α-Conotoxins are peptide neurotoxins isolated from the venom ducts of carnivorous marine cone snails that exhibit exquisite pharmacological potency and selectivity for various nicotinic acetylcholine receptor subtypes. As such, they are important research tools and drug leads for treating various diseases of the central nervous system, including pain and tobacco addiction. Despite their therapeutic potential, the chemical synthesis of α-conotoxins for use in structure-activity relationship studies is complicated by the possibility of three disulfide bond isomers, where inefficient folding meth

INTRODUCTION: Central nervous system (CNS) involvement is a common and less understood aspect of systemic lupus erythematosus (SLE). Microembolic signals (MES) have been reported in SLE. We conducted a prospective study to evaluate the frequency of MES among patients with CNS involvement and those without. The main aim of the study is to clarify the pathophysiology of the CNS involvement in SLE.
METHODS AND MATERIALS: Sixty eight patients with a diagnosis of SLE (60 females, 8 males) participated in the study. Both middle cerebral arteries were monitored using transcranial Doppler for 60 min

Some Lyme disease patients report debilitating chronic symptoms of pain, fatigue, and cognitive deficits despite recommended courses of antibiotic treatment. The mechanisms responsible for these symptoms, collectively referred to as post-Lyme disease syndrome (PLS) or chronic Lyme disease, remain unclear. We investigated the presence of immune system abnormalities in PLS by assessing the levels of antibodies to neural proteins in patients and controls. Serum samples from PLS patients, post-Lyme disease healthy individuals, patients with systemic lupus erythematosus, and normal healthy individu

Venoms of spiders and wasps are well recognized to present high affinity to the central nervous tissue of many mammalian species. Here we describe the effects of direct exposure of rat (Rattus norvegicus) brains to the crude and denatured venom of the Brazilian social wasp Polybia ignobilis. Lower doses of crude venom injected via intracerebroventricular (i.c.v.) inhibited the exploratory activity of animals, while higher doses provoked severe generalized tonic-clonic seizures, with hind limb extension. The status epilepticus lasted for few minutes leading the animals to respiratory depression

Altered expression or function of adhesion molecules on leukaemic blasts may contribute to the evolution and biological behaviour of acute leukaemia. This work studies the expression of CD54 and CD62L by lymphoid cells and the serum level of the shed form of L-selectin (sL-selectin) in children with acute lymphoblastic leukaemia (ALL) at initial diagnosis and after first remission, and their relationship to disease activity and subtype. The study is conducted on 20 children (age range 2-10 years) newly diagnosed with ALL and admitted to Alexandria University Children's Hospital. Ten apparently

The content of lactoferrin (LF) was studied in the liquor and blood of patients with tick-borne encephalitis (TBE) of the meningeal and focal types; additionally, the information density of the discussed parameter was assessed for evaluating the severity and degree of the inflammation process in the central nervous system (CNS). The LF level was determined in liquor of 37 samples obtained from TBE patients (main group) and of 10 persons with osteochondrosis (controls); it was also determined in the serum taken from 21 TBE patients and from 40 healthy donors by using the immune-enzyme analysis.

Neuroleptic-induced tardive dyskinesia has been linked to impaired iron homeostasis in the central nervous system attributed to increased iron levels. A chlorpromazine stimulatory effect upon iron uptake from 55Fe-citrate and 55Fe-transferrin by cortical synaptosome preparations of rats was recently demonstrated. The present work extends this study to other neuroleptic drugs such as thioridazine, haloperidol, clozapine and fluphenazine. Like chlorpromazine, thioridazine showed a stimulatory effect upon iron uptake from both iron donors whereas fluphenazine highly increased uptake from 55Fe-cit

Despite advances in medical therapy of congestive heart failure (CHF), morbidity and mortality for this disorder remain high. One factor that could contribute to the poor prognosis of CHF is Cheyne-Stokes respiration with central sleep apnea (CSR-CSA). This breathing disorder is a frequent complication of CHF, where it is associated with increased mortality. One reason for this higher mortality may be that apnea-related hypoxia and arousals from sleep can increase sympathetic nervous system activity (SNA), as manifested by increases in overnight urinary and daytime plasma norepinephrine concen

As central nervous system serotonergic dysregulation has been postulated to exist in both eating and aggression disorders, we hypothesized that anger attacks would be more common among patients with eating disorders than among control subjects. In addition, we wanted to examine possible relationships between the presence of anger attacks and the type or severity of the eating disorder. Subjects were 39 normal female volunteers and 132 female outpatients with anorexia nervosa, bulimia nervosa, or both. Forty-one (31%) of the patients met criteria for anger attacks compared with four (10%) of th

