Phase-Specific Dual-Site Beta Transcranial Alternating Current Stimulation Differentially Influences Functional Connectivity Associated With Motor Inhibition Performance.

Inhibitory control relies on coordinated beta-band activity within a fronto-basal ganglia network, which implements inhibition via downstream effects on (pre)motor areas. However, the causal role of beta synchrony in motor inhibition remains unclear. In this study, we employed dual-site transcranial alternating current stimulation (tACS) targeting the right inferior frontal gyrus (rIFG) and left primary motor cortex (lM1) to directly manipulate phase relationships in the beta band and assess their effects on both functional connectivity and motor inhibition. Fifty-two healthy participants rece

Effects of baclofen and lorazepam on interhemispheric inhibition in humans.

Transcallosal projections within the corpus callosum allow for the transfer of information between bilateral primary motor cortices (M1). Interhemispheric inhibition (IHI) is one approach to probe transcallosal communication using transcranial magnetic stimulation (TMS) whereby the motor evoked potential (MEP) elicited by TMS is reduced in amplitude when preceded by a conditioning TMS pulse delivered to the opposite M1. In a double-blinded, placebo-controlled study, 24 participants received 2.5 mg of lorazepam (GABAA agonist), 50 mg of baclofen (GABAB agonist) and a placebo in separate session

Parvalbumin neurons in inferior colliculus mediate maladaptive inhibitory compensation in age-related hearing loss.

BACKGROUND: Age-related hearing loss (ARHL) may be caused by the abnormal excitability of inferior colliculus (IC) neurons, and this mechanism may be related to the inhibitory compensation of parvalbumin (PV) neurons; however, its specific mechanism remains unclear. This study focused on PV neurons and investigated the inhibitory compensatory mechanism in the IC during ARHL. METHODS: We investigated hearing changes in C57BL/6J mice from young to middle-aged using auditory brainstem response and acoustic startle reflex tests, followed by analysis of cluster activity in excitatory and inhibitor

Frequency-specific network connectivity impairments linked to suicide attempts in major depressive disorder during the GO/NOGO task.

BACKGROUND: Major depressive disorder (MDD) is a main risk factor of suicide, emphasizing the urgent need for understanding the neurobiological mechanisms underlying suicide attempts (SAs) in depressive patients. We hypothesized that aberrant frequency-specific functional connectivity patterns underlying an executive and inhibition task might be associated with SA in depression. METHODS: The current study enrolled 143 subjects including 43 healthy controls and 87 patients with MDD (43 patients with SA and 44 without SA), who attended a GO/NOGO task during the magnetoencephalography recording.

NPTX2 transfection improves synaptic E/I balance and performance in learning impaired aged rats.

Excessive neural activity in the medial temporal lobe commonly associates with cognitive decline in elderly humans and also in rodents. An attractive model pathway to study synaptic mechanisms underlying age-dependent circuit hyperexcitability is the connection made by lateral entorhinal cortex cells onto the dentate gyrus (LEC→DG). Both structures are particularly affected by age and, importantly, in behaviorally characterized aged rats, learning impairment correlates with diminished feedforward inhibition of granule cells recruited by LEC inputs. In this rat model of aging, we evaluated how o

Lithium restores inhibitory function and neuronal excitability through GSK-3β inhibition in a bipolar disorder-associated Ank3 variant mouse model.

Bipolar disorder (BD) is a prevalent psychiatric condition characterized by mood dysregulation, psychosocial impairment, and an increased risk of suicide. The gene ANK3 has been identified as a risk locus for BD through multiple genome-wide association studies (GWAS). However, the mechanisms by which ANK3 variants influence BD pathophysiology and treatment response remain unclear. ANK3 encodes ankyrin-G, a protein that organizes the axon initial segment (AIS) and nodes of Ranvier by scaffolding ion channels and cell adhesion molecules to the cytoskeleton. Recent studies show that ankyrin-G int

Excitatory magnetic stimulation increased interhemispheric inhibition: a pharmacological and TMS-EEG study.

The inhibitory effect of interhemispheric signal propagation (ISP) is believed to be aligned with GABAB receptor mediated inhibitory neurotransmission and related to interhemispheric connectivity. Repetitive Transcranial magnetic stimulation (rTMS) is a safe and potent strategy for altering brain connectivity. However, it remains unclear if rTMS modulates ISP. With pretreatment of baclofen, a GABAB receptor agonist, this study characterized rTMS effects on ISP and the influence of GABAB receptor neurotransmission. ISP was measured with TMS and electroencephalography co-registration (TMS-EEG).

