Telomere-to-telomere genome of Stylosanthes guianensis uncovers symbiotic adaptation to phosphorus-deficient soils.

BACKGROUND: Stylosanthes guianensis, a representative tropical legume, exhibits remarkable adaptation to low-phosphorus acidic soils. As a symbiotic species, it forms root nodule associations with rhizobia to fix atmospheric nitrogen, potentially enhancing phosphate use efficiency. This study aims to decipher the mechanisms linking root nodule symbiosis to low-phosphate adaptation in Stylosanthes guianensis. RESULTS: We present the first gap-free, telomere-to-telomere genome of Stylosanthes guianensis (1.20 Gb), containing 82.28% repetitive sequences and 34,728 genes, with 99.30% BUSCO comple

Impact of Ethanol Stress on Yarrowia lipolytica for Sustainable Bioconversion of Agro-Food Oil Wastes into Lipases and Lipids.

Ethanol stress profoundly affects yeast metabolism, yet its integrated impact on lipase activity and lipid remodeling in Yarrowia lipolytica remains unexplored. Here, we investigated, for the first time, the combined effects of ethanol-induced stress on lipase production and fatty acid profiles in Y. lipolytica cultivated on two hydrophobic substrates: olive mill wastewater (OMW) and Waste Frying Oil (WFO). Ethanol was applied at increasing concentrations (3%, 5%, and 7% v/v), and the physiological responses were monitored over time (48, 72, and 96 h). Our results reveal a substrate-dependent

Comprehensive transcriptomic analysis in wild-type and ATM knockout lung cancer cells: Influence of cisplatin on oxidative stress-induced senescence.

Genetic mutations and impaired DNA repair mechanisms in cancer not only facilitate tumor progression but also reduce the effectiveness of chemotherapeutic agents, particularly cisplatin. Combination therapy has emerged as a promising strategy to overcome resistance. Comprehensive transcriptomic analyses, supported by integrated comparative bioinformatics and experimental approaches, are essential for identifying biomarkers and novel therapeutic targets underlying drug resistance. In this study, we performed overall survival and mutation analyses, examining 23 double-strand break repair protein

Trauma-induced insomnia: A novel model for trauma and sleep research.

Traumatic events have been increasingly recognized as important precipitants of clinically significant insomnia. Trauma is an extreme form of stressful life event that generates a sustained neurobiological response triggering the onset and maintenance of insomnia. Trauma may disrupt the normal sleep-wake regulatory mechanism by sensitizing the central nervous system's arousal centers, leading to pronounced central and physiological hyperarousal. The central concept of hyperarousal has been linked to both the pathogenesis of insomnia and to the neurobiological changes in the aftermath of trauma

Exploring the potential mechanism of Rhodomyrtus tomentosa (Ait.) Hassk fruit phenolic rich extract on ameliorating nonalcoholic fatty liver disease by integration of transcriptomics and metabolomics profiling.

Nonalcoholic fatty liver disease (NAFLD), as the commonest form of chronic liver disease, is accompanied by liver oxidative stress and inflammatory responses. Rhodomyrtus tomentosa (Ait.) Hassk fruit phenolic rich extract (RTE) possesses multiple pharmacological effects in management of chronic diseases. In this study, the liver-protective effect of RTE on mice with high-fat-diet (HFD)-induced NAFLD was investigated for the first time, and the underlying molecular mechanism was explored via integration of transcriptomics and metabolomics. The results showed that RTE mitigated liver damage, whi

Common genes and pathways involved in the response to stressful stimuli by astrocytes: A meta-analysis of genome-wide expression studies.

Astrocytes play pivotal roles in the brain and they become reactive under stress conditions. Here, we carried out, for the first time, an integrative meta-analysis of genome-wide expression profiling of astrocytes from human and mouse exposed to different stressful stimuli (hypoxia, infections by virus and bacteria, cytokines, ethanol, among others). We identified common differentially expressed genes and pathways in human and murine astrocytes. Our results showed that astrocytes induce expression of genes associated with stress response and immune system regulation when they are exposed to st

Iron-based nanomaterials for tumor therapy: structural engineering and functional mechanisms.

