Establishing and characterizing human periodontal ligament fibroblasts immortalized by SV40T-antigen and hTERT gene transfer.

The periodontal ligament (PDL) is a highly specialized tissue connecting the cementum with the tooth socket bone and affects the life span of the tooth. However, little is known about the precise characteristics and regenerative mechanism of PDL cells because of the absence of specific markers and cell lines. Therefore, we aimed to establish three immortalized human PDL fibroblast cell lines by using simian virus40 T-antigen (SV40T-Ag) and human telomerase reverse transcriptase (hTERT) transfection, expecting these cells to have the characteristics of primary cells. The transfected cells were

[Mitochondrial DNA mutations and related human diseases].

In the past two decades, it has been found that mitochondrial DNA (mtDNA) mutations are associated with a wide range of human diseases, from those affecting single organ to those with multi-system involvement. The purpose of this review is to explore the relationship between mtDNA mutations and human diseases. Four aspects are highlighted: characteristics of mitochondrial genetics, mtDNA mutations in human inherited diseases, role of somatic mtDNA mutations in aging and tumor, as well as diagnosis and treatment of mtDNA diseases.

Neighbourhood environments and later life flourishing: A systematic scoping review.

BACKGROUND: Enabling older people to live comfortably in their own neighbourhoods for longer, known as ageing-in-place, has become a policy priority to reduce pressures on health and social care systems amid global population ageing. However, research on place and health remains predominantly deficit-focused, emphasising illness and decline (e.g. frailty, depression, and anxiety). This review adopts a positive psychology perspective and a life-course framework to synthesise evidence on how neighbourhood environments support flourishing in later life, a multidimensional state encompassing happi

The association between neighborhood walkability and postpartum weight retention: A prospective cohort study.

OBJECTIVES: Postpartum weight retention is common and contributes to overweight and obesity. Living in a more walkable neighborhood may influence postpartum weight retention by encouraging physical activity and overall weight management. This study explores the potential association between neighborhood walkability and postpartum weight retention. STUDY DESIGN: The P3 Cohort is a prospective pregnancy cohort study. Participants were recruited at <32 weeks gestation and followed until 12 months postpartum. METHODS: The proportion of postpartum weight retention was calculated by dividing the d

Chronic health conditions and trajectories of depressive symptoms in late adulthood: role of age-friendly community.

BACKGROUND: Contextual factors influence how older adults perceive and adapt to chronic health conditions. We examined the impact of chronic health conditions on trajectories of depressive symptoms among older Chinese adults and investigated whether age-friendly communities buffer this effect. METHODS: We used panel data from the China Health and Retirement Longitudinal Study from 2011 to 2018, including 6122 adults aged ≥50 years (22 198 observations). Factor analysis of mixed data (FAMD) was performed to construct composite variables of community characteristics. The cross-level interaction

Fast-food and convenience outlets near schools in California: a comparison of private and public schools.

Food environment near schools (FENS) influence children's dietary habits and contribute to obesity. Socioeconomic characteristics of schools and school neighborhoods play a role in determining FENS. We compare the availability of fast-food restaurants (FFRs) and convenience stores (CSs) across schools' socioeconomic characteristics: whether the school is public or private and the school neighborhood's median household income. We obtained the number of FFRs and CSs within a 0.75-mile network buffer from schools' locations and the names of the outlets. Negative binomial regression models, strati

Mapping food access: how neighborhood deprivation shapes healthy food availability in the United Kingdom.

BACKGROUND: Limited access to affordable, nutritious food is a key structural determinant of health inequality. In the United Kingdom, food desert risk is often assumed to mirror socioeconomic deprivation, yet their alignment across small-area geographies remains unclear. OBJECTIVES: To characterize the spatial distribution of food desert risk across England, Wales, and Scotland; quantify its association and concordance with area-level deprivation; and identify deprivation domains most strongly linked to food desert risk. METHODS: We integrated the E-Food Desert Index (EFDI) with national Indi

Neighborhood Social Fragmentation and Excessive Body Weight Among Children-A View From a Portuguese Inner City.

This cross-sectional study examined the association between neighborhood social fragmentation and excessive body weight in childhood. The sample included 832 children aged 3-10 years living in the municipality of Porto, Portugal. Excessive body weight was classified using International Obesity Task Force sex- and age-specific cut-offs. Neighborhood social fragmentation was assessed using a composite index derived from 2021 Portuguese Census data at the statistical section level. Associations were estimated using sex-stratified multivariable logistic regression adjusted for relevant confounders

Neighborhood environment associations with cognitive function and structural brain measures in older African Americans.

