Dynamic changes in histone lysine lactylation during meiosis prophase I in mouse spermatogenesis.

Male germ cells, which are responsible for producing millions of genetically diverse sperm through meiosis in the testis, rely on lactate as their central energy metabolite. Recent study has revealed that lactate induces epigenetic modification in cells through histone lysine lactylation. Here, we report dynamic histone lactylation at histone H4-lysine 5 (K5), -K8, and -K12 during meiosis prophase I in mouse spermatogenesis. By profiling the genome-wide occupancy of histone H4-K8 lactylation (H4K8la), which peaks at zygotene, our data show that H4K8la mark is observed at the promoters of genes

The Role of the Sirtuin Family Histone Deacetylases in Acute Myeloid Leukemia-A Promising Road Ahead.

Acute myeloid leukemia (AML) corresponds to a heterogeneous group of clonal hematopoietic diseases, which are characterized by uncontrolled … processes, and cell survival. They are not only responsible for post-translational modification of histones but also play various biochemical functions and interact with other proteins regulating cell survival, such as p53. Thus, their role in key mechanisms of tumorigenesis makes them a worthwhile topic in AML. Different sirtuins have been shown to act o …

H2B.W2, a spermatocyte-specific histone variant, disrupts nucleosome stability, and reduces chromatin compaction.

Spermatogenesis is a highly regulated process that requires precise chromatin remodeling, which includes the incorporation of testis-specific histone variants. While several of these variants have been characterized, the role of H2B.W2, a member of the H2BW family, remains largely unclear. Here, we showed that H2B.W2 expression occurs mainly in spermatocytes, slightly later than its paralog H2B.W1. Cryo-electron microscopy analysis of H2B.W2-containing nucleosomes reveals a more relaxed conformation compared to canonical nucleosomes caused by weakened interactions between the outer DNA turn an

The histone-methyltransferase DOT1L cooperates with LSD1 to control cell division in blast-phase MPN.

Persistence of JAK2-mutated clones that may undergo clonal evolution and malignant transformation remains a challenge in myeloproliferative neoplasms (MPN), Novel therapeutic approaches to attenuate clonal evolution and progression to blast-phase are therefore urgently needed. LSD1 (KDM1A) inhibitors reduce symptoms and clonal burden in MPN, but whether these compounds may be effective in advanced disease stages remained so far elusive. Using a chromatin-focused CRISPR-Cas9 screen, we identified the histone methyltransferase DOT1L as a synthetic lethal target under pharmacologic LSD1 inhibitio

Histone lysine demethylases in breast cancer: molecular mechanisms, biological functions, and therapeutic intervention.

Breast cancer is a highly heterogeneous disease characterized by diverse molecular subtypes and complex pathogenesis. Recent advances in epigenetics have unveiled the crucial roles of lysine demethylases (KDMs) in modulating gene expression and chromatin dynamics, thereby influencing breast cancer progression, including metastasis, and therapeutic resistance. KDMs, which remove methyl groups from histone lysine residues, are mainly categorized into seven subfamilies (KDM1-7) based on their catalytic mechanisms and substrate specificities. Meanwhile, each subfamily exhibits distinct roles in br

Histone acetylation by SAGA complex but not by NuA4 complex is required for filamentation program in Candida albicans.

Candida albicans, a major human fungal pathogen undergoes filamentation from yeast to hyphal state under filamentation-inducing conditions. Gcn5 and Esa1 are key histone H3 and H4 acetyltransferases, respectively, encoded by the budding yeast and other eukaryotes. While Gcn5, a subunit of the SAGA complex, and Esa1, a subunit of the NuA4 complex, are critical for C. albicans virulence and hyphal induction, how the relative HAT activities impinge on hyphal gene expression during filamentation is less understood. We found that hyphal gene promoters are hyperacetylated at H3K9 and H4 upon filamen

Quercetin inhibits platelet activation and ER-stress mediated autophagy in response to extracellular histone.

BACKGROUND: Cellular histones are DNA-binding nuclear proteins involved in chromatin remodelling and regulation of gene expression. However, extracellular histones act as damage-associated molecular patterns (DAMPs) and contribute to multiorgan damage in conditions with sepsis and diseases with acute critical illnesses. Alongside, histones are associated with thrombocytopenia due to dysfunctional platelets that regulate hemostasis and thrombosis. There is no drug available to prevent histone-induced platelet toxicity. Therefore, we for the first time examined quercetin (QUE) as a novel therape…

Identification and validation of a histone modification-related gene signature to predict the prognosis of multiple myeloma.

BACKGROUND: Multiple myeloma (MM) is an incurable plasma cell malignancy with high heterogeneity. Current staging systems, including the International Staging System (ISS) and Revised ISS (R-ISS), have limited prognostic accuracy. Given the role of histone modifications in MM progression, we developed a histone modification-related (HMR) prognostic model to improve MM risk stratification. METHODS: Gene expression and mutation data were downloaded from the Gene Expression Omnibus database and the Cancer Genome Atlas. Prognostic HMR-related genes were identified through a combination of univari

Inhibition of histone-lysine N-methyltransferase G9a exacerbates alcoholic steatohepatitis (ASH) in mice.

