[The effect of donor conditioning with cyclosporin on the survival time of allogenic heart transplants in the rat].

The aim of the study was to investigate the influence of a pretreatment of the heart donor with cyclosporin (CsA) at 25 or 50 mg kg BW on day -1 on survival time of rat heart allografts in a donor-recipient combination that was different at their major histocompatibility complex (MHC). No further immunosuppression was used. The survival of CsA-pretreated heart transplants was significantly prolonged in allogeneic recipients compared with untreated controls and 3 out of 10 grafts (25 mg kg) and 4 out of 9 grafts (50 mg kg) survived permanently. These preliminary experimental findings are new, b…

Epigenetic programming via lysine-specific demethylase 1 regulates responses to immune checkpoint blockade.

Immune checkpoint blockade has revolutionized cancer therapy, but durable benefit remains limited by immune-cold tumor states, poor antigen presentation, T cell exhaustion, and adaptive resistance. Recent studies position lysine-specific demethylase 1 (LSD1) as a druggable epigenetic regulator that links these barriers across tumor and immune cells. Emerging evidence shows that targeting LSD1 can restore major histocompatibility complex class I antigen presentation, activate viral mimicry and interferon programs, modulate programmed death-ligand 1 in a context-dependent manner, influence CD8+ …

Sensing the enemy within: Vγ9Vδ2 T cells as metabolic sensors of intracellular parasites.

V gamma 9 V delta 2 (Vγ9Vδ2) T cells, the predominant γδ T cell population in human peripheral blood, uniquely recognize nonpeptidic phosphoantigens (pAgs) independent of major histocompatibility complex molecules. This sensing mechanism relies on intracellular pAg accumulation, which triggers heteromeric cooperation between transmembrane butyrophilin 3A1 (BTN3A1) and butyrophilin 2A1 (BTN2A1) receptors to activate the γδ T cell receptor. This review synthesizes current knowledge of Vγ9Vδ2 T cell immunobiology, focusing on responses to Plasmodium falciparum and Toxoplasma gondii. We examine ho…

The Role of Epigenetic Factors in the Pathogenesis of Psoriasis.

Psoriasis is a chronic inflammatory skin disease, the prevalence of which is increasing. Genetic, genomic, and epigenetic changes play a significant role in the pathogenesis of psoriasis. This review summarizes the impact of epigenetics on the development of psoriasis and highlights challenges for the future. The development of epigenetics provides a basis for the search for genetic markers associated with the major histocompatibility complex. Genome-wide association studies have made it possible to link psoriasis to genes and therefore to epigenetics. The acquired knowledge may in the future …

TCR Signals Controlling Adaptive Immunity against Toxoplasma and Cancer.

T cells play a crucial role in adaptive immunity by recognizing and eliminating foreign pathogens and abnormal cells such as cancer cells. T cell receptor (TCR), which is expressed on the surface of T cells, recognizes and binds to specific antigens presented by major histocompatibility complex (MHC) molecules on antigen-presenting cells (APCs). This activation process leads to the proliferation and differentiation of T cells, allowing them to carry out their specific immune response functions. This chapter outlines the TCR signaling pathways that are common to different T cell subsets, as wel…

However a significant frac … ne signalling factor STING and that dual targeting of EZH2 and STING induces interferon signalling, major histocompatibility complex expression and synergistically reduces tumor growth in a preclinical model. Strategies such as this stand to improve therapeutic opportunities for otherwise refractory tumor contexts.

Interaction of genetic and exposure factors in the prevalence of berylliosis.

Prevalence of berylliosis, a lung disorder driven by the activation of beryllium-specific T cells, is associated with a major histocompatibility complex (MHC) class II marker (HLA-DPB1Glu69) and with the type of industrial exposure. We evaluated the interaction between marker and exposure in a beryllium-exposed population in which the prevalence of berylliosis was associated with machining beryllium. The presence of the marker was associated with higher prevalence (HLA-DPB1Glu69-positive machinists 25%; HLA-DPB1Glu69-negative machinists 3.2%, P = 0.05) and predicted berylliosis independent of …

Peptide antigen treatment of naive and virus-immune mice: antigen-specific tolerance versus immunopathology.

