How to invade a susceptible host: cellular aspects of aspergillosis.

Diseases caused by Aspergillus spp. and in particular A. fumigatus are manifold and affect individuals suffering from immune dysfunctions, among them immunocompromised ones. The determinants of whether the encounter of a susceptible host with infectious propagules of this filamentous saprobe results in infection have been characterized to a limited extent. Several cellular characteristics of A. fumigatus that have evolved in its natural environment contribute to its virulence, among them general traits as well as particular ones that affect interaction with the mammalian host. Among the latter

Applying Phage Display Technology to Obtain Specific Peptides Blocking Host-Virus Interactions.

Phage display (PhD) has been proven to be a powerful tool for studying interactions when at least one partner of the interplay is of protein or peptide nature. In this chapter, PhD was utilized to search for peptides potentially binding onto the receptor-binding domain of spike proteins (S-RBD) of the SARS-CoV-2 virus. After three rounds of PhD selection, two peptides were identified to be able to dock onto the positions critical for S-RBD functions, validating the usefulness of PhD in the studies concerning host-virus interactions.

Long non-coding RNA U90926 modulates IFN-γ-stimulated gene transcription and cell-intrinsic anti-Cryptosporidium defense in intestinal epithelial cells.

Cryptosporidium infects the intestine in a wide variety of vertebrates, and intestinal epithelial cells provide the first line of defense against Cryptosporidium infection. Interferon gamma (IFN-γ) from immune cells infiltrated at the site of infection plays a key role in the epithelial cell-intrinsic defense. Nevertheless, the success of the parasite is the result of its ability to evade the host immune responses. Increasing evidence suggests that long noncoding RNAs (lncRNA) participate in host-pathogen interactions, but the underlying mechanisms are not fully understood. We previously demon…

Molecular evidence for bacterial and protozoan pathogens in hard ticks from Romania.

The aim of the present study was to provide a preliminary insight into the diversity of tick-borne pathogens circulating at the domestic host-tick interface in Romania. For this, feeding and questing ticks were analyzed by real-time polymerase chain reaction (PCR) for the presence of Anaplasma phagocytophilum, Anaplasma platys, Ehrlichia canis, Borrelia burgdorferi sensu latu, and by PCR and subsequent sequencing for Rickettsia spp., Babesia spp. and Theileria spp. A total of 382 ticks, encompassing 5 species from 4 genera, were collected in April-July 2010 from different areas of Romania; of

Linking disease and community ecology through behavioural indicators: immunochallenge of white-footed mice and its ecological impacts.

1. Pathogens and immune challenges can induce changes in host phenotype in ways that indirectly impact important community interactions, including those that affect host-pathogen interactions. 2. To explore host behavioural response to immune challenge, we exposed wild white-footed mice (Peromyscus leucopus) to an immunogen from an endemic, zoonotic pathogen, the spirochete Borrelia burgdorferi. White-footed mice are a major reservoir host of Lyme disease (LD) spirochetes in northeastern USA and an abundant member of forest communities. The activity patterns, foraging behaviour, and space use …

Clinical and mechanistic insights into Brucella and viral co-infection.

Co-infection of Brucella and viruses represents an emerging clinical and epidemiological challenge. Recent evidence reveals that Brucella-viral co-infections are characterized by atypical clinical manifestations and increased diagnostic complexity. These dual infections involve intricate pathogen-host and pathogen-pathogen interactions: viruses enhance Brucella susceptibility by subverting innate and adaptive immune responses, while Brucella compromises host immunity to create a permissive microenvironment for viral persistence. Furthermore, co-infection synergistically exacerbates tissue dama

Use of OmpU porins for attachment and invasion of Crassostrea gigas immune cells by the oyster pathogen Vibrio splendidus.

OmpU porins are increasingly recognized as key determinants of pathogenic host Vibrio interactions. Although mechanisms remain incompletely understood, various species, including the human pathogen Vibrio cholera, require OmpU for host colonization and virulence. We have shown previously that OmpU is essential for virulence in the oyster pathogen Vibrio splendidus LGP32. Here, we showed that V. splendidus LGP32 invades the oyster immune cells, the hemocytes, through subversion of host-cell actin cytoskeleton. In this process, OmpU serves as an adhesin/invasin required for β-integrin recognitio

Concurrent Host-Pathogen Transcriptional Responses in a Clostridium perfringens Murine Myonecrosis Infection.

To obtain an insight into host-pathogen interactions in clostridial myonecrosis, we carried out comparative transcriptome analysis of both the bacterium and the host in a murine Clostridium perfringens infection model, which is the first time that such an investigation has been conducted. Analysis of the host transcriptome from infected muscle tissues indicated that many genes were upregulated compared to the results seen with mock-infected mice. These genes were enriched for host defense pathways, including Toll-like receptor (TLR) and Nod-like receptor (NLR) signaling components. Real-time P…

Tuberculosis and Cellular Metabolism: Insights into the Crosstalk Between Macrophage Immunometabolism and Muscle Dysregulation.