Iron-based nanomaterials are transforming cancer therapeutics through their multifunctional properties, offering solutions to persistent clinical barriers such as systemic toxicity, tumour hypoxia, immune suppression, and resistance to standard treatments. Their intrinsic catalytic properties, particularly Fenton-like activity, enables in situ ROS generation, while magnetic responsiveness facilitates localized hyperthermia and synergistic therapeutic activation. Beyond direct cytotoxicity, these platforms trigger ferroptosis and oxidative stress mediated cell death, modulating the tumour micro

Uncertainty aware resilience enhancement of energy hub integrated distribution networks under heterogeneous disturbance events.

This study develops a 24 h mixed integer linear programming framework for resilient operation of an integrated electricity, gas, and heat distribution system with energy hubs under prescribed outages and operating uncertainty. Electrical and thermal demand and wind and photovoltaic shortfalls are represented through budgeted polyhedral adverse point scheduling. Five cases compare a no-event reference, a combined outage baseline, radial reconfiguration, energy storage, and coordinated electrical and thermal demand response on the IEEE 33-bus system. Energy storage provides the strongest overall

Mitochondrial transfer in sepsis: Dual role, mechanistic networks, and translational strategies.

Sepsis is a life-threatening clinical syndrome characterized by dysregulated host response, metabolic disturbance, and multiple organ dysfunction. Mitochondrial damage and bioenergetic failure are core pathological events driving sepsis progression. As a critical intercellular communication mechanism, mitochondrial transfer (MT) participates in mitochondrial quality control, energy homeostasis, and inflammatory regulation under septic stress. This review systematically summarizes the structural and functional mitochondrial injury in sepsis and endogenous quality control pathways. We focus on t

Atypical tetracyclines promote longevity and ferroptotic neuroprotection via translation attenuation.

Preclinical and clinical studies have reported neuroprotective and geroprotective effects of tetracyclines that are independent of their ant … e, inhibit the mitochondrial ribosome and attenuate cytosolic translation through activation of the Integrated Stress Response (ISR). In contrast, atypical tetracyclines such as 4-epiminocycline and 12-aminominocycline act as cytosolic-targeting tetracyclines (CytoTets), directly inhibiting the cytosolic ribosome, bypassing the ISR, and protecting neurons from ferroptotic c …

Validamycin A Induced Resistance in Rice against Rhizoctonia solani via the Salicylic Acid-Depended Signaling Pathway.

Validamycin A (VMA), the major active component of validamycin, provides effective control of rice sheath blight; however, the mechanisms underlying VMA-induced resistance to Rhizoctonia solani remain poorly understood. Here, we integrated antioxidant enzyme assays, phytohormone profiling, transcriptomics, and metabolomics to investigate VMA-induced immunity in rice. VMA improved oxidative stress tolerance by enhancing antioxidant enzyme activities and activating glutathione metabolism. Moreover, VMA established a primed multihormonal state characterized by elevated salicylic acid (SA), jasmon

The potential link between inflammatory bowel disease and male erectile dysfunction: mechanistic insights and novel therapeutic perspectives.

This review provides a comprehensive synthesis of the link between inflammatory bowel disease (IBD) and male erectile dysfunction (ED), with a distinct emphasis on underlying mechanisms and novel perspectives. We critically evaluate the evidence and then systematically elucidate the "gut-penis axis" detailing how gut-derived signals orchestrate a systemic inflammatory response that culminates in penile vascular dysfunction. A novel aspect of our work is the integration of psychological factors into a cohesive psychoneuroimmunological framework, linking stress, the cholinergic anti-inflammatory

PPARα regulates ER-lipid droplet protein Calsyntenin-3β to promote ketogenesis in hepatocytes.