BACKGROUND: Since older adults spend significant time in their neighborhood environment, environmental factors such as neighborhood socioeconomic disadvantage, high racial segregation, low healthy food availability, low access to recreation, and minimal social engagement may have adverse effects on cognitive function and increase susceptibility to dementia. DNA methylation, which is associated with neighborhood characteristics as well as cognitive function and white matter hyperintensity (WMH), may act as a mediator between neighborhood characteristics and neurocognitive outcomes. METHODS: In

Impact of neighborhood income inequality on adolescents' mental health. Results from the STARS study.

Over recent decades, there has been a simultaneous increase in income inequality and adverse mental health outcomes among adolescents in Western countries and especially in Sweden. Prior research on the relationship between income inequality and mental health has primarily focused on studies involving adults, yielding diverse findings regarding the nature of their association. Given the importance of relationships and comparisons to peers in immediate vicinity, we aimed to investigate the impact of neighborhood income inequality on mental health problems among seven-graders in Western Sweden.

Neighbourhood Deprivation and Gestational Diabetes Mellitus in Arizona From the AzPEARS Study.

BACKGROUND: The maternal population residing in rural areas, tribal lands or near the United States-Mexico border are at high risk for adverse pregnancy complications, notably gestational diabetes mellitus (GDM). Few studies have considered the socioeconomic status (SES) attributes of neighbourhoods on maternal health during gestation. Given that the national and global rate of GDM is rising, this analysis moves beyond individual-level factors and investigates the role of neighbourhood deprivation on GDM risk. OBJECTIVE: To investigate the relationship between neighbourhood deprivation and ri

Derivation of the MUSIe002-A human embryonic stem cell line.

The MUSIe002-A cell line was established from in vitro fertilization of human sperm and oocytes donated for research with informed consent. This cell line exhibited normal human embryonic stem cell (hESC) characteristics, including typical cell morphology, expression of all pluripotent stem cell markers, and potential to differentiate into three germ layers. A karyotyping analysis revealed 46 XY chromosome and cells that did not have mycoplasma contamination. MUSIe002-A represents a valuable unlimited cell source and is of potential interest for human in vitro stem cell based-models, genetic m

Challenges and Limits in Explaining and Acoustic Modeling of Voice Characteristics.

To this day, the assessment of human voices remains a challenge due to (i) inconsistencies in subjective ratings and (ii) the lack of objective measurements for the perceptual impressions of voice characteristics. This can lead to significant consequences in applied fields such as speech therapy, where the assessment of voices is crucial for a successful treatment. In this paper, we address the explanation of voice and its characteristics from two different angles: In a first study, 22 speech therapists in training assessed a set of 20 non-pathological voices regarding 20 voice characteristics

[Monthly dynamics of the antitoxic immunity against diphtheria and tetanus and of normal antibodies to fraction I of the plague microbe in adults].

Monthly characteristics of antitoxic immunity to diphtheria and tetanus toxoids in 3,334 adults in Ryazan were determined. As a result, the following differences were established: the characteristics of antidiphtheria immunity were somewhat higher in the autumn-winter period and dropped to the minimal level in the winter-spring period; the maximal characteristics of antitetanus immunity clearly coincided with the hot season, its minimal characteristics with the cold season. For the first time the frequency and titers of normal antibodies to fraction I of Pasteurella pestis in humans were deter

The human tail: a benign stigma. Case report.

Thirty-three cases of true human tails have been reported in the modern English literature. A new case is described and its radiological and pathological features are presented. A review of the literature and analysis of the pathological characteristics of this interesting lumbosacral stigma indicate that the true human tail is a benign condition not associated with any underlying cord malformation.

Direct reprogramming of human Sertoli cells into male germline stem cells with the self-renewal and differentiation potentials via overexpressing DAZL/DAZ2/BOULE genes.

We propose a new concept that human somatic cells can be converted to become male germline stem cells by the defined factors. Here, we demonstrated that the overexpression of DAZL, DAZ2, and BOULE could directly reprogram human Sertoli cells into cells with the characteristics of human spermatogonial stem cells (SSCs), as shown by their similar transcriptomes and proteomics with human SSCs. Significantly, human SSCs derived from human Sertoli cells colonized and proliferated in vivo, and they could differentiate into spermatocytes and haploid spermatids in vitro. Human Sertoli cell-derived SSC

Human blastoid as an in vitro model of human blastocysts.

Human development is a highly coordinated process, with any abnormalities during the early embryonic stages that can often have detrimental consequences. The complexity and nuances of human development underpin its significance in embryo research. However, this research is often hindered by limited availability and ethical considerations associated with the use of donated blastocysts from in vitro fertilization (IVF) surplus. Human blastoids offer promising alternatives as they can be easily generated and manipulated in the laboratory while preserving key characteristics of human blastocysts.