Peptide-specific down-regulation of T cell responses may represent a powerful tool to intervene in autoimmune diseases or graft rejections. It is therefore important to know whether peptide treatment tolerizes both naive and antigen-experienced memory T lymphocytes. Here we show that a major histocompatibility complex class I binding peptide, derived from the glycoprotein (GP33 peptide) of lymphocytic choriomeningitis virus (LCMV), specifically tolerized naive cytotoxic T lymphocytes (CTL) when administered three times intraperitoneally in incomplete Freund's adjuvants. However, in the presenc…

Genome sequence of a human tumorigenic poxvirus: prediction of specific host response-evasion genes.

Molluscum contagiosum virus (MCV) commonly causes asymptomatic cutaneous neoplasms in children and sexually active adults as well as persist … MCV possesses 59 genes that are predicted to encode previously uncharacterized proteins, including major histocompatibility complex class I, chemokine, and glutathione peroxidase homologs, which suggests that there are MCV-specific strategies for coexistence with the human host.

Immune response in mice that lack the interferon-gamma receptor.

Interferon-gamma (IFN-gamma) exerts pleiotropic effects, including antiviral activity, stimulation of macrophages and natural killer cells, and increased expression of major histocompatibility complex antigens. Mice without the IFN-gamma receptor had no overt anomalies, and their immune system appeared to develop normally. However, mutant mice had a defective natural resistance, they had increased susceptibility to infection by Listeria monocytogenes and vaccinia virus despite normal cytotoxic and T helper cell responses. Immunoglobulin isotype analysis revealed that IFN-gamma is necessary for…

Sequences and factors: a guide to MHC class-II transcription.

The expression of the class-II antigens of the major histocompatibility complex is tissue-specific, developmentally regulated in cells of the B lineage, and inducible by cytokine. Some of the molecular mechanisms that account for this regulation have recently been elucidated. DNA sequences upstream of human and mouse class-II genes that are critical for the transcription of these genes have been identified through transfection studies and transgenic analysis. Furthermore, a number of transcription factors that bind to important promoter motifs have recently been isolated. Examination of the cl…

Allogeneic H-2 antigen expression is insufficient for tumor rejection.

Murine A strain (KkDdLd) sarcoma I (SaI) tumor cells have been transfected with a cloned H-2Kb gene. Genetic control of Propionibacterium acnes-induced protection of mice against Babesia microti.

Using various strains of inbred mice, we found that Propionibacterium acnes-induced protection against the hemoprotozoan parasite Babesia microti was controlled by a dominant gene(s) not linked to the major histocompatibility gene (H2) complex of mice. P. acnes-induced resistance to infection was not merely an amplification of the normal immune response to B. microti, since innate resistance to infection was controlled by different genes. Expression of the nonspecific protection induced by P. acnes was found to be transferable with cells of bone marrow origin, and it did not necessarily accomp

A human autosomal phosphoglycerate kinase locus maps near the HLA cluster.

The human cDNA probe pPGK824 , of Singer-Sam et al. [Singer-Sam, J., Simmer , R. L., Keith, D. H., Shively , L., Teplitz , M., Itakura , K., Gartler , S. M. & Riggs, A. D. (1983) Proc. Natl. Acad. Sci. USA 80, 802-806] was used to isolate a genomic clone lambda PGK-1 containing a portion of an autosomal locus for phosphoglycerate kinase (PGK). A unique sequence subclone (pGK-1) of lambda PGK-1 was then used to map this locus to the region p23-q12 of human autosome 6--i.e., to the interval that contains the major human histocompatibility locus (HLA). Since an autosomal gene coding for testis-sp

How does the immune system learn to distinguish between good and evil? The first definitive studies of T cell central tolerance and positive selection.

Demonstration that immature CD4 + 8+ thymocytes contain T cell precursors that are subjected to positive and negative selection was the major step towards understanding how the adaptive immune system acquires the ability to distinguish foreign or abnormal (mutated or infected) self-cells from normal (healthy) cells. In the present review, the roles of TCR, CD4, CD8, and MHC molecules in intrathymic selection and some of the crucial experiments that contributed to the solution of the great immunological puzzle of self/nonself discrimination are described in an historical perspective. Recently,