Stroke Recovery-Related Changes in Cortical Reactivity Based on Modulation of Intracortical Inhibition.

BACKGROUND: Cortical excitation/inhibition dynamics have been suggested as a key mechanism occurring after stroke. Their supportive or maladaptive role in the course of recovery is still not completely understood. Here, we used transcranial magnetic stimulation (TMS)-electroencephalography coupling to study cortical reactivity and intracortical GABAergic inhibition, as well as their relationship to residual motor function and recovery longitudinally in patients with stroke. METHODS: Electroencephalography responses evoked by TMS applied to the ipsilesional motor cortex were acquired in patien

Enhanced Synaptic Inhibition in the Dorsolateral Geniculate Nucleus in a Mouse Model of Glaucoma.

Elevated intraocular pressure (IOP) triggers glaucoma by damaging the output neurons of the retina called retinal ganglion cells (RGCs). This leads to the loss of RGC signaling to visual centers of the brain such as the dorsolateral geniculate nucleus (dLGN), which is critical for processing and relaying information to the cortex for conscious vision. In response to altered levels of activity or synaptic input, neurons can homeostatically modulate postsynaptic neurotransmitter receptor numbers, allowing them to scale their synaptic responses to stabilize spike output. While prior work has indi

Interhemispheric interactions between the right angular gyrus and the left motor cortex: a transcranial magnetic stimulation study.

The interconnection of the angular gyrus of right posterior parietal cortex (PPC) and the left motor cortex (LM1) is essential for goal-directed hand movements. Previous work with transcranial magnetic stimulation (TMS) showed that right PPC stimulation increases LM1 excitability, but right PPC followed by left PPC-LM1 stimulation (LPPC-LM1) inhibits LM1 corticospinal output compared with LPPC-LM1 alone. It is not clear if right PPC-mediated inhibition of LPPC-LM1 is due to inhibition of left PPC or to combined effects of right and left PPC stimulation on LM1 excitability. We used paired-pulse

Functional Connectivity Hypersynchronization in Relatives of Alzheimer's Disease Patients: An Early E/I Balance Dysfunction?

Alzheimer's disease (AD) studies on animal models, and humans showed a tendency of the brain tissue to become hyperexcitable and hypersynchronized, causing neurodegeneration. However, we know little about either the onset of this phenomenon or its early effects on functional brain networks. We studied functional connectivity (FC) on 127 participants (92 middle-age relatives of AD patients and 35 age-matched nonrelatives) using magnetoencephalography. FC was estimated in the alpha band in areas known both for early amyloid accumulation and disrupted FC in MCI converters to AD. We found a fronto

4E-BP2-dependent translation in parvalbumin neurons controls epileptic seizure threshold.

The mechanistic/mammalian target of rapamycin complex 1 (mTORC1) integrates multiple signals to regulate critical cellular processes such as mRNA translation, lipid biogenesis, and autophagy. Germline and somatic mutations in mTOR and genes upstream of mTORC1, such as PTEN, TSC1/2, AKT3, PIK3CA, and components of GATOR1 and KICSTOR complexes, are associated with various epileptic disorders. Increased mTORC1 activity is linked to the pathophysiology of epilepsy in both humans and animal models, and mTORC1 inhibition suppresses epileptogenesis in humans with tuberous sclerosis and animal models

Transcranial Magnetic Stimulation-Electroencephalography Measures of Cortical Neuroplasticity Are Altered after Mild Traumatic Brain Injury.

While the potential long-term side effects of mild traumatic brain injury (mTBI) are becoming increasingly recognized, the associated neurophysiological mechanisms remain poorly understood. However, changes in cortical inhibitory function and neuroplasticity have been suggested as possible contributing factors. The current study applied transcranial magnetic stimulation (TMS) in conjunction with electroencephalography (combined TMS-EEG) to investigate further the effects of mTBI on these processes. In 17 patients with a history of mTBI and 15 healthy control subjects with no mTBI history, pair

Neurotransmitter- and Release-Mode-Specific Modulation of Inhibitory Transmission by Group I Metabotropic Glutamate Receptors in Central Auditory Neurons of the Mouse.

