton Depression Rating Scale. Seed-based connectiv

Chronic stress enhances neuropathic pain through activation of the anterior cingulate cortex-spinal dorsal horn pathway in rats.

Chronic stress is known to exacerbate neuropathic pain, yet the underlying mechanisms remain poorly understood. This study investigates the role of the anterior cingulate cortex (ACC) and its descending projections to the spinal dorsal horn (SDH) in mediating stress-induced pain facilitation. Using a chronic unpredictable mild stress (CUMS) model combined with a chronic constriction injury (CCI) model in male Sprague-Dawley rats, we assessed behavioral changes, neuronal activity, and molecular alterations. Our results demonstrate that CUMS significantly exacerbates mechanical allodynia in CCI

Increased Cortical Thickness in Alzheimer's Disease.

OBJECTIVE: Patients with Alzheimer's disease (AD) have diffuse brain atrophy, but some regions, such as the anterior cingulate cortex (ACC), are spared and may even show increase in size compared to controls. The extent, clinical significance, and mechanisms associated with increased cortical thickness in AD remain unknown. Recent work suggested neural facilitation of regions anticorrelated to atrophied regions in frontotemporal dementia. Here, we aim to determine whether increased thickness occurs in sporadic AD, whether it relates to clinical symptoms, and whether it occur in brain regions f

Effective Connectivity Network of Aberrant Prediction Error Processing in Auditory Phantom Perception.

Introduction: Prediction error (PE) is key to perception in the predictive coding framework. However, previous studies indicated the varied neural activities evoked by PE in tinnitus patients. Here, we aimed to reconcile the conflict by (1) a more nuanced view of PE, which could be driven by changing stimulus (stimulus-driven PE [sPE]) and violation of current context (context-driven PE [cPE]) and (2) investigating the aberrant connectivity networks that are engaged in the processing of the two types of PEs in tinnitus patients. Methods: Ten tinnitus patients with normal hearing and healthy co

Effective connectivity of default mode network subsystems and automatic smoking behaviour among males.

BACKGROUND: The default mode network (DMN) is not a single system, but rather is composed of smaller and distinct functional subsystems that interact with each other. The functional relevance of these subsystems in tobacco use disorder (TUD) and the neurobiological features associated with smoking motivation are still unclear; thus, we sought to assess causal or direct connectivity alterations within 3 subsystems of the DMN among people with TUD. METHODS: We recruited male smokers and nonsmokers. We conducted resting-state functional magnetic resonance imaging (rs-fMRI) and collected ratings

In Vivo Assessment of Astrocyte Reactivity in Patients with Progressive Supranuclear Palsy.

OBJECTIVE: Although astrocytic pathology is a pathological hallmark of progressive supranuclear palsy (PSP), its pathophysiological role remains unclear. This study aimed to assess astrocyte reactivity in vivo in patients with PSP. Furthermore, we investigated alterations in brain lactate levels and their relationship with astrocyte reactivity. METHODS: We included 30 patients with PSP-Richardson syndrome and 30 healthy controls; in patients, tau deposition was confirmed through 18F-florzolotau positron emission tomography. Myo-inositol, an astroglial marker, and lactate were quantified in th

Cognitive control & the anterior cingulate cortex: Necessity & coherence.

Influential theories of complex behaviour invoke the notion of cognitive control modulated by conflict between counterfactual actions. Medial frontal cortex, notably the anterior cingulate cortex, has been variously posited as critical to such conflict detection, resolution, or monitoring, largely based on correlative data from functional imaging. Examining performance on the most widely used "conflict" task-Stroop-in a large cohort of patients with focal brain injury (N = 176), we compare anatomical patterns of lesion-inferred neural substrate dependence to those derived from functional imagi

Identification of neurotransmitter imbalances in the cingulate cortex of NMOSD patients using magnetic resonance spectroscopy.

Cognitive impairment affects 29-67% of patients with neuromyelitis optica spectrum disorder. Previous studies have reported glutamate homeostasis disruptions in astrocytes, leading to imbalances in gamma-aminobutyric acid levels. However, the association between these neurotransmitter changes and cognitive deficits remains inadequately elucidated. Point RESolved Spectroscopy and Hadamard Encoding and Reconstruction of MEGA-Edited Spectroscopy techniques were utilized to evaluate gamma-aminobutyric acid, glutamate, glutathione levels, and excitation/inhibition balance in the anterior cingulate

Adolescent female valproic acid rats have impaired extra-dimensional shifts of attention and enlarged anterior cingulate cortices.

In order to develop better treatments for autism spectrum disorder (ASD) it is critical to understand the developmental trajectory of the disorder and the accompanying brain changes. This study used the valproic acid (VPA) model to induce ASD-like symptoms in rodents. Prior studies have demonstrated that VPA animals are impaired on executive function tasks, paralleling results in humans with ASD. Here, VPA adolescent female rats were impaired on a set-shifting task and had enlarged frontal cortices compared to control females. The deficits observed in the VPA female rats mirrors results in fem