Tuberculosis (TB) remains a leading cause of death from a single infectious agent, and its outcome is shaped not only by Mycobacterium tuberculosis (Mtb) itself, but also by the host's metabolic state. This review synthesises current understanding of how Mtb reprograms macrophage immunometabolism and how this reprogramming propagates to a systemic level, culminating in skeletal muscle dysregulation and TB-associated cachexia. We describe the molecular mechanisms by which Mtb subverts phagosomal maturation, the glycolytic (Warburg-like) switch governed by HIF-1α and accumulation of immunomodula

Systematic exploration of Escherichia coli phage-host interactions with the BASEL phage collection.

Bacteriophages, the viruses infecting bacteria, hold great potential for the treatment of multidrug-resistant bacterial infections and other applications due to their unparalleled diversity and recent breakthroughs in their genetic engineering. However, fundamental knowledge of the molecular mechanisms underlying phage-host interactions is mostly confined to a few traditional model systems and did not keep pace with the recent massive expansion of the field. The true potential of molecular biology encoded by these viruses has therefore remained largely untapped, and phages for therapy or other

Cooperative secretions facilitate host range expansion in bacteria.

The majority of emergent human pathogens are zoonotic in origin, that is, they can transmit to humans from other animals. Understanding the factors underlying the evolution of pathogen host range is therefore of critical importance in protecting human health. There are two main evolutionary routes to generalism: organisms can tolerate multiple environments or they can modify their environments to forms to which they are adapted. Here we use a combination of theory and a phylogenetic comparative analysis of 191 pathogenic bacterial species to show that bacteria use cooperative secretions that m

Haemophilus ducreyi Infection Induces Oxidative Stress, Central Metabolic Changes, and a Mixed Pro- and Anti-inflammatory Environment in the Human Host.

Few studies have investigated host-bacterial interactions at sites of infection in humans using transcriptomics and metabolomics. Haemophilus ducreyi causes cutaneous ulcers in children and the genital ulcer disease chancroid in adults. We developed a human challenge model in which healthy adult volunteers are infected with H. ducreyi on the upper arm until they develop pustules. Here, we characterized host-pathogen interactions in pustules using transcriptomics and metabolomics and examined interactions between the host transcriptome and metabolome using integrated omics. In a previous pilot …

High-Throughput Chip Assay for Investigating Escherichia coli Interaction with the Blood-Brain Barrier Using Microbial and Human Proteome Microarrays (Dual-Microarray Technology).

Bacterial meningitis in neonates and infants is an acute lethal disease and occurs in response to microbial exploitation of the blood-brain barrier (BBB), resulting in the intracranial inflammation. Several pathogens, such as Escherichia coli ( E. coli), can cause this devastating disease; however, the underlying molecular mechanisms by which these pathogens exploit the BBB remain incompletely understood. To identify important players on both the pathogen and host sides that govern the E. coli-BBB cell interactions, we took advantage of the E. coli and human proteome microarrays (i.e., HuProt)

A Mitocentric View of the Main Bacterial and Parasitic Infectious Diseases in the Pediatric Population.

Infectious diseases occur worldwide with great frequency in both adults and children. Both infections and their treatments trigger mitochondrial interactions at multiple levels: (i) incorporation of damaged or mutated proteins to the complexes of the electron transport chain, (ii) mitochondrial genome (depletion, deletions, and point mutations) and mitochondrial dynamics (fusion and fission), (iii) membrane potential, (iv) apoptotic regulation, (v) generation of reactive oxygen species, among others. Such alterations may result in serious adverse clinical events with great impact on children's

The roles of galectins in parasitic infections.

Galectins is a family of multifunctional lectins. Fifteen galectins have been identified from a variety of cells and tissues of vertebrates and invertebrates. Galectins have been shown to play pivotal roles in host-pathogen interaction such as adhesion of pathogens to host cells and activation of host innate and adaptive immunity. In recent years, the roles of galectins during parasite infections have gained increasing attention. Galectins produced by different hosts can act as pattern recognition receptors detecting conserved pathogen-associated molecular patterns of parasites, while galectin

Recurring principles of metabolic interactions in Enterococcus faecalis polymicrobial communities.

Enterococcus faecalis is a human gut commensal and major opportunistic pathogen that is frequently found within polymicrobial communities at biofilm-associated infection sites, including the catheterized urinary tract, surgical and chronic wounds, the gut, and oral and mucosal niches. Its interactions with co-isolated bacteria and fungi can alter biofilm formation, antimicrobial susceptibility, immune signaling, and disease severity, yet the molecular basis of these interactions, and whether the same mechanisms recur across anatomically distinct sites, remains incompletely understood. In this

MybA, a new player driving survival of the conidium of the human pathogen Aspergillus fumigatus.

Aspergillus fumigatus is an opportunistic human pathogen that causes various complications in patients with a weakened immune system functions. Asexual spores of A. fumigatus are responsible for initiation of aspergillosis. Long-term viability and proper germination of dormant conidia depend on trehalose accumulation, which protect the spores against thermal and oxidative stress. A putative Myb transcription factor, MybA has been recently found to be responsible for a variety of physiological and molecular roles ranging from conidiation, spore viability, trehalose accumulation, cell wall integ