Alcoholic steatohepatitis (ASH) is characterized by hepatic steatosis, inflammation, and alcohol-induced liver damage in humans. G9a, a histone methyltransferase, serves as a multipotent regulator of gene expression and plays a significant role in hepatic drug metabolism and cellular stress. Although epigenetic alterations occur during alcoholic liver disease progression, G9a's precise contribution to ASH pathogenesis remains unclear. Using a murine model of ASH, we demonstrate that G9a expression is downregulated during ASH progression, and G9a pharmacological inhibition exacerbates ASH sever

Mutations in histone lysine methyltransferase genes are associated with autoimmune cytopenias: a single-center study.

Epigenetic dysregulation is increasingly recognized as a contributor to autoimmunity and autoimmune cytopenias (AIC). Histone lysine methyltransferases (MTs) are key regulators of gene expression through epigenetic modification. Germline MT mutations are associated with immunodeficiency syndromes such as Kabuki syndrome, which frequently co-occurs with AIC, while somatic KMT2D mutations have been reported in cold agglutinin disease. This study aimed to (1) determine the frequency of mutations in histone methyltransferase genes (KMT2D, KMT2A, KMT2C, and KDM6A) among patients with AIC, and (2) c

Long-term histone lactylation connects metabolic and epigenetic rewiring in innate immune memory.

Trained immunity, a de facto innate immune memory characterized by enhanced responsiveness to future challenges, is underpinned by epigenetic and metabolic rewiring. In individuals vaccinated with Bacille Calmette-Guérin (BCG), lactate release was associated with enhanced cytokine responsiveness upon restimulation. Trained monocytes/macrophages are characterized by lactylation of histone H3 at lysine residue 18(H3K18la), mainly at distal regulatory regions. Histone lactylation was positively associated with active chromatin and gene transcription, persisted after the elimination of the trainin

Histone Lactylation Antagonizes Senescence and Skeletal Muscle Aging by Modulating Aging-Related Pathways.

Epigenetic alterations are among the prominent drivers of cellular senescence and/or aging, intricately orchestrating gene expression programs during these processes. This study shows that histone lactylation, plays a pivotal role in counteracting senescence and mitigating dysfunctions of skeletal muscle in aged mice. Mechanistically, histone lactylation and lactyl-CoA levels markedly decrease during cellular senescence but are restored under hypoxic conditions primarily due to elevated glycolytic activity. The enrichment of histone lactylation at promoters is essential for sustaining the expr

Histone methyltransferase G9a drives ferroptosis to aggravate vascular calcification by inhibiting SLC7A11 transcription.

Vascular calcification (VC) exacerbates the risk of cardiovascular morbidity and mortality in individuals with chronic kidney disease (CKD), and recent advances in pathogenesis have highlighted the significance of ferroptosis. The histone methyltransferase G9a participates in the regulation of different types of cell death including ferroptosis, but its role in VC needs to be further explored. Here, we found that G9a expression was elevated in microarray data obtained from CKD mouse VSMC specimens, and further experiments demonstrated similar results in CKD patients, mouse calcified arteries a

Targeting histone H2B acetylated enhanceosomes via p300/CBP degradation in prostate cancer.

Prostate cancer is driven by oncogenic transcription factor enhanceosomes comprising chromatin and epigenetic regulators. The lysine acetyltransferases p300 and CREB-binding protein (CBP) are key cofactors that activate enhancers through histone acetylation. Here we identify p300/CBP-mediated multisite histone H2B N-terminal acetylation (H2BNTac) as a defining feature of oncogenic enhanceosomes in androgen receptor (AR)-positive prostate cancer. p300/CBP are essential for AR and ETS transcription factor ERG transcriptional activity, and their dual degradation eliminates H2BNTac and histone H3

Nitric oxide delays floral transition in Arabidopsis by inhibiting histone deacetylases HDA5 and HDA6.

Nitric oxide (NO), a reactive small molecule, plays a critical role in various developmental and physiological processes in living organisms. Previous studies by our group revealed that NO delays flowering in Arabidopsis by increasing transcript levels of the flowering repressor FLOWERING LOCUS C (FLC). In this study, we further investigated the molecular mechanism by which NO regulates FLC expression. Genetic experiments demonstrated that NO-induced delayed flowering specifically depends on elevated FLC transcript levels. Chromatin Immunoprecipitation assays revealed that NO significantly enh

IGF2BP3 recognizes m6A to regulate histone-to-protamine replacement during mouse sperm development.

Post-meiotic development of spermatids is under the control of a sophisticated RNA metabolic network, wherein the N6-methyladenosine (m6A) modification of mRNA, and proteins that bind to it, exert crucial functions in regulating sperm development from spermatogonia to spermatocytes. However, which m6A recognition proteins are involved in male post-meiotic spermiogenesis, and via which regulatory mechanisms, remains largely unknown. Here, we uncover the involvement of the m6A reader protein IGF2BP3 in the regulation of post-meiotic spermatid development. Genetic ablation of Igf2bp3 results in s