Ketogenesis requires fatty acid flux from intracellular (lipid droplets) and extrahepatic (adipose tissue) lipid stores to hepatocyte mitoch … y changes in fatty acid oxidation, lipid-dependent mitochondrial respiration, and the mitochondrial integrated stress response. These findings define a function for CLSTN3β-dependent membrane contacts in hepatic lipid utilization and ketogenesis.

Design and Development of a Bioorthogonal, Visualizable and Mitochondria-Targeted Hydrogen Sulfide (H2 S) Delivery System.

Hydrogen sulfide (H2 S) is an important endogenous gasotransmitter, but the targeted delivery and real-time feedback of exogenous H2 S are still challenging. With the aid of density functional theory (DFT) calculations, we designed a new 1,3-dithiolium-4-olate (DTO) compound, which can react with a strained alkyne via the 1,3-dipolar cycloaddition and the retro-Diels-Alder reaction to generate carbonyl sulfide (COS) as the precursor of H2 S, and a thiophene derivative with turn-on fluorescence. Moreover, the diphenylamino substituent in DTO greatly increases the mitochondrial targeting of this

Engineering multifunctional bactericidal nanofibers for abdominal hernia repair.

The engineering of multifunctional surgical bactericidal nanofibers with inherent suitable mechanical and biological properties, through facile and cheap fabrication technology, is a great challenge. Moreover, hernia, which is when organ is pushed through an opening in the muscle or adjacent tissue due to damage of tissue structure or function, is a dire clinical challenge that currently needs surgery for recovery. Nevertheless, post-surgical hernia complications, like infection, fibrosis, tissue adhesions, scaffold rejection, inflammation, and recurrence still remain important clinical proble

Chloroplast Signaling Gates Thermotolerance in Arabidopsis.

Temperature is a key environmental variable influencing plant growth and survival. Protection against high temperature stress in eukaryotes is coordinated by heat shock factors (HSFs), transcription factors that activate the expression of protective chaperones such as HEAT SHOCK PROTEIN 70 (HSP70); however, the pathway by which temperature is sensed and integrated with other environmental signals into adaptive responses is not well understood. Plants are exposed to considerable diurnal variation in temperature, and we have found that there is diurnal variation in thermotolerance in Arabidopsis

Ribotoxic Stress, ZAK Activation and the Orchestration of Inflammatory Signaling Networks in Human Disease.

INTRODUCTION: Ribotoxic stress is an evolutionarily conserved intracellular surveillance mechanism that detects disturbances in translational fidelity and links ribosomal damage to inflammatory and stress-responsive signaling pathways. Diverse pathological stimuli, including microbial toxins, oxidative stress, metabolic dysfunction, and oncogenic translational overload, disrupt ribosome integrity and induce ribosome stalling and collision events. This review aims to summarize current advances in the molecular mechanisms underlying ribotoxic stress sensing, with particular emphasis on the ribos

Cigarette Smoke Extract Exposure Affects Innate Immune Response, Metabolic Rate, and Locomotor Activity of Drosophila melanogaster.

PURPOSE: Cigarette smoke (CS) disrupts innate immune homeostasis through oxidative stress and inflammatory signaling, contributing to respiratory diseases such as asthma and COPD. Because Drosophila melanogaster shares approximately 75% of disease-related human genes and possesses a well-characterized innate immune system without confounding adaptive responses, it represents a tractable model for investigating smoke-induced pathology. METHODS: This study examined the effects of developmental cigarette smoke extract (CSE) exposure on innate immune activation, locomotor activity, and metabolic

Impairment of ribosome maturation or function confers salt resistance on Escherichia coli cells.

We found that loss of integrity of the ribosome by removal of a putative ribosome maturation factor or a ribosomal protein conferred salt tolerance on Escherichia coli cells. Some protein synthesis inhibitors including kasugamycin and chloramphenicol also had a similar effect, although kasugamycin affected neither 16S rRNA maturation nor subunit association into a 70S ribosome. Thus, salt tolerance is a common feature of cells in which maturation or function of the ribosome is impaired. In these cells, premature induction of an alternative sigma factor, σ(E), by salt stress was observed. These