Neuromodulation mediated by metabotropic glutamate receptors (mGluRs) regulates many brain functions. However, the functions of mGluRs in the auditory system under normal and diseased states are not well understood. The medial nucleus of the trapezoid body (MNTB) is a critical nucleus in the auditory brainstem nuclei involved in sound localization. In addition to the classical calyx excitatory inputs, MNTB neurons also receive synaptic inhibition and it remains entirely unknown how this inhibition is regulated. Here, using whole-cell voltage clamp in brain slices, we investigated group I mGluR

Voluntary saccade inhibition deficits correlate with extended white-matter cortico-basal atrophy in Huntington's disease.

The ability to inhibit automatic versus voluntary saccade commands in demanding situations can be impaired in neurodegenerative diseases such as Huntington's disease (HD). These deficits could result from disruptions in the interaction between basal ganglia and the saccade control system. To investigate voluntary oculomotor control deficits related to the cortico-basal circuitry, we evaluated early HD patients using an interleaved pro- and anti-saccade task that requires flexible executive control to generate either an automatic response (look at a peripheral visual stimulus) or a voluntary re

Sedentary conditions and enhanced responses to GABA in the RVLM: role of the contralateral RVLM.

A sedentary lifestyle is a major risk factor for cardiovascular disease, and both conditions are associated with overactivity of the sympathetic nervous system. Ongoing discharge of sympathetic nerves is regulated by the rostral ventrolateral medulla (RVLM), which in turn is modulated by the primary excitatory and inhibitory neurotransmitters glutamate and γ-amino-butyric acid (GABA), respectively. We reported previously that sedentary conditions enhance GABAergic modulation of sympathoexcitation in the RVLM, despite overall increased sympathoexcitation. Thus the purpose of this study was to t

The stabilized supralinear network: a unifying circuit motif underlying multi-input integration in sensory cortex.

Neurons in sensory cortex integrate multiple influences to parse objects and support perception. Across multiple cortical areas, integration is characterized by two neuronal response properties: (1) surround suppression--modulatory contextual stimuli suppress responses to driving stimuli; and (2) "normalization"--responses to multiple driving stimuli add sublinearly. These depend on input strength: for weak driving stimuli, contextual influences facilitate or more weakly suppress and summation becomes linear or supralinear. Understanding the circuit operations underlying integration is critica

Enhanced GABAA-Mediated Tonic Inhibition in Auditory Thalamus of Rats with Behavioral Evidence of Tinnitus.

Accumulating evidence suggests a role for inhibitory neurotransmitter dysfunction in the pathology of tinnitus. Opposing hypotheses proposed either a pathologic decrease or increase of GABAergic inhibition in medial geniculate body (MGB). In thalamus, GABA mediates fast synaptic inhibition via synaptic GABAA receptors (GABAARs) and persistent tonic inhibition via high-affinity extrasynaptic GABAARs. Given that extrasynaptic GABAARs control the firing mode of thalamocortical neurons, we examined tonic GABAAR currents in MGB neurons in vitro, using the following three groups of adult rats: unexp

Medial olivocochlear efferent reflex inhibition of human cochlear nerve responses.

Inhibition of cochlear amplifier gain by the medial olivocochlear (MOC) efferent system has several putative roles: aiding listening in noise, protection against damage from acoustic overexposure, and slowing age-induced hearing loss. The human MOC reflex has been studied almost exclusively by measuring changes in otoacoustic emissions. However, to help understand how the MOC system influences what we hear, it is important to have measurements of the MOC effect on the total output of the organ of Corti, i.e., on cochlear nerve responses that couple sounds to the brain. In this work we measured

Vagal nerve stimulation blocks interleukin 6-dependent synaptic hyperexcitability induced by lipopolysaccharide-induced acute stress in the rodent prefrontal cortex.

The ratio between synaptic inhibition and excitation (sI/E) is a critical factor in the pathophysiology of neuropsychiatric disease. We recently described a stress-induced interleukin-6 dependent mechanism leading to a decrease in sI/E in the rodent temporal cortex. The aim of the present study was to determine whether a similar mechanism takes place in the prefrontal cortex, and to elaborate strategies to prevent or attenuate it. We used aseptic inflammation (single acute injections of lipopolysaccharide, LPS, 10mg/kg) as stress model, and patch-clamp recording on a prefrontal cortical slice