1. Blood pressure, heart rate and plasma catecholamine responses were examined in two groups of elderly subjects distinguished by blood pressure responses to standing. Subjects in the control group showed a fall of less than 15 mmHg in systolic blood pressure on standing; subjects in the orthostatic hypotension group had falls of more than 20 mmHg systolic and 10 mmHg diastolic blood pressure on standing. 2. The heart pressure response on standing showed no significant difference between the two groups. 3. The orthostatic hypotension patients had lower plasma noradrenaline concentrations than

To elucidate the pathophysiology of elevated blood pressure in hypercalcemic patients, we studied the plasma concentration of catecholamines and their major metabolites (as an index of sympathetic function) and the blood pressure response to norepinephrine infusion (vascular reactivity) in patients with primary hyperparathyroidism, in patients with primary hypertension, and in normal controls. In addition, we evaluated the hemodynamic response to calcium infusion in normotensive and hypertensive subjects. Plasma levels of both norepinephrine and epinephrine and the metabolites normetanephrine

Effects of guanfacine, a new centrally acting antihypertensive drug, on the central nervous system were studied in mice and rats, and findings compared with those of clonidine. In mice, guanfacine caused slight hypersensitivity and marked piloerection at 4 or 16 mg/kg, p.o. Clonidine exhibited the same symptoms as guanfacine, but at a higher dose elicited severe central excitation such as clonic convulsion. Such central depressive effects as hyposensitivity and catalepsy were, however, observed following administration of both drugs, at the same dose levels, to rats. Clonidine showed a tendenc

The purpose of this study was to investigate the association between low blood pressure (BP) with mild symptoms of orthostatism, sleep-disordered breathing (SDB) and tilt test results in 7- to 12-y-old children. A retrospective chart review of 301 children, ages 7 to 12 y, was initially performed to evaluate the frequency of abnormal BP measurements. Then a prospective study was performed on 7- to 12-y-old prepubertal children with SDB, looking for both abnormal BP and mild orthostatism. All children had polysomnography. Those identified with abnormal (high or low) BP measurements (called "BP

BACKGROUND: It is claimed that exaggerated blood pressure (BP) reactivity to behavioral stress may play a role in hypertension. This study aimed to clarify whether exaggerated BP reactivity during physical exercise is associated with any other abnormalities in ambulatory BP, autonomic nervous control, and target organ damage among middle-aged normotensive men.
MATERIAL/METHODS: A total of 54 sedentary men (39+/-4 years old) with normal BP were enrolled in this study. Twenty-seven with exaggerated BP reactivity to exercise (ER) were compared with 27 age-matched controls with normal BP reactivi

PTEN loss has been associated with poorer prognosis in many solid tumors. However, such investigation in lymphomas is limited. In this study, PTEN cytoplasmic and nuclear expression, PTEN gene deletion, and PTEN mutations were evaluated in two independent cohorts of diffuse large B-cell lymphoma (DLBCL). Cytoplasmic PTEN expression was found in approximately 67% of total 747 DLBCL cases, more frequently in the activated B-cell-like subtype. Nuclear PTEN expression was less frequent and at lower levels, which significantly correlated with higher PTEN mRNA expression. Remarkably, loss of PTEN pr

INTRODUCTION: Earlier antiretroviral therapy (ART) initiation in cryptococcal meningitis resulted in higher mortality compared with deferred ART initiation (1-2 weeks vs 5 weeks postmeningitis diagnosis). We hypothesized this was due to ART-associated immune pathology, without clinically recognized immune reconstitution inflammatory syndrome.
METHODS: Three macrophage activation markers and 19 cytokines/chemokines were measured from cryopreserved cerebrospinal fluid (CSF) and serum during the Cryptococcal Optimal ART Timing (COAT) trial. Comparisons were made between trial arms (early vs defe

The technique of microneurography has advanced our understanding of the sympathetic nervous system's role in the neurovascular control of blood pressure in humans, yet critical knowledge gaps remain, particularly across the premenopausal female lifespan. Historically, premenopausal females have been considered relatively protected from cardiovascular diseases, leading to a disproportionate focus on postmenopausal populations in cardiovascular research. However, emerging evidence contradicts this assumption: premenopausal females with hypertension exhibit 15-20% higher mortality rates than age-

Neuroinflammation, an immune process in the central nervous system (CNS), is a key contributor to a range of neurological diseases, including neurodegenerative disorders (e.g., Alzheimer's and Parkinson's disease), stroke, and depression, underscoring its significant pathological relevance. While maackiain (MAA) exhibits potent anti-inflammatory activity, its potential to mitigate neuroinflammation remains poorly understood. This study investigated the therapeutic effects of MAA on lipopolysaccharide (LPS)-induced neuroinflammation and its underlying mechanisms. In vitro, MAA significantly imp
